Abstract:We propose a novel parallelized fluorescence lifetime imaging microscopy (FLIM) method based on photon reassignment with a detector array and time-correlated single photon counting (TCSPC) array. The detector array composed of seven avalanche photon diodes (APDs) substitutes for original pinhole and detector to constitute an imaging scanning microscopy (ISM) system. The introduction of parallel detection can solve the pile-up effect caused by the dead-time in TCSPC measurement, which can enhance the effective photon detection efficiency and notably increase the fluorescence lifetime imaging speed. More importantly, the resolution and signal–noise ratio (SNR) can be enhanced based on photon reassignment. The simulation results embody the superiority of our FLIM system, and the experiments with different samples validate the improvement of the resolution and speed of fluorescence lifetime imaging. The experiment results show that the resolution of our method is enhanced by a factor of 1.5 and the imaging speed can be at least increased by a factor of 2.5 compared with conventional confocal TCSPC FLIM system. We envision that the proposed FLIM method based on parallel detection will allow for super-resolution fluorescence lifetime real-time imaging of live cell.

Abstract:Fluorescence lifetime microscopy has been widely used in quantifying cellular interaction or histopathological identification of different stained tissues. A novel, to the best of our knowledge, approach for high-throughput multiplexed fluorescence lifetime imaging is presented. To establish a high-throughput fluorescence lifetime acquisition system, a uniformed illumination optical focus array was generated by a novel computer-generated hologram algorithm based on matrix triple product. This, in conjunction with an array detector and multichannel time-correlated single-photon counting, enables the full use of the acquisition ability of each detector. By utilizing interval segmentation of photon time detection, a high-throughput multiplexed fluorescence lifetime imaging is achieved. Experimental results demonstrate that this method achieves a fivefold increase in the collection throughput of fluorescence lifetime and is capable of simultaneous dual-target fluorescence lifetime measurement.

Abstract:The recently proposed fluorescence emission difference (FED) microscopy has been demonstrated to be capable of breaking the diffraction barrier, which restricts the spatial resolution of far-field fluorescence microscopy. In this paper, we report a novel high-speed FED system that can realize real-time super-resolution imaging. By replacing the conventional nanopositioning stage with a galvo mirror, the temporal resolution of FED is improved to nearly one frame per second, which is 100 times faster than that of best existing FED, while the system maintains a super-high spatial resolution of 150 nm, which is far beyond the diffraction barrier. Therefore, the high-speed FED is suitable for large-area observations while avoiding photobleaching. Detailed theoretical analysis, simulations, and experimental real-time resolution tests on 100 nm nanoparticles and biological cells are reported.

Abstract:ABSTRACT Flow cytometry is widely recognized as the gold-standard technique for the analysis and enumeration of heterogeneous cellular populations and has become an indispensable tool in diagnostics, 1 rare-cell detection 2 and single-cell proteomics. 3 Although contemporary flow cytometers are able to analyse many thousands of cells per second, with classification based on scattering or fluorescence criteria, the vast majority require unacceptably large sample volumes, and do not allow the acquisition of spatial information. Herein, we report a sheathless, microfluidic imaging flow cytometer that incorporates stroboscopic illumination for blur-free fluorescence and brightfield detection at analytical throughputs in excess of 60,000 cells/s and 400,000 cells per second respectively. Our imaging platform is capable of multi-parametric fluorescence quantification and subcellular (co-)localization analysis of cellular structures down to 500 nm with microscopy image quality. We demonstrate the efficacy of our approach by performing challenging high-throughput localization analysis of cytoplasmic RNA granules in yeast and human cells. Results suggest significant utility of the imaging flow cytometer in the screening of rare events at the subcellular level for diagnostic applications.

Abstract:Single-pixel cameras based on the concepts of compressed sensing (CS) leverage the inherent structure of images to retrieve them with far fewer measurements and operate efficiently over a significantly broader spectral range than conventional silicon-based cameras. Recently, photonic time-stretch (PTS) technique facilitates the emergence of high-speed single-pixel cameras. A significant breakthrough in imaging speed of single-pixel cameras enables observation of fast dynamic phenomena. However, according to CS theory, image reconstruction is an iterative process that consumes enormous amounts of computational time and cannot be performed in real time. To address this challenge, we propose a novel single-pixel imaging technique that can produce high-quality images through rapid acquisition of their effective spatial Fourier spectrum. We employ phase-shifting sinusoidal structured illumination instead of random illumination for spectrum acquisition and apply inverse Fourier transform to the obtained spectrum for image restoration. We evaluate the performance of our prototype system by recognizing quick response (QR) codes and flow cytometric screening of cells. A frame rate of 625 kHz and a compression ratio of 10 recognition rate of the QR code. An imaging flow cytometer enabling high-content screening with an unprecedented throughput of 100,000 cells/s is also demonstrated. For real-time imaging applications, the proposed single-pixel microscope can significantly reduce the time required for image reconstruction by two orders of magnitude, which can be widely applied in industrial quality control and label-free biomedical imaging.

