Abstract:Synthesizing polycationic polymers directly from existing drugs overcomes the drug-loading limitations often associated with pharmacologically inert nanocarriers. We recently described nanocarriers formed from a first-generation polyamine analogue, bis(ethyl)norspermine (BENSpm), that could simultaneously target polyamine metabolism while delivering therapeutic nucleic acids. In the current study, we describe the synthesis and evaluation of self-immolative nanocarriers derived from the second-generation polyamine analogue PG-11047. Polyamines are absolutely essential for proliferation and their metabolism is frequently dysregulated in cancer. Through its effects on polyamine metabolism, PG-11047 effectively inhibits tumor growth in cancer cell lines of multiple origins as well as in human tumor mouse xenografts. Promising clinical trials have been completed verifying the safety and tolerance of this rotationally restricted polyamine analogue. We therefore used PG-11047 as the basis for Nano11047, a biodegradable, prodrug nanocarrier capable of targeting polyamine metabolism. Following exposure of lung cancer cell lines to Nano11047, uptake and intracellular degradation into the parent compound PG-11047 was observed. The release of PG-11047 highly induced the polyamine catabolic enzyme activities of spermidine/spermine N1-acetyltransferase (SSAT) and spermine oxidase (SMOX). By contrast, the activity of ornithine decarboxylase (ODC), a rate-limiting enzyme in polyamine biosynthesis and a putative oncogene, was decreased. Consequently, intracellular levels of the natural polyamines were depleted concurrent with tumor cell growth inhibition. This availability of Nano11047 as a novel drug form and potential nucleic acid delivery vector will potentially benefit and encourage future clinical studies.
Combined Dextran-Graft-Polyacrylamide/Zinc Oxide Nanocarrier for Effective Anticancer Therapy in vitro

Abstract:Introduction: Cancer chemotherapy faces two major challenges - high toxicity of active substances and tumor resistance to drugs. Low toxic nanocarriers in combination with anticancer agents can significantly increase the effectiveness of therapy. Modern advances in nanotechnology make it easy to create materials with the necessary physical and chemical properties. Methods: Two hybrid nanosystems of dextran-polyacrylamide/ zinc oxide nanoparticles (D-PAA/ZnO NPs) were synthesized in aqueous solution with zinc sulphate (D-PAA/ZnO NPs (SO42-)) and zinc acetate (D-PAA/ZnO NPs (-OAc)). The light absorption, fluorescence, dynamic light scattering and transmission electron microscopy for nanocomposite characterization were used. MTT, neutral red uptake and scratch assays were selected as fibroblasts cytotoxicity assays. Cytotoxicity was tested in vitro for normal fibroblasts, MAEC, prostate (LNCaP, PC-3, DU-145) and breast (MDA-MB-231, MCF-7) cancer cells lines. Immunocytochemical methods were used for detection of Ki-67, p53, Bcl-2, Bax, e-cadherin, N-cadherin and CD44 expression. Acridine orange was used to detect morphological changes in cells. Results: The radius of ZnO NPs (SO42-) was 1.5 nm and ZnO NPs (-OAc) was 2 nm. The nanosystems were low-toxic to fibroblasts, MAEC. Cells in the last stages of apoptosis with the formation of apoptotic bodies were detected for all investigated cancer cell lines. Proapoptotic proteins expression in cancer cells indicates an apoptotic death. Increased expression of E-cadherin and N-cadherin was registered for cancer cells line LNCaP, PC-3, DU-145 and MCF-7 after 48 h incubation with D-PAA/ZnO NPs (SO42-). Conclusion: The nanosystems were low-toxic to fibroblasts, MAEC. The D-PAA/ZnO NPs nanosystem synthesized using zinc sulphate demonstrates high cytotoxicity due to destruction of various types of cancer cells in vitro and potentially increases adhesion between cells. Thus, our findings indicate the selective cytotoxicity of D-PAA/ZnO NPs against cancer cells and can be potentially used for cancer treatment.

Abstract:The research for alternative administration methods for anticancer drugs, towards enhanced effectiveness and selectivity, represents a major challenge for the scientific community. In the last decade, polymeric nanostructured delivery systems represented a promising alternative to conventional drug administration since they ensure secure transport to the selected target, providing active compounds protection against elimination, while minimizing drug toxicity to non-target cells. In the present research, poly(glycerol sebacate), a biocompatible polymer, was synthesized and then nanostructured to allow curcumin encapsulation, a naturally occurring polyphenolic phytochemical isolated from the powdered rhizome of Curcuma longa L. Curcumin was selected as an anticancer agent in virtue of its strong chemotherapeutic activity against different cancer types combined with good cytocompatibility within healthy cells. Despite its strong and fascinating biological activity, its possible exploitation as a novel chemotherapeutic has been hampered by its low water solubility, which results in poor absorption and low bioavailability upon oral administration. Hence, its encapsulation within nanoparticles may overcome such issues. Nanoparticles obtained through nanoprecipitation, an easy and scalable technique, were characterized in terms of size and stability over time using dynamic light scattering and transmission electron microscopy, confirming their nanosized dimensions and spherical shape. Finally, biological investigation demonstrated an enhanced cytotoxic effect of curcumin-loaded PGS-NPs on human cervical cancer cells compared to free curcumin.

