Abstract:Health and environmental Impacts of graphene-based materials need to be thoroughly evaluated before their potential applications. Graphene has strong cytotoxicity toward bacteria. To better understand its antimicrobial mechanism, we compared the antibacterial activity of four types of graphene-based materials (graphite (Gt), graphite oxide (GtO), graphene oxide (GO), and reduced graphene oxide (rGO)) toward a bacterial model Escherichia coli. Under similar concentration and incubation conditions, GO dispersion shows the highest antibacterial activity, sequentially followed by rGO, Gt, and GtO. Scanning electron microscope (SEM) and dynamic light scattering analyses show that GO aggregates have the smallest average size among the four types of materials. SEM images display that the direct contacts with graphene nanosheets disrupt cell membrane. No superoxide anion (O-2(center dot-)) induced reactive oxygen species (ROS) production is detected. However, the four types of materials can oxidize glutathione, which serves as redox state mediator in bacteria. Conductive rGO and Gt have higher oxidation capacities than insulating GO and GtO. Results suggest that antimicrobial actions are contributed by both membrane and oxidation stress. We propose that a three-step antimicrobial mechanism, previously used for carbon nanotubes, is applicable to graphene-based materials. It includes initial cell deposition on graphene-based materials, membrane stress cared by direct contact with sharp nanosheets, and the ensuing superoxide anion-independent oxidation. We envision that physicochemical properties of graphene-based materials, such as density of functional groups, size, and conductivity, can be precisely tailored to either reducing their health and environmental risks or increasing their application potentials.

Abstract:Graphene is a monolayer of tightly packed carbon atoms that possesses many interesting properties and has numerous exciting applications. In this work, we report the antibacterial activity of two water-dispersible graphene derivatives, graphene oxide (GO) and reduced graphene oxide (rGO) nanosheets. Such graphene-based nanomaterials can effectively inhibit the growth of E. coli bacteria while showing minimal cytotoxicity. We have also demonstrated that macroscopic freestanding GO and rGO paper can be conveniently fabricated from their suspension via simple vacuum filtration. Given the superior antibacterial effect of GO and the fact that GO can be mass-produced and easily processed to make freestanding and flexible paper with low cost, we expect this new carbon nanomaterial may find important environmental and clinical applications.
Graphene oxide affects bacteriophage infection of bacteria by promoting the formation of biofilms

Abstract:Graphene oxide (GO) is increasingly used in a range of fields, such as electronics, biosensors, drug delivery, and water treatment, and the likelihood of its release into the environment is increasing correspondingly. GO is involved in the formation of biofilms and leads bacteria to over proliferate, but the effects of GO on bacteriophage infection remain unexplored. We noted bacterial overgrowth in experiments when GO was used to treat the bacterial culture medium, leading us to question whether bacterial proliferation caused by GO affects phage infection of target bacteria. Treating Pseudomonas aeruginosa with GO at a low dosage (0.02 mg/mL) led to biofilm expansion in LB medium. Biofilm formation in the presence of GO affected the ability of bacteriophages to kill bacteria and reproduce. Similarly, the presence of GO deposits increased the ratio of bacteria to phage, providing a favorable environment for bacterial growth. Additionally, increasing the positive electrical charge in the culture environment inhibited the rejection of bacteriophages by negatively charged GO, improving phage reproduction. Finally, adding GO to sewage in imitation field experiments significantly increased the bacterial diversity and richness in the sewage, stimulating a significant increase in the variety and number of bacteria. Collectively, these results indicate that GO hinders phage infection by providing a bacterial refuge. The results of this study provide valuable insights into how GO interacts with bacteriophages to explore the effects on bacterial growth.

