Abstract:Fraxinellone, a naturally occurring degraded limonoid, is mainly isolated from Dictamnus dasycarpus and Melia azedarach. In continuation of our program aimed at the discovery of plant natural products-based insecticidal agents, twenty-two new ethers from furyl-ring-based acylation derivatives were prepared by structural modifications of fraxinellone. Their insecticidal activity was evaluated against the pre-third-instar larvae of Mythimna separata Walker in vivo. Among them, compounds 5e and 5j exhibited more potent insecticidal activity than toosendanin with the final mortality rates greater than 60%. Their structure-activity relationships were also discussed.

Abstract:A total of 20 esters of fraxinellone C4/10-oxime were synthesized and determined by melting points, optical rotation, infrared spectra, proton nuclear magnetic resonance spectra, and high-resolution mass spectrometry spectra. Two steric configurations of compounds 7i and 8i were unambiguously confirmed by X-ray crystallography. Additionally, their pesticidal activities were assessed on two typical lepidopteran pests, Mythimna separata Walker and Plutella xylostella Linnaeus. Generally, all compounds exhibited less potent oral toxicity than toosendanin against third-instar larvae of P. xylostella. However, all compounds showed the growth inhibitory property against early third-instar larvae of M. separata. Notably, compounds 7m, 8b, 8k, 9, and 11 displayed more potent pesticidal activity than toosendanin. This demonstrated that introducing the C-4 carbonyl or oxime group on fraxinellone resulted in more promising derivatives than those bearing a C-10 carbonyl or oxime substituent.

Abstract:In continuation of a program aimed at the discovery and development of natural products-based insecticidal agents, two series of novel fraxinellone-based esters were synthesized by modification at the C-4 or C-10 position of fraxinellone and evaluated for their insecticidal activity against the pre-third-instar larvae of Mythimna separata in vivo. An efficient method for the stereoselective synthesis of 4 alpha-hydroxyfraxinellone from fraxinellonone was developed, and the steric configuration of 6h was unambiguously confirmed by X-ray crystallography. Among 37 compounds, some derivatives displayed potent insecticidal activity; especially compounds 6h, 6q, 6t, and 7q showed more promising insecticidal activity than toosendanin, a commercial botanical insecticide derived from Melia azedarach. This suggested that introduction of the fluorine atom on the phenyl ring could lead to a more potent compound than one possessing chlorine or bromine. Meanwhile, introduction of the heterocyclic fragments at the C-4 or C-10 position of fraxinellone was essential for their insecticidal activity. This will pave the way for further design, structural modification, and development of fraxinellone as an insecticidal agent.

Abstract:Two series of novel fraxinellone-based thioethers containing 1,3,4-oxadiazole moiety were prepared as insecticidal agents against the oriental armyworm, Mythimna separata Walker. The structural assignment was based on the spectroscopic and X-ray analysis data. Among all the target compounds, compounds 4b, 4k, 5b, 5j and 5k exhibited more potent insecticidal activity with final mortality rates (FMRs) of more than 65%, especially 4k with the FMR of 75.9%, when compared with toosendanin. Some interesting results of structure–activity relationships are also discussed.

Abstract:A series of novel fraxinellone derivatives modified at the C-1 or C-8 position in the B ring were prepared as insecticidal agents against the pre-third-instar larvae of oriental armyworm, Mythimna separata Walker at 1 mg/mL. Five key steric configurations of compounds 2, 3, and 8f,g,j were further determined by single-crystal X-ray diffraction. It was found that the kinds and the amount of the reduction products of fraxinellone were related to the molar ratio between the reduction agent Red-Al and the substrate fraxinellone. Among all of the derivatives, compounds 2 and 8i,j,o displayed more promising insecticidal activity than their precursors fraxinellone and toosendanin. The preliminary structure-activity relationships revealed that the lactone (B-ring) of fraxinellone contributed to the observed insecticidal activity; the double bond at the C-2 position of fraxinellone was not necessary for the insecticidal activity; conversion of the oxygen atom of carbonyl group on the lactone of fraxinellone to a sulfur one does not improve the insecticidal activity; introduction of electron-withdrawing groups on the phenyl ring of 8f, to the benzoyloxy series, could result in more potent compounds.

Abstract:Fraxinellone (1) is a naturally occurring degraded limonoid isolated from Meliaceae and Rutaceae plants. As a potential natural-product-based insecticidal agent, fraxinellone has been structurally modified to improve its activity. Furan ring of fraxinellone is critical in exhibiting its insecticidal activity, but with few modifications. Herein, C-ring-modified cyclopropyl analogues were semi-synthesized by Rh(II)-catalyzed cyclopropanation. The structures of the target compounds were well characterized by NMR and HRMS. The precise three-dimensional structural information of 3a was established by X-ray crystallography. Their insecticidal activity was evaluated against Mythimna separata Walker by a leaf-dipping method. Compound 3c exhibited stronger insecticidal activity than 1 and toosendanin against M. separata with teratogenic symptoms during the different periods, implying that cyclopropanation of the furan ring could strengthen the insecticidal activity of fraxinellone.

