Abstract:Staphylococcus aureus (S. aureus) is a major human pathogen that may produce a variety of toxins and cause staphylococcal food poisoning. In the present study, a direct loop-mediated isothermal amplification (LAMP) assay was developed and validated to detect S. aureus in food samples. Without prior cultural enrichment and DNA extraction steps, bacterial DNA was released by heating at 100°C for 5 min and directly subjected to LAMP assay. Using a set of LAMP primers recognizing six distinct regions of nuc gene, the developed direct LAMP assay was highly specific, and the analytical sensitivity was determined to be 7.6 × 102 CFU/mL. Moreover, with the pre-mixed LAMP reagents stored at -20°C, the entire assay should be finished within 40 min. These features greatly simplified the operating procedure and made the direct LAMP a powerful tool for rapid and on-site detection of S. aureus in food samples.

Abstract:Staphylococcus aureus (S. aureus) is a versatile and virulent pathogen and has become one of the leading causes of life-threatening bloodstream infections, such as sepsis and endocarditis. The morbidity and mortality of S. aureus bacteremia remain high in recent years. Rapid identification of S. aureus can lead to faster initiation of appro-priate antibiotic treatment, thereby reducing treatment costs and mortality. Blood culture is the gold standard for diagnosing bacteremia, but it is time-consuming and poorly sensitive. Herein, vancomycin modified magnetic nanoparticles (VMPs) and separation glue were introduced to enrich S. aureus in blood in a direct and convenient manner, and then the bacteria were detected by droplet digital PCR (ddPCR). The nanoparticle-based culture-free method can concentrate and detect S. aureus from whole blood within 3 h, significantly shortening the detection time compared with the traditional culture method (3-5 days). The limit of detection of this approach for S. aureus was as low as 10 CFU/mL, which significantly improved the diagnostic rate in blood bacteremia. The developed VMPs-ddPCR strategy could be a promising tool for identifying S. aureus, guiding treatment decisions in the early stages of bloodstream infections, and predicting efficacy and thereby reducing mortality.

Abstract:Staphylococcal infection is one of the most pressing problems in modern medicine due to the increasing antibiotic resistance with the overuse of antibiotics. Conventional methods for identification and antimicrobial susceptibility testing (AST) generally take 3-7 days and require skilled technicians. Rapid identification of Staphylococcus aureus directly from positive blood culture media using quantum dots as fluorescence probes

Abstract:The subcultivation of positive blood cultures for organism identification is a time-consuming process. We explored the use of fluorescent quantum dots (QDs) as probes for the improved detection and identification of Staphylococcus aureus directly from positive blood culture media. Immunofluorescence probes were synthesized by linking biotin-conjugated QDs to streptavidin-conjugated IgG molecules. This QDs-IgG complex is capable of binding anti-SPA antibodies which specifically bind to a cell surface protein of S. aureus. The specificity of the method was confirmed using reference strains. Among 372 positive blood cultures, 73 containing gram-positive cocci in clusters were investigated. Relative to the standard culture method, the immunofluorescence assay showed 90.5% (19/21) sensitivity and 100% (52/52) specificity for S. aureus. These results suggest that this immunofluorescence assay allows the rapid identification of S. aureus directly from specimens.

Abstract:Staphylococcus aureus (S. aureus), a Gram-positive bacterium, is often found on the skin, nasal passages, and mucous membranes of healthy individuals, where it acts as a commensal pathogen. Therefore, it is crucial to have a rapid and sensitive detection method to reduce the spread of S. aureus infections effectively. This study developed a reliable and rapid detection protocol to identify S. aureus DNA in skin infections. The assay exhibited no cross-reactivity with four other bacterial strains and demonstrated a sensitivity of 10 2 CFU/mL. When tested on 24 simulated samples, the MB-LAMP assay results were consistent with those of qPCR, but with a significantly reduced testing time of approximately 30 minutes. Furthermore, this detection scheme can also rapidly detect other pathogens such as Salmonella, Monocytogenes, and Vibrio parahaemolyticus.