Abstract:The antibiotics-independent antimicrobial activity of graphene oxide (GO) is of great importance since antibiotic therapy is facing great challenges from drug resistance. However, the relations of GO size with its antimicrobial activity and how the size regulates the antibacterial mechanisms are still unknown. Herein, we fabricated four GO suspensions with different sizes and demonstrated the parabolic relationship between GO size and its antibacterial activity against the Gram-positive cariogenic bacterium Streptococcus mutans. More interestingly, we found out how GO size regulated the nano-bio interaction-based physical antibacterial mechanisms. Increasing the size reduced the cutting effect but enhanced the cell entrapment effect, and vice versa. In conclusion, GO size affects its edge density and lateral dimension, further regulates its physical antibacterial mechanisms in different orientations and ultimately determines its activity. These findings provide a deep understanding of GO antibacterial property and may guide the design and development of GO for clinical use.
Interaction with teichoic acids contributes to highly effective antibacterial activity of graphene oxide on Gram-positive bacteria

Abstract:Graphene oxide (GO) has high-efficient antibacterial activity to diverse pathogenic bacteria. However, the detailed antibacterial mechanism of GO is not fully clear. Herein the antibacterial properties of GO against model Gram-positive (Gram+) (Staphylococcus aureus and Staphylococcus epidermidis) and Gram-negative (Gram−) bacteria (Pseudomonas aeruginosa and Escherichia coli) were compared by plate count method. Results showed that 4 mg/L of GO induced the mortality of Gram+ and Gram− bacteria by > 99% and < 25%, respectively. GO had greater adsorption affinity to teichoic acids, the unique components existing in the cell wall of Gram+ bacteria, mainly via π−π interaction. The adsorption efficiency of teichoic acids was 27 times higher than that of peptidoglycan when they were simultaneously exposed to 100 mg/L GO. The superior adsorption of teichoic acids onto GO increased one order of magnitude of atlA expression, the autolysin related gene. As a result, these accelerated bacterial death by hydrolyzing peptidoglycan in cell walls. Exogenous addition of 50 mg/L teichoic acids could impair 4–5 fold of antibacterial activity of GO against S. aureus. These new findings illuminate the antibacterial mechanism of GO against Gram+ bacteria, which paves the way for the further application of graphene-based materials in water disinfection and pathogen control.

Abstract:Duo to their superior physicochemical properties, graphene and its derivatives (GDs), such as graphene oxide (GO) and reduced graphene oxide (rGO), have attracted extensive research interests around the world. In recent years, antibacterial activities of GDs have aroused wide concern and substantial works have been done. However, the underlying antibacterial mechanisms still remain controversial. Antibacterial activities of GDs vary with various factors, such as size, number of layers, oxygen-containing groups, and experimental surroundings. We assume that combination types between graphene oxide and substrate may affect the antibacterial activity. Therefore, in this work, GO was fixed on the titanium surface with three kinds of combination types including drop with gravitational effects (GO-D), electrostatic interaction (GO-APS) and electrophoretic deposition (GO-EPD), and the antibacterial activities in vitro were systematically investigated. Results showed that combination types affected the ability of GO for preventing Staphylococcus aureus (S. aureus) from gathering, sharpness of wrinkles or edges and reactive oxygen spices (ROS) levels. Once S. aureus are in the form of separation without aggregation, GO can effectively interact with them and kill them with sharp wrinkles or edges and high ROS levels. GO-EPD could effectively prevent S. aureus from gathering, own sharp wrinkles or edges and could generate higher ROS levels. As a result, GO-EPD exhibited optimal antibacterial activity against S. aureus, followed by GO-APS and GO-D.

