Abstract:Choosing some representative kinds of green tea as the materials, the experiment established the calibra- tion models of three main sapor components——tea polyphenols (TPP), amino acids (AA) and theine from the tea soup by using NIR Quantitative analysis. The tea polyphenols(TPP) model: the standard error of calibration(SEE), correlation coefficient(r)of calibration set(C-set), standard error of cross-validation (SEP) and validation set (V-set) were: 1.1455、 0.8792、1.0600、0.8623. The AA model: its SEE, SEE -r, SEP and SEP -r were: 0.2518、0.9728、0.2597、0.9722. The theine model: its SEE, SEE-r, SEP, and SEP-r were: 0.5553、0.9648、0.5315、0.9554. It indicated that by external cross validation, correlation coefficient(R) of true content of samples and forecasted content of samples: R of TPP was 0.8935, R of AA was 0.9541, R of theine was 0.9386. The models could satisfy the analysis requirements of tea components. The proposed method was proved to be a rapid method for mensurating components of tea soup.
Rapid Quantitative Determination of Fritillaria Thunbergii Miq Mixed into Fritillaria Cirrhosa D. Don by Near- Infrared Diffuse Reflectance Spectroscopy

Abstract:Objective: To set up a new method for rapid quantitative determination of Fritillaria thunbergii Miq (FTM) mixed into Fritillaria cirrhosa D. Don (FCD) by combining near-infrared diffuse reflectance spectroscopy (NIRS) with Partial Least Square (PLS). Methods: Collect 41 powder samples with different rate mixed with FTM and FCD, and scan their NIR spectra. Then use Partial Least Square (PLS) to correlate these data and get the bestfitted model. Final use this model to analysis prediction samples. Results: The correlation coefficient (r) for the prediction samples is 0.9997,the recovery is 97.96%-100.9% ,the RSD is 0.81%. Conclusions: The new method proposed could be used directly to determine the mixture of Chinese traditional medicine materials which are similar in appearance and chemical components. It is fast, convenient and accurate.
A Method for the Quantitative Analysis of a Key Component in Complex Mixtures Using Raman Spectroscopy Based on Peak Decomposition

Abstract:Raman spectroscopy has been increasingly used in industrial production processes for analytical purposes. However, the quantitative analysis of complex mixtures based on their Raman spectra remains a problem, especially when not all of the spectra of pure components in the mixture are available. In this work, a method for the quantitative analysis of a key component in complex mixtures based on peak decomposition is introduced. The proposed method only requires the spectrum of the main component in the mixture before analysis. The spectra of the mixture are decomposed to the contributions of the main component and a group of parametric Lorentzian functions. Then, the representative peaks of the key component are extracted. Finally, a calibration model is established. This method is applied to a complex mixture containing m -xylene, o -xylene, p -xylene, ethylbenzene, methylbenzene, three trimethylbenzenes, non-aromatic hydrocarbons, etc. to determine the concentration of o -xylene in the mixture. Experiments demonstrate that the proposed method performs better than the commonly-used partial least squares (PLS), especially when the number of training samples is very small.

Abstract:Three spectrum regions (6100-4250 cm -1, 61 00 -5450 cm -1 and 4600-4250 cm -1, 7500-5450 cm -1and 4600-4250 cm -1) and four pretreatment methods (straight line subtraction, multiplicati ve scattering correction, vector normalization; first derivation ) were compare d to form a model for testing fat content in milled rice by using near infrared reflectance spectroscopy (NIRS). It was found that spectrum regions and pretreat ment methods had large effects on the model construction. In order to estimate the forecast result of the model, the feasibility of testing fat content in mil led rice by using NIRS and its application in breeding was discussed.

Abstract:At present, the terahertz time-domain spectroscopic information of each component in multi-composition compounds detection is comprehensively combined. The lower resolution level of mixed spectra has posed many difficulties in signal analysis due to the overlapping characteristic information in the mixed ones. In this paper, to compare the performances of Nonnegative Matrix Factorization (NMF), Self-modeling Mixture Analysis (SMMA) and Multivariate Curve Resolution-Alternating Least Squares (MCR-ALS) on complex systems, a binary mixture and a ternary mixture are employed during THz-TDS testing. The position of the absorption peak (PK) and the correlation coefficient are used to evaluate the decomposition effects. The experimental results show that the component spectra resolved by MCR-ALS demonstrate good consistency with the sample components. Further, MCR-ALS presents excellent results in comparison with NMF and SMMA in terms of decomposing precision and computing speed. MCR-ALS thus appears a promising algorithm to resolve the THz multi-way data, and may be useful for many applications, such as medicine quality assurance and unknown component identification in the general area of terahertz science and technology.
Raman spectral quantitative analysis based on peaks-decomposition

