Abstract:The clathration of dextromethorphan hydrobromide(DH)withβ-cyclodextrin and its derivatives was studied with fluorescance spectrometry.Through fixing the concentration of dextromethorphan hydrobromide and changing the concentration ofβ-cyclodextrin and its derivatives,the formation of inclusion compounds were verified by the change of the emission wavelength and enhancement of fluorescence intensity.The binding constant was calculated according to Lineweaver-Burk plot.The experimental results showed that three kinds of cyclodextrins had obvious effect on dextromethorphan hydrobromide under pH =7.4,forming inclusion compounds with a inclusion ratio of 1∶1.

Abstract:The variation of the I-1/I-3 ratio of the fluorescence of pyrene in polystyrene-based quaternary ammonium salt (PA) and sodium carboxymethylamylose (NaCMA) aqueous solutions was measured as a function of sodium perfluorooctanoate (SPFO) concentration. Association of the anionic fluorocarbon surfactant with cationic polyelectrolyte (PA) to form mixed micelles occurred at the surfactant concentrations far below its normal critical concentration, while no interaction between SPFO and anionic polyelectrolyte (NaCMA) was detected. The driving force for the formation of mixed micelles of SPFO and PA is electrostatic interactions.

Abstract:The interaction of New herbicide ZJ0777[C19H18BrN3O3 ] and Bovine Serum Albumin has been studied by fluorescence quenching method and synchronous fluorescence.The result has showed that the BSA has reacted with ZJ0777 and formed a certain new compound which belongs to static fluorescence quenching.According to Lineweaver-Burk equation,KLB(287 K:1.955×103 L·mol-1;298 K:2.441×103 L·mol-1;308 K:3.041×103 L·mol-1),the forming constants of the compound at different temperatures and the thermodynamic parameters(△H=15.566 kJ·mol-1;△S=117.24,117.08,117.22 J·K-1;△G=-18.082,-19.325,-20.537 kJ·mol-1) at correspondence temperatures are obtained.The latter shows that binding power between them is mainly the hydrophobic interaction and the number of binding site is 1.The effect of ZJ0777 on the conformation of BSA is analyzed by syschronous fluorescence.

Abstract:The polyunsaturated fatty acid docosahexaenoic acid (DHA, c22:6, n-3) is found at a level of about 50% in the phospholipids of neuronal tissue membranes and appears to be crucial to human health. Dipalmitoyl phosphatidylcholine (DPPC, 16:0/16:0 PC), 1-palmitoyl-2-oleoyl phosphatidylserine (POPS) and the DHA containing 1-stearaoyl-2-docosahexenoyl phosphatidylserine (SDPS) were used to make DPPC (60%)/POPS (29%)/SDPS (11%) bilayers with and without 10 mol% chlorpromazine (CPZ), a cationic, amphiphilic phenothiazine. The T1 relaxation measurements make it clear that the saturated acyl chains carbons (palmitic, stearic and most of the oleic chain) and the choline head group are not affected by CPZ addition. The observed increased signal intensity and T1-values of DHA indicate reduced mobility of C4 and C5 due to CPZ binding. 31P NMR spectra confirm that CPZ binding to the phosphatidylserines in the bilayer enhances phospholipid head group mobility.
Interactions between Rhodamine Dyes and Model Membrane Systems—Insights from Molecular Dynamics Simulations

Abstract:Background: rhodamines are dyes widely used as fluorescent tags in cell imaging, probing of mitochondrial membrane potential, and as P-glycoprotein model substrates. In all these applications, detailed understanding of the interaction between rhodamines and biomembranes is fundamental. Methods: we combined atomistic molecular dynamics (MD) simulations and fluorescence spectroscopy to characterize the interaction between rhodamines 123 and B (Rh123 and RhB, respectively) and POPC bilayers. Results: while the xanthene moiety orients roughly parallel to the membrane plane in unrestrained MD simulations, variations on the relative position of the benzoic ring (below the xanthene for Rh123, above it for RhB) were observed, and related to the structure of the two dyes and their interactions with water and lipids. Subtle distinctions were found among different ionization forms of the probes. Experimentally, RhB displayed a lipid/water partition coefficient more than two orders of magnitude higher than Rh123, in agreement with free energy profiles obtained from umbrella sampling MD. Conclusions: this work provided detailed insights on the similarities and differences in the behavior of bilayer-inserted Rh123 and RhB, related to the structure of the probes. The much higher affinity of RhB for the membranes increases the local concentration and explains its higher apparent affinity for P-glycoprotein reconstituted in model membranes.

