Abstract:Luteinizing hormone receptor knockout (LHRKO) mice exhibit bilateral cryptorchidism. The cryptorchid testes are located at slightly above the bladder neck indicating a defect in the inguinoscrotal phase of testicular descent. The maldescent is associated with diminished testosterone levels due to LHR inactivation, which is fully corrected by testosterone replacement therapy (TRT). Testosterone acts directly on the gubernaculum to modulate mesenchymal cell differentiation and development, which appears to be crucial in transinguinal testis descent. In order to better understand the complexity of androgen actions, we examined the global gene-expression profile of androgen responses in the gubernaculum using microarray analysis. Twelve-day old LHRKO and wild type (WT) male mice were intramuscularly injected either with 5 mg testosterone propionate (TP) in sesame oil or sesame oil alone as control. The animals were sacrificed 6 and 24 hr later and the gubernacula were harvested. The total RNA was isolated, labeled using Affymetrix IVT Express Kit and hybridized to Affymetrix Mouse Genome MOE430 2.0 array chip. Array data were normalized by the robust multichip average algorithm using the Partek Genomic Suite program to identify changes in gene level expression using a 2-way ANOVA comparison between differential expression in LHRKO and WT mice and the effects of TRT. The differentially expressed genes were selectively verified by quantitative PCR. To explore the biological relevance of the differentially expressed genes, Ingenuity Pathway Analysis was used to identify and compare genes that were altered with a false discovery rate corrected p-value <0.05 for the pathway and network analysis. The numbers of genes at 6 and 24-hr TP treatment in LHRKO mice that were upregulated (52 and 46, respectively) or down-regulated (26 and 27, respectively) were similar to those in WT mice. The key pathways that responded to TP treatment at 6 hr are primarily associated with connective tissue development and function, while at 24 hr, the skeletal and muscular system development and function pathways are influenced the most. Comparing LHRKO to WT in sesame oil treated animals, there were 19 genes upregulated and 8 genes down-regulated. The expression of these genes in LHRKO mice was completely reversed by TP treatment. Only one of the down-regulated genes, relaxin family peptide receptor 2, is known to be critical in mediating androgen signaling to stimulate transinguinal testicular descent. The association of the remaining expressed genes with testicular descent is novel and their roles remains to be elucidated. Further characterization of these genes using LHRKO mice will facilitate the identification of underlying mechanisms by which androgen controls transinguinal testicular descent.Research supported by: R01 HD057501-01 (ZML), Norton Health Trust Grant 29-10 (MEF).