Abstract:Introduction of the halogen atom or the acyl group at the C-ring of fraxinellone was investigated. Some unexpected halogenation products were obtained with the different chlorination/bromination reagents, and their possible reaction mechanisms were also proposed. Seven key steric structures of 2a’, 2b, 2b’, 2c’, 3a, 3b, and one isomer (5’α-Cl) of 2a were further confirmed by single-crystal X-ray diffraction. Especially compounds 2a, 2a’, 3a and 3c exhibited more potent insecticidal activity than toosendanin. Some structure-activity relationships of tested compounds were also described.

Abstract:Fraxinellone, a degraded limonoid, has been mainly isolated from some renewable plants in Meliaceae and Rutaceae. In a continuous effort to discover new natural products-based pesticides, two series of fraxinellone derivatives containing N-phenylpyrazole moiety were designed, synthesized and evaluated for their pesticidal activities against Mythimna separata Walker and Plutella xylostella Linnaeus. Two structures of compounds 7g and 8k were unambiguously determined by X-ray diffraction further. The bioassay showed that over half of the target compounds exhibited better insecticidal activity against M. separata than the precursor fraxinellone. Among all the target compounds, the compounds 7g-i and 8g-j exhibited more potent insecticidal activity than toosendanin, a commercial botanical pesticide. Furthermore, the compound 8g displayed more promising larvicidal activity with the LC50 value of 0.31 mu mol mL(-1) than the toosendanin against P. xylostella. The structure activity relationship (SAR) revealed that introduction of polyhalogenated phenylpyrazole ring on furyl-ring of fraxinellone could lead more potent compounds both against M. separata and P. xylostella than that of monohalogenated phenylpyrazole ring or electron-donating groups substituted phenylpyrazole ring.

Abstract:In continuation of our program aimed at the discovery and development of natural products-based insecticidal agents, twenty-three new fraxinellone-based hydrazone derivatives were smoothly prepared from fraxinellone via regioselectively allylic oxidation in the presence of selenium dioxide or chromium trioxide under microwave irradiation and subsequent condensation with hydrazides or hydrazines. Their insecticidal activity was evaluated against the pre-third-instar larvae of Mythimna separata Walker in vivo. Especially compounds 6d and 7a displayed the most pronounced insecticidal activity compared with toosendanin, a commercial botanical insecticide derived from Melia azedarach.

Abstract:In order to develop novel spirocyclic tetronic acid lead compounds, a series of new spirocyclic tetronic acid derivatives containing oxime ether moiety was synthesized and bioassayed. The structures of 16 target com-pounds were characterized by 1H NMR spectra, 13C NMR spectra and high-resolution mass spectrometer(HRMS). Preliminary bioassays indicated that most of the title compounds displayed excellent insecticidal activity against Aphis fabae and Nilaparvata lugens at 100 mg/L. In particular, compound 6k showed the similar activity(LC50= 6.87 mg/L) against A. fabae as the spirotetramat(LC50=4.56 mg/L) and had better effects(LC50=1.64 mg/L) against N. lugens in comparison with spirotetramat(LC50=7.90 mg/L). The study showed that compound 6k exhibited more promising and broad-spectrum insecticide activity and may serve as a new insecticidal agent for sucking pests.

Abstract:Furan ring of limoninoids is critical in exhibiting insecticidal activity. Herein, fraxinellone (1) was used as a template of furan-containing natural products and a series of its derivatives was synthesized by selective bromination in good yields on gram-scale and following Suzuki-Miyaura or Sonogashira coupling reactions in moderate to good yields. Bromination of limonin (9) was also accomplished without altering other functional groups in high yield. Furthermore, an evaluation of insecticidal activity against the instar larvae of Mythimna separata showed that derivatives 2, 3b, 3g, 5a, 5d and 5h displayed more potent insecticidal activity than 1 and toosendanin.

Abstract:Sulfimides and sulfoximines are highly relevant for medicinal chemistry and crop protection, as the resulting products can reveal interesting bioactivities. Herein, we report the design and synthesis of a series of novel 2,4-diphenyl-1,3-oxazolines containing sulfiliminyl and sulfoximinyl moieties. The acaricidal and insecticidal activities of the new compounds were evaluated and indicated that these compounds exhibited excellent acaricidal activities against spider mite larvae and eggs. The LC(50 )values of 6a-7, 6b-3, 6b-4, 6c-2, and 6c-4 against spider mite larvae were about 4 to 6 times lower than that of the commercial insecticide etoxazole (0.0221 mg L-1), and the LC50 value of 6a-4 against spider mite eggs was 0.0006 mg L-1, which was 10 times lower than that of etoxazole (0.0063 mg L-1). At the same time, most of the compounds showed insecticidal activity though their structure-activity relationships that were different. Oxazolines containing an N-cyano sulfiliminyl moiety at the para position of the 4-phenyl group exhibited better insecticidal activities against cotton bollworm and corn borer than etoxazole, whereas the compounds containing groups derived from sulfiliminyl and sulfoximinyl had weak insecticidal activities. This research again proved that the substituent type at the para site of the 4-phenyl moiety has a decisive role on the biological activity and insecticidal spectrum.