Abstract:Graphene oxide (GO) is a promising precursor to produce graphene-family nanomaterials for various applications. Their potential health and environmental impacts need a good understanding of their cellular interactions. Many factors may influence their biological interactions with cells, and the lateral dimension of GO sheets is one of the most relevant material properties. In this study, a model bacterium, Escherichia coli ( E. coli ), was used to evaluate the antibacterial activity of well-dispersed GO sheets, whose lateral size differs by more than 100 times. Our results show that the antibacterial activity of GO sheets toward E. coli cells is lateral size dependent. Larger GO sheets show stronger antibacterial activity than do smaller ones, and they have different time- and concentration-dependent antibacterial activities. Large GO sheets lead to most cell loss after 1 h incubation, and their concentration strongly influences antibacterial activity at relative low concentration (<10 μg/mL). In contrast, when incubating with small GO sheets up to 4 h, the inactivation rate of E. coli cells continues increasing. The increase of small GO sheet concentration also results in persistent increases in their antibacterial activity. In this study, GO sheets with different lateral sizes are all well dispersed, and their oxidation capacity toward glutathione is similar, consistent with X-ray photoelectron spectroscopy and ultraviolet-visible absorption spectroscopy results. This suggests the lateral size-dependent antibacterial activity of GO sheets is caused by neither their aggregation states, nor oxidation capacity. Atomic force microscope analysis of GO sheets and cells shows that GO sheets interact strongly with cells. Large GO sheets more easily cover cells, and cells cannot proliferate once fully covered, resulting in the cell viability loss observed in the followed colony counting test. In contrast, small GO sheets adhere to the bacterial surfaces, which cannot effectively isolate cells from environment. This study highlights the importance of tailoring the lateral dimension of GO sheets to optimize the application potential with minimal risks for environmental health and safety.

Abstract:Amino-functionalized graphene oxide (GO-NH2) was prepared for enhanced antibacterial properties through excellent photothermal efficiency. This nanosheet could easily target the bacterial surface through electrostatic attraction. Upon white light irradiation, GO-NH2 with superior antibacterial efficacy could inhibit the growth of Gram-negative and Gram-positive bacteria, resulting in an increase to 32-times more than the antibacterial activity of GO alone. Through studies of the antibacterial mechanism, it was found that the nanosheets could destroy the cytomembrane of bacteria, resulting in cytoplasmic leakage. Moreover, the analysis of cell proliferation indicated that the nanosheets had good biocompatibility. This work indicates that graphene oxide could be used as a new nanostructured carbon material to construct an antimicrobial agent for photothermal therapy in the biomedical field.

Abstract:Antibiotic resistance in microbial pathogens has become a serious health problem in the world. The increasing spread of hospital acquired infections especially in immunocompromised and cancer patients caused by multidrug-resistant (MDR) microbial pathogens is restricting the choices for impressive antibiotic therapy. So many efforts have been made to develop new compounds with antimicrobial activity. In recent years, nanoparticles, particularly graphene oxide (GO) nanoparticles have found many applications in various fields, including antibacterial action, pathogens bio detection, cancer therapy, and drug and gene delivery. The use of graphene oxide as an antibacterial agent for the treatment of infections with multidrug resistance is growing due to the unique physicochemical properties as wide surface area, excellent electrical and thermal conductivity, and biocompatibility. To reduce toxicity and increase the efficiency of graphene oxide as an antimicrobial agent, different surface modification and functionalization with inorganic nanostructures, biomolecules and polymers were developed. In this review article, we give our overview of the progress made on the graphene oxide nanocomposites as a new generation of antimicrobial agents.

Abstract:The development of new antibacterial agents that are highly effective are of great interest. Herein, we present a recyclable and synergistic nanocomposite by growing both iron oxide nanoparticles (IONPs) and silver nanoparticles (AgNPs) on the surface of graphene oxide (GO), obtaining GO-IONP-Ag nanocomposite as a novel multifunctional antibacterial material. Compared with AgNPs, which have been widely used as antibacterial agents, our GO-IONP-Ag shows much higher antibacterial efficiency toward both Gram-negative bacteria Escherichia coli (E. coli) and Gram-positive bacteria Staphylococcus aureus (S. aureus). Taking the advantage of its strong near-infrared (NIR) absorbance, photothermal treatment is also conducted with GO-IONP-Ag, achieving a remarkable synergistic antibacterial effect to inhibit S. aureus at a rather low concentration of this agent. Moreover, with magnetic IONPs existing in the composite, we can easily recycle GO-IONP-Ag by magnetic separation, allowing its repeated use. Given the above advantages as well as its easy preparation and cheap cost, GO-IONP-Ag developed in this work may find potential applications as a useful antibacterial agent in the areas of healthcare and environmental engineering.
Effect of graphene oxide nanosheets on visible light-assisted antibacterial activity of vertically-aligned copper oxide nanowire arrays

