Abstract:Over the past few years, ratiometric fluorescent nanoprobes have garnered substantial interest because of their self-calibration characteristics. This research developed a ratiometric fluorescent sensor to detect phosphate. Through encapsulating luminescent materials, gold nanoclusters (AuNCs) and carbon dots (CDs) into a zeolitic imidazolate framework-8 (ZIF-8), the fluorescence signal of AuNCs was enhanced, while that of CDs was suppressed. After phosphate was added, it could decompose ZIF-8, and AuNCs and CDs were released, which weakened the fluorescence signal of the AuNCs while restoring that of the CDs. Thereby, this makes CDs/AuNCs@ZIF-8 a potential fluorescent sensor for phosphate determination. The ratiometric sensor had facile synthesis, good selectivity, and a low detection limit. Therefore, this sensor was an effective tool for the detection of phosphate.

Abstract:Phosphate pollution leads to deterioration in water quality, posing a serious threat to human health. Therefore, it is important to develop a highly selective and sensitive fluorescent probe for phosphate detection. Here, we report a novel ratiometric fluorescent probe, Fluorescein@NH2-UiO-66 (denoted as Flu@NH2-UiO-66), for the trace detection of phosphate in water. Specifically, during the in-situ solvothermal synthesis of Flu@NH2-UiO-66, fluorescein molecules were encapsulated into the cavities of the metal-organic framework. Furthermore, the encapsulation amount of fluorescein was controlled by adjusting the acidity of the system. The Flu@NH2-UiO-66 (60 H+) sample, prepared with a 60:1 molar ratio of acetic acid to the metal center exhibited distinct dual fluorescence signal peaks. The probe showed a highly selective fluorescence response to phosphate. Within a range of 0-20 μM phosphate concentration, the probe demonstrated excellent linear detection capability with a detection limit of 0.37 μM. Moreover, the mechanism of fluorescence enhancement can be attributed to the addition of phosphate, which greatly increases the UV absorbance of the probe. This study developed a novel ratiometric fluorescent probe capable of rapid, sensitive, and stable detection of trace phosphate, which is of great significance for environmental management.

Cell-Compatible Nanoprobes for Imaging Intracellular Phosphatase Activities.

Abstract:Phosphatases play an important role in cell biology, but only a few probes are suitable for selectively imaging phosphatase activity in live cells, because the current probes require cell fixation or exhibit considerable cytotoxicity. Herein, we show that conjugating a d-peptide to a quinazolinone derivative generates cell-compatible, biostable probes for imaging the phosphatase activity inside live cells. Moreover, our results show that inhibiting ectophosphatases is a critical factor for imaging intracellular phosphatases. As the first example of using selective inhibitors to ensure intracellular function of molecular probes, this work illustrates a facile approach to design molecular probes for profiling the activities of enzymes in a spatial, selective manner in a complicated environment.

Compared with other Zr-based NMOF probes for phosphate, reduced number of connections for ZrO clusters with the ligand in PCN-224 obtained by a linker-elimination strategy simultaneously provide more active recognition sites for phosphate, which effectively improves the sen-sitivity of the Zr-based NMOF probes. The detection limit of the probe is only 54 nM. Additionally, the accuracy of the ratiometric detection based on this probe was further proved by the detection of phosphate in human serum and drinking water.

Abstract:Developing fluorescent sensors for small-molecule phosphates offers opportunities in optically detecting biorelevant reactions and events. However, remains elusive how to identify phosphates from other anions, such as carboxylates and sulfates, because current synthetic phosphate receptors lack selectivity. Here we report the construction of multicolor fluorescent sensor that can identify phosphates from other analogous anions. The key design principle is to take advantage of the highly sensitive conformation-dependent emissive wavelength of diphenyl-9,14-dihydrodibenzo[a,c]phenazine fluor-ophores to sense the minor structural differences between phosphates and other anions, for example, sulfates and carboxylates. The effect of structural factors such as spacer length and urea versus thiourea has been investigated by comparing the optical properties and binding affinities with the phosphate receptors. This strategy provides a simple and robust fluorescent sensing solution to optically analyze small-molecule phosphates with anti interference performance.