Abstract:Mitochondrial targeting drug delivery systems have made unprecedented progress in tumor treatment. Nevertheless, the stability of systemic circulation and the effectiveness of tumor accumulation are the basis for achieving tumor subcellular targeting. This study aims to overcome the biological barrier while improving the mitochondria-targeted effect of nanoparticles based on natural polysaccharides. Novel polysaccharide-based nanoparticles, with tumor microenvironment-responsive charge-reversal and mitochondrial targeting abilities, were prepared in our study. Curcumin (Cur) was loaded into the core of a positively charged chitosan oligosaccharide (COS) derivative with mitochondrial targeting ability, and a negatively charged shell based on angelica sinensis polysaccharide (AS) derivative was wrapped in the surface of the core. At the same time, the pH-sensitive borate ester bond was formed between the shell and the core. In vitro experiments showed that mitochondrial-targeted core-shell nanoparticles achieved charge-reversal and release more Cur in the acidic tumor microenvironment. After entering into the tumor cells, the lysosomes escape was effectively realized, and more Cur was transmitted to the mitochondria. This process led to the enhancement of the cytotoxicity, the reduction of the mitochondrial membrane potential and the activation of the apoptotic pathway. The results of in vivo experiments showed that the core-shell nanoparticles efficiently delivered the drug to the tumor site and significantly prolonged the retention time of the drug in the tumor tissue. At the same time, it had excellent antitumor activity and in vivo safety for tumor-bearing nude mice.

Abstract:Polyphenols and their derivates, a kind of natural product distributed in herb plants, vegetables, and fruits, are the most abundant antioxidants in the human diet and have been found to display cancer-preventative effects in several epidemiological studies. The scientific community has also validated the anti-cancer bioactivities and low toxicities of polyphenolic compounds, including flavones, tannins, phenolic acids, and anthocyanins, through in vitro and in vivo studies. However, the low stability, weak targeting ability, poor solubility, and low bioavailability of pure polyphenolic agents have significantly impaired their treatment efficacy. Nowadays, nano-based technology has been applied to surmount these restrictions and maximize the treatment efficacy of polyphenols. In this review, we summarize the advantages and related mechanisms of polyphenols in cancer treatment. Moreover, aiming at the poor solubility and low bioavailability of pure polyphenols in vivo, the advantages of nano-based delivery systems and recent research developments are highlighted. Herein, particular emphasis is mainly placed on the most widely used nanomaterials in the delivery of natural products, including liposomes, micelles, and nanogels. Finally, we present an overview and the challenges of future implementations of nano-based delivery systems of polyphenolic compounds in the cancer therapeutic field.

Abstract:Efficient delivery of antitumor drugs is an important area in the scientific community because of their effective distribution, high bioavailability, and improved therapeutic value. Herein, a biomimetic carrier using albumin nanoparticles (HSANP)and exosomes (EXO) was developed for the sustained release of carboplatin (CP). The formation of the nanocarrier was confirmed using FTIR, UV–Vis, fluorescence spectroscopy, DLS, and FESEM. The incubation time required for the binding of biomolecules to CP was investigated, and then CP loads on HSANP, EXO, and HSANP.EXO. The release behavior of the synthesized nanocarriers with average hydrodynamic diameters of 525 ± 11 nm, 453 ± 23 nm, and 347 ± 8 nm for CP.EXO, CP@HSANP, and CP.EXO@HSANP, respectively, was studied. The CP.EXO@HSANP system exhibited prolonged release of CP compared to the other systems. The release mechanism and kinetic parameters of the system were calculated. Furthermore, the viability of MCF7 cancer cells was assessed by MTT assay, which showed higher activity of CP.EXO@HSANP (CC50 = 0.42 mM) than that of free CP (CC50 = 0.83 mM), CP.EXO (CC50 = 0.61 mM), and CP@HSANP (CC50 = 0.96 mM). In addition, apoptotic induction by AO/PI staining indicated that CP.EXO@HSANP system affects cancer cells, MCF7, while having no significant impact on normal cells, HDF. In contrast, other systems induced more necrotic cell death in both cancer and normal cells than CP.EXO@HSANP system. Overall, biomimetic nanocarriers represent a promising approach for the sustained release of antitumor drugs, with higher efficacy and apoptotic induction.

