Abstract:Objective To investigate the clinical effects of reduced glutathione on the treatment of chronic active hepatitis B in children.Methods Thirty-six children with chronic active hepatitis B were recruited and treated with reduced glutathione,clinic parameters and hepatic functions were analyzed 14~28 days after treatment.Results Liver function parameters such as serum ALT,AST and bilirubin were significantly decreased after systemic treatment compared with those at the beginning of clinical trial.Clinical significances were also improved.Conclusion Reduced glutathione is effective to treat chronic hepatitis B in children and correct liver function abnormalities.
Effects of different dialyzers on oxidative stress for maintenance hemodialysis patients

Abstract:Objective To explore the effect of different dialyzers on the oxidative stress in maintenance hemodialysis(MHD) patients.Methods Thirty MHD patients were randomly divided into three groups(n = 10). The patients were dialyzed with regenerated cellulose membrane(650 group),polysulfone membrane(F6 group) and vitamin E-modified membrane(CL-E group).Ten healthy persons(CON group) and ten uremia patients without dialysis(NHD group) were taken as control.Blood samples were collected on an empty stomach in all the groups for the biochemical tests and for the plasma concentrations of malonaldehyde(MDA),total antioxidation capacity(T-AOC),advanced oxidation protein products(AOPPs),and vitamin E(Vit E) before and after dialysis. Results The average age and the level of C reactive protein in the NHD,650,F6 and CL-E groups were significantly higher than those in the CON group(P<0.05).The hemoglobin level in the NHD group was significantly lower than that in CON group(P < 0.05).The levels of MDA and AOPPs in the NHD group were significantly higher than those in the CON group(P<0.05),but there was no significant difference between the two groups in the levels of T-AOC and Vit E.There were few changes of AOPPs in 650,F6,and CL-E groups before and after dialysis(P>0.05).The T-AOC level was significantly decreased in 650,F6,and CL-E groups after dialysis(P<0.01).The level of MDA was remarkably decreased in CL-E group after dialysis(P<0.05), but there were no obvious changes of MDA in 650 and F6 groups before and after dialysis.The level of Vit E in CON group([5.02±3.80]ug/mL) and NHD group([5.19±4.54]ug/mL) were significantly higher than that in CL-E group before dialysis([0.92±0.23]ug/mL,p<0.05);the level of Vit E in CL-E group after dialysis ([6.26±6.45]ug/mL) was significantly increased as compared with the level before dialysis(P<0.05). Conclusion The oxidative stress is present in the MHD patients.CL-E can improve the oxidative stress in MHD to some extent.'
The effect of reduced L-glutathione supplementation on TNF-α, hs-CRP, and neutrophil-lymphocyte ratio in maintenance hemodialysis patients

Abstract:Objective: Cardiovascular disease is the main cause of mortality in patients with chronic kidney disease stage 5 on dialysis (CKD-5D). High sensitivity C-reactive protein (hs-CRP), tumor necrosis factor-α (TNF-α), and neutrophil-lymphocyte ratio (NLR) are several inflammatory parameters associated with high cardiovascular events in CKD-5D. The main aim of this study was to evaluate the effect of reduced L-glutathione supplementation on serum hs-CRP, TNF-α, and NLR in patients with CKD-5D. Patients and methods: This study is a quasi-experimental research with one group pretest-posttest design. Subjects included were patients with CKD-5D who routinely underwent hemodialysis therapy two times a week in Hasan Sadikin General Hospital. Serum hs-CRP, TNF-α, and NLR levels were obtained before and after the intervention of reduced L-glutathione supplementation dosing of one thousand milligrams a day for four weeks. Statistical analysis was then conducted using the Wilcoxon test. Results: There were 26 hemodialysis patients included in the study, with a median age of 43 years and a male predominance. There was a significant decrease in serum TNF-α level after reduced L-glutathione supplementation for 4 weeks, 5.40 (-10.80-0.00) pg/mL, p = 0.002. However, there was no statistically significant decrease in either serum hs-CRP level, 0.40 (-0.70-0.80) mg/L (p = 0.656), or NLR with a difference of 0.55 (0.30-1.00) p = 0.055. Conclusions: Exogenous oral reduced glutathione supplementation for four weeks significantly reduced TNF-α level, but no significant decrease in hs-CRP level and NLR in patients with CKD-5D.
Effects of Trimetazidine on the Oxidative Stress in Maintenance Hemodialysis Patients

