Abstract:Residual injury of atrazine to the succeeding crops has been frequently reported. It is necessary to find a solution for the detoxification of atrazine contaminated soil. A high-efficient bacterial strain Arthrobacter sp. C2 for atrazine degradation was isolated in this study. The genomic information of the isolate C2, and its degradation characteristics and potential application in detoxification of atrazine contaminated soil were investigated. The results indicated that the isolate C2 genome contained 4,305,216 bp nucleotides, three plasmids, and 4705 coding genes. The degradation rates of atrazine at levels of 1, 10, 100 mg/L by the isolate C2 were 0.34, 1.94, 18.64 mg/L/d, respectively. The optimum temperature and pH for the isolate C2 to degrade atrazine were 30 degrees C and 7.0-9.0. Based on the metabolites detected by UPLC-TOF-MS/MS and genome annotation of the isolate C2, a common metabolic pathway of atrazine was proposed as that atrazine is firstly dechlorinated into hydroxyatrazine, and subsequently to N-isopropylammelide via dealkylation, and ultimately deaminated to cyanuric acid. Introduction of the isolate C2 into soil can enhance degradation of atrazine and thus eliminate the toxic effect of this herbicide on wheat growth. Abstract:BackgroundThe isolation of atrazine-degrading microorganisms with specific characteristics is fundamental for bioaugmenting the treatment of wastewater containing atrazine. However, studies describing the specific features of such microorganisms are limited, and further investigation is needed to improve our understanding of bioaugmentation.Results and conclusionsIn this study, strain Arthrobacter sp. ZXY-2, which displayed a strong capacity to degrade atrazine, was isolated and shown to be a potential candidate for bioaugmentation. The factors associated with the biodegrading capacity of strain ZXY-2 were investigated, and how these factors likely govern the metabolic characteristics that control bioaugmentation functionality was determined. The growth pattern of Arthrobacter sp. ZXY-2 followed the Haldane–Andrews model with an inhibition constant (Ki) of 52.76 mg L−1, indicating the possible augmentation of wastewater treatment with relatively high atrazine concentrations (> 50 ppm). Real-time quantitative PCR (RT-qPCR) results showed a positive correlation between the atrazine degradation rate and the expression levels of three functional genes (trzN, atzB, and atzC), which helped elucidate the role of strain ZXY-2 in bioaugmentation. In addition, multiple copies of the atzB gene were putatively identified, explaining the higher expression levels of this gene than those of the other functional genes. Multiple copies of the atzB gene may represent a compensatory mechanism that ensures the biodegradation of atrazine, a feature that should be exploited in future bioaugmentation applications.

Strain ZXY-2 demonstrated excellent atrazine degradation performance and saline tolerance. Abstract:Atrazine has been a pollutant of great environmental concern due to its high potential to contaminate soil and waters. In north China, atrazine has widely applied in corn fields and is still in use. Isolation of a high-efficiency atrazine degrader whose degradation ability is not inhibited by organic matters and fertilziers in the soils is of practical importance for bioremediation of contaminated soils. A bacterial strain (DAT1) capable of utilizing atrazine as a sole nitrogen source for growth was isolated from an agricultural soil in Hebei Province, China. The strain DAT1 was identified as an Arthrobacter species and harbored degradation genes trzN, atzB and atzC on plasmid DNA. The strain demonstrated a very high efficiency of atrazine biodegradation with a broad optimum pH and temperature ranges and could be enhanced by addition of both carbon and nitrogen sources, suggesting its huge potential for remediation of atrazine-contaminated sites. This work might provide some new insights on atrazine remediation and contributes to the limited pool of knowledge on the function of Arthrobacter species.
Isolation and Characterization of Atrazine-Degrading Arthrobacter Sp. AD26 and Use of This Strain in Bioremediation of Contaminated Soil

