Abstract:The formation of two-dimensional (2D) oriented porous organic cage crystals (consisting of imine-based tetrahedral molecules) on various substrates (such as silicon wafers and glass) by solution-processing is reported. Insight into the crystallinity, preferred orientation, and cage crystal growth was obtained by experimental and computational techniques. For the first time, structural defects in porous molecular materials were observed directly and the defect concentration could be correlated with crystal growth rate. These oriented crystals suggest potential for future applications, such as solution-processable molecular crystalline 2D membranes for molecular separations.

Abstract:Shape‐persistent organic cages are an intriguing class of molecular porous materials. Through hierarchical molecular design, size and shape of the intrinsic molecular voids are controlled by dynamic covalent chemistry, while pore structure and topology are governed by noncovalent alignment in the solid state. However, the predictable and reliable crystallization of organic cages is still challenging since long‐range superstructures are solely based on weak and rather unidirectional supramolecular interactions. In this tutorial review, we provide a general classification of porous solid‐state materials and discuss specific design principles regarding the dynamic covalent reactions, the small‐molecule building blocks and solid‐state engineering. Furthermore, we introduce the most important analytical techniques for porous materials with a special focus on organic cages.

Abstract:Reticular chemistry offers the possibility of systematic design of porous materials with different pores by varying the building blocks, while the emerging porous organic cage (POC) system remains generally unexplored. A series of new POCs with dimeric cages with odd–even behaviors, unprecedented trimeric triangular prisms, and the largest recorded hexameric octahedra have been prepared. These POCs are all constructed from the same tetratopic tetraformylresorcin[4]arene cavitand by simply varying the diamine ligands through Schiff-base reactions and are fully characterized by X-ray crystallography, gas sorption measurements, NMR spectroscopy, and mass spectrometry. The odd–even effects in the POC conformation changes of the [2 + 4] dimeric cages have been confirmed by density functional theory calculations, which are the first examples of odd–even effects reported in the cavitand-based cage system. Moreover, the "V" shape phenylenediamine linkers are responsible for the novel [3 + 6] triangular prisms. The window size and environment can be easily functionalized by different groups, providing a promising platform for the construction of multivariate POCs. Use of linear phenylenediamines led to record-breakingly large [6 + 12] truncated octahedral cages, the maximum inner cavity diameters and volumes of which could be readily modulated by increasing the spacer length of the phenylenediamine linkers. Modular Self-Assembly of a Low-Symmetry Helicene Racemate into Robust Yet Facilely Recyclable Noncovalent Porous Crystals

Abstract:Porous frameworks constructed via noncovalent interactions show wide potential in molecular separation, gas adsorption, etc. However, it remains a major challenge to prepare these materials from low-symmetry molecular building blocks. Herein, we report a facile strategy to fabricate noncovalent porous crystals through modular self-assembly of a low-symmetry helicene racemate. The P and M enantiomers in the racemate first stack into right- and left-handed triangular prisms, respectively, and subsequently the two types of prisms alternatively stack together into a hexagonal network with one-dimensional channels with a diameter of 14.5 Å. Remarkably, the framework reveals high stability upon heating to 275 °C, majorly due to the abundant π-interactions between the complementarily engaged helicene building blocks. Such porous framework can be readily prepared by fast rotary evaporation, and is easy to recycle and repeatedly reform. The refined porous structure and enriched π-conjugation also favor the selective adsorption of a series of small molecules.

Abstract:Molecular cages contain an internal cavity designed to encapsulate other molecules, resulting in applications in molecular separation, gas storage, and catalysis. Introducing chirality in cage molecules can improve the selective separation of chiral molecules and add new functionalities due to the realisation of chiral photophysical properties. It has recently been shown that solid-state supramolecular interactions between achiral cages can result in the formation of chiral cavities. Here, we develop a computational technique to predict when achiral cages form chiral cavities in the solid-state through the combination of atomistic calculations and coarse-grained modelling to predict the crystalline phase behaviour. Our focus is on the achiral cage B11, which contains rotatable arene rings on the vertices of the cage that can form propeller-like orientations, inducing a chiral cavity. We show that by using dimer pair calculations, we can inform coarse-grained models to predict the packing of the cage. Our results reveal how the supramolecular interactions drive chirality in the achiral cages without the need for a chiral guest. These findings are a first step towards understanding how we can design chirality through supramolecular interactions by using abstract coarse-grained models to inform design principles for targeted solid-state phase behaviour.

