Abstract:As one of the most abundant biopolymers, lignin is a widely available resource. However, its potential largely remains untapped, with most of it ending up as waste from industries like paper production, pulp processing, and bio-refining. The research undertaken in this study focused on the extraction of lignin from agroforestry waste using a deep eutectic solvent (DES) as a carrier for alpha-amylase immobilization, resulting in high stability and reusability. Several techniques, including Nuclear Magnetic Resonance (NMR), Scanning Electron Microscopy (SEM), Energy-Dispersive X-ray Spectroscopy (EDS), and the Brunauer-Emmett-Teller (BET) method were employed to examine the structure and morphology of both the extracted lignin and the immobilized enzyme. The temperature used to recover lignin by DES would affect immobilization efficiency and enzyme loading by influencing its specific surface area, pore size, and volume distribution. Investigations using Nuclear Overhauser Effect Spectroscopy (NOESY) uncovered that the hydroxyl groups in G, H, and S units and the beta-O-4 structure of lignin primarily serve as binding sites for enzyme molecules. Immobilized alpha-amylase demonstrated a higher pH and thermal stability level, with an optimal pH of 7.0 and temperature of 100 degrees C, compared to the free enzyme, which exhibited optimal activity at a pH of 6.5 and temperature of 90 degrees C. Importantly, immobilized alpha-amylase retained >80 % of its initial activity even after 28 days at room temperature, and it maintained 70 % of its activity after being reused 12 times. These findings strongly suggest that lignin derived from agroforestry residues holds promising potential as a future versatile immobilization material, a prospect integral to society's sustainable development.
Evaluation of Organosolv Pretreatment on the Structural Characteristics of Lignin Polymers and Follow-Up Enzymatic Hydrolysis of the Substrates from Eucalyptus Wood.

Abstract:In the present study, Eucalyptus was subjected to organosolv pretreatment processes with aqueous 2-propanol at 200–220°C to obtain lignin with benign characters for its valorization and digestible substrates for bioethanol production. Results showed that different delignification ratios (64.00%–81.26%) and molecular weights (Mw=610–2680g/mol) of lignin fractions were dissociated from various pretreatment conditions, and the glucose yields of all the pretreated substrates significantly increased to 54.65–88.59% as compared to that of raw material (9.20%). Additionally, the amounts of β-O-4, β-β, and β-5 substructures were regularly decreased with the increased temperature and time. By contrast, the structural characteristics of the lignin fractions obtained with 70% 2-PrOH/water were less-altered than those with 50% 2-PrOH/water pretreatment. Remarkably, lignin with the lowest molecular weight (Mw=610g/mol) and less-altered chemical structure was achieved at 200°C for 120min, which is beneficial to subsequent catalytic degradation of lignin.

Abstract:Herein, sodium hydroxide (NaOH) pretreatment of lignocellulose is performed to investigate the structural evolution of lignin, cellulose, and hemicellulose. It is found that most of hemicellulose and part of cellulose are hydrolyzed/converted to monosaccharide, alcohols, and carboxylic acids. Over half of cellulose is stabilized because the reducing end groups of cellulose convert to stable carboxylic groups. In addition, lignin is converted to aromatic monomers (predominately vanillin and acetovanillone) with a highest yield of 6.1 wt%, along with dimers and oligomers with Mw less than 10 000 Da. According to heteronuclear single quantum coherence analysis, no common interlinkages (β‐O‐4, β‐β, β‐5, etc.) appear in lignin oligomers due to lignin fragments condensed during the alkaline pretreatment. Notably, the acidification process is also the cause of lignin condensation since high aromatic monomers yield of 15 wt% (based on original lignin in pine) is achieved by a postoxidation of alkaline solution, while few no aromatic monomers generated by using the isolated lignin oligomers. The direct oxidation of lignocellulose in alkaline solutions to aromatic monomers demonstrates the potential application of lignocellulose alkaline pretreatment for high‐value chemicals production.

