Abstract:Abstract Black phosphorus (BP) has increasingly attracted scientific attention since its first applications in biomedicine due to its unique properties and excellent biocompatibility. In particular, its layer‐dependent bandgap, moderate carrier mobility, large surface‐area‐to‐volume ratio, biodegradability, intrinsic photoacoustic properties, and biocompatibility make it an ideal candidate for use in photothermal therapy, photodynamic therapy, drug delivery, 3D printing, bioimaging, biosensing, and theranostics, which are reviewed here. In addition, the article discusses strategies to overcome challenges related to surface instability due to chemical degradation, a major obstacle for its application. This review not only provides a comprehensive summary on BP preparation and biomedical applications but also summarizes recent research and future possibilities.

Abstract:Ascribe to the unique two-dimensional planar nanostructure with exceptional physical and chemical properties, black phosphorous (BP) as the emerging inorganic twodimensional nanomaterial with high biocompatibility and degradability has been becoming one of the most promising materials of great potentials in biomedicine. The exfoliated BP sheets possess ultra-high surface area available for valid bio-conjugation and molecular loading for chemotherapy. Utilizing the intrinsic near-infrared optical absorbance, BPbased photothermal therapy in vivo, photodynamic therapy and biomedical imaging has been realized, achieving unprecedented anti-tumor therapeutic efficacy in animal experiments. Additionally, the BP nanosheets can strongly react with oxygen and water, and finally degrade to non-toxic phosphate and phosphonate in the aqueous solution. This manuscript aimed to summarize the preliminary progresses on theranostic application of BP and its derivatives black phosphorus quantum dots (BPQDs), and discussed the prospects and the state-of-art unsolved critical issues of using BP-based material for theranostic applications.

Abstract:Black phosphorus (BP) or phosphorene, a new superstar among two-dimensional (2D) materials, has sparked huge scientific interest since its discovery in 2014. BP offers unique characteristics including high drug loading efficiency, excellent photodynamic and photothermal properties and good biocompatibility. These characteristics expand versatility of BP in nanomedicine. Although the outlook of BP seems promising, its practical biomedical applications are still at the very initial stage especially in comparison to other thoroughly investigated inorganic nanomaterials. This paper reviews BP structure and properties as well as its preparation approaches with the emphasis on techniques to improve BP stability and biocompatibility for their further usage in physiological environment. Meanwhile, recent progress made in various biomedical research fields from bioimaging to biosensing is discussed. Last, but not least, current challenges and prospects for BP in biomedicine are briefly examined, which will be useful to guide future developments of BP.

Abstract:Black phosphorus (BP), emerging as a new member of two-dimensional nanomaterials, has attracted growing research interests for its amazing photoelectric properties and promising application in electronic devices. Recently, BP has been confirmed to be a desirable candidate for phototherapy against cancer, including photothermal therapy and photodynamic therapy. By regulating the number of layers, the bandgap of BP nanosheets (NSs) can be finely tuned to present near infrared light triggered phototherapeutic behaviors. Furthermore, the exfoliated nano-sized BP also exhibits excellent tumor-targeting property as a nanomedicine via the enhanced permeability and retention effect. With biodegradable nature and outstanding therapeutic performance, BP is highly expected to be developed as novel anti-cancer agents as well as a potential carrier for advanced cancer theranostics. In this review, on the basis of summarizing the recent advances of BP in biomedical applications, the size and layer effects of BP on its targeting effect and phototherapeutic performance are discussed. Then, the rationally designed multifunctional nanoplatforms based on BP are introduced. And, the remaining challenges and prospects of nano-BP for clinic applications against cancer are discussed and outlooked.

Abstract:Black phosphorus (BP), a new type of two-dimensional nanomaterial, has attracted crucial attention in recent years owing to its excellent properties and great potential in various chemical, physical, and biological fields. In this study, BP nanosheets loaded with Au nanoparticles (BP-Au NSs) are obtained by a one-step facile synthetic method. The Au nanostructures can not only enhance the photothermal efficiency of the nanocomposites, but also endow BP-Au NSs with the potential to act as effective surface-enhanced Raman scattering (SERS) substrates for Raman biodetection. Cancer photothermal therapy (PTT) has been carried out in vitro and in vivo using BP-Au NSs as nanoagents. Under irradiation by an 808 nm laser, BP-Au NSs are capable of producing sufficient hyperthermia to destroy cancer cells, and the transplanted tumors in most of the tumor-bearing mice disappeared; BP-Au NSs are more effective than bare BP nanosheets. The PTT effect can also be monitored by a Raman technique that benefits from the high SERS activity of the BP-Au NSs. The molecular fingerprint features of breast tumors before and after PTT treatment were clearly identified using SERS analysis. The theranostic applications of BP-Au NSs exhibit promising potential in biomedicine.