Abstract:The existing calibration algorithms for Raman spectral quantitative analysis,such as principal component regression,partial least squares and support vector machine need a large number of training samples and the regression model may have unsatisfactory generalization ability.Indirect hard modeling(IHM)is a recently introduced method for spectral quantitative analysis,which is used to analyze spectra with highly overlapping band and nonlinear variation.This method can be used to build a calibration model with high generalization ability only by a few training samples.However,all the spectra of pure-components in the mixture is required in IHM method.Obviously,this condition is difficult to achieve in practical applications.To overcome the limitation of IHM,a novel method based on Voigt function for spectral quantitative analysis,i.e.,direct hard modeling(DHM),was presented.The novel method did not require the pure-component spectra.By DHM algorithm,the characteristic peaks of every pure component in the spectra of mixture could be fixed,and then a linear calibration model could be built based on the areas of unique peaks and the concentration of pure-components.The DHM algorithm was successfully demonstrated while testing on the quantitative analysis of xylene mixture in a PX unit.Experimental results showed that the DHM algorithm had the advantages of robustness and fewer training samples.

Abstract:To overcome the shortcoming of traditional Raman spectral decomposition method that requires a priori information of pure spectra, a new Raman spectral decomposition algorithm based on multivariate curve resolution and Lorentz function constraint was proposed. This method was based on the bilinear model for mixture spectra, and only the mixture spectral matrix was needed to perform the analysis. It first decomposes the mixture spectra with principal component resolution, and then the evolving factor analysis was performed to find the distribution of the mixture spectral eigenvalues evolves along with the pixel direction, so that it could determine the number of components in the mixture, the range of their peaks and the initial estimation 14 their spectra. Finally an alternative least squares method with Lorentz function constraint was proposed to optimize the pure spectra, during the process, it first fits the pure spectra with Lorentz function, and then based on the fitted spectra, it regresses the spectra and the coefficient matrix with non-negative alternative least squares method. The above optimization process was repeatedly performed until the fit error for the mixture spectra was within the threshold. In the experimental part, the proposed algorithm was adopted to decompose the Raman spectra of the circulating fluid in an adsorption tower of a p-xylene unit, with which the relative pure spectra was obtained in the mixture fluid. Finally, with the repeated quantitative analysis experiments for its main component of p-xylene, we prove the effectiveness for the proposed algorithm.

Abstract:Digital image processing has been applied on various fields such as classification and qualitative analysis. In this work, a very simple quantitative approach was proposed for the first time. Based on the digital grayscale images of three-dimensional fluorescence spectra, several wavelet moment invariants were calculated, and used to establish the linear models for the quantitative analysis. This approach was applied to the quantitative analysis of Tryptophan, Tyrosine and Phenylalanine in mixture samples, and the correlation coefficients R2 of the obtained linear models were more than 0.99, which were supported by the strict statistical parameters as well as leave-one-out and Jackknife cross-validations. Our study indicates that the selected wavelet moment invariants are immune from the noise and background signals, and the quantitative analysis can be performed accurately based on the overlapping peaks of compounds in mixture. This proposed approach provides a novel pathway for the analysis of three-dimensional spectra.

Blind Spectral Unmixing in Terahertz Domain Using Nonnegative Matrix Factorization

Abstract:Recovering component spectra from terahertz measurements of unknown mixtures has been studied in this paper using nonnegative matrix factorization (NMF). NMF mathematically decomposes the spectra data into two nonnegative matrixes which describe the component spectra and the corresponding fractional abundance. Two basic algorithms in the class of this method, NMF and NMF with smoothness constraint (cNMF), were adopted to resolve the terahertz absorption spectra matrix obtained from a ternary mixture with varying compositions of Nitrofurantoin, L-Leucine and D-Tyrosine. The quality of the decomposition results was evaluated. The performance of the two algorithms on extracting component terahertz spectra was compared. The optimal result reached by cNMF in this study implies the capability of the NMF method for blind terahertz spectral unmixing. The attempt made in our work helps to further investigate unknown mixtures by terahertz spectroscopy.
Study on the Influence of Temperature on Near Infrared Spectra

Abstract:Near-infrared (NIR) spectroscopy technique is one of qualitative and quantitative analysis techniques that were developed quickly in recent years. However, the characteristic of NIR spectral region determines its low absorbance, low sign-to-noise and peak overlapping, which results in the phenomenon that th NIR spectrum is affected by sample source, sample varieties, sample state, location and sample temperature and uncertainty of spectrum. In this paper, the influence of temperature on NIR spectra was studied and calibration and validation models of chlorophyll at different temperatures were built. The results show that temperature does have effect on the model precision. High precision could be achieved at 10 and 20 degrees C and the former was the best though using more principal components. Calibration and validation precisions of models were poor when it was 25 degrees C. At the same time, spectra obtained at different temperatures were classified using discriminate analysis. It was found that the spectra were misclassified exception ones obtained at 20 degrees C which meant that there were not clear differences between the spectra exception the ones at 20 degrees C. The primary research work about the condition and application of detecting leaves was studied which helps to provide theory foundation for proposing temperature modification model in the future.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.