Abstract:Objective To investigate the effects of trimetazidine on the oxidative stress in maintenance he-modialysis (MHD) patients. Methods Eighty-six MHD patients and 30 healthy volunteers were recruited in the study. The activity of glutathione peroxidase (GSHPx) was measured by colorimetry and superoxide dismutase (SOD) in serum was measured by hydroxylamine method. The levels of serum malondialdehyde (MDA) were meas-ured by thiobarbituric acid reaction. Serum advanced oxidation protein products (AOPP) levels were measured by enzyme-linked immunosorbent assay (ELISA). All MHD patients were randomly divided into two groups, treatment group (n = 46) and control group (n = 40), who had undergone hemodialysis for at least three months before the study and were in a stable clinic status without signs of infection or disease activity. In the treatment group,20 mg of trimetazidine was taken orally three times each day for twenty-four weeks, when the parameters for oxidative stress were studied. The levels of GSHPx. SOD, MDA and AOPP in serum were measured before and after the treat-ment. Results At the initiation of the investigation, the serum levels of GSHPx [(584.37±215.70) μmol/L, (580.87±309.12) μmol/L vs (769.06±302.46) μmol/L] and SOD [(347.87±82.09) kU/L, (348.16±75.33) kU/L vs (428.34±15.23)kU/L] in the M HD patients were significantly lower than those in the normal eontrol group (P < 0.01), whereas the content of MDA [(4.94±1.32) nmol/L, (4.97±1.61) nmol/L vs (3.56±0.46)nmol/L] and AOPP [(120.95±59.24) μg/L,(121.76±69.12) μg/L vs (47.69±20.15) μg/L] in MHD patients was higher than those in the control group( P < 0.05 and P <0.01, respectively). After treatment for twelve weeks, the scores of GSHPx and SOD were significantly increased in the treatment group compared to that before treatment (P <0.01). However, the contents of the MDA and AOPP decreased. There were significant differences in the levels of GSHPx, SOD,MDA and AOPP between the two groups of MHD patients after the treatment with trim-etazidine. Conclusions Trimetazidine in maintenance hemodialysis patients appears to be associated with an im-provement of oxidative stress.
Preliminary application of ascorbate-rich dialysate on ameliorating oxidative stress in maintenance hemodialysis patients

Abstract:Purpose To observe the effects of ascorbate (vitamin C, Vit C) dialysate on oxidative stress in maintenance hemodialysis (MHD) patients, particularly in comparison with those of intravenous ascorbate supplementation. Methods (1) Single dialysis session: 8 MHD patients performed three successive dialysis sessions, and each course was taken as a group. They were respectively] 1control group; 2iv-AA group; 3D-AA group(using ascorbate-containing acid dialysate of 2 g/L). Blood samples were drawn every hour during each course for determining plasma total ascorbic acid (TAA), ratio of dehydroascorbic acid(DHAA) to TAA(DHAA/TAA), vitamin E(Vit E), malondialdehyde(MDA) in both plasma and erythrocytes. (2) 4-week application of ascorbate-rich dialysate : Twenty-four MHD patients were randomly divided into control group(n= 12) and experimental group (n= 11 with use of acid dialysate containing ascorbic acid 0. 8 g/L). The above-mentioned oxidative stress markers, oxidized low density lipoprotein (oxLDL), advanced oxidation protein products (AOPP), serum C-reactive protein(CRP), serum ferritin(SF) and hematological indices were examined before and after the 4-week period. Results (1) Plasma TAA gradually decreased during the course of dialysis in control group meanwhile MDA showed a moderate and significant increase[ postdialysis (8. 47 ± 3. 45) μmol/L vs pre-dialysis (5. 12 ± 1. 15) μmol/L,P<0. 05]. A TAA concentration peak protruded in the curve right after injection of ascorbic acid, accompanied by dramatic reduction of DHAA/TAA. However, a precipitous fall of TAA soon followed and ultimately reverted to the similar level at the beginning of dialysis. Plasma TAA concentration remained table and VitE level slightly elevated in D-AA group[post-dialysis (26. 7 ± 2. 8) μmol/L vs pre-dialysis (22. 4 ± 0. 9) μmol/ L, P<0. 001]. (2) Plasma TAA augmented in experimental group at the end of the 4-week investigation[at the end (83. 8 ± 56. 5) μmol/L vs at the start (32. 6 ± 25. 2) μmol/L, P<0. 05], and plasma oxLDL value became lower in the experimental group. Conclusions Plenty of plasma ascorbic acid losses in hemodialysis while oxidative stress is exacerbated. Ascorbic acid supplementation via either intravenous injection or extracorporeal circuit will alleviate loss of this antioxidant and help attenuate oxidative stress. but internal exquisite change will probably be avoided only by the latter means.
Effect of Reduced Glutathione on Diabetic Cardiomyopathy Patients with Heart Failure