Abstract:Black phosphorus nanostructures (nano-BPs) include BP nanosheets, BP quantum dots, and BP nanoparticles. Since first being discovered in 2014, nano-BP has become one of the most popular nanomaterials. Nano-BP has many unique properties, such as excellent surface activity, tunable bandgap, high carrier mobility, moderate on/off ratio, excellent biocompatibility, good biodegradation, etc., all of which make nano-BP particularly attractive in biomedicine and biosensing. This review article comprehensively summarizes recent advances in synthesis, functionalization, biomedicine, and biosensing applications of nano-BP. Different methods are first introduced, such as mechanical cleavage, liquid-phase ultrasonic exfoliation, electrochemical exfoliation, solvothermal treatment, and acoustic-microfluidic stripping, for making the nano-BP. Then two strategies are emphasized to enhance ambient stability of nano-BP, namely physical encapsulation and chemical modification. Next, how to develop nano-BP as advanced imaging agents, nanocarriers, and nanomedicine for bioimaging (fluorescence imaging, thermal imaging, and photoacoustic imaging) and disease treatment (phototherapy and photo/chemical/immune synergistic therapy) is demonstrated. The biosensing applications on nano-BP is introduced, including electrochemical biosensor, fluorescence biosensor, chemiluminescence biosensor, electrogenerated chemiluminescence biosensor, and colorimetric biosensor. Finally, the current challenges and future perspectives on nano-BP in bioapplications are discussed.

Abstract:2D materials have gained spectacular status across various scientific and technological disciplines owing to their exceptional unique properties. The very recent member of 2D family, Black Phosphorus monolayers, known as Phosphorene have attracted recent scientific attention since its first exfoliation and appreciable rediscovery in 2014. Compared to other 2D materials and graphene analogs, it has outstanding properties like tunable band gap, good carrier mobility, excellent ON-OFF current ratio, potent in vivo biocompatibility and non-toxic biodegradability. Although the outlook of this material seems to be a promising candidate for future biomedical technology, its practical applications are still highly challenging. Unveiling those challenges by proper characterization and functionalization makes this material a mile stone for future theranostic and biomedicine scenario. This review has given precise attention to familiarize with the unique fundamental properties of black phosphorus, which makes it an excellent platform for future biomedical applications. Also underlines various synthesis procedures applicable for BP nanosheets and quantum dot synthesis. Its various biomedical applications including biosensors, cancer therapy, imaging and photothermal/photo acoustic/photodynamic therapy, drug delivery, neuronal regeneration, 3D printing scaffold etc., are subsequently reviewed. Furthermore this review briefly focused on the toxicity of this emerging material.

Due to its puckered structure, BP exhibits strong in‐plane anisotropy unlike other layered materials. Abstract:Low-dimensional black phosphorus (BP) is a class of nanomaterial derived from layered semiconductor BP which has gained tremendous attention in a variety of fields, owing to its uncommon structural features and appealing physical properties. More surprisingly, it has addressed current biomedical obstacles due to its orthorhombic puckered honeycomb crystal structure and unique properties such as tunable direct-bandgap, high carrier mobility, and exceptional photo-responsiveness. However, few reviews have focused on the interactions of low-dimensional BP’s physical properties with its biomedical performances. Herein, we discuss the physical properties of low-dimensional BP and potential biomedical applications associated with these physical properties. Moreover, different preparation methods, surface modification techniques, and future challenges, as well as future outlooks, are presented. This comprehensive review will provide a clear understanding of the relationship between low-dimensional BP’s physical properties and biomedical performances, with the ultimate goal of better knowledge of utilizing BP.

Abstract:Black phosphorus (BP) has exhibited excellent biocompatibility and high photothermal conversion efficiency under near-infrared light, which makes it very promising for photothermal therapy. However, practical applications are highly hampered because it lacks a targeting property and rapidly degrades in cancer cells, especially in response to strong intracellular oxidative stress. Here, we reported that the mitochondrial targeting peptide functionalized black phosphorus nanosheets covered with an acid-labile polymer shell (doubly functionalized black phosphorus (DFBP) nanosheets) exhibited good stability. DFBP nanosheets not only have excellent ability of accumulating in tumor tissue via surface charge switching but also can target mitochondria. The doubly functionalized black phosphorus nanosheets resulted in robust cancer cell uptake but very poor normal cell accumulation. In vivo, the BP nanosheets could highly accumulate in a tumor and specifically target mitochondria, generating enough hyperthermia under near-infrared light, leading to cell death. This work provides a powerful way to ablate a tumor selectively with negligible side effects.