Abstract:Quinacrine is a versatile drug that is widely recognized for its antimalarial action through its inhibition of the phospholipase enzyme. It also has antianthelmintic and antiprotozoan activities and is a strong DNA binder that may be used to combat multidrug resistance in cancer. Despite extensive cell-based studies, a detailed understanding of quinacrine's influence on the cell membrane, including permeability, binding, and rearrangement at the molecular level, is lacking. Herein, we apply microcavity-suspended lipid bilayers (MSLBs) as in vitro models of the cell membrane comprising DOPC, DOPC:Chol(3:1), and DOPC:SM:Chol(2:2:1) to investigate the influence of cholesterol and intrinsic phase heterogeneity induced by mixed-lipid composition on the membrane interactions of quinacrine. Using electrochemical impedance spectroscopy (EIS) and surface-enhanced Raman spectroscopy (SERS) as label-free surface-sensitive techniques, we have studied quinacrine interaction and permeability across the different MSLBs. Our EIS data reveal that the drug is permeable through ternary DOPC:SM:Chol and DOPC-only bilayer compositions. In contrast, the binary cholesterol/DOPC membrane arrested permeation, yet the drug binds or intercalates at this membrane as reflected by an increase in membrane impedance. SERS supported the EIS data, which was utilized to gain structural insights into the drug-membrane interaction. Our SERS data also provides a simple but powerful label-free assessment of drug permeation because a significant SERS enhancement of the drug's Raman signature was observed only if the drug accessed the plasmonic interior of the pore cavity passing through the membrane. Fluorescent lifetime correlation spectroscopy (FLCS) provides further biophysical insight, revealing that quinacrine binding increases the lipid diffusivity of DOPC and the ternary membrane while remarkably decreasing the lipid diffusivity of the DOPC:Chol membrane. Overall, because of its adaptability to multimodal approaches, the MSLB platform provides rich and detailed insights into drug-membrane interactions, making it a powerful tool for in vitro drug screening.
Terpyridine-substituted, fluorescent polymers and their chelation with zinc ion: The ligand-to-metal ratio and optical properties

Abstract:Soluble, fluorescent, terpyridine-substituted, conjugated polymers were prepared and characterized. The polymer chains included a defined oligo(phenylenevinylene) fragment, on which the terpyridine-functional group was attached. The polymers were blue-fluorescent with emission peaks at 400–427 nm in tetrahydrofuran solutions. Upon chelation with the Zn(II) cation, the emission maxima were shifted to a longer wavelength by as much as 113 to 506–526 nm. A model compound was also prepared to aid the structural characterization. The ratio of terpyridine to Zn2+ in the polymer complex was found to be 1:1 on the basis of spectroscopic evidence, which included mass spectrometry, 1H NMR, and Job titration. © 2006 Wiley Periodicals, Inc. Abstract:Elucidating the interaction mechanism between nanomaterials and cell membranes is critical for cytotoxicity mechanisms and the design of safer biomedicines. Recently, graphene oxide quantum dots (GOQDs) were shown to induce disruption in the cell membrane. Little is known, however, about why this occurs and what microscopic interactions are important. Here, we investigate the continuous-time interaction of model cell membrane with GOQDs via atomic force microscopy/surface plasmon resonance. The binding of particles to the model membranes is a concentration-dependent and reversible process, which is significantly hampered by cholesterol, ganglioside GM1 and protein corona. In addition, the molecular mechanism of GOQD-cell membrane interaction is analyzed using molecular dynamics simulation: GOQDs attach rapidly to the surface of the cell membrane and then induce the deformation of lipid bilayers, subsequently physically extracting lipid molecules, particularly cholesterol and ganglioside GM1. Hydrogen bonding plays a dominant role in the interaction of GOQDs and the membrane, which is mainly derived from the hydroxyl, carbonyl and phosphate groups of membranes. These findings might have implications in studies of the cytotoxicity of nanomaterials and in novel biomedicine design.

However, studies of complex systems often require an additional introduction of a specific quencher molecule acting in combination with a fluorophore to provide structural or thermodynamic information. In this review, we outline the fundamentals and summarize the latest progress in applications of different classes of fluorescent probes and their specific quenchers, aimed at studies of protein folding and protein‐membrane interactions Fluorescence spectroscopy provides numerous methodological tools for structural and functional studies of biological macromolecules and their complexes. All fluorescence‐based approaches require either existence of an intrinsic probe or an introduction of an extrinsic one. Moreover, studies of complex systems often require an additional introduction of a specific quencher molecule acting in combination with a fluorophore to provide structural or thermodynamic information. Here, we review the fundamentals and summarize the latest progress in applications of different classes of fluorescent probes and their specific quenchers, aimed at studies of protein folding and protein‐membrane interactions. Specifically, we discuss various environment‐sensitive dyes, FRET probes, probes for short‐distance measurements, and several probe‐quencher pairs for studies of membrane penetration of proteins and peptides. The goals of this review are: (a) to familiarize the readership with the general concept that complex biological systems often require both a probe and a quencher to decipher mechanistic details of functioning and (b) to provide example of the immediate applications of the described methods.

