Expression Analysis of Lncrna As Prognostic Biomarkers in Colon Adenocarcinoma

Abstract:Introduction: Long non-coding RNAs (lncRNAs) have important biological functions and can be used as prognostic biomarkers in cancer. To identify a lncRNA as prognostic signature in colon adenocarcinoma (COAD). Methods: In this study, the lncRNA profiles in 135 COAD patients from The Cancer Genome Atlas (TCGA) database were analyzed. The associations of the differentially expressed miRNAs with patient survival and other clinical characteristics were examined with t-test and Cox proportional regression model. Finally, a prognostic lncRNA signature was generated and examined with Kaplan–Meier survival and univariate Cox regression analyses. Results: A total of 9 out of 12727 lncRNAs were significantly negatively correlated with overall survival of patients with COAD (log-rank test, P < 0.05). Another 173 lncRNAs were found to be significantly associated with disease free survival (DFS, log-rank test, P < 0.05). Among the 173 lncRNAs, nine lncRNAs (ENSG00000175701.6, ENSG00000227290.1, ENSG00000237250.3, ENSG00000241158.2, ENSG00000247675.2, ENSG00000248049.2, ENSG00000255503.1, ENSG00000269293.2 and ENSG00000269386.1) were further selected by fitting a penalized Cox proportional hazards model to predict the DFS of patients with COAD. The prediction power of the nine lncRNAs was evaluated through the receiver operating characteristic (ROC) curve and AUC for the nine-lncRNA is 0.665. The patients were categorized into high- and low-risk groups based on their nine-lncRNA profiles. Patients with high-risk scores had worse DFS than those with low risk scores (log-rank test P = 0.03). Conclusion: Our findings showed that the nine-lncRNA signature might be a novel biomarker for the accurate prognosis prediction of COAD patients.