Abstract:Objective: To observe the effect of reduced glutathione(GSH) on diabetic cardiomyopathy patients with heart failure.Methods: 70 diabetic cardiomyopathy patients with heart failure were randomly divided into control group(n=34)and treatment group(n=36).Patients in control group were treated by routine medicine therapy(Betaloc,spironolactone,hydrochlorothiazide,digoxin and Valsartan tablets);Patients in treatment group were treated by combination of reduced glutathione tablets and routine medicine.The total effective rate,glycemic control,TGF-β1 and cardiac structure were observed after 8 weeks of treatment.Results: The total effective rate in treatment group(83.3%) was higher than that in control group(61.8%)(P0.05);FBG,PBG,HbA1c were decreased significantly(P0.05 or P0.01) after treatment,and HbA1c in treatment group was significantly lower than that in control group significantly(P0.05).TGF-β1 in treatment group markedly was decreased(P0.05) after 8 weeks,also decreased significantly compared with the control group(P0.05).LAD,LVD were declined,EF increased(P0.05) after treatment,and that LVD and EF ameliorated more significantly that in the treatment group(P0.05).Conclusions: For diabetic cardiomyopathy patients with heart failure,the therapy of reduced glutathione tablets and routine medicine can effectively relieve clinical symptoms,improve blood glucose,reduce blood TGF-β1,and improve left ventricular structure and function.
A Randomized Controlled Trial of Reduced Glutathione in the Treatment of Acute Renal Failure

Abstract:Objective To evaluate the effectiveness and safety of reduced glutathione in the treatment of acute renal failure.Methods Twenty-three patients with acute renal failure were divided into the treatment group (n=10) and the control group (n=13) by a simple random method.Patients in the treatment group received intravenous reduced glutathione 1 200 mg daily,whereas patients in the control group were not treated with reduced glutathione.The therapeutic course for the two groups was 4 weeks.Serum creatinine and urea nitrogen were determined before treatment as well as at the end of Week 1,2,3 and 4.Proximal and distal renal tubular functions were evaluated at the end of the treatment.The time when clinical symptoms were improved was recorded and adverse drug reactions were monitored.Results The durations of nausea and vomiting as well as the oliguria stage in the treatment group were obviously shorter than those of the control group. The serum creatinine level in the treatment group decreased more markedly than that of the control group.At the end of the treatment,the renal tubular function in the treatment group was better than that of the control group.Conclusion Reduced glutathione in the treatment of acute renal failure contributes to the early recovery of renal function.However,more high- quality and large-scale randomized controlled trials are needed.
Reduced Glutathione for Prevention of Renal Outcomes in Patients Undergoing Selective Coronary Angiography or Intervention.