Abstract:The interaction between chloramphenicol (CHL) and neuroglobin (Ngb) has been investigated by using fluorescence, synchronous fluorescence, UV-Vis and circular dichroism (CD) spectroscopy. It has been found that CHL molecule can quench the intrinsic fluorescence of Ngb in a way of dynamic quenching mechanism, which was supported by UV-Vis spectral data. Their effective quenching constants (KSV) are2.2×104,2.6×104,and 3.1×104 L⋅mol−1at 298 K, 303 K, and 308 K, respectively. The enthalpy change (ΔH) and entropy change (ΔS) for this reaction are 26.42 kJ⋅mol−1and 171.7 J⋅K−1, respectively. It means that the hydrophobic interaction is the main intermolecular force of the interaction between CHL and Ngb. Synchronous fluorescence spectra showed that the microenvironment of tryptophan and tyrosine residues of Ngb has been changed slightly. The fluorescence quenching efficiency of CHL to tyrosine residues is a little bit more than that to tryptophan residues of Ngb. Furthermore, CD spectra indicated that CHL can induce the formation of α-helix of Ngb.
Subcellular distribution of fluoranthene in Chlorella vulgaris with the presence of cetyltrimethylammonium chloride

Abstract:This study explored the possible mechanism of the joint toxicity of binary mixtures of cetyltrimethylammonium chloride (CTAC) and fluoranthene (Flu) to the green alga Chlorella vulgaris by examining the subcellular distribution of Flu within the alga. The joint action of CTAC (100 μg L(-1)) and Flu (0-200 μg L(-1)) on the algae changed from a synergetic effect (0-50 μg L(-1)) to an antagonistic effect (50-200 μg L(-1)) with an increase of the Flu concentration. The Flu uptake was enhanced by the presence of CTAC through the intracellular detection of Flu. Furthermore, the highest amount of Flu bound to the cytosol, whereas the least amount bound to the cellular debris when synergistic effect was observed at 2.5 μg L(-1) Flu. However, the highest amount of Flu bound to the cellular debris, whereas the least amount bound to the organelles when antagonistic effect was displayed at 200 μg L(-1) Flu. The different subcellular distribution of Flu may affect the uptake of the highly toxic CTAC by the algae in the binary mixture, and consequently lead to a different level of CTAC toxicity. The abovementioned results indicate that the subcellular distribution of chemicals can be used to elucidate possible mechanisms for the joint toxicity of their binary mixtures to aquatic organisms.

Abstract:Cell membrane is a thin semi-permeable membrane that surrounds the cytoplasm and protects the homeostasis of internal components of a cell, which is also involved in a variety of molecular biological processes such as cell adhesion, ion conductivity and cell signaling. Model cell membrane may provide an efficient alternative for studying these processes. Hence, studying the interactions between medical nanoparticles and model cell membrane may play an important role in evaluating the biosafety of nanoparticles and promoting the biomedical applications of nanoparticles. In this paper, we reviewed the recent progresses of simulation studies of the interactions between medical nanoparticles and model cell membrane. And on this basis, we proposed the future research directions considering the focuses of modern membrane biophysics.

Abstract:Fluorescent cell membrane probes (1a-d) were designed from bioactive oxazolo [4,5-b]pyridine unit. The probes were connected by oxazolo [4,5-b]pyridine and benzofuran (1a), triphenyl (1b), coumarin (1c), diethylaminobenzene (1d), respectively, with a double bond. The optical properties of 1a-d with different solvents were tested, they had obvious solvatochromism in different polar solvents, Large stokes shifts (63–204 nm) and significant fluorescent enhancement in large multilamellar vesicles (MLV). After 6 h of continuous illumination, the remaining absorption of the probes was 81–97%. The cytotoxicity test indicated that the survival rate of HeLa and L929 cells was 94–119% after co-cultured with probes 1a-d for 6 h. Laser confocal experiments further illustrated that the probes could quickly (<1 min) combine with the cell membrane. Among them, probe 1a worked in the green channel, while probes 1b, 1c and 1d employed in the red channel. Moreover, the retention time of probes 1a and 1c on cell membrane was found to 2–3 h.