Abstract:Zero-dimensional (0D) black phosphorus quantum dots (BPQDs), the new derivatives of black phosphorus (BP) nanomaterials, have attracted considerable attention since they were first prepared in 2015. Compared to traditional two-dimensional (2D) BP nanosheets, BPQDs exhibit some unique properties and demonstrate great potential for a broad range of applications, especially in the field of biomedicine. Due to the rapid development and substantial research interest in this area, it is urgent to review the current advances, challenges and near-future possibilities of BPQD-related biomedical research, which will benefit the further development of this field. This review is mainly focused on the latest progress of BPQD related applications in the biomedical field, including photothermal therapy (PTT), photodynamic therapy (PDT), drug delivery, biological imaging, etc. The challenges and future prospects are also discussed.

Abstract:Flake-shaped nanohybrids based on black phosphorus (BP) have been developed as multifunctional theranostic nanoplatforms for drug delivery, phototherapy and bioimaging. In this work, we report a facile strategy for fabrication of black phosphorus-Au nanoparticle hybrids (BP-AuNPs), which reveal an extraordinary near-infrared (NIR) photothermal transduction efficiency and drug delivery capacity. The applications of the nanocomposites as therapeutic agents for high-performance chemo-photothermal tumor therapy are accomplished in vitro and in vivo. BP-AuNPs also exhibit wonderful surface-enhanced Raman scattering (SERS) activity under NIR laser excitation with a low Raman background, allowing BP-AuNPs to be used as a promising two-dimensional (2D) fingerprint nanoprobe for bio-SERS analysis. The cellular component identification and label-free live-cell bioimaging based on this type of 2D SERS substrate are generally investigated, which open up promising new perspectives in nanomedicine, including diagnosis, imaging and therapy.

Abstract:Since their discovery, Black Phosphorus (BP)-based nanomaterials have received extensive attentions in the fields of electromechanics, optics and biomedicine, due to their remarkable properties and excellent biocompatibility. The most essential feature of BP is that it is composed of a single phosphorus element, which has a high degree of homology with the inorganic components of natural bone, therefore it has a full advantage in the treatment of bone defects. This review will first introduce the source, physicochemical properties, and degradation products of BP, then introduce the remodeling process of bone, and comprehensively summarize the progress of BP-based materials for bone therapy in the form of hydrogels, polymer membranes, microspheres, and three-dimensional (3D) printed scaffolds. Abstract:Black phosphorus (BP) is one of the emerging versatile nanomaterials with outstanding biocompatibility and biodegradability, exhibiting great potential as a promising inorganic nanomaterial in the biomedical field. BP nanomaterials possess excellent ability for valid bio-conjugation and molecular loading in anticancer therapy. Generally, BP nanomaterials can be classified into BP nanosheets (BPNSs) and BP quantum dots (BPQDs), both of which can be synthesized through various preparation routes. In addition, BP nanomaterials can be applied as photothermal agents (PTA) for the photothermal therapy (PTT) due to their high photothermal conversion efficiency and larger extinction coefficients. The generated local hyperpyrexia leads to thermal elimination of tumor. Besides, BP nanomaterials are capable of producing singlet oxygen, which enable its application as a photosensitizer for photodynamic therapy (PDT). Moreover, BP nanomaterials can be oxidized and degraded to nontoxic phosphonates and phosphate under physiological conditions, improving their safety as a nano drug carrier in cancer therapy. Recently, it has been reported that BP-based PTT is capable of activating immune responses and alleviating the immunosuppressive tumor microenvironment by detection of T lymphocytes and various immunocytokines, indicating that BP-based nanocomposites not only serve as effective PTAs to ablate large solid tumors but also function as an immunomodulation agent to eliminate discrete tumorlets. Therefore, BP-mediated immunotherapy would provide more possibilities for synergistic cancer treatment.

Abstract:In article 1603276, J. Shi, H. Zhang, L. Mei, and co-workers describe black phosphorus nanosheets (BP NSs), which could lead to an enhanced antitumor effect, by a triple-response combined therapy strategy. When they enter the cancer cell, the BP NSs can produce a local high temperature initiated by an 808 nm laser. Release of loaded doxorubicin can be triggered by the laser and heat, leading to a combined photothermal–chemotherapy effect, which effectively kills the cancer cell. Moreover, under the influence of chloroquine, the degradation of BP NSs in the lysosome can be inhibited. The drug is then concentrated in the cell nucleus.