Extrusion process to enhance the pretreatment effect of ionic liquid for improving enzymatic hydrolysis of lignocellulosic biomass

Abstract:Extrusion process using 1-ethyl-3-methylimidazolium acetate ([EMIM]Ac) in co-solvent, dimethyl sulfoxide (DMSO), was conducted to improve the pretreatment effect on the enzymatic saccharification of pussy willow (Salix gracilistyla Miq.). The use of a twin screw extruder capable of continuous processing increased the solid loading to 50% in [EMIM]Ac and its co-solvent DMSO. The water-soluble fraction was increased by increasing the [EMIM]Ac amount in co-solvent and the extrusion temperature but decreasing the screw rotating speed and solid loading. The surface morphologies of the extruded product showed remarkable disruption of the cell wall structures and fibrillation with increasing amounts of [EMIM]Ac in co-solvent. With increasing [EMIM]Ac amount in co-solvent and extrusion temperature and with decreasing screw rotating speed and solid loading, the crystalline structure of cellulose was more disrupted, resulting in the increase in saccharification yield. The highest yield of glucose and xylose was 99.0% and 99.5%, respectively, from the pretreated products using only [EMIM]Ac under solid loading of 15% at 160 °C extrusion temperature and 5 rpm screw rotating speed. The initial saccharification rate was faster in the pretreated product precipitated in alcohol and acetone than in water.

Abstract:Electrochemical oxidation provides a green process for lignin depolymerization. In this work, depolymerization mechanisms of alkali and organosolv lignin on a PbO2 anode have been explored in details through 2D‐HSQC NMR, in‐situ IR spectroelectrochemistry and depolymerized product analysis. It not only clarifies lignin structure changes, but also reveals the selectivity in bond cleavage and product distribution in the oxidation process. To better understand the process and mechanism of lignin structural changes and relative product formation during PbO2 anode electrolysis, a Ti/Sb−SnO2/PbO2 composite electrode has been prepared and utilized for the alkaline electrolytic oxidation of both alkali lignin (AL) and bagasse organosolv lignin (OL). By combining electrochemical measurements, in‐situ FTIR, 2D‐HSQC NMR, and product distribution analysis, we not only investigate the impact of influence factors on the process of lignin depolymerization, but also reveal the depolymerization mechanism from the whole process of lignin structural changes in details. Under certain electrolysis conditions, the highest vanillin yield, 10.1 mg g−1, has been obtained through the electrolysis of alkali lignin, whereas the highest syringaldehyde yield of 4.0 mg g−1 has been achieved during the electrolysis of organic solvent lignin. In particular, AL mainly performs depolymerization through the selective cleavage C−C bonds to produce a series of G unit monomer products of vanillin, acetovanillone, and vanillic acid. By contrast, the depolymerization of OL shows significant selectivity in H units. Particularly, the production of the major H unit monomer product of 4‐vinylphenol has been confirmed by IR spectroelectrochemical studies from the oxidation and decarboxylation of p‐coumaric acid unit.

A Novel and Highly Efficient Polymerization of Sulfomethylated Alkaline Lignins Via Alkyl Chain Cross-Linking Method

Abstract:A new family of water-soluble lignosulfonate polymers with ultrahigh molecular weight (Mw) was developed based on alkali lignin (AL) as starting material in a one-pot reaction in two steps: sulfomethylation of AL as raw material led to AL-S and this material was subsequently cross-linked via alkylation with 1,6-dibromohexane (alkAL-S). Gel permeation chromatography showed a significant increase of Mw from 5200 Da of AL-S to 201 000 Da of alkAL-S with high degree of alkylation. Fourier transform infrared spectroscopy (FTIR) and proton nuclear magnetic resonance spectroscopy and functional group analysis confirmed the efficient polymerization by a nucleophilic substitution reaction mechanism. Additionally, alkAL-S with high Mw showed unexpected viscosity-reducing effect, stability and good rheological properties on a low-rank coal-water slurry (CWS), which are even better than those obtained by naphthalene sulfonate formaldehyde (NSF) as additive. The adsorption properties of the new products were also characterized via a quartz crystal microbalance combined with dissipation monitoring (QCM-D method). Cross-linked structure, large steric hindrance from high Mw and suitable amphiphilic properties of alkAL-S polymers contribute together to the highly improved dispersion performances for CWS.
Possible Dissolution Mechanism of Alkali Lignin in Lactic Acid-Choline Chloride under Mild Conditions.