Abstract:Abstract Several efforts have been extensively accomplished for the amelioration of the cancer treatments using different types of new drugs and less invasives therapies in comparison with the traditional therapeutic modalities, which are widely associated with numerous drawbacks, such as drug resistance, non-selectivity and high costs, restraining their clinical response. The application of natural compounds for the prevention and treatment of different cancer cells has attracted significant attention from the pharmaceuticals and scientific communities over the past decades. Although the use of nanotechnology in cancer therapy is still in the preliminary stages, the application of nanotherapeutics has demonstrated to decrease the various limitations related to the use of natural compounds, such as physical/chemical instability, poor aqueous solubility, and low bioavailability. Despite the nanotechnology has emerged as a promise to improve the bioavailability of the natural compounds, there are still limited clinical trials performed for their application with various challenges required for the pre-clinical and clinical trials, such as production at an industrial level, assurance of nanotherapeutics long-term stability, physiological barriers and safety and regulatory issues. This review highlights the most recent advances in the nanocarriers for natural compounds secreted from plants, bacteria, fungi, and marine organisms, as well as their role on cell signaling pathways for anticancer treatments. Additionally, the clinical status and the main challenges regarding the natural compounds loaded in nanocarriers for clinical applications were also discussed. Graphical abstract

Abstract:The search for safe and effective anticancer therapies is one of the major challenges of the 21st century. The ineffective treatment of cancers, classified as civilization diseases, contributes to a decreased quality of life, health loss, and premature mortality in oncological patients. Many natural phytochemicals have anticancer potential. Pentacyclic triterpenoids, characterized by six- and five-membered ring structures, are one of the largest class of natural metabolites sourced from the plant kingdom. Among the known natural triterpenoids, we can distinguish lupane-, oleanane-, and ursane-types. Pentacyclic triterpenoids are known to have many biological activities, e.g., anti-inflammatory, antibacterial, hepatoprotective, immunomodulatory, antioxidant, and anticancer properties. Unfortunately, they are also characterized by poor water solubility and, hence, low bioavailability. These pharmacological properties may be improved by both introducing some modifications to their native structures and developing novel delivery systems based on the latest nanotechnological achievements. The development of nanocarrier-delivery systems is aimed at increasing the transport capacity of bioactive compounds by enhancing their solubility, bioavailability, stability in vivo and ensuring tumor-targeting while their toxicity and risk of side effects are significantly reduced. Nanocarriers may vary in sizes, constituents, shapes, and surface properties, all of which affect the ultimate efficacy and safety of a given anticancer therapy, as presented in this review. The presented results demonstrate the high antitumor potential of systems for delivery of pentacyclic triterpenoids.

Bio-Mediated Synthesis and Characterisation of Silver Nanocarrier, and Its Potent Anticancer Action

Abstract:Discovery of a potent drug nanocarrier is crucial for cancer therapy in which drugs often face challenges in penetrating efficiently into solid tumours. Here, biosynthesis of silver nanoparticles (AgNPs) using a waste material, Garcinia mangostana (GM) fruit peel extract is demonstrated. The best condition for AgNPs synthesis was with 0.5 g of peel extract, 7.5 mM silver nitrate at 45 °C, ~pH 4 for 16 h. The synthesized AgNPs were spherical and 32.7 ± 5.7 nm in size. To test its efficiency to be used as drug carrier, plant-based drug, protocatechuic acid (PCA) was used as a test drug. AgNPs loaded with PCA (AgPCA) resulted in 80% of inhibition at 15.6 μg/mL as compared to AgNPs which only killed 5% of HCT116 colorectal cells at same concentration. The IC50 of AgNPs and AgPCA for HCT116 were 40.2 and 10.7 μg/mL, respectively. At 15.6 μg/mL, AgPCA was not toxic to the tested colon normal cells, CCD112. Ag-based drug carrier could also potentially reduce the toxicity of loaded drug as the IC50 of PCA alone (148.1 μg/mL) was higher than IC50 of AgPCA (10.7 μg/mL) against HCT116. Further, 24-h treatment of 15.6 μg/mL AgPCA resulted in loss of membrane potential in the mitochondria of HCT116 cells and increased level of reaction oxygen species (ROS). These could be the cellular killing mechanisms of AgPCA. Collectively, our findings show the synergistic anticancer activity of AgNPs and PCA, and its potential to be used as a potent anticancer drug nanocarrier.