Abstract:Increasing demand for understanding the vast heterogeneity of cellular phenotypes has driven the development of imaging flow cytometry (IFC), that combines features of flow cytometry with fluorescence and bright field microscopy. IFC combines the throughput and statistical advantage of flow cytometry with the ability to discretely measure events based on a real or computational image, as well as conventional flow cytometry metrics. A limitation of existing IFC systems is that, regardless of detection methodology, only two-dimensional (2D) cell images are obtained. Without tomographic three-dimensional (3D) resolution the projection problem remains: collapsing 3D information onto a 2D image, limiting the reliability of spot counting or co-localization crucial to cell phenotyping. Here we present a solution to the projection problem: three-dimensional imaging flow cytometry (3D-IFC), a high-throughput 3D cell imager based on optical sectioning microscopy. We combine orthogonal light-sheet scanning illumination with our previous spatiotemporal transformation detection to produce 3D cell image reconstruction from a cameraless single-pixel photodetector readout. We further demonstrate this capability by co-capturing 3D fluorescence and label-free side-scattering images of single cells in flow at a velocity of 0.2 m s-1, corresponding to a throughput of approximately 500 cells per second with 60,000 voxels (resized subsequently to 106 voxels) for each cell image at a resolution of less than 1 micron in X, Y, and Z dimensions. Improved high-throughput imaging tools are needed to phenotype-genotype recognized heterogeneity in the fields of immunology, oncology, cell- and gene- therapy, and drug discovery.

Abstract:3D light‐sheet microscopy is a powerful tool to obtain content‐rich information of cell populations. However, the limited frame rate of high‐quantum‐efficiency cameras hinders its application in high‐throughput flow cytometry. Herein, a high‐throughput 3D‐imaging flow cytometry technique based on light‐sheet microscopy that can screen thousands of cells per second is introduced. This method enables fast, parallel optofluidic scanning of cells by performing 1D acoustofluidic focusing on multiple cells under wide‐field light‐sheet microscopy with a single objective lens. Multicolor 3D‐imaging flow cytometry with cell‐line samples at a record detection throughput of over 2000 cells s−1 is demonstrated. Furthermore, an unprecedentedly large‐scale 3D‐morphology‐based flow cytometric analysis at an order of 105 cells is demonstrated. Results show that the system is able to capture subcellular structures even at high throughputs and obtain cellular information that is overlooked under 2D‐imaging cytometry. High‐throughput 3D‐imaging flow cytometry is realized by parallel light‐sheet imaging of cells flowed with 1D acoustofluidic focusing. With a maximum detection throughput of over 2000 cells s−1, the technology enables large‐scale (>105 cells) 3D‐morphology‐based analysis for practical 3D‐imaging flow cytometry.

Abstract:Single-pixel imaging (SPI) exploits the sparsity structure of natural images and enables image acquisition with far fewer measurements than the number of pixels. However, current SPI devices suffer from low imaging speed and an extremely time-consuming image reconstruction operation. Photonic time-stretch (PTS) technique is demonstrated to enable high-speed structured-illumination pattern generation, which significantly accelerates the imaging speed of conventional SPI devices. Meanwhile, to achieve fast image reconstruction, phase-shifted sinusoidal illumination patterns are used to acquire the Fourier spectrum of images and then a simple inverse fast Fourier transform (IFFT) operation is required for image reconstruction, which is quite computationally efficient. This paper presents a high-speed real-time compressive imaging system and its applications to imaging flow cytometry. Not only is a 625-kHz frame rate demonstrated, which is three orders of magnitude higher than that of a conventional SPI device, but the reconstruction process is remarkably accelerated as well, which enables our prototype system to operate in real time.

