Characterization of A Model of Ovariectomy-Induced Knee Osteoarthritis in Guinea Pigs

Abstract:Purpose: The aim of present study was to characterize the phenotypes of a model of primary osteoarthritis with syndrome of kidney deficiency after post-menopause induced by bilateral ovariectomy in guinea pigs. Methods: Fifty-four of guinea pigs were randomly divided into control, ovariectomized (OVX), and sham operation (sham) groups, at the ages of 3-month-old and 5-month-old, respectively. Surgical removal of bilateral ovaries was performed to induce post-menopause. The kidney deficiency was determined by behavior observation, body weight alterations, and a series of biochemical parameters, including serum levels of cyclic adenosine monophosphate (cAMP), cyclic guanosine monophosphate (cGMP), and the ratio changes of cAMP and cGMP. Then continuous observation for changes of vaginal exfoliated cells and detection of serum concentrations of estradiol (E2) in estrous cycle was performed to validate postmenopausal status. The lesions of knee were characterized by the gross appearances of stifle knee-joint and the microscopic phenotypes of knee osteoarthritis. Results: The normal estrous cycle disappeared in both 3- and 5-moth old OVX groups(graphic1).The animals in OVX groups exhibited apparent signs and symptoms of kidney deficiency, including listlessness, gray hairs, shaking chills with hot flashes, loss of appetite, loose stool, and slower increase of body weight. Serum levels of E2, cAMP and ratio of cAMP/cGMP were significantly decreased, and cGMP level significantly increased, suggesting a status of yang deficiency of kidney in animals in OVX(graphic4–graphic5). Quantitative evaluation for synovitis by macroscopic scores, modified Mankin's scores, and ten-point semiquantitative scores showed significantly increased , suggesting existence of knee osteoarthritis, compared to controls (P<0.05). On average, OVX animals at 5-month old showed higher synovitis scores than OVX animals at 3-month old(graphic2–graphic3). Conclusions: The bilateral removal of the ovaries in female Hartley guinea pigs can be used to induce primary knee osteoarthritis with syndrome of kidney deficiency after post-menopause.
