Abstract:The copper-catalyzed sulfenylation of indoles with aryl iodide and sulfur powder has been investigated both experimentally and theoretically. This protocol provides a direct and facile approach to prepare 3-sulfenylindoles with moderate to excellent yields and good functional-group tolerance. The in-situ IR analysis provided evidence for that NaOAc could promote the synthesis of diphenyl disulfide by the coupling of aryl iodide and sulfur powder. According to DFT calculations, the coupling pathway involving the intermediate N-methyl-3-iodoindole is more favored than the direct coupling by the CH activation of indole. The N-methyl-3-iodoindole was identified as a crucial intermediate in the catalytic cycle.

Abstract:A palladium-catalyzed direct C-H bond sulfonylation of indoles with the insertion of sulfur dioxide is achieved through a three-component reaction of 1-(pyridin-2-yl)indoles, DABCO·(SO2)2, and aryldiazonium tetrafluoroborates under mild conditions. Diverse 2-sulfonated indoles are generated by using 10 mol % of palladium(II) bromide as the catalyst at room temperature. This synthetic approach is efficient by merging palladium catalysis and insertion of sulfur dioxide via a radical process. 2-Pyrimidinyl can be used as the directing group well as for the C-H bond sulfonylation. Additionally, the directing group can be easily removed.

Abstract:We describe here a Cu-catalyzed and 4-OH-TEMPO-mediated sequential dehydrogenation/aza-Michael addition/annulation cascade reaction for the construction of N-alkyl 2-arylindoles from facilely available saturated ketones and 2-arylethynylanilines. This reaction shows high regioselectivity and tolerates a variety of functional groups. Moreover, 3-alkyl-substituted indoles can also be achieved when using 2-alkylethynylanilines as starting materials.

Abstract:An efficient and convenient Rh(III)-catalyzed double aryl C(sp2)-H bond and N-H activation and annulation reaction is reported for the synthesis of indolo[2,1-a]isoquinolines in the presence of the Cu(OAc)2 oxidant under heating conditions. Distinct from previous works with other arylamine donors, one molecule of 1,3-diarylurea can serve as a precursor of two molecules of arylamine in the reaction with diaryl-substituted alkynes.

Abstract:A novel method for the synthesis of C2-quaternary indole-3-ones via palladium-catalyzed one-pot transformation of 2-alkynyl arylazides is described. Abstract:A three‐component alkylsulfonylation/cyclization/ hydrolysis relay reaction of 1‐acryloyl‐2‐cyanoindoles with 4‐alkyl Hantzsch esters and Na2S2O5 to access a series of alkylsulfonyl pyrrolo[1,2‐a]indolediones derivatives involving in situ sulfur dioxide insertion is established. With this method, a variety of primary and secondary alkyl radicals can be used for trapping sulfur dioxide and in situ formed corresponding alkylsulfonyl radical intermediates, followed by alkyl sulfonyl radical addition, cyclization and hydrolysis. In addition, this reaction goes through once selective cleavage of σ carbon‐carbon bond and following successively formation of three new chemical bonds, in which cyano group acts as the dual roles, including the radical acceptor and carbonyl source.

Abstract:An efficient method for the synthesis of 1H-indole-3-sulfonates via palladium-catalyzed tandem reactions of 2-alkynyl arylazides with sulfonic acids has been developed. The desired products were obtained in good to excellent yields under mild reaction conditions. The reactions were shown to proceed very fast, in most cases, within 10 min.

Abstract:A four-component reaction of 2-vinylbenzoic acids, aryldiazonium tetrafluoroborates, the sulfur dioxide surrogate DABCO(SO2)(2), and nitriles under photocatalysis in the presence of visible light is developed. This multicomponent reaction works efficiently, promoted by BF3OEt2 with the insertion of sulfur dioxide under mild conditions, leading to sulfonated 1-isoindolinones in moderate to good yields. This route is highly selective, and several competitive pathways are not observed under these conditions. A possible mechanism involving photocatalysis and a Lewis acid is proposed.

Abstract:C-H bond sulfonylation of anilines with the insertion of sulfur dioxide under metal-free conditions is described. 2-Sulfonylanilines are generated in moderate to good yields through a three-component reaction of anilines, DABCO(SO2)(2), and aryldiazonium tetrafluoroborates under mild conditions. No metal catalysts or additives are needed for this transformation. This direct C-H functionalization is highly efficient, and broad functional group tolerance is observed. A radical process is believed to be involved. In the reaction process, the arylsulfonyl radical and the tertiary amine radical cation generated in situ from DABCO(SO2)(2), and aryldiazonium tetrafluoroborate are the key intermediates. Additionally, the tertiary amine radical cation acts as the electron carrier through a single electron transfer process.

Abstract:A formal (3+2) annulation reaction between isatin-derived N-tosylhydrazones and o-(trimethylsilyl)aryl triflates allows facile synthesis of spiro[indazole-3,3'-indolin]-2'-ones in high to excellent yields. Furthermore, indazolo[2,3-c]quinazolin-6(5H)-ones could be accessed from the same starting materials in one-pot fashion via an annulation/rearrangement pathway.

Abstract:A facile strategy for the synthesis of valuable indolines has been developed, involving the palladium(II)/Brønsted acid co-catalyzed annulation of readily available (2-aminophenyl) methanols and sulfoxonium ylides. The protocol allows for the direct utilization of the OH group as a leaving group, tolerates alkyl and aryl groups on the N atom of the aniline moiety, operates under mild reaction conditions, and exhibits good efficiency.

Abstract:The chiral unsymmetrical α,α-diarylmethine skeleton is widely found in natural products and synthetic pharmaceuticals. The catalytic asymmetric C-H insertion reaction of simple, unprotected indoles and pyrroles with α-aryl sulfur ylides provides a direct route to their stereoselective preparation. Nevertheless, controlling the reactivity and stereoselectivity of this reaction has presented a significant challenge. In this study, we present the highly enantioselective asymmetric formal C-H insertion reactions of N-H free indoles and pyrroles with sulfoxonium ylides, yielding chiral gem-heterodiarylmethine frameworks with good to excellent yields and enantioselectivities. Importantly, experimental mechanistic studies and DFT calculations indicate that the stereoselectivity originates from dynamic kinetic resolution, where multiple noncovalent interactions play a pivotal role, with C-C bond formation as the rate-determining step. The findings in this study may pave the way for the development of catalytic asymmetric C-H insertion reactions of sulfur ylides with other carbon nucleophiles.

Abstract:A highly efficient palladium-catalyzed domino coupling reaction of ortho-iodoaryl allene with sodium sulfonates under mild conditions is described. This novel method provides a practical protocol to access diverse indole- and benzofuran-containing sulfones by simultaneous construction of C(sp2)-C(sp2) bond and a C(sp3)-S bonds in one pot. The salient features of this transformation include simple operations, broad substrate scope, and good functional group tolerance.

Abstract:2-Amino-3-(arylthio)indoles were conveniently synthesized via the Rh(ii)-catalyzed C-S/N-C coupling reaction between 3-diazoindol-2-imines and thioesters. The products could be further oxidized to 2-amino-3-(arylsulfonyl)indoles by m-chloroperbenzoic acid and the N-sulfonyl group could be easily removed by reduction with SmI2.
