Abstract:AIM: To observe the effects of hypoxia on the levels of nitric oxide (NO), endothelin (ET-1) and the expression of inducible nitric oxide synthase (iNOS) mRNA in human umbilical vein endothelial cells (HUVECs), and further investigate the mechanism of hypoxic pulmonary hypertension. METHODS: On the basis of the HUVECs culture model, the methods of nitrate reductase and radioimmunoassay were used to determine the changes of NO and ET-1 in the medium secreted by HUVECs, and the expression of iNOS mRNA was analyzed by semi quantitative RT-PCR after exposure to hypoxia (3% O_2) for 6, 12 or 24 h. RESULTS: The contents of NO_2-/NO_3-and ET-1 in hypoxia group in the medium was significantly higher than that in control group at different time points (P0.05). Also, iNOS mRNA expression increased significantly (P0.05). CONCLUSION: Hypoxia stimulates the release of NO and ET-1 from HUVECs, also induces iNOS-mRNA expression. The change of NO may be the result of iNOS mRNA upregulation induced by hypoxia.
[Relationship Between Expression of Endothelial Nitric Oxide Synthase and NADPH Oxidase in Lungs of Mice Exposed to Chronic Hypoxia].

Abstract:OBJECTIVE:To explore the relationship between the expression of endothelial nitric oxide synthase (eNOS) and NADPH oxidase (NOX) in the lungs of mice treated by chronic hypoxic exposure. METHODS:Thirty male wild-type (WT) C57Bl/6 mice and thirty male eNOS-knockout (KO) C57BL/6 mice were randomly divided into normoxic groups (exposed to normoxia for 7 days or 21 days), hypoxic groups (exposed to 10% oxygen for 7 days or 21 days), and treatment groups (exposed to 10% oxygen and orally administrated 10 mmol/L 4-hydroxy TEMPO in drinking water for 7 days or 21 days) (n=6 in each group). The remodeling of the small pulmonary arteries was evaluated by the percentage of media wall thickness (MT%). The weight ratio of right ventricle to left ventricle plus septum (RV/[LV+S]) was calculated to evaluate the hypertrophy of right ventricle. Real-time PCR was used to measure the mRNA expression of NOX2, NOX4, and eNOS in mouse lungs. ELISA was used to determine the concentration of reactive oxygen species (ROS) in mouse lungs. RESULTS:In WT mice and KO mice, the hypoxic groups had significantly increased pulmonary vascular remodeling and RV/[LV+S] compared with the normoxic and treatment groups (P<0.05), but there were no significant differences between the normoxic and treatment groups (P>0.05). In WT mice, the hypoxic and treatment groups had significantly lower ROS concentrations than the normoxic group (P<0.05), but there were no significant differences between the hypoxic and treatment groups (P>0.05). In WT mice, the mRNA expression of eNOS, NOX2, and NOX4 was significantly higher in the hypoxic group than in the normoxic group (P<0.05), and 4-hydroxy TEMPO reversed their over-expression. In the normoxic group, the KO mice had significantly higher NOX2 and NOX4 mRNA expression than the WT mice (P<0.05); in KO mice, the hypoxic group showed no significant changes in NOX4 mRNA expression (P>0.05), but had significantly reduced NOX2 mRNA expression (P<0.05), as compared with the normoxic group; the treatment group had reduced expression of NOX2 mRNA expression and increased NOX4 mRNA expression (P<0.05), as compared with the hypoxic group. CONCLUSIONS:eNOS plays a key role in the regulation of expression of NOX2 and NOX4 in the lungs exposed to hypoxia. It suggests that NOX and eNOS may physically interact with one another in pulmonary vascular remodeling induced by chronic hypoxia.
Expressions of NOX4 and eNOS in pulmonary vasculature in mouse model of chronic hypoxia