Abstract:A bacterial strain (AD26) capable of utilizing atrazine as a sole nitrogen source for growth was isolated from an industrial wastewater sample by enrichment culture. The 16S rRNA gene sequencing identified AD26 as an Arthrobacter sp. PCR assays indicated that AD26 contained atrazine-degrading genes trzN and atzBC. The trzN gene of AD26 only differs from the trzN of Arthrobacter aurescens TC1 by one base (A-->T at 907) and one amino acid (Met-->Leu at 303). The specific activity of trzN of AD26 in crude atrazine-containing minimal media than two well characterized atrazine-degrading bacteria, Pseudomonas sp. ADP and Arthrobacter aurescens TC1. After incubating for 48 h at 30 degrees C, the OD(600) of AD26 reached 2.6 compared with 1.33 of ADP. AD26 was capable of degrading 500 mg/L of atrazine in minimal medium at 95% in 72 h, while the degradative rates by TC1 and ADP were only 90% and 86%, respectively. A bioremediation trial of contaminated soil has indicated that AD26 can degrade as high as 98% of atrazine contained in soil (300 mg/kg) after incubating for 20 d at 26 degrees C, nominating this strain as a good candidate for use in bioremediation programs.

Abstract:A new atrazine-degrading strain named ZXY-1 was isolated in the present study. ZXY-1 was identified as Pseudomonas based on the physio-biochemical characteristics and 16s rRNA gene sequence analysis. The newly isolated strain had a strong ability to biodegrade atrazine of 100 mg/L completely within 11 h, with a degrading rate up to 9.09 mg/( L·h) . The optimal conditions for atrazine degradation were captured: temperature 30 ℃, pH 8.0, velocity of shaker 150 r/min and inoculums 3％. Atrazine biodegradation, under the optimum conditions, fitted the first-order kinetic equation when the initial atrazine concentration ranged from 20 mg/L to 80 mg/L, and the zero-order kinetic equation when the initial atrazine concentration ranged from 100 mg/L to 250 mg/L. The kinetic analysis indicated that the strain ZXY-1 cell growth perfectly suited Haldane model withμmax, Ks and Ki of 0.696 h-1 , 98.55 and 142.6 mg/L, respectively. Overall, this study provided an efficient bacterium and approach that might serve as a viable bioremediation solution to treat the atrazine wastewater.

Abstract:Atrazine, a widely used herbicide, is increasing the agricultural production effectively, while also causing great environmental concern. Efficient atrazine-degrading bacterium is necessary to removal atrazine rapidly to keep a safe environment. In the present study, a new atrazine-degrading strain ZXY-1, identified as Pseudomonas, was isolated. This new isolated strain has a strong ability to biodegrade atrazine with a high efficiency of 9.09 mg/L/hr.Temperature, p H, inoculum size and initial atrazine concentration were examined to further optimize the degradation of atrazine, and the synthetic effect of these factors were investigated by the response surface methodology. With a high quadratic polynomial mathematical model(R 2 = 0.9821) being obtained, the highest biodegradation efficiency of 19.03 mg/L/hr was reached compared to previous reports under the optimal conditions(30.71°C, pH 7.14, 4.23%(V/V) inoculum size and 157.1 mg/L initial atrazine concentration).Overall, this study provided an efficient bacterium and approach that could be potentially useful for the bioremediation of wastewater containing atrazine.
Isolation, Screening and Identification of Atrazine-degrading Bacterium FM326(Arthrobacter Sp.)

Abstract:The triazine herbicide atrazine(2-chloro-4-ethylamino-6-isopropyl-amino-s-triazine) is one of the most used pesticides. Atrazine is principally used for control of certain annual broadleaf and grass weeds, primarily in corn but also in sorghum, sugarcane, and other crops. Although atrazine has relatively low persistency in soil, its remanets have been found in many higher concentrations even years after application due to widespread use. Microbial remediation is an effective and economic method to control the environment that has been polluted by atrazine. From herbicide contaminated soil, 5 atrazine degrading bacterial strains were isolated by enrichment culture, and 6 atrazine-degrading bacterial strains were isolated by direct coating method. In the 11 strains, FM326 was screened out as the most highly efficient degradation bacteria by degradation test and could use atrazine as the sole carbon and nitrogen sources for growth. The degradation efficiency of strain FM326 was up to 97% within 96 hours while atrazine concentration was 1 000 mg·L-1. Strain FM326 was identified as Arthrobacter sp. according to physiological and biochemical tests and the phylogenic tree established by means of 16S rDNA sequencing. Moreover, strain FM326 was aerobic bacteria, its optimum temperature and pH value for growth was 30~35 ℃ and 5~9, respectively. Strain of highly efficient degrading atrazine was found from soils, and which provide a basis for the treatment of contaminated soils by atrazine for the further research.
Bioaugmentation with Arthrobacter Sp. Strain DAT1 for Remediation of Heavily Atrazine-Contaminated Soil

