Neuregulin 1 Is a Novel Oocyte Maturation Regulatory Factor Expressed in Granulosa Cells During the Ovulation Process in Mice.

Abstract:LH acts on granulosa cells to induce the expression of EGF-like factors, amphiregulin (AREG), epiregulin and betacellulin (Park et al., 2004). Since their receptor (EGF receptor, ErbB1) is expressed in granulosa cells and cumulus cells, EGF-like factors act on both cell types to induce changes in granulosa cell function, cumulus cell-oocyte complex (COC) expansion and oocyte maturation. When murine granulosa cells or COCs were cultured with AREG, the expression of genes that were induced specifically during ovulation process were detected, but the levels of expression were much lower than those observed in vivo (Shimada et al., 2006), suggesting that other LH-induced factors secreted from granulosa cells are required to induce ovulation in vivo. To find other factors, we examined the expression and functional roles of additional ErbB family members. The EGF receptors, ErbB2 and ErbB3, were expressed but not dramatically changed in granulosa cells and cumulus cells of eCG/hCG-treated mice, whereas ErbB4 was down regulated by hCG. However, hCG induced the phosphorylation of these expressed receptors. Because ErbB2 lacks a ligand binding domain and because we found high expression of neuregulin 1 (Nrg1) that selectively binds ErbB3, in cumulus cells from our micro-array database, we focused on the expression and functions of NRG1. RT-PCR analyses showed that Nrg1 mRNA increased in granulosa cells within 2 hr after hCG stimulation in an ERK1/2 dependent manner. The secreted form of NRG1, as well as intact form, were also detected at 2 hr after hCG stimulation, suggesting that the NRG1-ErbB3 pathway is functional during the ovulation process. In in vitro culture of granulosa cells or COCs, the addition of NRG1 induced the phosphorylation of PKB/AKT but not ERK1/2 in a dose dependent manner, whereas AREG up-regulated both PKB/AKT and ERK1/2 pathway. When both NRG1 and AREG were added to the culture medium, the phosphorylation of ERK1/2 increased markedly as compared with that by either ligand alone. Microarray analyses of mouse granulosa cells showed that AREG induced significantly the expression of 276 genes, 22 of which were also induced by NRG1. Star and Sphk1 were increased by each ligand in granulosa cells and cumulus cells of COCs, and the expression levels were enhanced by the addition of both ligands. Sphk1 encodes sphingosine 1 kinase to make sphingosine 1 phosphate, a known ligand for GPR3 that is expressed on oocytes and negatively regulates meiotic resumption. To determine the role of Sphk1 expressed by NRG1 in oocyte maturation, COCs were cultured with NRG1 and/or sphingosine 1 kinase inhibitor (SKI-II). The addition of NRG1 suppressed spontaneous meiotic resumption, but SKI-II overcame the inhibitory effect. NRG1 also suppressed AREG-induced meiotic resumption at 4 hr culture, but not at 16 hr, suggesting that NRG1 delayed the meiotic resumption. The matured oocyte cultured with both NRG1 and AREG had significantly higher developmental competence as compared with that of oocytes cultured with AREG alone. From these results, we show for the first time that NRG1 expressed in granulosa cells by LH has at least dual roles during ovulation process; one is to enhance AREG-induced progesterone production (increase of Star expression) in granulosa cells and the other is to regulate oocyte meiotic maturation optimizing the developmental competence via a synthesis of sphingosine 1 phosphate.