Abstract:Objective To investigate the pulmonary vascular endothelial nitric oxide synthase(eNOS)and NOX4 expressions in mice following chronic hypoxia.Methods C57BL/6J male mice were exposed to either normoxia or hypoxia(10±0.5% O2) for 1,3,7 days or 2 weeks.HE staining was performed to observe pulmonary vascular remodeling of chronic hypoxia in mice.Immunohistochemisty applied for detecting eNOS and α-SMA protein.The real-time PCR was used to investigate eNOS and NOX4 mRNA.Results Mice exposed to hypoxia exhibited significant pulmonary vascular remodeling characterized by thickening of the vascular wall and muscularization of previously nonmuscularized more distal small vessels.The expressions of mRNA of eNOS or NOX4 were significantly increased in the lungs in chronically hypoxic mice compared with normoxia controls.The immunohistochemical staining using an isoform-specific eNOS antibody demonstrated significant increase in eNOS expression in the lung vascular endothelium.Conclusion Both NOX4 and eNOS may play important roles in the pathogenesis of chronic hypoxia pulmonary hypertension.
The relationship of portal hemodynamics to nitric oxide and endothelin levels in patients with liver cirrhosis

Abstract:To study the relationships of portal hemodynamics to vasoactive substances - nitric oxide (NO) and endothelin (ET) in cirrhosis patients, 48 patients with liver cirrhosis (LC) and 32 normal controls (NC) were studied, including 18 patients with compensated LC and 30 patients with decompensated LC. Portal venous and splenic venous blood flow (PVBF and SVBF) were measured by color duplex doppler ultrasonography, and the blood coucentrations of ET and NO were detected. After analyzing the correlation between PVBF and SVBF to NO(ET), the results suggest that protal hy- perdynamic changes, which exist in cirrhosis patients from beginning to the end, were possibly due to the high NO level and the low sensitiving to ET, and it is useful for the clinical treatment.
Influence of Nitric Oxide and Endothelin on Levels of Liver Function and Portal Hyperdynamics in Patients with Liver Cirrhosis

Abstract:Objective:To investigate the influence of nitric oxide(NO) and endothelin(ET)levels on liver function and portal hyperdynamics in patients with liver cirrhosis.Methods:The plasma levels of NO and ET of 51 cirrhotic patients[who were divided into group A(n=19),B(n=21) and C(n=11) according to the liver function] and 31 normal controls were measured by Griess and radioimmunoassay,respectively.Color Doppler ultrasonography was used to measure the portal hypernamics(including portal venous and splenic venous blood flow) and automatic biochemistry analyzer was used to measure the liver function.Results:The NO and ET level in the plasma of the cirrhotic patients(including Child-Pugh A,B and C)was significantly higher than that of the controls(P(0.01)).The NO and ET level in the plasma of the cirrhotic patients increased with the degeneration of the liver function and the difference was obvious between different grades(P0.01).The ratio of NO/ET in the plasma of the cirrhotic patients(including Child-Pugh A,B and C)was significantly higher than that of the controls(P0.05).The difference of NO/ET levels was significant between group A and group C(P0.01),but the difference was not obvious between group B and group C(P0.05).The plasma level of portal venous and splenic venous blood flow(PVBF and SVBF) was significantly higher in the cirrhotic patients than that in the controls(P0.01).The increase was more obvious in group C.The difference was significant between group C and group A(P0.01)but there was no difference between group C and group A.The plasma of NO and ET had a positive correlation with portal venous and splenic venous blood flow(PVBF and SVBF) in the cirrhotic patients,and it had a positive correlation with PVBF and SVBF in the controls.The plasma levels of NO and ET were significantly higher in ascites patients than those in the controls(P0.01)and the no ascites patients (P0.05).Conclusions:The serum levels of NO and ET are of great value in the evaluation of the liver function and the plasma levels of NO and ET may play an important role in the pathogenesis and promotion of portal hyperdynamics in patients with cirrhosis.