Abstract:ObjectivesTo evaluate the effect of reduced glutathione on renal outcomes following the selective coronary angiography and/or intervention. BackgroundContrast agents can cause an acute reduction in renal function that may be due to the generation of reactive oxygen species. The role of antioxidants in prevention of this renal impairment is controversial. Methods and ResultsWe conducted a randomized, placebo-controlled trial of reduced glutathione in 825 patients who underwent selective coronary angiography and/or intervention. Patients were assigned to reduced glutathione 1800mg (n=416) or placebo (n=411) intravenously. Contrast-induced acute kidney injury was defined by an absolute increase of serum creatinine 0.5mg/dl (44.2mol/L) or a relative increase of 25% measured 48hours after the procedure. The incidence of contrast-induced acute kidney injury was 5.07% in the glutathione group and 4.97% in the control group (relative risk, 1.04; 95% confidence interval, 0.59-1.83; P=0.886). Change in serum malondialdehyde was (-) 1.011.69nmol/ml in the glutathione group and (-) 0.67 +/- 1.55nmol/ml in the control group (P=0.054), and change in serum total antioxidant capacity level was also similar in both groups (0.91 +/- 2.06nmol/ml and 0.79 +/- 2.18nmol/ml, respectively; P=0.936). ConclusionsA single dose of reduced glutathione does not reduce the risk of contrast-induced acute kidney injury in patients undergoing coronary angiography and/or intervention. Multivariate analysis of oxidative stress in maintenance hemodialysis patients

Abstract:Objectives To investigate the oxidative stress in maintenance hemodialysis(MHD)patients and analyze the multiple factors of oxidative stress.Methods A total of 152 patients receiving hemodialysis were selected in this study,and 30 healthy volunteers were selected in the control group.Malondialdehyde(MDA),total antioxidant capacity(TAC),serum creatinine (Scr),C reactive protein (CRP),Uric Acid (UA) and albumin (ALB) were detected between the two groups.Meanwhile,the related information of MHD patients was selected,such as dialysis ages、dosage of Erythropoietin (EPO) and so on.The data were analyzed by multiple linear regression.Results The level of MDA in MHD patients was higher than that in control group(P ＜0.05),and the level of TAC in MHD patients was lower than that in control group(P ＜ 0.05).For MHD patients,microinflammation,use of intravenous iron,dialysis ages and Scr were positively correlated with MDA.Moreover,the standardized partial regression coefficient of microinflammation was the largest among all factors.ALB,UA and EPO were negatively correlated with MDA and positively correlated with TAC.Conclusions The oxidative stress in MHD patients are more severe than that in healthy people.Microinflammation is the most influential factor on oxidative stress of MHD patients.
Markers of oxidative stress in patients undergoing maintenance hemodialysis:establishment and evaluation

Abstract:Objective To investigate the levels of markers of oxidative stress in patients undergoing maintenance hemodialysis (MHD) and observe how they changed in single hemodialysis session. Efforts were made to establish a series of methods to reflect the real situation of oxidative stress in MHD patients, with evaluating their clinical significance. Methods 49 MHD patients and 18 healthy people were enrolled for measuring and comparing their redox markers, namely plasma total ascorbic acid (TAA), dehydroascorbic acid (DHAA), ratio of DHAA to TAA (DHAA/TAA), advanced oxidation protein products (AOPP) and malondialdehyde (MDA) in both plasma and erythrocytes. Besides, these markers were determined during the hemodialysis for dynamic records. Correlation analyses between plasma AOPP, MDA and C reactive protein (CRP) were performed in MHD patients. Results Plasma MDA and AOPP, especially the latter, were significantly higher in MHD patients than in the healthy (6.8±2.2 μmol/L vs 3.9±0.5μmol/L, P 0.05 and 395.2±98.7 μmol/L vs 182.3±43.0 μmol/L, P 0.01). The other redox markers in patients appeared comparable to those in control group. During hemodialysis, plasma TAA gradually decreased while AOPP and MDA seemed to rise up slowly, and other parameters almost remained the same. No correlations were found between plasma AOPP, MDA and CRP. Conclusion Vitamins are prone to be disturbed by various endogenous or exogenous factors, and the normal ranges of their plasma concentrations are considerably wide, thus, plasma AOPP and MDA might be more eligible and reliable to delineate the status of oxidative stress.
Effect of Reduced Glutathione in Treatment of Non-alcoholic Fatty Liver Disease