Abstract:In the present work, the effect of graphene oxide (GO) nanosheets on the antibacterial activity of CuO nanowire arrays under visible light irradiation is shown. A combined thermal oxidation/electrophoretic deposition technique was employed to prepare three-dimensional networks of graphene oxide nanosheets hybridized with vertically aligned CuO nanowires. With the help of standard antibacterial assays and X-ray photoelectron spectroscopy, it is shown that the light-activated antibacterial response of the hybrid material against gram-negative Escherichia coli is significantly improved as the oxide functional groups of the GO nanosheets are reduced. In order to explore the physicochemical mechanism behind this behavior, ab-initio simulations based on density functional theory were performed and the effect of surface functional groups and hybridization were elucidated. Supported by the experiments, a three-step photo-antibacterial based mechanism is suggested: (i) injection of an electron from CuO into rGO, (ii) localization of the excess electron on rGO functional groups, and (iii) release of reactive oxygen species lethal to bacteria. Activation of new photoactive and physical mechanisms in the hybrid system makes rGO-modified CuO nanowire coatings as promising nanostructure devices for antimicrobial applications in particular for dry environments.
Killing Dental Pathogens Using Antibacterial Graphene Oxide

Abstract:Dental caries and periodontal diseases have a close relationship with microbes such as Streptococcus mutans, Porphyromonas gingivalis and Fusobacterium nucleatum. Graphene oxide (GO), as the derivative of graphene, plays an important role in many areas including biology and medicine. In particular, it has been known as a promising antimicrobial nanomaterial. In this study, we focused on the antimicrobial property of GO against dental pathogens. With the utilization of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) reduced test, colony forming units (CFU) counting, growth curve observation, live/dead fluorescent staining, and confocal laser scanning microscopy (CLSM), we found GO nanosheets were highly effective in inhibiting the growth of dental pathogens. Transmission electron microscopy (TEM) images revealed that the cell wall and membrane of bacteria lost their integrity and the intracellular contents leaked out after they were treated by GO. Therefore, GO nanosheets would be an effective antibacterial material against dental pathogens and the potential applications in dental care and therapy are promising.

Abstract:Recently, graphene oxide (GO) based materials have shown great potential in the treatment of implant bacterial infections due to its inherent antibacterial activity. However, the effect of GO-based materials on biological systems particularly the antibacterial mechanisms is still not clear. In this study, GO, NaBH4 treated GO (GO-Y), copper decorated GO (GO-Cu, GO-Cu-GO) composited coatings were prepared on the surface of silicon dioxide (SiO2) substrate by spin-coating and chemical in-situ formation. The growth of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) on GO-Y, GO-Cu, and GO-Cu-GO were significantly inhibited, especially on GO-Cu and GO-Cu-GO coatings. The implied antibacterial mechanism of GO-Cu-Cu coatings was further studied and discussed. The enhanced antibacterial performance of GO-Cu-GO coatings has significant potential application in preventing clinical implant bacterial infections. Moreover, the systematic study of various antibacterial effects also enriches our knowledge of the possible antibacterial mechanisms of graphene-based materials.

Superior Antibacterial Activity of Zinc Oxide/graphene Oxide Composites Originating from High Zinc Concentration Localized Around Bacteria.