Abstract:Objective: To ascertain the role of nitric oxide(NO) in hemodynamic dysfunction in patients with liver cirrhosis. Methods: The plasma levels of NO and nitric oxide synthase (NOS) of 28 cirrhotic patients and 16 normal controls were measured by Griess and colorimetric methods. Meanwhile, cardiac output (CO), cardiac index (CI) and systemic vascular resistance (R) were determined. Results: Higher plasma levels of NO and NOS activity were observed in cirrhotic patients than those in normal controls (52.34 vs 36.95 μmol/L, P 0.05; 32.69 vs 21.24 μmol·L -1 ·min -1 , P 0.05). The worse the liver function, the higher the levels of plasma NO. In Child Pugh C cirrhotic patients, increased CO, CI (9.62 v s 4.84 L/min, P 0.01; 5.36 vs 3.62 L·min -1 ·m -2 , P 0.01) and decreased R (8.61 vs 12.12 Pa·s·m -3 , P 0.01) were found when compared with controls. The levels of NO had a positive correlation with CO and CI ( r =0.672, P 0.05; r =0.704, P 0.05) and had a negative correlation with R ( r =-0.586, P 0.05). Conclusion: NO is overproduced in patients with liver cirrhosis, and it might play an important role in hemodynamic dysfunction. [

Abstract:To investigate the relationship between hypertensive left ventricular hypertrophy (LVH) and levels of endothelin (ET) and nitric oxide (NO), and to provide an experimental basis for prevention and treatment of hypertensive LVH. Fifty eight hypertensive patients and 14 healthy controls were studied. All patients were examined by echocardiography. Left ventricular mass (LVM) and left ventricular mass index (LVMI) were calculated using Devereux RB formula. Hypertensive patients were divided into a LVH (+) group (n= 21) and a LVH (-) group (n=37), and the levels of endothelin and nitric oxide in the peripheral venous blood were measured. The mean ET level was significantly higher in the LVH (+) group than in LVH (-) group (p < 0.05), but the NO level was significantly lower in the LVH (+) group. The ET/NO ratio was significantly higher in the LVH (+) group than in LVH (-) group (p< 0.01). For the stepwise multiple regression analysis, the LVMI of hypertensive patients served as a dependent variable, and age, sex, BMI, MAP, ET, NO, and ET/NO served as independent variables. Only MAP, ET, and NO were found to have significant correlation to hypertensive LVH. ET had a significant positive correlation, and NO a significant negative relation to LVMI, but ET/NO showed no correlation to hypertensive LVH. ET and NO are involved in hypertensive LVH; the independent action of ET and NO in the pathogenesis of hypertensive LVH may weaken the relation between ET/NO and hypertensive LVH.
Endogenous nitric oxide regulates heme oxygenase/carbon monoxide system in intrapulmonary arteries of hypoxic rats

Abstract:The study aimed to investigate the regulation of heme oxygenase/carbon monoxide system of pulmonary arteries by endogenous nitric oxide in hypoxic rats so as to explore the regulatory mechanism responsible for hypoxic pulmonary vascular structural remodeling. Twenty-one rats were randomly divided into three groups: normoxia group, hypoxia group and hypoxia +L-NAME group. Immunohistochemistry technique was used to assess the abundance and localization of heme oxygenase-I in pulmonary arterial smooth muscle cells. At the same time the levels of carbon monoxide in plasma and lung tissue homogenates were detected by spectrophotometer. The expression of heme oxygenase-1 by pulmonary arteries smooth muscle cells of hypoxic rats increased significantly compared to that in normoxic rats (P < 0.01). However, when L-NAME was used, expression of heme oxygenase-1 by pulmonary artery smooth muscle cells of hypoxic rats decreased significantly compared to that in rats of hypoxia group (P < 0.01). The levels of carbon monoxide in plasma and lung tissue homogenates of hypoxia rats also increased significantly in hypoxic rats as compared to those of normoxic rats (P < 0.0l). But they were markedly reduced in rats of hypoxia +L-NAME group compared to hypoxia group (P < 0.01). The results suggest that endogenous nitric oxide might regulate pulmonary circulation via carbon monoxide pathway under hypoxia.
Relationship Between Plasma Carbon Monoxide and Hyperdynamic Circulation of Cirrhotic Rats