Abstract:This study delves into the biomolecular mechanisms underlying the antitumoral efficacy of a hybrid nanosystem, comprised of a silver core@shell (Ag@MSNs) functionalized with transferrin (Tf). Employing a SILAC proteomics strategy, we identified over 150 de-regulated proteins following exposure to the nanosystem. These proteins play pivotal roles in diverse cellular processes, including mitochondrial fission, calcium homeostasis, endoplasmic reticulum (ER) stress, oxidative stress response, migration, invasion, protein synthesis, RNA maturation, chemoresistance, and cellular proliferation. Rigorous validation of key findings substantiates that the nanosystem elicits its antitumoral effects by activating mitochondrial fission, leading to disruptions in calcium homeostasis, as corroborated by RT-qPCR and flow cytometry analyses. Additionally, induction of ER stress was validated through western blotting of ER stress markers. The cytotoxic action of the nanosystem was further affirmed through the generation of cytosolic and mitochondrial reactive oxygen species (ROS). Finally, in vivo experiments using a chicken embryo model not only confirmed the antitumoral capacity of the nanosystem, but also demonstrated its efficacy in reducing cellular proliferation. These comprehensive findings endorse the potential of the designed Ag@MSNs-Tf nanosystem as a groundbreaking chemotherapeutic agent, shedding light on its multifaceted mechanisms and in vivo applicability.

Dual drug-loaded nano-platform for targeted cancer therapy: toward clinical therapeutic efficacy of multifunctionality

Abstract:Abstract Background Poor targeting and penetration of chemotherapy drugs in solid tumors, and the development of resistance to chemotherapeutic agents are currently hindering the therapy of breast cancer; meanwhile, breast cancer metastasis is one of the leading causes of death in breast cancer patients. With the development of nanotechnology, nanomaterials have been widely used in tumor therapy. Results A multi-functional nano-platform containing gambogic acid (GA) and paclitaxel (PTX) was characterized by a small size, high encapsulation efficiency, slow release, long systemic circulation time in vivo, showed good targeting and penetrability to tumor tissues and tumor cells, and exhibited higher anti-tumor effect and lower systemic toxicity in BALB/c mice bearing 4T1 tumor. GA not only overcame the multidrug resistance of PTX by inhibiting P-glycoprotein (P-gp) activity in MCF-7/ADR cells, but also inhibited MDA-MB-231 cells migration and invasion, playing a crucial role in preventing and treating the lung metastasis of breast cancer caused by PTX; meanwhile, the synergistic anti-tumor effect of GA and PTX has also been verified in vitro and in vivo experiments. Conclusion Our data described the better recognition and penetration of tumor cells of R9dGR-modified versatile nanosystems containing GA and PTX, which exerted one stone three birds clinical therapeutic efficacy of multifunctionality.

Bioinspired neuromelanin-like Pt(iv) polymeric nanoparticles for cancer treatment

Abstract:To expand the chemotherapeutic potential of platinum complexes, different approaches have been followed, two of the most relevant being their administration as the prodrug Pt( iv ) and encapsulation in nanocarriers. Herein, we demonstrate how neuromelanin may become a good bioinspiration for the synthesis of nanoparticles (NPs), combining both approaches. For this, complex PtBC reacts with sodium periodate, inducing a melanization process and the formation of nanoparticles. In vitro results on non-malignant human fibroblast cells (1Br3G), human cervical cancer, murine glioma (GL261), and human ovarian cancer confirmed its therapeutic efficacy. The role of the Pt( iv ) ion on the cytotoxicity effects was confirmed by comparison with the results obtained for a family of nanoparticles obtained with nordihydroguaiaretic acid under the same experimental conditions. Finally, intranasal administration of the NPs in orthotopic glioblastoma multiforme murine models in female C57BL/6 mice showed excellent in vivo biodistribution and tolerability. Overall, this innovative approach represents a step toward more specific and less toxic therapies in the field of cancer chemotherapy.