Abstract:In this study, several representative deep eutectic solvents (DESs) were designed to evaluate the solubility for alkali lignin (AL). It was found that DESs with lactic acid (LA) as hydrogen bond donors (HBDs) had good solubility for AL, in which lactic acid(LA) : choline chloride(ChCl) 10 : 1 showed excellent solubility with more than 17 wt% under a relatively mild condition of 60 degrees C. The results of gel permeation chromatography (GPC), FTIR and H-1 and HSQC NMR spectroscopies revealed an important possible dissolution mechanism of AL in LA-ChCl, that is, AL could be depolymerized under the action of LA when dissolved in LA-ChCl. Then, a new connection would form between the phenolic groups on the lignin fragments and ChCl, which is similar to that between ChCl and LA in DES, leading to an increase in the molecule weight of lignin. The new connections could be easily broken under the action of heat (150 degrees C) or microwave to the redispersion of lignin fragments. The results would provide a theoretic base for the high-value application of lignin in bioresources.

Abstract:Novel organosolv pretreatments of biomass using biobased green solvents have attracted increasing attention to realize sustainable biorefinery. We have demonstrated that the feasibility of dimethyl isosorbide/water co-solvent in fractionating lignocellulosic biomass ( Eucalyptus ) for multiple products under a mild condition (120 °C, 60 min). To take a further step towards revealing the effects of catalysts, we explored various Brønsted (e.g., H 2 SO 4 , HCl, and p -TsOH), Lewis acid catalysts (e.g., AlCl 3 and FeCl 3 ) and their combinations in this co-solvent system. An interesting finding was that the delignification efficiency of AlCl 3 was lower than that of H 2 SO 4 (69.9% vs 91.2%), while the cellulose conversion efficiency of the pretreated pulp was similar (∼80%). It is hypothesized that the presence of Al 3+ could inhibit the binding of lignin to cellulase, which was supported by the quantum chemistry calculations. Results revealed the presence of cation− π interactions between Al 3+ and lignin with interaction energy (ΔE) was 1022.04 kJ/mol and 1003.04 kJ/mol for S and G type of lignin, respectively. In addition, the pretreatment with H 2 SO 4 -AlCl 3 co-catalyst could achieve a high lignin removal efficiency of 93.0% and almost complete enzymatic hydrolysis of obtained cellulose pulp (∼100%). This research demonstrated that the co-catalyst strategy effectively improved the overall performance of the solvent-based biorefinery.

Abstract:In this study, the alkaline catalyst (NaOH)-aided sulfolane organosolv pretreatment was first developed to reduce the recalcitrance of willow (Salix matsudana cv. Zhuliu) for effective saccharification and lignin valorization in subsequent processes. The pretreatment notably removed lignin, minimized cellulose loss, and increased the accessibility of cellulose. The sulfolane/water (50/50, w/w) pretreatment with 4% NaOH at 140 degrees C for 1.5 h increased the glucose saccharification yield by 14.2-fold of the yield from the untreated willow. The recovered lignin had high purity (similar to 97.0%), well-preserved molecular weight (4300-5010 g/mol), and low polydispersity (similar to 2.0). NMR analysis of the recovered lignin also indicated that it retained the representative linkage structures (e.g., beta-O-4, beta-beta linkages). Based on these observations, the recovered lignin showed great potential as an intermediate in the production of functional biomaterials and/or green biofuels.
Alkaline sulfonation and thermomechanical pulping pretreatment of softwood chips and pellets to enhance enzymatic hydrolysis