Abstract:Objective To explore the relationship between the plasma level of carbon monoxide and the hyperdynamic circulation of cirrhotic rats. Methods S.D. rats were divided into cirrhotic ( n =10) and control ( n =10) groups. Liver cirrhosis was induced by percutaneous administration of carbon tetrachloride. Mean arterial pressure (MAP, kPa),heart rate (HR,beat/min) and portal pressure (PP,cm H 2O) were measured by indwelling catheters. Plasma CO was measured by Chalmers method, and plasma NO 2/NO 3 was determined by the reduction of cadmium column. Heme oxygenase (HO) activity in splanchnic organs from cirrhotic rats and controls was also determined. Results Typical features of cirrhosis were histologically observed in carbon tetrachloride treated animals. The cirrhotic group presented a significant increase in plasma CO [( 18.69 ± 1.86 )μmol/L vs. ( 10.27 ± 1.21 )μmol/L, P 0.01 ], plasma NO [( 43.12 ± 3.25 )μmol/L vs. ( 27.75 ± 2.72 )μmol/L, P 0.01 ], and PP [( 16.67 ± 0.63 ) vs. ( 8.82 ± 0.29 ) cm H 2O, P 0.01 ] but a significant decrease in MAP [( 15.92 ± 0.74 )kPa vs. ( 18.93 ± 0.71 )kPa, P 0.01 ] in comparison with the controls. No differences were observed regarding HR. In a single regression analysis, significant correlations were detected between plasma CO and NO ( r = 0.57 , P 0.05 ), between plasma CO and MAP ( r= -0.67 ,P 0.05 ), and between plasma NO and MAP ( r= -0.74 ,P 0.01 ). In comparison with controls, the activity of HO was significantly increased in the spleens (80% increase), intestines (100% increase), and mesenteries (330% increase) from cirrhotic rats. Conclusions Carbon monoxide may act as a new messenger to play a role in hyperdynamic circulation of cirrhotic rats.

Abstract:Objective To investigate the impact of endothelium-derived nitric oxide synthase (eNOS) on rats with hypoxic pulmonary vascular remodeling. Methods 24 rats were randomly divided into two groups: normoxic (n=12) and hypoxic (n=12). The relative medial thickness (RMT) of small and medium muscularized pulmonary arteries was determined under light microscope. Immunohistochemical technique was used to assess the abundance and localization of eNOS in pulmonary arteries. Results RMT of small and medium pulmonary arteries in rats of hypoxic group was markedly higher than that of normoxic group (P0.01). eNOS was expressed in pulmonary artery endothelial cells of both groups, more marked in normoxic group than in hypoxic group (P0.01). There was a negative correlation between RMT and eNOS expression in small and medium pulmonary arteries(r=-0.8041 and -0.8757,P0.01 respectively). Conclusion The decrease of eNOS in pulmonary arteries is involved in the hypoxic pulmonary vascular remodeling.
Nitric Oxide Regulation of Myocardial O-2 Consumption and Hep Metabolism

Abstract:NO and O(2) compete at cytochrome-c oxidase, thus potentially allowing NO to modulate mitochondrial respiration. We previously observed a decrease of myocardial phosphocreatine (PCr)/ATP during very high cardiac work states, corresponding to an increase in cytosolic free ADP. This study tested the hypothesis that NO inhibition of respiration contributes to this increase of ADP. Infusion of dobutamine + dopamine (DbDp, each 20 microg.kg(-1).min(-1) iv) to more than double myocardial oxygen consumption (MVo(2)) in open-chest dogs caused a decrease of myocardial PCr/ATP measured with (31)P NMR from 2.04 +/- 0.09 to 1.85 +/- 0.08 (P < 0.05). Inhibition of NO synthesis with N(omega)-nitro-L-arginine (L-NNA), while catecholamine infusion continued, caused PCr/ATP to increase to the control value. In a second group of animals, L-NNA administered before catecholamine stimulation (reverse intervention of the first group) increased PCr/ATP during basal conditions. In these animals L-NNA did not prevent a decrease of PCr/ATP at the high cardiac work state but, relative to MVo(2), PCr/ATP was significantly higher after L-NNA. In a third group of animals, pharmacological coronary vasodilation with carbochromen was used to prevent changes in coronary flow that might alter endothelial NO production. In these animals L-NNA again restored depressed myocardial PCr/ATP during catecholamine infusion. The finding that inhibition of NO production increased PCr/ATP suggests that during very high work states NO inhibition of mitochondrial respiration requires ADP to increase to drive oxidative phosphorylation.
Study on relationship between pulmonary hypoxia and lesion of liver function in elderly patients with chronic obstructive pulmonary disease