Abstract:A previously isolated atrazine-degrading bacterium, Arthrobacter sp. strain DAT1, was used to investigate its potential of remediating soil with heavy contamination. Change of atrazine-degrading genetic potential was estimated by quantitative PCR targeting trzN, atzB and atzC genes. Growth of the strain DAT1 was monitored using terminal restriction fragment length polymorphism (TRFLP) analysis. The results indicated that this bioremediation process could achieve a high removal rate in 2-3 days. Continuous increase in the number of trzN, atzB and atzC genes and the relative abundance of the strain DAT1 was observed during the bioaugmentation process.This work might provide some new insights toward bioremediation of heavily atrazine-contaminated soil. Abstract:Atrazine residues running off the fields and entering water resources are a major threat to food security and the ecosystem. In this study, a psychrotrophic functional strain named KN0901 to remove atrazine residues was screened. KN0901 could degrade 30 mg·L -1 atrazine in 4 days at 15oC with 10 5 CFU·mL -1 incubation. The phylogenetic results showed KN0901 belonged to Paenarthrobacter sp. PCR results showed that the functional genes consist of trzN , atzB , and atzC , suggesting atrazine was transformed to cyanuric acid by KN0901. KN0901 could degrade atrazine without adding exogenous carbon and nitrogen sources. What's more, KN0901 could tolerate extreme low temperature (5oC) and high atrazine concentration (100 mg·L -1 ). When growth and degradation curves were compared, the results indicated the length of lag time showed significant correlation to atrazine degradation rate. The hydroponic experiments showed that the toxicity of atrazine was significantly reduced with KN0901 treatment. The study provided an effective, economic, and eco-friendly bioremediation measure to address atrazine contamination.

Abstract:High salt concentration and salinity fluctuations in wastewater challenge the efficiency of microbial strains used for cleanup of pollutants. In this study, it was investigated that the new isolated Arthrobacter sp. W1 degraded mixed phenolic compounds under complex salt conditions. The results showed that Arthrobacter sp. W1 was able to utilize various phenolic compounds as carbon source under high salt conditions. It can degrade phenol and p -cresol mixture at 10% NaCl, although rates of degradation and cell growth were lower compared to 5% NaCl. The presence of trace p -cresol significantly inhibited phenol biodegradation. When salinity fluctuations were between 1% and 10% NaCl, strain W1 was able to degrade substrates and survived. It was also suggested that the presence of salts (i.e., NaCl, KCl, Na 2 SO 4 , and K 2 SO 4 ) had almost no effects on the microbial growth and biodegradation process. Therefore, Arthrobacter sp. W1 would be a promising candidate for bioremediation of phenolic compounds under complex salt conditions.

Abstract:Agricultural waterways are often contaminated with herbicide and antibiotic residues due to the widespread use of these chemicals in modern agriculture. The search for resistant bacterial strains that can adapt to and degrade these mixed contaminants is essential for effective in situ bioremediation. Herein, by integrating chemical and transcriptomic analyses, we shed light on mechanisms through which Paenarthrobacter sp. AT5, a well-known atrazine-degrading bacterial strain, can adapt to sulfamethoxazole (SMX) while degrading atrazine. When exposed to SMX and/or atrazine, strain AT5 increased the production of extracellular polymeric substances and reactive oxygen species, as well as the rate of activity of antioxidant enzymes. Atrazine and SMX, either alone or combined, increased the expression of genes involved in antioxidant responses, multidrug resistance, DNA repair, and membrane transport of lipopolysaccharides. Unlike atrazine alone, co-exposure with SMX reduced the expression of genes encoding enzymes involved in the lower part of the atrazine degradation pathway. Overall, these findings emphasize the complexity of bacterial adaptation to mixed herbicide and antibiotic residues and highlight the potential of strain AT5 in bioremediation efforts. ENVIRONMENTAL IMPLICATION The coexistence of different toxic chemicals in the natural environment poses a significant challenge to the successful implementation of in situ bioremediation. To overcome this challenge, it is crucial to identify bacterial strains that are resistant to and capable of degrading combined pollutants. Herein, through the integration of chemical and transcriptomic analyses, we provide mechanistic evidence for the ability of Paenarthrobacter sp. AT5 to adapt to sulfamethoxazole while degrading atrazine. This research contributes to the understanding of bacterial adaptation to and degradation of combined pollutants and paves the way for the development of more effective bioremediation strategies.