Cage molecules are solution cast on various substrates to form amorphous thin films, with the structures tuned by tailoring the cage chemistry and processing conditions. General and Modular Synthesis of Covalent Organic Cages for Efficient Molecular Recognition

Abstract:Cage-type structures based on coordination and dynamic covalent chemistry have the characteristics of facile and efficient preparation but poor stability. Chemically stable organic cages, generally involving fragment coupling and multi-step reactions, are relatively difficult to synthesize. Herein, we offer a general and modular strategy to customize covalent organic cages with diverse skeletons and sizes. First, one skeleton (S) module with three extension (E) modules and three reaction (R) modules are connected by one- or two-step coupling to get the triangular monomer bearing three reaction sites. Then one-pot Friedel-Crafts condensation of the monomer and linking module of paraformaldehyde produces the designed organic cages. The cage forming could be regulated by the geometrical configuration of monomeric blocks. The S-E-R angles in the monomer is crucial; only 120o (2,4-dimethoxyphen as reaction module) or 60o (2,5-dimethoxyphen as reaction module) angle between S-E-R successfully affords the resulting cages. By the rational design of the three modules, a series of organic cages have been constructed. In addition, the host-guest properties show that the representative cages could strongly encapsulate neutral aromatic diimine guests driven by solvophobic interactions in polar solvents, giving the highest association constant of (2.58 ± 0.18) × 105 M-1.

Abstract:Porous organic molecular materials are a subclass of porous solids that are defined by their modular, molecular structures, and the absence of extended covalent or coordination bonding in the solid‐state. As a result, porous molecular materials are soluble and they can be processed into different forms, such as mixed matrix membranes. The structure of the porous modules can be fine‐tuned for specific applications, such as gas isotope separations, and in some cases the solid‐state properties of these materials can be defined by the structure of the porous molecule as viewed in isolation. In this review, the authors focus on the design of porous organic molecular materials and how their properties can be tuned for specific applications by using crystal engineering techniques. The authors distinguish between strategies where porosity is defined largely by the molecule itself, for example, in porous organic cages, and cases where porosity is generated by the solid‐state crystalline assembly. They emphasize the importance of computational techniques in the de novo design of functional, porous organic molecular materials, and how molecular modeling is applied to understand the properties of these materials.

Abstract:How molecules pack has vital ramifications for their applications as functional molecular materials. Small changes in a molecule's functionality can lead to large, non-intuitive, changes in their global solid-state packing, resulting in difficulty in targeted design. Predicting the crystal structure of organic molecules from only their molecular structure is a well-known problem plaguing crystal engineering. Although relevant to the properties of many organic molecules, the packing behaviour of modular porous materials, such as porous organic cages (POCs), greatly impacts the properties of the material. We present a novel way of predicting the solid-state phase behaviour of POCs by using a simplistic model containing the dominant degrees of freedom driving crystalline phase formation. We employ coarse-grained simulations to systematically study how chemical functionality of pseudo-octahedral cages can be used to manipulate the solid-state phase formation of POCs. While presenting a lower computational cost route for predicting molecular crystal packing, coarse-grained models also allow for the development of design rules.

Abstract:The physical properties of 3-D porous solids are defined by their molecular geometry. Hence, precise control of pore size, pore shape, and pore connectivity are needed to tailor them for specific applications. However, for porous molecular crystals, the modification of pore size by adding pore-blocking groups can also affect crystal packing in an unpredictable way. This precludes strategies adopted for isoreticular metal-organic frameworks, where addition of a small group, such as a methyl group, does not affect the basic framework topology. Here, we narrow the pore size of a cage molecule, CC3, in a systematic way by introducing methyl groups into the cage windows. Computational crystal structure prediction was used to anticipate the packing preferences of two homochiral methylated cages, CC14-R and CC15-R, and to assess the structure-energy landscape of a CC15-R/CC3-S cocrystal, designed such that both component cages could be directed to pack with a 3-D, interconnected pore structure. The experimental gas sorption properties of these three cage systems agree well with physical properties predicted by computational energy-structure-function maps.

Cage versus sheet: Probing the Determinants of Protein - Cucurbit[7]uril Crystalline Architectures

Abstract:The donut-shaped cucurbit[n]urils (Qn) are a class of rigid macrocyclic receptor with protein recognition capabilities. Qn encapsulation of amino acid side chains can enable protein assembly. Recently, cucurbit[7]uril (Q7) has been applied as a molecular glue for organizing protein building blocks into crystalline architectures. Q7 co-crystallization with dimethylated Ralstonia solanacearum lectin (RSL*) has yielded novel crystalline architectures. Co-crystallization of RSL* and Q7 yields either cage- or sheet-like architectures which may be modulated via protein engineering. However, questions remain as to the factors dictating the formation of one architecture over another (cage versus sheet). Here, we make use of an engineered RSL*-Q7 system which co-crystallizes as the cage or sheet assembly with easily-distinguished crystal morphologies. Using this model system, we probe how the crystallization conditions dictate which crystalline architecture is adopted. Protein-ligand ratios and the sodium concentration were identified as key determinants for the growth of the cage versus sheet assemblies.