Abstract:To assess the impact of alkalinity on sulfonation and the enzyme-mediated hydrolysis of softwood cellulose, Lodgepole pine chips were impregnated with 8% sodium sulfite and increasing loadings of sodium carbonate before thermomechanical pulping. It was apparent that alkali addition enhanced lignin sulfonation with an additional 4% loading of sodium carbonate proving optimal. TEM indicated that sulfonation predominantly occurred within the secondary-cell-wall lignin, increasing cellulose accessibility to the cellulase enzymes. Although increasing alkalinity did not significantly enhance lignin sulfonation, likely due to the lower acetyl content of the softwood chips, it increases mannan solubilization. Despite their smaller particle size, softwood pellets were more poorly sulfonated, probably due to their higher lignin content and lower amount of acid groups. This more condensed lignin structure was confirmed by 2D-NMR and GPC analyses which indicated that the EMAL derived from softwood pellets contained less native β-O-4 linkages and had a higher molecular weight.

Abstract:This study emphasized the synergistic production of bioprocessible lignin and carbohydrates during a sequential liquid hot water and alkali pretreatment of lignocellulose, facilitating their subsequent individual fermentation. Increasing the dose of alkaline lignin from 0 to 8 g/L inhibited cell growth in anaerobic digestion, with varying levels of inhibition observed in the following order: hydrolytic bacteria < acidogens < acetogens. Alkali pretreatment was adapted to maximize yields of bioprocessible lignin liquor without compromising utilization of the carbohydrates. Increasing the NaOH dose from 50 to 200 mg/g-feedstock monotonically improved lignin yields, but further increases in alkali loading led to a decline in lignin recovery. Volatile fatty acids production from anaerobic digestion of the carbohydrate moiety consistently increased with higher NaOH doses. The optimal conditions for maximizing lignin yields were determined to be 105 °C for 30 min, with NaOH loading in the range of 150–200 mg/g-feedstock, resulting in approximately 80 % lignin recovery, of which 35 % was biologically utilizable. Liquid hot water treatment prior to alkali pretreatment was confirmed as necessary to preserve carbohydrates of 0.1 g/g-feedstock at a low temperature of 70 °C. These findings are crucial for economically producing bioprocessible lignin without carbohydrate loss, a key step towards achieving full lignocellulose valorization.

Abstract:High cost and high viscosity of ionic liquid restricted its commercial application in pretreatment of lignocellulose. Water and ethanol were used as additive in [EMIM][OAc] to pretreat corn cob at moderate temperature (< 100 °C). It was found that enzyme hydrolysis (EH) sugar yield was increased with the increase of IL content. The largest EH sugar yield of 68.8% was obtained when pure IL was used. However, for [EMIM][OAc]/ethanol, the EH sugar yield as high as 66.9% was gained when the IL content was 80%, which was comparable to that for pure IL pretreatment. In addition, Kamlet-Taft parameter was calculated to characterize the polarity solvency of binary liquid phase, to illustrate the underlying reason for the increase of EH sugar and the lignin removal. Finally, to demonstrate the crystalline and microstructure change after pretreatment, XRD and SEM were performed for the raw materials and the pretreated samples.
Exploring chemical reactions to enhance thermal and dispersion stability of kraft and organosolv lignin

Abstract:Lignin has been overlooked and used as a waste for long due to its complex and partially hydrophobic structure. Many efforts have been carried out to overcome these deficiencies and apply it as a high-value product, which are insufficient to reach the full potential of lignin in various advanced applications, since they require with procedures for the obtaining of more specific and fine-tuned chemical structures. This work focuses on the obtaining of differently structured hydrophilic lignins derived both from Kraft and organosolv isolation processes. The chemical structures of the different lignin types were studied, and the effect of the structural differences in the modification processes and their subsequent properties analyzed, valorizing their potential application for diverse purposes. The carboxymethylation and sulfomethylation reactions were carried out with the aim of enhancing the polarity of the lignin samples, while the methylation reaction aimed to obtain lignins with higher stability. The physicochemical analyses of the samples, carried out by FTIR, GPC, 31 P NMR, 13 C NMR, and HSQC NMR, verified the effectiveness of the chemical reactions and conditions selected, obtaining lignins with lower hydroxyl content, due to their substitution and insertion of carboxymetyl, sulfomethyl and methyl groups, therefore obtaining more condensed, aromatic and oxygenated aromatic carbon structures. While the methylation reaction was the most efficient in substituting the OH groups, due to its non-selectivity, OL showed higher modification yields than KL. In terms of the thermal and morphological properties, analyzed by DLS and TGA respectively, it was observed that the modified samples showed lower Z potential values, along with higher conductivity, being the sulfomethylated organosolv lignin the one showing the best results, which was also the one with the smallest particle size and polydispersity index. Finally, all the modified samples showed higher T 50% values, suggesting a better stability towards degradation.

