Abstract:Label-free vibrational imaging by stimulated Raman scattering (SRS) provides unprecedented insight into real-time chemical distributions. Specifically, SRS in the fingerprint region (400–1800 cm −1 ) can resolve multiple chemicals in a complex bio-environment. However, due to the intrinsic weak Raman cross-sections and the lack of ultrafast spectral acquisition schemes with high spectral fidelity, SRS in the fingerprint region is not viable for studying living cells or large-scale tissue samples. Here, we report a fingerprint spectroscopic SRS platform that acquires a distortion-free SRS spectrum at 10 cm −1 spectral resolution within 20 µs using a polygon scanner. Meanwhile, we significantly improve the signal-to-noise ratio by employing a spatial-spectral residual learning network, reaching a level comparable to that with 100 times integration. Collectively, our system enables high-speed vibrational spectroscopic imaging of multiple biomolecules in samples ranging from a single live microbe to a tissue slice.
Spectrometer-free Vibrational Imaging by Retrieving Stimulated Raman Signal from Highly Scattered Photons

Abstract:In vivo vibrational spectroscopic imaging is inhibited by relatively slow spectral acquisition on the second scale and low photon collection efficiency for a highly scattering system. Recently developed multiplex coherent anti-Stokes Raman scattering and stimulated Raman scattering techniques have improved the spectral acquisition time down to microsecond scale. These methods using a spectrometer setting are not suitable for turbid systems in which nearly all photons are scattered. We demonstrate vibrational imaging by spatial frequency multiplexing of incident photons and single photodiode detection of a stimulated Raman spectrum within 60 μs. Compared to the spectrometer setting, our method improved the photon collection efficiency by two orders of magnitude for highly scattering specimens. We demonstrated in vivo imaging of vitamin E distribution on mouse skin and in situ imaging of human breast cancerous tissues. The reported work opens new opportunities for spectroscopic imaging in a surgical room and for development of deep-tissue Raman spectroscopy toward molecular level diagnosis.

Abstract:Coherent Raman microscopy provide label-free imaging by interrogating the intrinsic vibration of biomolecules. Nevertheless, trade-off between high chemical-specificity and high imaging-speed currently exists in transition from spectroscopy to spectroscopic imaging when capturing dynamics in complex living systems. Here, we present a novel concept in dual-comb scheme to substantially beat this trade-off and facilitate high-resolution broadband coherent anti-Stokes Raman spectroscopic imaging based on down-converted, automatically varying delay-time in spectral focusing excitation. A rapid measurement of vibrational microspectroscopy on sub-microsecond scale over a spectral span ~700 cm-1 with solid signal-to-noise ratio provides access to well-resolved molecular signatures within the fingerprint region. We demonstrate this high-performance spectroscopic imaging of spatially inhomogeneous distributions of chemical substances as well as the carotenoids accumulation in plant cells. This technique offers an unprecedented route for broadband CARS imaging with hundreds of kHz frame rate using available 1-GHz oscillators.

Abstract:Coherent anti-Stokes Raman scattering (CARS) spectroscopy with time-delayed ultrashort pulses and a single-pixel photodetector has shown great potential for spectroscopic imaging and transient studies in chemistry and biological research. However, those systems rely on mechanical delay lines or two asynchronous optical combs with inflexible repetition frequencies, technically limiting their acquisition speeds. Here, we demonstrate a hybrid dual-comb CARS system involving a broadband fiber laser and a highly-flexible, frequency-modulated electro-optic comb. We achieve multiplex CARS spectra (2800-3200 cm-1), with a moderate resolution (22 cm-1), at a maximum refresh rate of 1 MHz, limited by the radio-frequency synthesizer we use. Fast spectroscopic CARS imaging is demonstrated for liquid mixtures. Our system enables spectral measurements in the high-wavenumber C-H stretching region at a record speed that is an order of magnitude higher than state-of-the-art systems, which may open up new opportunities for fast chemical sensing and imaging.

Abstract:We report an all-fiber free-running bidirectional dual-comb laser system for coherent anti-Stokes Raman scattering spectroscopy based on spectral focusing. The mode-locked oscillator is a bidirectional ring-cavity erbium fiber laser running at a repetition rate of ∼114MHz. One output of the bidirectional laser is wavelength-shifted from 1560 to 1060 nm via supercontinuum generation for use as the pump source. We have been able to record the Raman spectra of various samples such as polystyrene, olive oil, polymethyl methacrylate (PMMA), and polyethylene in the C-H stretching window. We believe that this all-fiber laser design has promising potential for coherent Raman spectroscopy and also label-free imaging for a variety of practical applications.