Abstract:Inorganic and organic phosphates—including orthophosphate, nucleotides, and DNA—are some of the most fundamental anions in cellular biology, regulating numerous processes of both medical and environmental significance. The characteristic long lifetimes of emitting lanthanides, including the brighter europium(III) and terbium(III), make them ideally suited for the development of molecular probes for the detection of phosphates directly in complex aqueous media. Moreover, given their high oxophilicity and the exquisite sensitivity of their quantum yields to their hydration number, those luminescent lanthanides are perfect for the detection of phosphates. Herein we discuss the principles that have guided the recent developments of molecular probes selective for inorganic or organic phosphates and how these lanthanide complexes facilitate the study of numerous biological processes.

Abstract:Phosphate is widely used in industry and agriculture fields. However, excess accumulation of PO43− causes several adverse effects on the human body and ecological environment. Consequently, it is important to develop a simple method for the detection of PO43− concentration in the ecological environment and in vivo. Herein, two caffeic acid derivative-based fluorescence probes (BAM-HM and BAM-HH) were developed for the detection of phosphate. The BAM-HM probe could detect phosphate via fluorescence enhancement at 500 nm, with the detection limit being 0.612 μM. Meanwhile, the BAM-HH probe showed a significant turn-on signal at 450 nm after the addition of phosphate, and the detection limit was calculated to be 0.318 μM. The sensing mechanism was determined by 1H NMR and MS. Furthermore, the two probes (BAM-HM and BAM-HH) were applied for PO43−detection in living cells and water samples.

Abstract:Alkaline phosphatase (ALP) is an important biomarker for diagnostics, and excessive level of ALP is closely related to a variety of pathological processes; hence the development of convenient and sensitive methods for detecting ALP is of great significance for medical sciences and diagnostics. Herein, we report a fluorescent probe for detecting ALP via excited state intramolecular proton transfer (ESIPT). The probe was synthesized by coupling a phosphate group onto flavone-based fluorophore. For this probe, the phosphate group serves both as the blocking agent for ESIPT and the responsive moiety toward ALP. Due to the shielding of the hydroxyl group by the phosphate moiety, no ESIPT process occurs, the tautomer emission of the probe is therefore quenched; while in the presence of ALP, ALP cleaves phosphate group and generates hydroxyl group, thereby enabling the ESIPT and generating strong ESIPT-based emission. This probe exhibits high selectivity for ALP detection with a very low detection limit of 0.032 U/L. It is capable of detecting ALP in biological fluid like serum. Furthermore, it is of little cytotoxicity and can be easily internalized into cells for endogenous ALP imaging. Therefore, the probe herein displays strong potential for applications in biological diagnostics and pathological analysis. (C) 2015 Elsevier B.V. All rights reserved.

Abstract:Herein, a new fluorescent probe, AE-Phos, is reported for detecting the ALP activity with the combined advantages of aggregation-induced emission (AIE) and excited state intramolecular proton transfer (ESIPT). Further detailed fluorescence experiments demonstrated that AE-Phos exhibited excellent selectivity and sensitivity, a large Stokes shift, and a fast response towards ALP. Furthermore, AE-Phos was applied to imaging the ALP activity in different cell lines quantitatively.

Abstract:A new bis(salamo)-like compound H 3 L was designed and synthesized. With the addition of Zn 2+ ions to the H 3 L solution, the fluorescence intensity increased significantly and the color changed from blue-cyan to bright green. Based on ESI-MS, UV-Vis spectroscopy and DFT calculations, a plausible mechanism for the interaction of the compound H 3 L with Zn 2+ ions is proposed. The in situ complex sensor L-Zn 2+ formed by the coordination of the compound H 3 L with Zn 2+ ions has been used for various anion recognition. The results showed that the sensor L-Zn 2+ has high selectivity and sensitivity to phosphates. The fluorescence experiments of the compound H 3 L and sensor L-Zn 2+ at different pH values showed that fluorescence response is good in neutral and weak base environment. Simple test paper strips and the detection in real water samples were applied for the detection of phosphates in complex environment.

Abstract:A ratio fluorescence nanoprobe was constructed by simple mixing BCNO QDs with 8-hydroxyquinoline-5-sulfonic acid (HQSA), which had an obvious fluorescence peak at 420 nm and a weak fluorescence peak at 500 nm, corresponding to the BCNO QDs and HQSA, respectively. This fluorescence probe takes stable fluorescence of BCNO QDs as an internal standard, based on HQSA chelating enhanced fluorescence and specificity of phosphate in the presence of Mg 2+ , which realizes a rapid and sensitive detection of phosphate with good linearity in the range 0.3–50 μM and 50–100 μM and a detection limit of 0.073 μM. The recovery is between 94.1 and 111% and the relative standard deviations (RSDs) below 10%. At the same time, we took color photos of the reaction solution under 310-nm UV lamp with smartphones for visual detection through RGB data image analysis, which make the detection easier and faster. The proposed method provides a new strategy for the intelligent online detection of other targets in complex environment samples.