Abstract:In this study, an alkaline-catalyzed sulfolane/water solvent system was developed for isolating high-purity and antioxidative lignin from willow (Salix matsudana cv. Zhuliu). Optimization of the pretreatment conditions such as temperature, sulfolane/water ratio, and alkaline catalyst (NaOH) dosage were comprehensively investigated for effective lignin extraction from willow. The 44.4% of lignin was recovered from the biomass with 54% of delignification in 50/50 (w/w) sulfolane/water system at 170 °C. As the addition of the alkaline catalyst (NaOH) increased to 4%, the delignification yield was increased up to 94% with about 70% of lignin recovery yield. The recovered lignin was comparatively investigated with its control, milled wood lignin (MWL). The β-O-4 linkages and phenolic hydroxyl were well preserved in the extracted lignin fractions with the sulfolane/water system. Furthermore, excellent radical scavenging ability was observed with the extracted lignins by sulfolane/water pretreatments owing to rich phenolic hydroxyl groups in the lignins. Hence, systematical investigation on the lignin properties and potential applications under sulfolane organosolv pretreatment would promote the utilization of lignin in biorefinery processes.

Structural Changes of Lignin after Ionic Liquid Pretreatment

Abstract:Changes of lignin's structure were investigated resulting from an ionic liquid ([Bmim]Cl, 1-butyl-3-methylimidazolium chloride salt) pretreatment. The purified lignin was pretreated by [Bmim]Cl under the following conditions: the ionic liquid to lignin mass ratio was 10:1, the temperature was 85 degrees C, and the processing time was 2 h and 4 h. The chemical structure of lignin was studied via Ultraviolet (UV) spectra, Fourier Transform infrared (FT-IR) spectra, Nuclear Magnetic Resonance Spectroscopy (C-13-NMR), Gel Permeation Chromatography (GPC), X-ray photoelectron spectroscopy (XPS), and Thermogravimetric Analysis (TG). The total content of phenolic hydroxyl increased with increased time. Moreover, the contents of [OHI] and [OHII] (types of phenolic hydroxyl groups) groups were disproportionately increased. The main structure of lignin still was present after the [Bmim]Cl pretreatment. The beta-O-4 linkages were broken apart. The degree of lignin degeneration increased with increased time, after being pre-treated with [Bmim]Cl. Simultaneously, a condensation reaction also took place during the pretreatment. Preparation of nano disperse dyes using sulfomethylated lignin: Effects of sulfonic group contents

Abstract:The molecular simulation software was firstly applied to analyze the adsorption of sulfomethylated lignin (SAL) on dye surfaces. Then, SALs with different sulfonic group contents were prepared and characterized by FTIR, NMR, EA and GPC measurements using alkali lignin (AL) as raw materials and sodium sulfite as sulfonating agents. Next, SAL 1.53 was determined to the optimum dispersant by TSI, particle size and thermal storage stability measurements, which had the smallest particle size of 173 nm and highest stability, comparable to the commercial Reax 85A lignin dispersant and basically satisfying the requirement of nano disperse dyes used in the digital printing technology. QCM, AFM and zeta potential results indicated that as the sulfonic group content of SAL increased, the adsorption mass, rigidity of the adsorbed layer, adsorption force and absolute zeta potential value all showed a gradually increasing tendency due to an enhanced hydrophilicity, and thus a decreased intermolecular agglomeration and an increased molecular chain stretching degree. A maximum was observed for SAL 1.53 . This research not only provided a novel approach to the preparation of high-performance lignin dispersants for nano disperse dyes, but also would broaden the high value-added industrial applications of biomass lignin into the digital printing and dyeing field.