Abstract:Objective To study the relationship between hypoxia and lesion of liver function in elderly patients with chronic obstructive pulmonary disease(COPD).Methods The peripheral white blood cell count,liver function and arterial blood gas analysis were measured in elderly patients with COPD,and student verification of the parameters in every group were made.The association between the parameters of blood gases and liver function were analyzed by linear regression.Results These parameters of peripheral white blood cell count,liver function and arterial blood gas analysis were remarkably improved(P0.01);PaO2 was increased and PaCO2 was reduced in elderly patients with COPD after treatment(P0.01).The analysis of correlation in elderly patients with COPD suggested that pH,PaO2 had significant negative correlation with levels of ALT,AST;PaCO2 had significant positive correlation with levels of ALT,AST(P0.01).Conclusions It is remarkably harmful for liver function of COPD combined with hypoxia or retention of carbon dioxide and acid intoxication in elderly patients;Treatment of anti-infection,protection of liver function and support therapy of nutrition can control hypoxia.

The Relationship Between Endothelin,endothelin to Nitric Oxide Ratio and Myocardial Perfusion in Patients with Cardiological Syndrome X

Abstract:Objective The purpose of this study was to evaluate the relation between myocardial perfusion and endothelin(ET-1) and ET-1/NO ratio in patients with cardiological syndrome X.Methods Twenty patients with cardiological syndrome X(group A) and fifteen healthy volunteers as control(group B) were involved.Blood sample was taken at rest and maximum exercise respectively.99Tcm-MIBI myocardial scintigrams were performed in all patients.Qualitative and semi-quantitative methods were used to evaluate the characteristics of myocardial perfusion in patients with cardiological syndrome X.Results(1)ET-1 and ET-1/NO in group A increased significantly(P0.001 and P0.05) and ET-1/NO decreased significantly in group B(P0.05) at maximum exercise compared to at rest.ET-1 and ET-1/NO in group A were significantly higher than that in group B at maximum exercise(P0.05).(2)At maximum exercises,ET-1,ET-1/NO were positively correlated with myocardial perfusion defect total score(amounts of defect segments multiply defect score),(r=0.607,P0.01;r=0.892,P0.001,respectively)in group A.Conclusions(1)The significant increases of ET-1 and ET-1/NO indicate endothelial dysfunction in syndrome X.(2)The increases of ET-1 and ET-1/NO were positively correlated with myocardial ischemia in cardiological syndrome X.ET-1 and ET-1/NO were important medium of myocardial ischemia and indicate the perfusion level a certain extent.
Impact of Endogenous Nitric Oxide on the Hydrogen Sulfide/cystathionine-Γ-lyase System in Pulmonary Artery of Hypoxic Rats

Abstract:Objective To explore the impact of endogenous nitric oxide(NO) on the H 2S/cystahionine γ lyase(CSE) system in pulmonary artery of hypoxic rats.Methods Ninteen rats were randomly divided into 3 groups:hypoxic group ( n =7), hypoxic + L-NAME group ( n =6) and control group ( n =6). After 21 days, mean pulmonary artery pressure (mPAP) of each rat was evaluated and the ratio of right ventricular mass to left ventricular plus septum mass was detected. Meanwhile, the production of NO in pulmonary tissue, the plasma concentration of H 2S and the activity of CSE in pulmonary tissue were detected.Results Mean PAP and RV/LV+SP significantly increased in hypoxic rats as compared with those of normal controls ( P 0.01). Meanwhile, compared with controls, the production of NO in pulmonary tissue, the plasma concentration of H 2S and the activity of CSE in pulmonary tissue markedly decreased in hypoxic rats( P 0.01, respectively). However, mPAP and RV/LV+SP were significantly increased in hypoxic + L-NAME groups as compared with hypoxic groups( P 0.05), at the same time, the production of NO in pulmonary tissue was markedly decreased ( P 0.01 , respectively). However, the plasma concentration of H 2S and the activity of CSE in pulmonary tissue in hypoxic + L-NAME group increased significantly as compared with those of hypoxic group( P 0.05).Conclusion The endogenous nitric oxide might inhibit H 2S production in pulmonary artery of hypoxic rats.
[Hypoxia Induces Fibrosis of Corpus Cavernosum Smooth Muscle in SD Rats].