Abstract:By using podophyllotoxin as a phytoinsecticidal lead compound, 15 novel aromatic esters of 4'-demethyl-4-deoxypodophyllotoxin were semisynthesized and preliminarily tested for their insecticidal activity against the pre-third-instar larvae of Mythimna separata Walker in vivo for the first time. Among all of the tested compounds, especially two compounds, 4m and 4o, containing a pyridinyl group, for which final corrected mortality rates against M. separata at 1 mg/mL were 62.9 and 59.2%, respectively, showed the most promising and pronounced insecticidal activity as compared with toosendanin, a commercial insecticide derived from Melia azedarach. The quantitative structure-activity relationships (QSAR) of compounds 4a-4o showed that the relative number of benzene rings and final heat of formation were very important descriptors to their insecticidal activity.

Abstract:A series of novel symmetrical and asymmetrical 2,5-disubstituted-1,3,4-oxadiazoles and their intermediates were synthesized. A convenient procedure for preparation of aromatic nitrile and ester intermediates is reported. The insecticidal activities of oxadiazoles against Dropsophila melanogaster were recorded, and analysis of structure-activity relationships showed that the highest occupied molecular orbital energy (E(H)) was the main factor to affect the bioactivities.

Abstract:To search for improved insecticidal compounds based on β-dihydroagarofuran sesquiterpenoids, forty-four β-dihydroagarofuran acetal derivatives were designed and synthesized. Insecticidal activities and structure-activity relationship of these target compounds were evaluated. Some of the newly synthesized β-dihydroagarofuran acetal compounds were found to show higher insecticidal activity against sixth-instar larvae of Mythimna separate. Especially, compounds 2.2.9, 2.2.10, 2.2.11, 2.3.4, 2.3.6, 2.3.7, 2.5.4, 2.5.7 had great insecticidal activities with lower LD50 than that of the positive control celangulin-V (110.13 μg/g). It deserves mentioning that compound 2.2.11 showed the lowest LD50 (60.33 μg/g) among these compounds. Structure-activity relationship results suggested that the substituent groups of 1-, 6- and 9-positions of the target structures could greatly affect the insecticidal activity. Especially, when the substituent groups of 6-position were n-propyl, n-butyl, allyl, propargyl, o-fluorobenzyl, and p-fluorobenzyl, the compounds showed outstanding insecticidal activities.

Abstract:In search of environmentally benign insecticides with high activity, low toxicity and low resistance, a series of novel anthranilic(isophthalic) acid esters was designed and synthesized based on the structure of ryanodine modulating agent. All the compounds were characterized by H-1 NMR spectra, elemental analysis or high resolution mass spectrometry(HRMS). The preliminary results of biological activity assessment indicate that some of the title compounds exhibit certain but unremarkable insecticidal activity against Mythimna separata Walker at 200 mg/L and fungicidal activities against five funguses at 50 mg/L.

All of the compounds were characterized and confirmed by IR, H-1 NMR, MS and elemental analysis. The single crystal structure of 14d was determined by X-ray diffraction. The bioassay tests showed that the synthesized compounds exhibited good insecticidal activities against Mythimna separata Walker and Culex pipiens pallens.

Abstract:On the basis of etoxazole, a series of novel 2-(2,6-difluorophenyl)-4-(4-substituted phenyl)-1,3-oxazolines containing a sulfur ether moiety were designed and synthesized via the key intermediate N-(1-(4-(bromomethyl)phenyl)-2-chloroethyl)-2,6-difluorobenzamide. The bioassay results showed that most of these designed target compounds exhibited excellent acaricidal activity against both the eggs and larvae of Tetranychus cinnabarinus, especially the eggs. Among compounds with high activity against the eggs of mites, the LC50 values of 2, 11, 17, and 19 were 0.0003, 0.0002, 0.0005, and 0.0008 mg L-1, respectively, much lower than that of etoxazole (0.0089 mg L-1). Compound 2 was chosen to evaluate the acaricidal activity in the field, and the results displayed that at a concentration of 22 mg kg(-1), 2 had a much better control effect than etoxazole against both T. cinnabarinus and P. latus on eggplant. Some compounds also showed good insecticidal activities against oriental armyworm and mosquito. On the basis of our research, the newly found structure activity relationship may guide the development of new acaricides/pesticides that are required in the agriculture market.