Abstract:New materials with good antibacterial activity and less toxicity to other species attract numerous research interest. Taking advantage of zinc oxide (ZnO) and graphene oxide (GO), the ZnO/GO composites were prepared by a facile one-pot reaction to achieve superior antibacterial properties without damaging other species. In the composites, ZnO nanoparticles (NPs), with a size of about 4 nm, homogeneously anchored onto GO sheets. The typical bacterium Escherichia coli and HeLa cell were used to evaluate the antibacterial activity and cytotoxicity of the ZnO/GO composites, respectively. The synergistic effects of GO and ZnO NPs led to the superior antibacterial activity of the composites. GO helped the dispersion of ZnO NPs, slowed the dissolution of ZnO, acted as the storage site for the dissolved zinc ions, and enabled the intimate contact of E. coli with ZnO NPs and zinc ions as well. The close contact enhanced the local zinc concentration pitting on the bacterial membrane and the permeability of the bacterial membrane and thus induced bacterial death. In addition, the ZnO/GO composites were found to be much less toxic to HeLa cells, compared to the equivalent concentration of ZnO NPs in the composites. The results indicate that the ZnO/GO composites are promising disinfection materials to be used in surface coatings on various substrates to effectively inhibit bacterial growth, propagation, and survival in medical devices.
Synthesis, Characterization, and Bactericidal Evaluation of Chitosan/Guanidine Functionalized Graphene Oxide Composites

Abstract:In response to the wide spread of microbial contamination induced by bacterial pathogens, the development of novel materials with excellent antibacterial activity is of great interest. In this study, novel antibacterial chitosan (CS) and polyhexamethylene guanidine hydrochloride (PHGC) dual-polymer-functionalized graphene oxide (GO) (GO-CS-PHGC) composites were designed and easily fabricated. The as-prepared materials were characterized by Fourier transform infrared (FTIR), X-ray photoelectron spectrometer (XPS), field emission scanning electron microscopy (FE-SEM), transmission electron microscope (TEM), thermogravimetric analysis (TGA) and Raman spectroscopy. Their antibacterial capability towards bacterial strains was also studied by incubating both Gram-negative bacteria and Gram-positive bacteria in their presence. More significantly, the synergistic antibacterial action of the three components was assayed, and the findings implied that the as-prepared GO-CS-PHGC shows enhanced antibacterial activity when compared to its single components (GO, CS, PHGC or CS-PHGC) and the mixture of individual components. Not only Gram-negative bacteria but also Gram-positive bacteria are greatly inhibited by GO-CS-PHGC composites. The minimum inhibitory concentration (MIC) value of GO-CS-PHGC against E. coli was 32 μg/mL. With the powerful antibacterial activity as well as its low cost and facile preparation, GO-CS-PHGC has potential applications as a novel antibacterial agent in a wide range of biomedical uses.
Functionalized graphene oxide nanosheets with unique three-in-one properties for efficient and tunable antibacterial applications

Abstract:Developing antibiotics-independent antibacterial agents is of great importance since antibiotic therapy faces great challenges from drug resistance. Graphene oxide (GO) is a promising agent due to its natural antibacterial mechanisms, such as sharp edge-mediated cutting effect. However, the antibacterial activity of GO is limited by its negative charge and low photothermal effect. Herein, the amino-functionalized GO nanosheets (AGO) with unique three-in-one properties were synthesized. Three essential properties (positive charge, strong photothermal effect, and natural cutting effect) were integrated into AGO. The positive charge (30 mV) rendered AGO a strong interaction force with model pathogen Streptococcus mutans (330 nN). The natural cutting effect of 100 µg·mL−1 AGO caused 27% loss of bacterial viability after incubation for 30 min. Most importantly, upon the near-infrared irradiation for just 5 min, the three-in-one properties of AGO caused 98% viability loss. Abstract:Graphene materials have unique structures and outstanding thermal, optical, mechanical and electronic properties. In the last decade, these materials have attracted substantial interest in the field of nanomaterials, with applications ranging from biosensors to biomedicine. Among these applications, great advances have been made in the field of antibacterial agents. Here, recent advancements in the use of graphene and its derivatives as antibacterial agents are reviewed. Graphene is used in three forms: the pristine form; mixed with other antibacterial agents, such as Ag and chitosan; or with a base material, such as poly (N-vinylcarbazole) (PVK) and poly (lactic acid) (PLA). The main mechanisms proposed to explain the antibacterial behaviors of graphene and its derivatives are the membrane stress hypothesis, the oxidative stress hypothesis, the entrapment hypothesis, the electron transfer hypothesis and the photothermal hypothesis. This review describes contributions to improving these promising materials for antibacterial applications.
Surface activation of graphene oxide nanosheets by ultraviolet irradiation for highly efficient anti-bacterials