Abstract:We report the use of helicene with an intrinsic helical molecular structure to prepare covalent organic cages via imine condensation. The organic cages revealed a [3+2]-type architecture containing a triple-stranded helical structure with three helicene units arranged in a propeller-like fashion with the framework integrally twisted. Such structural chirality was retained upon dissolution in organic solvents, as indicated by a strong diastereotopy effect in proton NMR and unique Cotton effects in circular dichroism spectra. Further study on chiral adsorption showed that the chiral organic cages possess considerable enantioselectivity toward a series of aromatic racemates.

Abstract:A variety of organic cages with different geometries have been developed during the last decade, most of them exhibiting a single cavity. In contrast, the number of organic cages featuring a pair of cavities remains scarce. These structures may pave the way towards novel porous materials with emergent properties and functions.We herein report on rational design of a three-dimensional hexaformyl precursor 1 , which exhibits two types of conformers, i.e . Conformer-1 and -2, with different cleft positions and sizes. Aided by molecular dynamics simulations, we select two triamino conformation capturers (denoted CC). Small-sized CC-1 selectively capture Conformer-1 by matching its cleft size, while the large-sized CC-2 is able to match and capture both conformers. This strategy allows the formation of three compounds with twin cavities, which we coin diphane. The self-assembly of diphane units results in superstructures with tunable proton conductivity, which reaches up to 1.37×10 -5 S cm -1 .

Abstract:The first two cage based crystalline covalent organic frameworks, cage-COF-1 and cage-COF-2, were constructed from a prism-like three-aldehyde-containing molecular cage. The cage contains two horizontal phloroglucinol and three vertical triazine moieties forming three identical V-shaped cavities. By reacting with p-phenylenediamine and 4,4'-biphenyldiamine, the two cage-COFs were formed with a hexagonal skeleton and possess a unique structure. Due to the pillared cage nodes, the linkers are hanging with their π-surfaces but not C-H sites exposed to the pore, and enjoy certain rotational dynamics as suggested by 13C CP/MAS NMR. The antidirection of the diimine linkages leads to rippled layers which pack in unique ABC mode through alternate stacking of the cage twosided faces in both AB and AC layers. Such packing forms trigonal channels along c axis which are interconnected in ab plane due to the large open space created across the hanging linkers, resembling the porous characteristics of 3D COFs. The cage-COFs have a permanent porosity and can adsorb CO2 facilitated by the intrinsic cage cavities that serve as prime adsorption sites. The unprecedented cage-COFs not only merge the borderline of 2D and 3D COFs but also bridge porous organic cages to extended crystalline organic frameworks.

Abstract:Among the various types of materials with intrinsic porosity, porous organic cages (POCs) are distinctive as discrete molecules that possess intrinsic cavities and extrinsic channels capable of facilitating molecular sieving. However, the fabrication of POC membranes remains highly challenging due to the weak noncovalent intermolecular interactions and most reported POCs are powders. In this study, we constructed crystalline free-standing porous organic cage membranes by fortifying intermolecular interactions through the induction of intramolecular hydrogen bonds, which was confirmed by single-crystal X-ray analysis. To elucidate the driving forces behind, a series of terephthaldehyde building blocks containing different substitutions were reacted with flexible triamine under different conditions via interfacial polymerization (IP). Furthermore, density functional theory (DFT) calculations suggest that intramolecular hydrogen bonding can significantly boost the intermolecular interactions. The resulting membranes exhibited fast solvent permeance and high rejection of dyes not only in water, but also in organic solvents. In addition, the membrane demonstrated excellent performance in precise molecular sieving in organic solvents. This work opens an avenue to designing and fabricating free-standing membranes composed of porous organic materials for efficient molecular sieving.

Abstract:Supramolecular self-assembly of biomolecules provides a powerful bottom-up strategy to build functional nanostructures and materials. Among the different biomacromolecules, protein cages offer various advantages including uniform size, versatility, multi-modularity, and high stability. Additionally, protein cage crystals present confined microenvironments with well-defined dimensions. On the other hand, molecular hosts, such as cyclophanes, possess a defined cavity size and selective recognition of guest molecules. However, the successful combination of macrocycles and protein cages to achieve functional co-crystals has remained limited. In this study, we demonstrate electrostatic binding between cationic pillar[5]arenes and (apo)ferritin cages that results in porous and crystalline frameworks. The electrostatically assembled crystals present a face-centered cubic (FCC) lattice and have been characterized by means of small-angle X-ray scattering and cryo-TEM. These hierarchical structures result in a multiadsorbent framework capable of hosting both organic and inorganic pollutants, such as dyes and toxic metals, with potential application in water-remediation technologies.