Abstract:OBJECTIVE:To explore the effect of hypoxia on the fibrosis of corpus cavernosum smooth muscle cells (CCSMC) in SD rats.METHODS:CCSMCs were cultured in vitro, identified by immunohistochemistry, and then exposed to hypoxia at the concentration of 1% O2 for 12, 24, 48 and 72 hours. Those exposed to normal oxygen concentration for the corresponding lengths of time were used as the control. The relative expressions of TGF-131, type I collagen and type DI collagen were determined by RT-PCR.RESULTS:The in vitro cultured CCSMCs grew well, and the anti-a-smooth muscle actin monoclonal antibodies were positive on immunohistochemical staining. The relative expression levels of TGF-beta1, type I collagen and type mI collagen were positively correlated with the time of hypoxia interference within 48 hours, and did not increase further with prolonged exposure.CONCLUSION:When exposed to hypoxia, the relative expressions of TGF-beta1, type I collagen and type mI collagen in the CCSMCs of SD rats increased with the length of time, and reached the peak at 48 hours. Hypoxia can cause fibrosis of CCSMCs in SD rats.
[Clinical significance of circulating hypoxia inducible factor-1α and hemeoxygenase-1 in patients with liver cirrhosis].

Abstract:OBJECTIVE:To explore the levels of circulating hypoxia inducible factor-1α (HIF-1α) and hemeoxygenase-1 (HO-1) in liver cirrhosis and to explore their diagnostic values as noninvasive methods. METHODS:The levels of circulating HIF-1α and HO-1 were quantitatively detected in 34 patients with liver cirrhosis and 10 healthy controls by ELISA from May 2012 to May 2013. The diagnostic values were analyzed by ROC curve and their correlation with clinicopathological characters were also compared. RESULTS:The serum levels of HIF-1α and HO-1 were significantly higher than those in healthy controls ((10.99 ± 0.24) vs (5.79 ± 0.84) µg/L, (63.04 ± 1.87) vs (16.35 ± 2.07) µg/L, both P < 0.01). No significant difference existed among Child-Pugh classification. The expressions of HIF-1α and HO-1 had a linear correlation between each other, and closely related with rebleeding and portal thrombosis during a one-year follow-up (all P < 0.05). The area under ROC curve of HIF-1α and HO-1 was 0.894 and 0.994 respectively. With 9.45 µg/L as a critical point for HIF-1α, the diagnosis of liver cirrhosis might be predicted with a positive predictive value of 91.2%. And HO-1 >31.86 µg/L predicted the diagnosis of liver cirrhosis with a sensitivity of 97.1% and a specificity of 100%. CONCLUSION:The serum levels of HIF-1α and HO-1 may serve as useful molecular markers for a noninvasive diagnosis of liver cirrhosis.
[Interaction Between Endogenous Nitric Oxide and Hydrogen Sulfide in Pathogenesis of Hypoxic Pulmonary Hypertension].

Abstract:OBJECTIVETo investigate the interaction between nitric (NO) / nitric oxygenase (NOS) and hydrogen sulfide (H(2)S)/ cystathionine-gamma-lyase (CSE) system in the pathogenesis of hypoxic pulmonary hypertension.METHODS25 rats were randomly divided into four groups: hypoxic group (n=7 ), hypoxic + L-NAME group (n=6 ), hypoxic + PPG group (n=6) and control group (n=6 ). After 21 days, pulmonary artery mean pressure (mPAP) of each rat was evaluated, and the plasma concentration of H(2)S and NO was measured. Meanwhile, the activities of CSE in pulmonary tissue in hypoxic, hypoxic + L-NAME and control groups were detected, respectively, and expressions of NOS in pulmonary arteries in hypoxic, hypoxic + PPG and control groups were also detected by immunohistochemistry technique.RESULTSmPAP was significantly increased in hypoxic rats as compared with normal controls. Meanwhile, compared with controls, the production of NO and H(2)S in plasma, the activity of CSE in pulmonary tissue and expression of NOS in pulmonary arteries were markedly decreased in hypoxic rats. However, mPAP was significantly increased in hyopxic + L-NAME group as compared with hypoxic groups, and at the same time, the plasma concentration of NO was markedly decreased. However, the plasma concentration of H(2)S and the activity of CSE in pulmonary tissue in hyopxic+L-NAME group were increased significantly as compared with those of hypoxic group. PPG also worsened pulmonary hypertension of hypoxic rats, however,it increased endogenous production of NO and the expression of NOS of pulmonary arteries obviously.CONCLUSIONThere is a negative feed back effect between NO/NOS system and H(2)S/CSE system in development of hypoxic pulmonary hypertension. They might interact with each other and therefore play an important regulating role in hypoxic pulmonary hypertension.