Abstract:Alkaline phosphatase (ALP) is associated with many diseases, and its accurate detection is of great significance. Fluorescent compounds with aggregation-induced emission (AIE) feature show beneficial advantages for serving as fluorescent probes. Herein, an AIE-active "turn on" probe for ALP detection was synthesized through incorporating a strong electron-withdrawing group (cyano) in the middle and the recognition moiety phosphate group at the end, thereby rendering a D-A-D structure with a relatively high conjugation degree and good water solubility. It was found that the probe TPE-CN-pho is highly sensitive to ALP in aqueous solution. In the presence of ALP, the hydrophilic phosphate group on the probe is rapidly removed, resulting in a decrease in water solubility and subsequent formation of aggregates, thereby achieving aggregation-induced emission. Moreover, the probe TPE-CN-pho has also been successfully applied to imaging ALP in living cells.

Abstract:A tetracationic perylene probe (probe 1) was designed and synthesized. Probe 1 was used for the real-time fluorescence turn-on assay of alkaline phosphatase (ALP) activity and inhibitor screening. Probe 1 monomer fluorescence could be very efficiently quenched by ATP through the formation of an ATP/probe 1 complex. ALP triggered the degradation of ATP, the breakdown of the ATP/probe 1 complex, and the recovery of the probe 1 monomer fluorescence. In the presence of an ALP inhibitor, a decrease in fluorescence recovery was observed.

Abstract:A novel coumarin-based compound DPAC with two dipicolylamine (DPA) arms as the chelator sites was designed and synthesized. The compound DPAC exhibits a highly selective response to Cu 2+ ions with a distinctly emission-quenching phenomenon. Moreover, the in situ formed complex DPAC-Cu 2+ was used for the detection of pyrophosphate (PPi). The binding manner of probe DPAC-Cu 2+ with PPi in 1 : 1 stoichiometry was supported by the Benesi-Hildebrand fitting, ESI-MS and HPLC analysis. The linear range of PPi concentration was 1-4 μ M, and the detection limit was 0.53 μ M. The competing experiments illustrated that the probe DPAC-Cu 2+ had good sensitivity and selectivity for PPi than other anions, including ATP, ADP, AMP, and Pi in CH 3 CN : HEPES (3 : 2, v / v , pH = 7.20 ) buffer. Further, cell fluorescence imaging experiments indicated that the probe DPAC-Cu 2+ had a potential to be used to detect PPi in vivo.

Abstract:Phosphate is not only an important indicator for aquatic ecosystems, but also plays vital roles in biosystems. A new strategy for ultrasensitive and selective detection of phosphate is fabricated based on a new insight found in this paper, which a lower concentration of surfactant sodium dodecylbenzenesulfonate (SDBS) can greatly induce fluorescence resonance energy transfer (FRET) from ciprofloxacin (CIP) to Eu3+ in the CIP-Eu3+ complex. Surfactant SDBS does not act as a sensitizer for enhancing the fluorescence intensity of the system, while acts as a sensitizer of FRET and make the native fluorescence of CIP quenched completely (switch off). Eu3+ ions can coordinate with the oxygen-donor atoms of phosphate, which weakens FRET from CIP to Eu3+ and results in the fluorescence recovery of CIP (turn on). The multi-complex of the CIP-Eu3+-phosphate has more sensitive fluorescent response than that of the reported coordination nanoparticle-based fluorescent probes. The LOD (S/N=3) of this sensing system can attain 4.3 nM. The possible interferential substances existed in environmental samples, such as 17 common metal ions, 11 anions and fulvic acid investigated do not interfere with the phosphate detection. This sensing system has been successfully applied for the phosphate detection in environmental samples such as wastewater, surface water and atmospheric particulates. This work not only develops a fluorescent probe for the phosphate detection, but also provides a new strategy for designing fluorescent probes based on FRET or coordination nanoparticle-based.

Abstract:A simple pH fluorescent probe based on new fluorophorepyrido[1,2-a]benzimidazole derivative has been synthesized and characterized. This probe responds to acidic pH (pKa =5.18) with high quantum yield (phi)=0.96), fast response (within 1 min), high selectivity and sensitivity.