Abstract:Background: Staphylococcus is a genus of Gram-positive bacteria, known to cause food poisoning and gastrointestinal illness in humans. Additionally, the emergence of methicillin-resistant S. aureus (MRSA) strains has caused a major health care burden worldwide. Cronobacter is a group of Gram-negative bacteria that can survive in extreme dry conditions. Cronobacter sakazakii is known to contaminate powdered infant formula and cause life-threatening infections in neonates. Vibrio is a genus of human-pathogenic Gram-negative bacteria that can cause foodborne illness by consuming undercooked or raw seafood. Vibrio parahaemolyticus can cause serious gastrointestinal disease in humans. Thus, rapid identification of Staphylococcus spp., Cronobacter spp., and Vibrio spp. is crucial for the source tracking of contaminated food, as well as to measure the transmission dynamics of these bacterial pathogens causing foodborne diseases and outbreaks. Objective: This single-laboratory performance evaluation study used the VITEK MS system to evaluate the potential of MALDI-TOF MS technology for rapid identification of S. aureus-like, C. sakazakii-like, and V. parahaemolyticus-like isolates of public health importance. Method: A total of 226 isolates recovered from various food, environmental surveillance samples, and other sources were identified by bioMérieux VITEK 2 and VITEK MS systems as Staphylococcus spp., Cronobacter spp., and Vibrio spp. Five American Type Culture Collection (ATCC) reference Gram-positive and Gram-negative bacterial isolates were also tested to complete the study. In addition, for some Staphylococcus spp. isolates, whole genome sequencing (WGS) and DNA sequencing of 16S rRNA partial region were also performed for species identification. Results: The VITEK MS system was able to provide species identification to all 96 isolates of Staphylococcus spp. and to all 29 isolates of Vibrio spp. examined with a high confidence value (99.9%). Similarly, species identification was observed for the majority of spots (245 of 303) for the 101 Cronobacter spp. isolates (∼82.0%) with a high confidence value (99.9%), and genus level identification was noticed for the rest of the Cronobacter spp. isolates (18.0%; 58 of the 303 spots) analyzed. Species identification data generated by VITEK 2 system were comparable to data obtained by the VITEK MS system. Conclusions: The VITEK MS system is a reliable high-throughput platform that can rapidly identify Staphylococcus, Vibrio, and Cronobacter to the genus level, as well as S. aureus, C. sakazakii, V. parahaemolyticus, and other closely related foodborne isolates and bacterial isolates from additional sources, in most cases. Highlights: The VITEK MS system can be used in the rapid genus and species identification of human-pathogenic Staphylococcus spp., Cronobacter spp., and Vibrio spp. isolates.

Abstract:Staphylococcus aureus (S. aureus) is one of the most common pathogens for nosocomial and community infections, which is closely related to the occurrence of pyogenic and toxic diseases in human beings. In the current study, a lab-built microchip capillary electrophoresis (microchip CE) system was employed for the rapid determination of S. aureus, while a simple-to-use space domain internal standard (SDIS) method was carried out for the reliable quantitative analysis. The precision, accuracy, and reliability of SDIS were investigated in detail. Noted that these properties could be elevated in SDIS compared with traditional IS method. Remarkably, the PCR products of S. aureusnuc gene could be identified and quantitated within 80 s. The theoretical detection limit could achieve a value of 0.066 ng/μL, determined by the using SDIS method. The current work may provide a promising detection strategy for the high-speed and highly efficient analysis of pathogens in the fields of food safety and clinical diagnosis.

Rapid detection and differentiation of Staphylococcus colonies using an optical scattering technology

Abstract:Staphylococcus species are a major pathogen responsible for nosocomial infections and foodborne illnesses. We applied a laser-based BARDOT (bacterial rapid detection using optical scattering technology) for rapid colony screening and detection of Staphylococcus on an agar plate and differentiate these from non-Staphylococcus spp. Among the six growth media tested, phenol red mannitol agar (PRMA) was found most suitable for building the Staphylococcus species scatter image libraries. Scatter image library for Staphylococcus species gave a high positive predictive value (PPV 87.5-100%) when tested against known laboratory strains of Staphylococcus spp., while the PPV against non-Staphylococcus spp. was 0-38%. A total of nine naturally contaminated bovine raw milk and ready-to-eat chicken salad samples were tested, and BARDOT detected Staphylococcus including Staphylococcus aureus with 80-100% PPV. Forty-five BARDOT-identified bacterial isolates from naturally contaminated foods were further confirmed by tuf and nuc gene-specific PCR and 16S rRNA gene sequence. This label-free, non-invasive on-plate colony screening technology can be adopted by the food industries, biotechnology companies, and public health laboratories for Staphylococcus species detection including S. aureus from various samples for food safety and public health management. Abstract:The detection of Staphylococcus aureus (S. aureus) is very important as it is responsible for bacterial infectious diseases and food poisoning. In this paper we explored the application of fluorescently labelled vancomycin to specifically bind and detect S. aureus. In view of the specificity of vancomycin towards bacterial cell surfaces, we utilized Cy5 to label vancomycin (Cy5-Van) for the identification of S. aureus. Our experiments were designed to examine in greater detail the specificity of the reaction between Cy5-Van and S. aureus. Detection parameters such as the derivatization conditions, concentrations of buffer, pH value, response performance of Cy5-Van to bacterial surface, injection time and reversed-polarity time have been investigated and optimized. To develop a simple and quick assay for the detection of S. aureus at low concentrations, we propose to use the Cy5-Van for labeling the S. aureus coupled with an on-line multiple-concentration in microchip electrophoresis. Under the optimized conditions, the detection of S. aureus was achieved within 150s with limit of detection (S/N=3) of 981 CFU/mL, and 350-fold enhancement was obtained for S. aureus as compared to using the no concentration step. It is self-evident that this approach has great potential in the future for the analysis of S. aureus.
Evaluation Of An In-House Maldi-Tof Ms Rapid Diagnostic Method For Direct Identification Of Micro-Organisms From Blood Cultures