Abstract:In the present study, 3-(fluorobenzylideneamino)-6-chloro-1-(3,3-dimethylbutanoyl)-phenyl-2,3-dihydroquinazolin-4(1H)-one (FDQL) derivatives have been designed and synthesized to study the interaction between fluorine substituted dihydroquinazoline derivatives with human serum albumin (HSA) using fluorescence, circular dichroism and Fourier transform infrared spectroscopy. The results indicated that the FDQL could bind to HSA, induce conformation and the secondary structure changes of HSA, and quench the intrinsic fluorescence of HSA through a static quenching mechanism. The thermodynamic parameters, ΔH, ΔS, and ΔG, calculated at different temperatures, revealed that the binding was through spontaneous and hydrophobic forces and thus played major roles in the association. Based on the number of binding sites, it was considered that one molecule of FDQL could bind to a single site of HSA. Site marker competition experiments indicated that the reactive site of HSA to FDQL mainly located in site II (subdomain IIIA). The substitution by fluorine in the benzene ring could increase the interactions between FDQL and HSA to some extent in the proper temperature range through hydrophobic effect, and the substitution at meta-position enhanced the affinity greater than that at para- and ortho-positions.

Abstract:Although there are some proposed explanations for fluorescence enhancement of metal ions, a phenomenon with wide applications in biosensors and chemosensors, current understanding of the quantum–mechanical origin of this photophysical behaviour is limited. To address this issue, we selected thymolphthalein-based probe (T) reported recently as model system, and investigated the photochemical and photophysical properties of T and T-Zn(II) complex. The geometry structures of T and its Zn(II) complex were optimized, and the experimental absorption spectrum were well reproduced through density functional theory (DFT) methods. Furthermore, the excited state intramolecular proton transfer (ESIPT) mechanisms of T were systematically investigated. The constructed potential energy surfaces (PESs) of T at ground (S0) and first exited (S1) states indicate that, after photo-excitation, the intramolecular single-proton transfer reaction occurs more readily with the H atom removing from O to neighboring N. Furthermore, thermodynamic analysis of the T-Zn(II) complex shows that mononuclear complex (C1) is the most thermodynamically stable structure, and the intramolecular single proton transfer process of C1 is thermodynamically unfavorable at the S1 state. Time-dependent DFT calculations show that the fluorescence emission is from the S1 states of the single-proton transfer tautomer of T, as predicted from Kasha's rule, in comparison, the fluorescence emission of Zn(II) complex is from the S1 states of T-Zn(II) rather than its single-proton transfer tautomer. The inhibitor of Zn(II) on the single proton transfer reaction plays an important role on fluorescence enhancement.

Microscopic Interactions of Melatonin, Serotonin and Tryptophan with Zwitterionic Phospholipid Membranes

Abstract:The interactions at the atomic level between small molecules and the main components of cellular plasma membranes are crucial for elucidating the mechanisms allowing for the entrance of such small species inside the cell. We have performed molecular dynamics and metadynamics simulations of tryptophan, serotonin, and melatonin at the interface of zwitterionic phospholipid bilayers. In this work, we will review recent computer simulation developments and report microscopic properties, such as the area per lipid and thickness of the membranes, atomic radial distribution functions, angular orientations, and free energy landscapes of small molecule binding to the membrane. Cholesterol affects the behaviour of the small molecules, which are mainly buried in the interfacial regions. We have observed a competition between the binding of small molecules to phospholipids and cholesterol through lipidic hydrogen-bonds. Free energy barriers that are associated to translational and orientational changes of melatonin have been found to be between 10-20 kJ/mol for distances of 1 nm between melatonin and the center of the membrane. Corresponding barriers for tryptophan and serotonin that are obtained from reversible work methods are of the order of 10 kJ/mol and reveal strong hydrogen bonding between such species and specific phospholipid sites. The diffusion of tryptophan and melatonin is of the order of 10-7 cm2/s for the cholesterol-free and cholesterol-rich setups.

Abstract:The polyunsaturated fatty acid docosahexaenoic acid (DHA, 22:6, n-3) is found at a level of about 50% in the phospholipids of neuronal tissue membranes and appears to be crucial to human health. Dipalmitoyl phosphatidylcholine (DPPC, 16:0/16:0 PC) and the DHA containing 1-stearoyl-2-docosahexenoyl phosphatidylserine (SDPS) were used to make DPPC (60%)/SDPS (40%) bilayers with and without 10 mol% chlorpromazine (CPZ), a cationic, amphiphilic phenothiazine.

Abstract:The donor-acceptor design is a classic method of synthesizing new fluorescent molecules. In this study, a series of new fluorescent compounds (1-10) were synthesized based on 2-(3,5-bis(trifluoromethyl)phenyl)-quinazoline acceptor and various amino donors. The fluorescent emissions of1-10cover the spectrum from 414 nm to 597 nm in cyclohexane solutions with various amino donors on 4- or 7-positions of quinazoline. Ultimately, compounds1and2presented the highest photoluminescence quantum yield (QY) over 80%, while compound10provided the largest Stokes shift (161 nm) in cyclohexane. Most of them have strong emissions in aggregated states such as in nanoparticles, in powders, in crystals and in films. Mechanochromic properties were observed for compounds1,2,4and7. Furthermore, blue OLEDs were fabricated by using compound2or7as the active layer.