Abstract:Objective:To assess the effect of reduced glutathione(GSH)on non-alcoholic fatty liver disease.Methods:80 patients with non-alcoholic fatty liver disease were divided into two groups randomly,the treatment group(40 patients)received GSH 1 200 mg intravenously per day for 3～4 weeks and the control group(40 patients)received vitamin C 2 000 mg intravenously per day for 3～4 weeks.The changes of liver function in the two groups were compared.Results:The levels of serum ALT, AST,γ-GT in the treatment group were all obviously reduced at the end point compared with the baseline.The levels of serum ALT in the control group was reduced,but the levels ofserum AST,r-GT were no obviously changed.The total effective rate of the treatment group and the control group were 87.5% and 30.0%,respeclively(P < 0.01). Conclusion:GSH is an effective and safe remedy for the treatment of non-alcoholic fatty liver disease.
The effect of low molecular weight heparin as an antioxidant in chronic kidney disease

Abstract:Objective To identify the anti-oxidative effect of low molecular weight heparin (LMWH) in chronic kidney disease (CKD). Method Forty CKD patients at stage 3-5 were randomly divided into two groups, glutathione (GSH) group (0.9% NaCl 250ml + Glutathione 1.8g/d, intravenous injection for 10 days) and LMWH group (Fraxiparine 0.4ml/d, subcutaneous injection for 10 days). Plasma malondialdehyde (MDA), advanced oxidation protein product (AOPP) and glutathione peroxidase activity (GSH-PX) were measured at the beginning and the end of glutathione or LMWH administration. Results Plasma AOPP decreased and plasma GSH-PX increased significantly after the treatment in both LMWH and GSH groups (P0.05). The decrease of plasma AOPP was more significant in GSH group than in LMWH group (2.86±0.82 vs. 4.89±0.66μmol/l, P0.05). After the treatment, plasma MDA did not change significantly in LMWH group, but decreased (5.09±1.44 vs. 4.61±1.30nmol/ml, P0.05) in GSH group. There were no apparent side effects in both groups. Conclusion LMWH ameliorates oxidative stress state without apparent side effects. The anti-oxidative effect of LMWH is slightly lower than that of GSH.

Protective effect of reduced glutathione on mitochondria in the kidney of diabetic rats

Abstract:Objective To explore the renoprotective effect of reduced glutathione on streptozotocin-induced diabetic rats and its possible mitochondrial mechanism in the kidneys.Methods After the diabetic rat models were induced by intraperitoneal injection of streptozotocin (STZ),the diabetic rats were randomly divided into non-treated group (DM),and reduced glutathione-treated group (DM + GSH).The normal non-diabetic rats were served as the control group (NC).Eight weeks later,the excretion of urinary albumin excrection rate (UAER),serum creatinine (SCr) and serum urea nitrogen (BUN) were detected.Morphological changes of the kidney were observed by optics microscope.The levels of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in serum and renal cortex were detected.Meanwhile,kidney mitochondrial membrane potential and mitochondrial swelling were measured between the different groups.Results Compared with the NC group,the excretion of UAER,SCr and BUN were significantly increased (all P ＜ 0.05),and pathological changes of diabetic nephropathy occured in the DM group.The levels of MDA in serum (μmol/L) and renal cortex (μmol/g) were significantly elevated (5.59±1.028 vs 2.97±0.77,4.80±0.83 vs 2.98±0.75,all P ＜ 0.01),and the activity of SOD in serum (U/ml) and renal cortex (U/mg) was significantly reduced in the DM group(89.13±22.73 vs 124.1±9.27,46.05±10.24 vs 89.89±17.62,all P ＜0.01).The kidney mitochondrial membrane potential was significantly decreased (495.79±124.71 vs 965.77±246.48,P ＜ 0.05),and mitochondrial swelling was weaken in the DM group.Compared with the DM group,the excretion of UAER,SCr and BUN were significantly decreased (all P ＜ 0.05),and above abnormal pathological changes were improved in the DM + GSH group.The levels of MDA in serum (μmoL/L) and renal cortex (μmol/g) were significantly reduced (4.15±0.59 vs 5.59±1.03,3.39±0.61 vs 4.80±0.83,all P＜0.05),and the activity of SOD in serum(U/ml) and renal cortex(U/mg) was significantly elevated in the DM + GSH group (112.92±8.93 vs 89.13±22.73,83.15±16.75 vs 46.05±10.24,all P＜0.05).The kidney mitochondrial membrane potential was significantly elevated (715.97±188.65 vs 495.79±124.71,P ＜ 0.05) and mitochondrial swelling was reinforced.Conclusion Reduced glutathione could ameliorate the function of mitochondria in the kidneys of STZ-induced diabetic rats to show nephroprotective effect.
Effect of Reduced Glutathion on the Life Span of Red Blood Cells of Patients with Uremia