Abstract:This study aims at resolving the spectral profiles of pure components from the mixture THz data. A method based on simple-to-use interactive self-modeling mixture analysis (SIMPLISMA), one of self-modeling curve resolution (SMCR) techniques, was put forward to deal with two-way THz spectral data of unknown mixtures for spectral resolution of single species. SMCR requires pure variables which have intensity contributions from only one component for extracting the content information. However, for the spectra of many materials in THz region (about 0.1–3THz), pure variables are usually unavailable for the overlap of absorption peaks or the presence of baseline. To settle this problem, inverted second derivative was employed as an intermediate step to optimize the determination of pure variable. To test the effect, the proposed method was performed on simulated and experimental THz spectra of multicomponent systems. It is demonstrated that SIMPLISMA combined with inverted second derivative spectra is a promising tool to deconvolve multicomponent systems contributing to the identification of ingredients in unknown mixture in THz region.

Abstract:Terahertz (THz) radiation is the electromagnetic spectrum lying between the microwave and infrared portions, and covers wavelength between 3mm and 30um. Terahertz spectroscopy has been used in qualitative and quantitative analysis since molecular vibrations and rotations of a great large number of materials lie in this region. Many techniques, such as principal component regression (PCR) and partial least square regression (PLSR), have been applied in quantitative analysis of terahertz time domain spectroscopy. However, those "soft-modeling" methods, though effective in many cases, all require a large number of samples and have poor extrapolation capability, and the utilization is also restricted in some cases (samples lack or requirement of extrapolation). A parametric method of spectrum, Indirect Hard Modeling (IHM), was proposed to resolve terahertz spectral matrix and quantify the mixtures. Savitzky-Golay method was used to eliminate the influence of noise. Moreover, the features of terahertz spectra were considered and the artificial baseline was also resolved as a signal spectrum to improve spectrum extraction accuracy. Subsequently, the analysis of IHM was carried out, and the ability of prediction and accuracy of the model in the case of low training samples were discussed. In order to validate the capability of the proposed method, quaternary systems of Rifampicin, Lactose monohydrate, microcrystalline cellulose and magnesium stearate were tested. The regression correlation coefficient (R) and root mean square error (RMSE) were employed to estimate the performance of the model. Finally, the comparison of IHM and PLSR were carried out, and theoretical analysis and experimental results indicated that the number of samples required by IHM modeling could be reduce to two during calibration, at the same time, the accuracy of quantitative and extrapolation capabilities were also improved.
Component Spectra Extraction from Terahertz Measurements of Unknown Mixtures.

Abstract:The aim of this work is to extract component spectra from unknown mixtures in the terahertz region. To that end, a method, hard modeling factor analysis (HMFA), was applied to resolve terahertz spectral matrices collected from the unknown mixtures. This method does not require any expertise of the user and allows the consideration of nonlinear effects such as peak variations or peak shifts. It describes the spectra using a peak-based nonlinear mathematic model and builds the component spectra automatically by recombination of the resolved peaks through correlation analysis. Meanwhile, modifications on the method were made to take the features of terahertz spectra into account and to deal with the artificial baseline problem that troubles the extraction process of some terahertz spectra. In order to validate the proposed method, simulated wideband terahertz spectra of binary and ternary systems and experimental terahertz absorption spectra of amino acids mixtures were tested. In each test, not only the number of pure components could be correctly predicted but also the identified pure spectra had a good similarity with the true spectra. Moreover, the proposed method associated the molecular motions with the component extraction, making the identification process more physically meaningful and interpretable compared to other methods. The results indicate that the HMFA method with the modifications can be a practical tool for identifying component terahertz spectra in completely unknown mixtures. This work reports the solution to this kind of problem in the terahertz region for the first time, to the best of the authors' knowledge, and represents a significant advance toward exploring physical or chemical mechanisms of unknown complex systems by terahertz spectroscopy.