Abstract:We propose an all-fiber-generated dual-soliton pulses based scheme for the background-free detection of coherent anti-Stokes Raman spectroscopy under the spectral focusing mechanism. Due to the strong birefringence and high nonlinearity of a polarization-maintaining photonic crystal fiber (PM-PCF), two redshifted soliton pulses can be simultaneously generated relying on high-order dispersion and nonlinear effects along two eigenpolarization axes in the anomalous dispersion regime, while allowing feasible tunability of the frequency distance and temporal interval between them. This proposed scheme, termed as DS-CARS, exploits a unique combination of slight frequency-shift and advisable temporal walk-off of this two soliton pulses to achieve robust and efficient suppression of nonresonant background with compact all-fiber coherent excitation source. Capability of the DS-CARS is experimentally demonstrated by the background-free CARS spectroscopy and unambiguous CARS microscopy of polymer beads in the fingerprint region.

Abstract:Rapid multiplex coherent anti-Stokes Raman scattering (CARS) spectroscopy in the high-wavenumber (HW) region shows great advantages in real-time dynamic process visualizations, clinical diagnosis, abundant microplastic assessment, etc. Fourier transform CARS (FT-CARS) improves the acquisition speed of multiplex CARS to the order of tens of kHz, yet typical approaches to utilize the intrinsic coherence of ultrabroadband pulses notably impede the attainable Raman vibrations in the HW region. Here, a novel delay-spectral focusing dual-comb (DC) CARS scheme is proposed for rapid HW Raman detection based on two fiber combs with 100 MHz repetition rates. By combining the particular advantages of DC asynchronous optical sampling and spectral focusing instantaneous single molecular vibration excitation, the Raman spectrum directly maps to DC relative delay, releasing the coherence constraint of excitation sources. Thus, an Er comb and a Yb comb, as pump and Stokes excitation pulses, respectively, flexibly match the Raman vibration in the HW region. Further, a rapid delay focusing method with intracavity electro-optic modulation is applied to actively control the DC relative delay scanning in the Raman shift region of interest. With these efforts, the spectral acquisition rate is improved more than 1000-fold up to 40 000 spectra/s, while keeping spectral resolution (∼10 cm–1) and the signal-to-noise ratio (∼260) stable along with active acquisition rate tuning. Combined with a microfluidic device, high-speed measurement in the HW region of 15 μm microbeads in flow is demonstrated by the system, reaching a high throughput of around 2150 events/s and more than 99% classification consistency. The prospect of rapid acquiring multiplex CARS spectra without the sacrifice of spectral resolution or the need for the coherence of the pulse sources offers huge potential in rapid monitoring circumstances, such as flow cytometry and microspectroscopic imaging in biomedicine.

Abstract:Rapid coherent Raman hyperspectral imaging shows great promise for applications in sensing, medical diagnostics, and dynamic metabolism monitoring. However, the spectral acquisition speed of current multiplex coherent anti-Stokes Raman scattering (CARS) microscopy is generally limited by the spectrometer integration time, and as the detection speed increases, the signal-to-noise ratio (SNR) of single spectrum will decrease, leading to a terrible imaging quality. In this Letter, we report a dual-comb coherent Raman hyperspec-tral microscopy imaging system developed by integrating two approaches, a rapid delay-spectral focusing method and deep learning. The spectral refresh rate is exploited by focusing the relative delay scanning in the effective Raman excitation region, enabling a spectral acquisition speed of 36 kHz, approximate to 4 frames/s, for a pixel resolution of 95 x 95 pixels and a spectral bandwidth no less than 200 cm(-1). To improve the spectral SNR and imaging quality, the deep learning models are designed for spectral preprocessing and automatic unsupervised feature extraction. In addition, by changing the relative delay focusing region of the comb pairs, the detected spectral wavenumber region can be flexibly tuned to the high SNR region of the spectrum. Abstract:Coherent anti‐Stokes Raman scattering performed with a synchronously pumped dual optical parametric oscillator allows to image high wavenumber and cell silent regions simultaneously without motion artefacts. Imaging was performed using an emulsion composed of water, heavy water, and oil, in addition to flowing plastic beads and deuterated HeLa cells. The system is sensitive enough to separate deuterated and regular lipids inside a human cell and allows to effectively perform SHG/THG microscopy due to built‐in group delay dispersion control. Coherent anti‐Stokes Raman scattering microscopy probing two independently selectable vibrational frequencies across the vibrational spectrum is demonstrated. A one‐box femtosecond laser source provides three synchronised 100–200 fs pulses of 77 MHz repetition rate, one at a fixed wavelength of 1025 nm from an Yb oscillator and two independently tuneable ones, over 680–960 nm and 960–1300 nm, respectively, from two optically synchronised parametric oscillators. Combined with spectral focusing, the source allows addressing the fingerprint, cell silent and C–H stretch vibrational regions simultaneously, avoiding the motion artefacts seen in sequential imaging. We demonstrate the microscopy method on water/heavy water/oil emulsions, plastic beads and deuterated lipids inside cells.