Novel polyurea/polyurethane nanocapsules loaded with a tambjamine analog to improve cancer chemotherapy delivery and safety in lung cancer

Abstract:Background The clinical potential of many promising anti-tumor drugs is restricted by their intrinsic physicochemical properties and lack of selectivity towards cancer cells, leading to insufficient drug concentration at the tumor site. To overcome these hurdles, we have developed a new drug delivery system based on polyurea/polyurethane nanocapsules (NCs), which might protect the loaded drug from premature degradation and specifically release it inside cancer cells. Polyurea/polyurethane polymers present amphoteric properties, thus, these NCs are anionic at basic pH and cationic at acid pH. Tumor microenvironment is slightly acidic (6.8-7) in comparison to healthy tissue (7.2-7.4); hence, NCs became cationic in the tumor area, what facilitates their internalization into tumor cells. Methods In order to study this novel delivery system, an analog of tambjamine has been loaded in these amphoteric NCs and their selectivity and antitumor properties have been analyzed in vitro comparing two pHs, one acidic (6.8) and another basic (7.5) by proliferation assays, flow cytometry and microscopy. Additionally, the toxicity and efficacy of this delivery approach has been evaluated in an orthotopic lung cancer mouse model. Results In vitro results showed higher levels of NCs uptake in tumor cells at pH 6.8, resulting in greater cytotoxic effects compared to those at pH 7.5. Toxicity studies in vivo showed a good tolerability to repeated administrations and furthermore, the encapsulation of the drug reduces its toxicity compared to the free compound, allowing its intravenously administration. Preliminary data show a tendency limiting tumor growth, suggesting that NCs have reached the tumor site and the drug has been released. Conclusions Altogether, we have developed a promising new drug delivery system based on polyurea/polyurethane nanocapsules loaded with a tambjamine analog in order to be specifically delivered in cancer cells through reductive conditions, facilitating drug transport to tumor, administration and improving drug safety. Abstract:Cancer forms exhibiting poor prognosis have been extensively researched for therapeutic solutions. One of the conventional modes of treatment, chemotherapy shows inadequacy in its methodology due to imminent side-effects and acquired drug-resistance by cancer cells. However, advancements in nanotechnology have opened new frontiers to significantly alleviate collateral damage caused by current treatments via innovative delivery techniques, eliminating pitfalls encountered in conventional treatments. Properties like reduced drug-clearance and increased dose efficacy by the enhanced permeability and retention effect deem nanoparticles suitable for this application. Optimization of size, surface charge and surface modifications have provided nanoparticles with stealth properties capable of evading immune responses, thus deeming them as excellent carriers of chemotherapeutic agents. Biocompatible and biodegradable forms of polymers enhance the bioavailability of chemotherapeutic agents, and permit a sustained and time-dependent release of drugs which is a characteristic of their composition, thereby providing a controlled therapeutic approach. Studies conducted <i>in vitro</i> and animal models have also demonstrated a synergism in cytotoxicity given the mechanism of action of anticancer drugs when administered in combination providing promising results. Combination therapy has also shown implications in overcoming multiple-drug resistance, which can however be subdued by the adaptable nature of tumor microenvironment. Surface modifications with targeting moieties can therefore feasibly increase nanoparticle uptake by specific receptor-ligand interactions, increasing dose efficacy which can seemingly overcome drug-resistance. This article reviews recent trends and investigations in employing polymeric nanoparticles for effectively delivering combination chemotherapy, and modifications in delivery parameters enhancing dose efficacy, thus validating the potential in this approach for anticancer treatment.
A Convergent Synthetic Platform for Dual Anticancer Drugs Functionalized by Reduced Graphene Nanocomposite Delivery for Hepatocellular Cancer.

Abstract:Hepatocellular carcinoma (HCC) is widespread cancer with a high degree of morbidity and mortality in individuals worldwide and a serious concern for its resistance to present chemotherapy drugs. In this investigation, the combination of cisplatin (CPT) and metformin (MET) to kill the HepG2 and caco-2 cells was developed into a new pH-responding magnetic nanocomposite based on reduced graphene oxide. Polyhydroxyethyl methacrylic (PHEA) was then linked employing grafting from approach to the reduced graphene oxide by ATRP polymerization (Fe3O4@rGO-G-PSEA). FT-IR, SEM, XRD, DLS, and TGA analyses evaluated physicochemical characteristics of the nanocomposite. In addition, the cellular uptake property of the nanocomposites was examined by the HepG2 cells. The outcomes of cell viability results indicate that the nanoparticles loaded with MET&CPT showed the lowest concentration rate of HepG2 and Caco-2 cells compared to the drug-loaded single nanocomposite groups and free drugs. The histological analysis has demonstrated relatively safe and does not produce different stress such as swelling and inflammation of the mice organs. Our results show the enhancement in cytotoxicity in HepG2 and Cocoa-2 cells by MET and CPT graphene oxide-based nanocomposite by promoting apoptotic response. Moreover, Fe3O4@rGO-G-PSEA showed potent in vivo antitumor efficacy but showed no adverse toxicity to normal tissues. Together, this study can provide insight into how surface embellishment may tune these nanocomposites' tumor specificity and provide the basis for developing anticancer efficacy.