Abstract:Objective To study the effect of reduced glutathione(GSH) on the life span of red blood cells of patients with anemia caused by chronic renal function failure.Methods 40 cases with anemia caused by chronic renal function failure were randomly divided into 2 groups: treatment group and control group;r-HuEPO50U/kg was given to all the patients subcutaneously,twice a week,for 16 weeks,and at the same time,GSH20mg/kg was given to the patients in treatment group;before the treatment and in the 6th, 12th week after treatment,51Cr labeled erythrocytes determination was applied to detect the red blood cell life span;2ml of blood was taken from each patient in both groups before treatment and in the 4th,8th and 12th week to detect the levels of Hb,RBC and HCT.Results: The life span of red blood cell of the patients in treatment group was longer than that in control group(p0.05);the level of Hb,RBC and HCT increased obviously(p0.01) while that of the patients in treatment group increased much significantly than that of the patients in control group(p0.05).Conclusions GSH can significantly prolong the life span of red blood cells of the patients with anemia caused by chronic renal function failure,and can greatly improve the curative effect of r-HuEPO in treating renal anemia.

Abstract:Objective To study the effect of reduced glutathione on antioxidant function in patients with chronic cor pulmonale at acute exacerbation stage.Methods 80 patients with acute exacerbation of chronic pulmonary heart disease from January 2016 to June 2017 were investigated in our hospital.According to the random number table method,were randomly divided into study group and control group.The patients in control group were given expectorant,antiasthmatic and regular oxygen therapy.The study group was treated with reduced glutathione on the basis of the treatment of the control group.Before and after the treatment,the SOD,GSH-Px,LPO,MDA and CAT level of the two groups were compared and analyzed,and the curative effect of the patients after treatment the investigation and analysis of status and prognosis.Results After treatment,the levels of SOD,GSH-Px and CAT in patients in the study group were significantly increased,the levels of LPO and MDA were significantly lower,and there was a significant difference between the two groups.The average right ventricular pressure and mean pulmonary arterial water pressure in the study group decreased significantly,and the study group was significantly lower than the control group,the two groups were statistically significant.The total effective rate of the treatment group (87.50％) was significantly higher than that of the control group (70％),and the difference between the two groups was statistically significant.The incidence of acute renal failure in the study group (7.50％) was significantly lower than that in the control group (25％),and there was no significant difference in the incidence of MODS and death between the two groups.Conclusion Reduced glutathione can significantly improve the antioxidant function in patients with chronic cor pulmonale at acute exacerbation stage,improve the curative effect and improve the prognosis of patients with chronic pulmonary heart disease,and is worthy of wide application.
[The effect of hemodialysis on some parameters of the antioxidant system in the blood of patients with chronic renal failure]