Abstract:The purpose of this study is to enhance the biodegradability of atrazine with FH-1 and NJ-1 alone by selecting the mixing ratio, optimizing the culture medium and conditions. The results showed that FH-1 and NJ-1 have the best biodegradation effect on atrazine being mixed in a volume ratio of 3:2. In a single factor experiment, sucrose and NH4Cl provided carbon and nitrogen sources for the mixed bacteria. Subsequently, composition of fermentation medium was further optimized using Box-Behnken design of response surface methodology. Based on the results, growth of mixed bacteria and biodegradation of atrazine performed best effects with a biodegradation rate of 85.6% when sucrose and NH4Cl amounts were 35.30 g/L and 10.28 g/L. The optimal medium condition was 10% inoculum of mixed bacteria, with initial atrazine concentration of 50 mg/L, neutral or weakly alkaline pH value, 30°C. The biodegradation rate reached 97.4%, 11.8% higher than the unoptimized condition.

Abstract:One strain that can grow with atrazine as sole nitrogen source, W4 was isolated and screened from sludge of workshop in the pesticide factory under 10°C. This strain was identified as Pseudomonas through 16S rDNA basic sequence detection and comparison. The optimum degradation conditions were optimized as following: the initial pH is 79, and the carbon source is sucrose and lactose. Under the optimum conditions, degradation kinetics of atrazine follows zero-order kinetics equation at the initial concentration of 34 mg/L, 22 mg/L and 10 mg/L, and degradation kinetics of atrazine follows first-order kinetics equation at the initial concentration of 5 mg/L. GC/MS analysis shows that atrazine can be hydrolyzed and dehalogenated by strain W4; the biodegradation product- hydroxyatrazine is confirmed.

Abstract:An aerobic bacterium named strain BH was isolated from soil samples based on its bensulfuronmethyl-degrading characteristics using continuous enrichment cultures. The cells of the strain were non-motile, gram-positive short rods. Colonies formed on agar medium were round, smooth, sticky, white-yellow in colour and of butyrous consistency. Analyses of nutritional utilization in Biolog microplates, conventional phenotypic characteristics and 16S rRNA gene sequencing were consistent with assigning strain BH to the genus Brevibacterium. Growth of the cells and their ability to degrade bensulfuronmethyl were simultaneously monitored under different liquid medium conditions during 7 days of incubation. They degraded bensulfuronmethyl from 100 to 70.6 mg l−1 in mineral M9 medium and exhibited more effective degradation in the presence of yeast extract, completely removing an initial concentration of 100 mg l−1 and at best 80% of an initial concentration of 200 mg l−1. Further studies are required to determine the potential use of the isolate in the disposal of bensulfuronmethyl residues in agriculture and industry.

Abstract:The objective of this study was to investigate the impact of glucose supplementation on the soil microbiota inoculated with the atrazine-degrading Arthrobacter strain DAT1. Soil microcosms with different treatments were constructed for biodegradation tests. The impact of glucose supplementation on atrazine degradation capacity of the strain DAT1 and the strain's survival and growth were assessed. The densities of the 16S rRNA gene and the atrazine-metabolic trzN gene were determined using quantitative PCR. The growth of the strain DAT1 and the bacterial community structure were characterized using terminal restriction fragment length polymorphism. Glucose supplementation could affect atrazine degradation by the strain DAT1 and the strain's trzN gene density and growth. The density of the16S rRNA gene decreased during the incubation period. Glucose supplementation could alter the bacterial community structure during the bioaugmentation process. Glucose supplementation could promote the growth of the autochthonous soil degraders that harbored novel functional genes transforming atrazine. Further study will be necessary in order to elucidate the impact of exogenous carbon on autochthonous and inoculated degraders. This study could add some new insights on atrazine bioremediation.