Abstract:The distinctive vibrational features in terahertz (THz) spectroscopy characterize a "fingerprint" of the single-component molecular substance. However, due to componential spectral overlapping and baseline drift, the identification and quantification of multicomponent mixtures are quite challenging for THz spectral analysis. A systematic and feasible strategy has been proposed by combining machine learning with THz spectroscopy for both qualitative and quantitative analysis. After the component number was effectively determined by singular value decomposition (SVD), nonnegative matrix factorization (NMF) and self-modeling mixture analysis (SMMA) were applied to extract componential THz spectra. The difficulties of NMF and SMMA encountered in handling ternary mixtures were solved. The results show component spectra extracted by SMMA are highly consistent with the experimental spectra of pure substances after standardization to correct baseline drift, which greatly facilitates rapid identification of compositions in mixtures. Additionally, compared to back-propagation neural network (BPNN), support vector regression (SVR) predict the contents of each individual component with high robustness and the decision coefficient R 2 greater than 0.949. Fingerprint terahertz spectroscopy enhanced by machine learning provided an effective strategy for mixture analysis in practical applications.

[Study on Rapid Quality Analysis Method of Shengxuebao Mixture].

Abstract:A rapid analysis method based on ultraviolet-visual(UV-Vis) spectroscopy, near infrared(NIR) spectroscopy and multivariable data analysis was established for quality evaluation of Shengxuebao Mixture. The contents of eight active ingredients of Shengxuebao Mixture including albiflorin, paeoniflorin, 2, 3, 5, 4'-tetra-hydroxy-stilbene-2-O-β-D-glucopyranoside, specnuezhenide,ecliptasaponin D, emodin, calycosin-7-glucoside and astragaloside Ⅳ were simultaneously detected by using this method. HPLC-UV-MS was used as a reference method for determining the contents of these ingredients. Partial least squares(PLS) analysis was implemented as a linear method for multivariate models calibrated between UV spectrum/NIR spectrum and contents of 8 ingredients. Finally, the performance of the model was evaluated by 24 batches of test samples. The results showed that both UV-Vis and NIR models gave a good calibration ability with an R~2 value above 0.9, and the prediction ability was also satisfactory, with an R~2 value higher than 0.83 for UV-Vis model and higher than 0.79 for NIR model. The overall results demonstrate that the established method is accurate, robust and fast, therefore, it can be used for rapid quality evaluation of Shengxuebao Mixture.
Design of an FT-NIR Spectrometer for Online Quality Analysis of Traditional Chinese Medicine Manufacturing Process

Abstract:As a fast, non-destructive analysis method, Fourier transform (FT) near-infrared (NIR) spectroscopy is very suitable and effective for online quality analysis of traditional Chinese medicine (TCM) manufacturing process. In this thesis, the theoretics of FT-NIRS was analyzed and an FT-NIR spectrometer with 4 cm(-1) resolution in the 12500-5000 cm(-1) frequency range was designed. The spectrometer was based on a Michelson interferometer with Bromine tungsten lamp as the NIR light source and InGaAs detector to collect the interference signal. Each element was designed and chosen to provide maximum sensitivity in the NIR spectral region. A fiber-optic flow cell system was used to realize online analysis of traditional Chinese medicine. The performance of the spectrometer was evaluated and the feasibility of using FT-NIR spectrometer to get absorption spectra of traditional Chinese medicine was demonstrated.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Abstract:Quantification of each constituent within a multicomponent system is very important.Modern spectrometers such as Raman spectrometry (RS),Fourier transform infrared (FT-IR) spectroscopy,ultraviolet-visible(UV-Vis) spectroscopy,nuclear magnetic resonance (NMR),mass spectrometry (MS) help to provide a colorful view inside a sample by acquiring spectra with abounding peaks.However,it has been extremely challenging for them to simultaneously quantify each constituent in a mixture by sample spectra,due to the complexity of the spectra and the burden of attributing peaks for each constituent.In this study,we introduce an effective and reliable strategy named quantitative principal component analysis(qPCA) to calculate the proportion of each constituent within a mixture without any manual spectral inspection for the first time.By using spectra of pure constituent as references,a spectrum of multicomponent system can be automatically resolved by PCA and then proportion of each constituent can be calculated with our calculating method.Both computer modeling and RS,FT-IR,UV-Vis,1H-NMR,ESI-MS experiments have demonstrated this strategy suitable to accomplish quantification task for multicomponent systems.

Abstract:Traditional Chinese medicines (TCMs) have been used in China to prevent and cure diseases for centuries. Due to the complexity of TCMs, fast and highly efficient techniques are required to develop their quality control. Near infrared spectroscopy (NIRS) has been widely applied in TCM analysis for both nonpolar and polar components. This review describes an overview of experimental conditions (spectra pretreatments and multivariate techniques) investigated in NIRS for TCM components that are related to individual compounds such as lipids, essential oils, phenolics, alkaloids, saponins and carbohydrates. Their potential in the separation of TCMs using relevant applications with a particular focus on quantitative analysis is also demonstrated.