Abstract:Purpose. Bloodstream infections are major causes of morbidity and mortality among hospitalized patients worldwide. Early identification of micro-organisms from blood culture can facilitate earlier optimization of treatment. The objective of this study was to assess an in-house method based on a new matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) platform (Clin-TOF MS) for direct organism identification.Methodology. We studied the performance of the in-house method for direct identification and the conventional sub-culture method in parallel. Identification from subcultures was analysed with Bruker MS as the reference method.Results. A total of 666 blood cultures with a single micro-organism that flagged positive after no more than a 3-day incubation period were collected. The identification accuracy of the in-house Clin-TOF MS method for direct identification and the sub-culture method was 88.6 and 100 %, respectively. The in-house method exhibited better performance for Gram-negative bacteria than for Gram-positive bacteria (93.3 vs 81.6 %). The accuracy rate for anaerobes was 100% (3/3). The lowest accurate identification rate was for yeast; this was only 20 %. Lytic Anaerobic/F (LAF) and Plus Aerobic/F (PAF) provided the highest accurate identification rates, and it was noteworthy that the accuracy rate for FAN Aerobic (FA) was 82 %, which is higher than previously reported and showed that the method was effective.Conclusion. Our study provides an effective sample preparation method for the direct identification of pathogens from positive blood culture vials via Clin-TOF MS at a very low cost of about $0.5 per sample and with a short turnaround time of about 20 min. This will help clinicians make precise diagnoses and provide targeted prescriptions, reducing the risk of the potential development of resistance.
Fluorescent Identification and Detection of Staphylococcus Aureus with Carboxymethyl Chitosan/cds Quantum Dots Bioconjugates.

Abstract:A fast and sensitive method based on fluorescent carboxymethyl chitosan/CdS quantum dots (CMCS-CdS QDs) composites was developed for specific detection of Staphylococcus aureus in food and the environment. Fluorescent CMCS-CdS QDs were prepared in aqueous solution through a green method. A human immunoglobulin (IgG) antibody was then bioconjugated to the QDs in the presence of 1-ethyl-3-(3)-dimethylaminopropyl carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS) to make a novel type of mono-dispersed water-soluble fluorescent bioprobes. The fluorescent bioprobes were employed to identify S. aureus by incubating them with the bacteria for a certain time and observing the marked cells under fluorescence microscopy after removing free fluorescent QDs. Fluorescence microscopy images showed the S. aureus cells were successfully recognized by the bioprobes. Several other bacteria commonly found in environment such as Escherichia coli and Bacillus subtilis were also incubated with the bioprobes to test their specificity. It was found that the novel QDs-CMCS-IgG bioprobes had specific identification to S. aureus cells. Fluorescence measurement using a luminescence spectrometer could be applied to quantify S. aureus cells. The fluorescence intensity of the samples at 600 nm was proportional to the cell concentration in the range of 10(3)-10(7) cfu/ml, and the detection limit was as low as 900 cfu/ml. Considering the simplicity and cost-efficiency of this method, its application in the identification and quantification of bacteria in clinical, food and environmental samples is anticipated. Abstract:Clostridium difficile infection(CDI)is a global public health issue and one of the most common pathogenic infections in the community and hospitals, causing varying degrees of diarrhea and pseudomembranous colitis.The current CDI diagnosis is based on the detection of clostridium difficile(CD)associated toxins or toxin encoding genes in patients′ stool, but these traditional microbiological techniques have limitations that may delay disease diagnosis.In clinical and research, CD has a distinct odor "horse barn odor" , which is caused by the volatile organic compounds(VOCs). VOCs in human breath, blood, urine and feces have recently received increased attention as potential noninvasive diagnostic biomarkers for a wide range of diseases, including infectious diseases and digestive tract disorders.It was discovered that VOCs from CD, with their potential characteristic VOCs, contributed to the rapid diagnosis of CDI.In this paper, we review VOCs and their application in the rapid detection of CDI.