Abstract:The application of Atrazine in soil has always been a main problem in agriculture because its residuals may maintain in the soil for a long term. In this paper, two strains of Atrazine degrading bacteria (Klebsiella sp. FH-1 and Arthrobacter sp. NJ-1) were used to make biological compound microbial inoculum to repair the Atrazine contaminated typical black soil in Northeast China. Grain chaff was chosen as the optimal carrier material for microbial inoculum. The dynamic changes of Atrazine were detected by gas chromatography. The half-life of Atrazine in soil containing microbial inoculum was shortened from 9.8 d to 4.2 d. The Atrazine sensitive crops grown in the repaired soil showed increased stem length, root length, and emergence rate. The effects of microbial remediation on the original bacterial and fungal biota in the typical black soil in Northeast China were analyzed using the metagenomic approach. Results showed that Atrazine inhibited the original bacteria and fungi populations. The total numbers of bacterial and fungal species in the soil were partially recovered by adding the microbial inoculum. Two genera (Sphingosinicella and Sphingomonas) were the dominant bacteria. The beneficial bacterial biota was recovered and the number of species of the beneficial bacteria was higher than that in the original soil after adding the microbial inoculum. The dominant fungi included genera Guehomyces and Chaetomella. There was a total of 113 unclassified fungal genera (22.6% of 499), indicating the potential utility of the unclassified fungal species in the assessment of the soil contamination by Atrazine.
Biodegradation of Atrazine by the Novel Klebsiella Variicola Strain FH-1

Abstract:Bacterial strain FH-1 with high efficiency of degrading Atrazine is separated by means of enrichment culture from the soil applied with Atrazine for many years. FH-1, recognized as Klebsiella variicola based on phylogenetic analysis of 16S rDNA sequences, can grow with Atrazine which is the sole nitrogen source. In fluid inorganic salt medium, the optimal degradation temperature, pH value, and initial concentration of Atrazine are 25°C, 9.0, and 50 mg L–1, respectively, and the degradation rate of Atrazine by strain FH-1 reached 81.5% in 11 d of culture. The degrading process conforms to the kinetics equation of pesticide degradation. Among the metal ions tested, Zn2+ (0.2 mM) has the most significant effect of facilitation on the degradation of Atrazine. In the fluid medium with Zn2+, the degradation rate of Atrazine is increased to 72.5%, while the Cu2+ (0.2 mM) inhibits the degradation of Atrazine. The degradation products of Atrazine by strain FH-1 were identified as HEIT (2-hydroxyl-4-ethylamino-6-isopropylamino-1,3,5-triazine), MEET (2-hydroxyl-4,6-bis(ethylamino)-1,3,5-triazine), and AEEO (4,6-bis(ethylamino)-1,3,5-triazin-2(1H)-one) by HPLC-MS/MS. Three genes (atzC, trzN, and trzD) encoding for Atrazine degrading enzymes were identified by PCR and sequencing in strain FH-1. This study provides additional theoretical support for the application of strain FH-1 in bioremediation of fields polluted by Atrazine.

Abstract:AbstractAtrazine toxicity is one of the limiting factors inhibiting sensitive plant growth. Previous studies showed that atrazine-degrading bacteria could alleviate atrazine toxicity. However, there is limited information on how atrazine-degrading bacteria and plant growth-promote bacteria alleviate atrazine toxicity in soybeans. Therefore, the current study aimed to explore the atrazine removal, phosphorus utilization, and the oxidative stress alleviation of atrazine-degrading bacterium Arthrobacter sp. DNS10 and/or inorganic phosphorus-solubilizing bacterium Enterobacter sp. P1 in the reduction of atrazine toxicity in soybean. The results showed that atrazine exposure to soybean seedlings led to significant inhibition in growth, atrazine removal, and phosphorus utilization. However, the co-inoculatied strains significantly increased seedlings biomass, chlorophyll a/b contents, and total phosphorus in leaves accompanied by great reduction of the atrazine-induced antioxidant enzymes activities and malonaldehyde (MDA) contents, as well as atrazine contents in soil and soybeans under atrazine stress. Furthermore, transcriptome analysis highlighted that co-inoculated strains increased the expression levels of genes related to photosynthetic-antenna proteins, carbohydrate metabolism, and fatty acid degradation in leaves. All the results suggest that the co-inoculation mitigates atrazine-induced oxidative stress on soybean by accelerating atrazine removal from soil and phosphorus accumulation in leaves, enhancing the chlorophyll contents, and regulating plant transcriptome. It may be suggested that co-inoculation of atrazine-degrading bacteria and inorganic phosphorus-solubilizing bacteria can be used as a potential method to alleviate atrazine toxicity to the sensitive crops.Graphical Abstract