Abstract:A comprehensive investigation of anti-bacterial properties of graphene oxide (GO) and ultraviolet (UV) irradiated GO nanosheets was carried out. Microscopic characterization revealed that the GO nanosheet-like structures had wavy features and wrinkles or thin grooves. Fundamental surface chemical states of GO nanosheets (before and after UV irradiation) were investigated using x-ray photoelectron spectroscopy and ultraviolet photoelectron spectroscopy. Minimum inhibitory concentration (MIC) results revealed that UV irradiated GO nanosheets have more pronounced anti-bacterial behavior than GO nanosheets and standard antibiotic, kanamycin. The MIC of UV irradiated GO nanosheets was 0.125 μg ml⁻¹ for Escherichia coli and Salmonella typhimurium, 0.25 μg ml⁻¹ for Bacillus subtilis and 0.5 μg ml⁻¹ for Enterococcus faecalis, ensuring its potential as an anti-infective agent for controlling the growth of pathogenic bacteria. The minimum bactericidal concentration of normal GO nanosheets was determined to be two-fold higher than its corresponding MIC value, indicating promising bactericidal activity. The mechanism of anti-bacterial action was evaluated by measuring the enzymatic activity of β-D-galactosidase for the hydrolysis of o-nitrophenol-β-D-galactopyranoside.

Abstract:Graphene oxide (GO) is an excellent bacteria-killing nanomaterial. In this work, macroscopic applications of this promising nanomaterial by fixing GO sheets onto cotton fabrics, which possess strong antibacterial property and great laundering durability, are reported. The GO-based antibacterial cotton fabrics are prepared in three ways: direct adsorption, radiation-induced crosslinking, and chemical crosslinking. Antibacterial tests show that all these GO-containing fabrics possess strong antibacterial property and could inactivate 98% of bacteria. Most significantly, these fabrics can still kill >90% bacteria even after being washed for 100 times. Also importantly, animal tests show that GO-modified cotton fabrics cause no irritation to rabbit skin. Hence, it is believed that these flexible, foldable, and re-usable GO-based antibacterial cotton fabrics have high promise as a type of new nano-engineered antibacterial materials for a wide range of applications.

Abstract:Antibiotics are currently the most used antibacterial treatment for killing bacteria. However, bacteria develop resistance when continually overexposed to antibiotics. Developing antimicrobial agents that can replace existing antibiotics is essential because antibiotic-resistant bacteria have resistance mechanisms for all current antibiotics and can promote nosocomial infections. To address this challenge, in this study, we propose graphene oxide/copper (GO/Cu) nanocomposites as antibacterial materials that can replace the existing antibiotics. GO/Cu nanocomposites are characterized by transmission electron microscopy and scanning electron microscopy. They show that copper (Cu) nanoparticles are well-grown on the graphene oxide sheets. Additionally, a microdilution broth method is used to confirm the efficacy of the antimicrobial substance against methicillin-resistant Staphylococcus aureus (MRSA) and Pseudomonas aeruginosa (P. aeruginosa), which are frequently implicated in nosocomial infections. Specifically, 99.8% of MRSA and 84.7% of P. aeruginosa are eliminated by 500 µg/mL of GO/Cu nanocomposites. Metal nanocomposites can eradicate antibiotic-resistant bacteria by releasing ions, forming reactive oxygen species, and physically damaging the bacteria. This study demonstrates the potential of antibacterial GO/Cu nanocomposites in eradicating antibiotic-resistant bacteria.