Abstract:Given the significant threat to human health posed by Staphylococcus aureus, rapid and accurate quantification of S. aureus is essential to prevent and diagnose foodborne diseases and to ensure the safety of food consumption. Here, we developed a novel, simple，and label-free biosensor for quantitatively detecting S. aureus by integrating an allosteric aptamer probe and a rolling circle amplification technique. Pathogen S. aureus-specific binding could induce the structural change of the allosteric aptamer beacon to expose the primer for the padlock probe. Then, the primer could trigger the RCA reaction to produce a long-chain singles-tranded DNA harboring repeating Thioflavin T (ThT) aptamer, leading to powerful fluorescence signals. The assay demonstrates sensitivity, specificity, and accuracy, showing its potential as an alternative tool for pathogen detection in different fields, such as food safety monitoring.

Abstract:Objective To establish a clinical method for early detection of the bloodstream infection bacteria based on matrix-assisted laser ionization time of flight mass spectrometry (MALDI-TOF MS).Methods After consulting the Laboratory Information System statistics and domestic related literature,We chose 10 kinds of bacteria (Escherichia coli.,Pseudomonas aeruginosa,Acinetobacter baumannii,Klebsiella pneumoniae,Enterococcus faecium,Enterococcus faecalis,Staphylococcus aureus and Staphylococcus epidermidis,Proteus mirabilis,Streptococcus pneumoniae) as target.The MALDI-TOF MS was established using simulated bacterial infection blood samples.From March to May 2017,33 blood samples of suspected sepsis patients from Emergency Department,General Hospital of PLA,were tested.Results The MALDI-TOF MS whose detecting sensitivity was 100CFU/ml had the same negative detection rate with the blood culture (27cases/27cases,100％).Besides the 2 samples of Morganella morganii infection and Staphylococcus hominis infection were out of the range,the results of the remaining 4 positive samples were consistent (100％).Conclusion Compared to the blood culture and biochemical identification,MALDI-TOF MS can rapidly detect 10 kinds of bloodstream infection bacteria with high sensitivity and high accuracy.

Abstract:New and rapid antimicrobial susceptibility/resistance testing methods are required for bacteria from positive blood cultures. In this study, a multiplex-targeted liquid chromatography-tandem mass spectrometry (LC-MS/MS) assay was developed and validated for the detection of β-lactam, aminoglycoside, and fluoroquinolone resistance mechanisms in blood cultures growing Escherichia coli or Klebsiella pneumoniae complex. Selected targets were the β-lactamases SHV, TEM, OXA-1-like, CTX-M-1-like, CMY-2-like, chromosomal E. coli AmpC (cAmpC), OXA-48-like, NDM, VIM, and KPC; the aminoglycoside-modifying enzymes AAC(3)-Ia, AAC(3)-II, AAC(3)-IV, AAC(3)-VI, AAC(6′)-Ib, ANT(2′′)-I, and APH(3′)-VI; the 16S-RMTases ArmA, RmtB, RmtC, and RmtF; the quinolone resistance mechanisms QnrA, QnrB, AAC(6′)-Ib-cr; the wildtype quinolone resistance determining region of GyrA; and the E. coli porins OmpC and OmpF. The developed assay was evaluated using 100 prospectively collected positive blood cultures, and 148 negative blood culture samples spiked with isolates previously collected from blood cultures or isolates carrying less prevalent resistance mechanisms. The time to result was approximately 3 h. LC-MS/MS results were compared with whole-genome sequencing and antimicrobial susceptibility testing results. Overall, there was a high agreement between LC-MS/MS results and whole-genome sequencing results. In addition, the majority of susceptible and non-susceptible phenotypes were correctly predicted based on LC-MS/MS results. Exceptions were the predictions for ciprofloxacin and amoxicillin/clavulanic acid that matched with the phenotype in 85.9 and 63.7% of the isolates, respectively. Targeted LC-MS/MS based on parallel reaction monitoring can be applied for the rapid and accurate detection of various resistance mechanisms in blood cultures growing E. coli or K. pneumoniae complex.

Direct Identification of Staphylococcus Aureus in Blood Cultures by Coagulase Test

Abstract:Objective By utilizing the plasma-coagulase test to quickly identify the Staphylococcus aureus in positive blood cultures.Methods When the blood culture alarms,gather the positive blood cultures to take the Garm's stain immediately,and take the plasma coagulase test which smearing result is Staphylococcus,and in conventional cultivation and isolation,using the VITEK 2 Compact instruments to identify these pure cultures.Results From March to July 2010,there have been 100 positive blood cultures which smearing results were Staphylococcus,to take the plasma coagulase test,and then,the total 100 cultures,including 23 coagulase positive cultures,identified by VITEK 2 Compact instruments.All of the 23 coagulase positive cultures were Staphylococcus aureus,and of the 77 coagulase negative cultures,were coagulase negative staphylococci.Plasma coagulase test will be quickly completed in one hour.Conclusion Directly using the blood cultures to do the plasma-coagulase test can be used for the identification of the Staphylococcus aureus