Abstract:The dual-comb coherent anti-Stokes Raman scattering (CARS) technique has been proposed and developed for many years with various applications. It maps the spectral information to the time domain and resolves Raman peaks by scanning the relative delay of the pulse pair. One main drawback of this method is the low duty cycle. The span of scanning is far larger than needed, leading to a massive waste of time and energy. In this Letter, a dual-comb fiber laser system with fast repetition modulation ability is proposed and demonstrated. It can detect the relative delay via an asymmetric sum-frequency-mixing method and flip the repetition frequency difference at the exact time to increase the duty cycle by several hundred times. Consequently, it is expected that the detection efficiency is greatly enhanced compared to traditional dual-comb systems for CARS measurements.

Abstract:Spectroscopy of molecular mixtures with broadband femtosecond laser pulses often involves simultaneous excitation of multiple molecular species with close resonance frequencies. Interpreting the collective optical response typically requires Fourier analysis of time-resolved signals. We propose and demonstrate an alternative method of separating coherent Raman scattering signals from two molecular components with neighboring vibrational modes. We exploit "silence windows," which arise from coupling of the vibrational and rotational degrees of freedom in molecular dynamics. In silence windows, the detected signal stems solely from the minority species (here, CO2), while the strong resonant background from the dominant species (O2) is greatly suppressed.

Abstract:Though dual soliton is used as the Stokes source in coherent anti-Stokes Raman scattering (CARS) experiments, the mechanism behind it is still not fully investigated. In this paper, dual-soliton pulses generation with a highly birefringent photonic crystal fiber (PCF) is numerically explored and experimentally verified. The simulated pulse exhibits various characteristics in the temporal and spectral domains with respect to the input linear polarization angle, and can be classified into four regions based on distances in the temporal and spectral domain of the soliton pair. By tuning input power and polarization angle, a soliton pair with desirable spectral coverage and temporal delay for CARS applications can be flexibly generated before the two solitons overlap in time domain. The simulated results are then experimentally validated by comparing their temporal and spectral characteristics. In addition, the benefit of using dual-soliton sources, such as Stokes beam, in CARS experiments is also demonstrated. The results and methods presented in this work can improve CARS spectroscopy and microscopy techniques, and may also be beneficial to other dual-wavelength source applications.

Abstract:We present a simple and easily implementable scheme for multiplexed Coherent Anti-Stokes Raman Scattering (CARS) spectroscopy and microscopy using a single femtosecond pulse, shaped with a narrow spectral notch. We show that a tunable spectral notch, shaped by a resonant photonic crystal slab, can serve as a narrowband, optimally time-delayed probe, resolving a broad vibrational spectrum with high spectral resolution in a single-shot measurement. Our single-source, single-beam scheme allows the simple transformation of any multiphoton microscope with adequate bandwidth into a nearly alignment-free CARS microscope.

Abstract:Femtosecond coherent anti-Stokes Raman scattering (CARS) spectra suffer from low spectral resolution because of the broadband laser spectrum. In this paper, we propose a feasible scheme to achieve a high-resolution two-pulse CARS spectrum by shaping both the pump and probe pulses using rectangular amplitude modulation. We show that a narrowband hole in the CARS spectrum can be created by the amplitude-shaped laser pulse, the position of which is correlated with the Raman resonant frequency of the molecule. Thus, by observing holes in the CARS spectrum, we are able to obtain a high-resolution CARS spectrum and the energy-level diagram of the molecule.
Broadband coherent Raman scattering spectroscopy at 50,000,000 spectra/s

Abstract:Raman scattering spectroscopy is widely used as an analytical technique in various fields, but its measurement process tends to be slow due to the low scattering cross-section. In the last decade, various broadband coherent Raman scattering spectroscopy techniques have been developed to address this limitation, achieving a measurement rate of about 100 kSpectra/s. Here, we present a significantly increased measurement rate of 50 MSpectra/s, which is 500 times higher than the previous state-of-the-art, by developing time-stretch coherent Raman scattering spectroscopy. Our newly-developed system, based on a mode-locked Yb fiber laser, enables highly-efficient broadband excitation of molecular vibrations via impulsive stimulated Raman scattering with an ultrashort femtosecond pulse and sensitive time-stretch detection with a picosecond probe pulse at a high repetition rate of the laser. As a proof-of-concept demonstration, we measure broadband coherent Stokes Raman scattering spectra of organic compounds covering the molecular fingerprint region from 200 to 1,200 cm-1. This high-speed broadband vibrational spectroscopy technique holds promise for unprecedented measurements of sub-microsecond dynamics of irreversible phenomena and extremely high-throughput measurements.