Abstract:Direct contact of the blood with the dialysis membrane during hemodialysis elicits a series of changes in blood cells. White blood cell count and total lymphocyte number are reduced, neutrocytes are stimulated and degranulate, platelet adhesiveness is enhanced. Interactions of granulocytes with the dialysis membrane stimulate the production of ROS and activate aerobic reactions triggering oxidative stress. Red blood cells form the "first line of defense" during contact of the blood with the dialysis membrane. The effect of ROS on erythrocytes appears in the form of reduced osmotic resistance of their cellular membrane and susceptibility to disintegration which results in the release of small quantities of hemoglobin and significant reduction in the lifetime of these cells. Oxidative stress caused by contact of the blood with the dialysis membrane triggers defense mechanisms in the erythrocyte that protect against oxidative damage. Reports on the effects of hemodialysis on the antioxidant system of the blood in patients with chronic renal failure are conflicting and the present work was aimed at resolving some of the discord. The following questions were addressed: (1) Does hemodialysis affect the antioxidant system of the blood in patients with chronic renal failure? (2) Are there associations between parameters of hemodialysis and of the blood's antioxidant system? (3) Is supplementation with antioxidants needed in hemodialysed patients? The experiments were done at the Department of Biochemistry and Chemistry, Pomeranian Medical University. Plasma and erythrocytes were obtained from 42 patients aged 67.7 +/- 12.3 years, treated at the Department of Nephrology, Transplantology and Internal Medicine, Pomeranian Medical University, due to chronic renal failure caused by chronic glomerulonephritis (n=16), pyelonephritis (n=12), diabetes (n=3), arterial hypertension (n=3) or other cause (gout, nephrolithiasis, n=8). The patients were assigned to two equal groups: 1--with end-stage renal failure on hemodialysis; 2--eated conservatively. Hemodialysis was done three times per week, 4 hours per session, using Fresenius dialysers with polysulfone membranes and bicarbonate buffer with glucose. The control group comprised 21 healthy volunteers. All groups were matched for age and gender. A 10 mL heparinized blood sample was collected twice from group 1 patients before and after dialysis directly from the dialysis catheter and once after an overnight fast from group 2 patients and controls. Glutathione concentration was determined in whole blood. Plasma was prepared by centrifugation and MDA, Se, Cu, Zn, and Fe levels were measured. SOD, GSH-Px, and CAT activities, MDA, Se, Cu, Zn, and Fe levels were determined in erythrocytes. Statistical methods were applied to determine significance of differences between groups and correlations between parameters studied. Conclusions: (1) Hemodialysis leads to significant changes in the antioxidant system of the blood of patients with chronic renal failure. The effect is noticeable with antioxidant enzyme activities and concentrations of nonenzymatic components of the system. (2) The activity of erythrocyte GSH-Px decreases with time of dialysis potentiating the effects of oxidative stress. (3) Reduced erythrocyte levels of Se in dialysed patients correlate with lower GSH-Px activities. Selenium supplementation may be of benefit to the antioxidative defense mechanisms.
Enhancement of Antioxidant Capability of Cancer Patients During Chemotherapy by Reduced Glutathione

Abstract:BACKGROUND & OBJECTIVE:It is unknown how administration of reduced glutathione (GSH) affects chemotherapy of cancer patients. This study was designed to investigate the effect of GSH on lipid peroxidation, and activities of antioxidant enzyme among cancer patients with chemotherapy.METHODS:Sixty-two cancer patients with chemotherapy were enrolled randomly into AB or BA group in cross-over pattern. In AB group, combination of chemotherapy and GSH was administrated first, then following chemotherapy alone was given 21 or 28 days later. In group BA, chemotherapy alone was administrated first, then the combination therapy was given. Duration of chemotherapy was 2-5 days, 21-28 days for a cycle, depended on chemotherapy strategies. GSH was given as a 15 minute intravenous infusion at the dose of 1 500 mgx(m(2)xd)(-1) for 7 days from day 1. Serum samples were collected from the patients on the day just before the chemotherapy, the 7(th) day, and the 21(st) (if 21 days per cycle of the chemotherapy) or 28(th) day of treatment. Concentration of malondialdehyde (MDA), activity of glutathione peroxidase (GSH-Px), and total superoxide dismutase (T-SOD) of serum samples were analyzed biochemically.RESULTS:(1)Administration of chemotherapy significantly increased serum MDA level on the 7(th) day compared with that before chemotherapy (mean+/-SD,6.12+/-1.94 micromol/L versus 4.63+/-1.87 micromol/L,P< 0.01). The increased serum MDA level was restored partially (5.05+/-2.07)micromol/L on the 21(th) or 28(th) day, but still higher than that before chemotherapy (P< 0.05). (2)Serum activity of T-SOD and GSH-Px decreased on the 7(th) day (P< 0.01) and restored partially on the 21(th) or 28th day, but still lower than that before chemotherapy (T-SOD, P< 0.05;GSH-Px,P< 0.01).(3)Co-treatment of GSH prevents lipid peroxidation and depletion of antioxidant enzymes by chemotherapy partially but significantly (P< 0.01). (4)Similar results were obtained in both AB group and BA group.CONCLUSION:Chemotherapy depletes antioxidant capability of cancer patients and co- treatment of GSH might prevent such depletion.
Effects of Reduced Glutathione and Glycyrrhizin on Treatment of Chronic Hepatitis B