Abstract:To study endothelin-1 (ET-1) gene expression and its distribution in intrapulmonary arteries of chronically hypoxic rats. Methods: in situ hybridization studies were performed onpulmonary arteries in Wistar rats using biotinlabelled cRNA probe for ET-1. Results: Most of intrapulmonary arteries showed ET-1 mRNA positive expression (+) in endothelial cells of either one-week ortwo-week hypoxic rats, whereas only few intrapulmonary arteries did in those of control rats (P＜0.01). Furthermore, some intrapulmonary arteries expressed intense ET-1 mRNA signals (+ +) in endothelial cells. ET-1 was expressed (+) in pulmonary artery smooth muscle cells of either one-week ortwoweek hypoxic rats but no signal was observed in those of controls (P ＜ 0．01 and 0. 05,respectively). Conclusion: Chronic hypoxia alters ET-1 gene transcription mainly in the pulmonaryartery endothelial cells and then smooth muscle cells.

Abstract:Objective: To investigate the impact of nitric oxide (NO) on platelet-derived growth factor (PDGF) expression in intrapulmonary arteries of rats with hypoxic pulmonary vascular structural remodeling and to examine the possible mechanism responsible for hypoxic pulmonary hypertension. Methods: Twenty-five male Wistar rats were randomly divided into four groups: nomoxic group (n=6), hypoxic group (n=6), hypoxic + L-arginine group (hypoxic + L-Arg group) (n=7) and hypoxic + N (1) -L-nitro-arginine methyl ester group (hypoxic + L-NAME group) (n=6). Relative medial thickness (RMT) of small and median muscularized pulmonary arteries and the percentage of muscularized arteries in small pulmonary arteries were observed in lung sections of rats by using a light microscope. Immunohistochemistry technique was used to assess the abundance and localization of PDGF in intrapulmonary arteries of rats in four groups. Results: RMT of pulmonary arteries and the percentage of muscularized arteries in small pulmonary arteries were obviously increased in hypoxic rats as compared with normal controls (P<0.01). L-arginine significantly reduced RMT and the percentage of muscularized arteries of chronically hypoxic rats (P<0.01). L-NAME, however, markedly increased RMT and the percentage of muscularized arteries of chronically hypoxic rats (P<0.01). There were PDGF expressions in intrapulmonary artery endothelial cells and smooth muscle cells in four groups. PDGF expressions were augmented in intrapulmonary artery endothelial cells and smooth muscle cells in hypoxic rats as compared with normal controls (P<0.01). L-Arg significantly inhibited the PDGF expressions by intrapulmonary artery endothelial cells and smooth muscle cells in hypoxic rats. However, L-NAME markedly strengthened the PDGF expressions by intrapulmonary artery endothelial cells and smooth muscle cells. There was a positive correlation between RMT and PDGF expression of endothelial cells and smooth muscle cells of median pulmonary arteries (r=0.8636, 0.8264, P<0.01, respectively). There was also a positive correlation between RMT and PDGF expression of endothelial cells and smooth muscle cells of small pulmonary arteries (r=0.8898, 0.8688, P<0.01, respectively). Conclusion: Nitric oxide inhibited PDGF expression in intrapulmonary arteries of rats with hypoxic pulmonary vascular structural remodeling.