Abstract:The poor solubility of lignin in most solvents causes the biggest challenges in lignin valorization to improve overall biorefinery economics. In this work, humic acid (HA), a natural surfactant, was investigated with respect to its ability to assist in biomass delignification. After obtaining a baseline level of delignification using standard HA, a mechanism of action was proposed in which the lignin removal was driven by hydrophobic interactions. To explore this hypothesis, HA was modified in two ways. One batch was sulfonated to increase HA hydrophilicity, while the other was esterified to increase HA hydrophobicity. Treatment of the same substrates with the modified HA materials revealed that elevated hydrophobicity provided a further increase in the level of delignification. It was found that the solubility of lignin in the HA solution was increased with the enhancement of HA hydrophobicity from 12.2% (S-HA) to 16.8% (E-HA). Therefore, the initial hypothesis was confirmed that HA-derived delignification improvements appear to be a function of hydrophobic interactions. These results should aid further investigation into choosing surfactants (ideally those that are biobased) for utility in biorefinery processes.

Abstract:Lignin holds the key for maximizing value extraction from lignocellulosic biomass. This is currently hindered by the application of fractionation methods that significantly alter the lignin structure to give highly recalcitrant materials. For this reason, it can be highly beneficial to use less-severe fractionation conditions that allow for efficient extraction of lignin with retention of the β-aryl ether (β-O-4) content. Here, we present a detailed study on mild alcohol-based organosolv fractionation with the aim of understanding how to achieve a balance between efficiency of lignin extraction and the structure of the resulting lignin polymers, using walnut shells as model biomass. Monitoring different extraction conditions reveals how the structure of the extracted lignin changes depending on the extraction conditions in terms of molecular weight, alcohol incorporation, and H/G/S ratios. Moving from ethanol to n-pentanol, it was revealed that, in particular, alcohol incorporation at the benzylic α-position of β-aryl ether units not only plays a key role in protecting the β-O-4 linking motif but more importantly increases the solubility of larger lignin fragments under extraction conditions. This study shows that α-substitution already occurs prior to extraction and is essential for reaching improved extraction efficiencies. Furthermore, α-substitution with not only bulky secondary alcohols and tertiary alcohols but also chloride was revealed for the first time and the latter could be involved in facilitating α-alkoxylation. Overall, this study demonstrates how by tuning the fractionation setup and conditions, the resulting lignin characteristics can be influenced and potentially tailored to suit downstream demands.Benzylic alcohol incorporation allows for the extraction of lignin with high retention of β-aryl ether linking motifs.

Abstract:Lignins, heterogeneous assemblies of polyphenolic oligomers present in lignocellulosic plant biomass, are valorized mainly by combustion because of their low solubility in commonly used solvents. However, in addition to antioxidant, anti‐UV and antimicrobial properties, and marked interfacial properties are part of lignins physicochemical properties, thus making them good candidates for sustainable colloids. It is hypothesized that lignins solubility and emulsifying performances could be modulated by fractionation, functionalization, and formulation strategies. The importance of the solvent, and of the pH on lignins solubilization in aqueous phase was investigated. Accordingly, simple O/W emulsions stabilized by different lignins were designed and characterized. The whole results demonstrated that lignins solubility in water can be optimized using appropriate co‐solvent and pH readjustment resulting in a homogeneous dispersion with only 7.2 ± 1.8% wt/wt of insoluble lignins. Both fractionation and biocatalytic modification of lignins allowed increasing stabilization of the oil–water interface by limiting coalescence (23%–24% of relative increase of the D90) compared with non‐fractionated lignins (32%), thus advocating their potential use as multifunctional emulsion stabilizers. As a result, a direct link between the molar mass profile of lignins and their ability to stabilize the oil–water interface as well as to reduce emulsion's sedimentation could be established.