Abstract:Objective To study the therapeutic effects of reduced glutathione and glycyrrhizin on treatment of chronic hepatitis B, as well as on the levels of serum immunoglobulins. Methods From May 2005 to January 2006, 36 patients with chronic hepatitis B were selected and treated with reduced glutathione and glycyrrhizin. Peripheral blood was taken 14 ~ 28 days after combined treatment. Hepatic function parameters, immunoglobulins and clinic response were analyzed in these patients. Results After combined treatment with reduced glutathione and glycyrrhizin, liver function parameters, e.g. ALT, AST and bilirubin were significantly decreased than those at the beginning of treatment (P<0.05). Clinical symptoms were also significantly improved. Liver biopsies were performed in 8 patients before and after combined treatment, and demonstrated a marked decreased lymphocyte infiltration in liver lobules. However, serum immunoglobulins were not significantly changed after combined treatment(P>0.05). Conclusion Combined treatment with reduced glutathione and glycyrrhizin is effective to improve clinical symptoms of patients with chronic hepatitis B, control the disease development, improve the clinical symptoms, and down-regulate inflammation in liver tissue.
Effects of calcium dobesilate on microinflammation and oxidative stress status in maintenance hemodialysis patients

Abstract:Objective To investigate the effects of calcium dobesilate on nutrition,microinflammation and oxidative stress status in maintenance hemodialysis(MHD) patients.Methods Sixty-two MHD patients were selected,who had undergone hemodialysis for at least six months before the study and were in a stable clinic status without signs of infection or disease.Two tablets of calcium dobesilate were taken three times a day to the patients for six months.Results After treatment with calcium dobesilate for six months,the average level of dry body mass and mid-arm muscle circumference(MAMC) increased(P<0.05),while the score of modified subjective global assessment(SGA) decreased markedly after treatment(P<0.01).The average serum level of albumin(40±5) g/L vs(37±5) g/L were elevated(P<0.05) and pre-albumin(292±33) mg/L vs(247±23) mg/L and hemoglobin(95±14) g/L vs(88±12) g/L(P<0.01) were elevated with six months' treatment.The average serum level of C-reactive protein(C-RP),interleukin 6(IL-6),tumor necrosis factor-α(TNF-α),malondialdehyde(MDA) and total homocysteine(tHcy) were significantly lower than those of baseline(P<0.01).Kt/V was kept stable,but protein catabolic rate(PCR) rose significantly(P<0.01).Difference was not significant between serum lipid parameters before and after the treatment(P>0.05).Conclusion Supplement of calcium dobesilate in MHD patients appear to be associated with an improvement of nutrition,microinflammation and oxidative stress status.

Abstract:Objective To observe the effect of reduced glutathione(GSH) on cardiac function of diabetic cardiomyopathy(DCM) in rats.Methods SD rats were randomly divided into normal group(C group) and diabetic cardiomyopathy model group.Diabetic cardiomyopathy rat model was established by intraperitoneal injection of streptozptocin.The successful model rats were randomly divided into diabetic cardiomyopathy model group(DCM) and GSH-intervention group(DCM+GSH group).The echocardiography,glucose,lipid,biochemical indexes and pathology examination were detected after eight weeks.Results In comparison with the C group,DCM rats'blood glucose,LDL-C,TG,TC,SBP,DBP and MBP were increased with a statistically significant difference(P < 0.05).The difference of lower HR,LVEDD,LVEF and LVFS were significant(P < 0.05).LVESD was lower but the difference was not statistically significant(P > 0.05).In comparison with the C group,the blood glucose,TC of the DCM+GSH group were increased with the significant difference(P < 0.05).HR,LVEDD,LVEF and LVFS were lower with the significant difference(P < 0.05).Other indexes had no statistical significance(P > 0.05).Compared with the DCM group,the blood glucose of the DCM+GSH group was lower,but the difference was not statistically significant(P > 0.05).TG,SBP,DBP and MBP were decreased with statistically significant differences(P < 0.05).LVEDD,LVESD,HDL-C,LDL-C,TC,CK and LDH had no significant statistical change(P > 0.05).LVEF and LVFS increased with the significant difference(P < 0.05).Conclusion The reduced glutathione can improve the cardiac function of diabetic cardiomyopathy in rats to a certain extent.