Abstract:In this work, we developed a new method for high throughput and high content spectral imaging flow cytometry based on structured linear spot array excitation. This method leverages equally spaced laser spots for illumination, scanning of a single cell with cell movement, and the cell image is reconstructed by splitting and assembling PMT signals. To demonstrate this method, we first built an imaging flow cytometer with dual-laser and five imaging channels (Bright-field, FITC, PE, PI, APC). More specially, due to the excellent scalability of this method, for the first time, we demonstrated a high-throughput hyperspectral imaging flow cytometer by integrating a high speed 32-channel spectrometer. This system obtains 32 spectral images of 1 μm resolution at the cell flowing speed of 5 m/s with the maximum throughput up to 5000 eps.

Abstract:Optical microscopy is one of the most widely used diagnostic methods in scientific, industrial, and biomedical applications. However, while useful for detailed examination of a small number (< 10,000) of microscopic entities, conventional optical microscopy is incapable of statistically relevant screening of large populations (> 100,000,000) with high precision due to its low throughput and limited digital memory size. We present an automated flow-through single-particle optical microscope that overcomes this limitation by performing sensitive blur-free image acquisition and nonstop real-time image-recording and classification of microparticles during high-speed flow. This is made possible by integrating ultrafast optical imaging technology, self-focusing microfluidic technology, optoelectronic communication technology, and information technology. To show the system's utility, we demonstrate high-throughput image-based screening of budding yeast and rare breast cancer cells in blood with an unprecedented throughput of 100,000 particles/s and a record false positive rate of one in a million.

Abstract:Fourier ptychographic microscopy (FPM), as an emerging computational imaging method, has been applied to quantitative phase imaging with resolution bypassing the physical limit of the detection objective. Due to the weak illumination intensity and long image acquisition time, the achieved imaging speed in current FPM methods is still low, making them unsuitable for real-time imaging applications. We propose and demonstrate a high-speed FPM method based on using laser illumination and digital micro-mirror devices for illumination angle scanning. In this new, to the best of our knowledge, FPM method, we realized quantitative phase imaging and intensity imaging at over 42 frames per second (fps) with around 1 µm lateral resolution. The quantitative phase images have revealed membrane height fluctuations of red blood cells with nanometer-scale sensitivity, while the intensity images have resolved subcellular features in stained cancer tissue slices.

Abstract:In this work, we developed a new method for high throughput imaging flow cytometry, using diffractive optics elements to generate linear laser spot array for illumination, and single-pixel detectors for detection. The illumination spots are arranged in a line at equal intervals and form a small angle with the direction of the cell movement. When the cell passes through the illumination area, the two-dimensional information of the cell's fluorescence and scattered intensity profile is encoded into signals detected by the PMTs. Fluorescence and scattering imaging were experimentally demonstrated for beads and cells traveling at a velocity of 4.7 m/s in a microfluidic chip, with a resolution of 1 μm and a maximum throughput of 5000cell/s.

Abstract:Imaging flow cytometry is a powerful tool for single-cell analysis or sorting, combining high-throughput flow cytometry and high-content microscopy. However, its scalability and utility are limited by the fundamental trade-off between throughput, sensitivity, and resolution when cells are flowing quickly. To overcome these limitations, linear array spot excitation (LASE) sequentially illuminates vertical profiles of single cells at successive horizontal positions using a line of equally spaced dots. The cell images are subsequently decoded from signals detected by single-pixel photomultipliers (PMTs) to reconstruct label-free and fluorescence images of cells. A LASE imaging flow cytometer was built with two lasers and five imaging channels, achieving a theoretical maximum throughput of >5,000 cells/s and a resolution of 1.3 mm. Moreover, we conceptionally demonstrated a spectral imaging flow cytometer with 32 hyperspectral imaging channels for the first time. With its simplicity, scalability, and compatibility with conventional flow cytometers, LASE shows great promise for widespread biomedical applications.

Abstract:Image-based cellular assay advances approaches to dissect complex cellular characteristics through direct visualization of cellular functional structures. However, available technologies face a common challenge, especially when it comes to the unmet need for unraveling population heterogeneity at single-cell precision: higher imaging resolution (and thus content) comes at the expense of lower throughput, or vice versa. To overcome this challenge, a new type of imaging flow cytometer based upon an all-optical ultrafast laser-scanning imaging technique, called free-space angular-chirp-enhanced delay (FACED) is reported. It enables an imaging throughput (>20 000 cells s-1 ) 1 to 2 orders of magnitude higher than the camera-based imaging flow cytometers. It also has 2 critical advantages over optical time-stretch imaging flow cytometry, which achieves a similar throughput: (1) it is widely compatible to the repertoire of biochemical contrast agents, favoring biomolecular-specific cellular assay and (2) it enables high-throughput visualization of functional morphology of individual cells with subcellular resolution. These capabilities enable multiparametric single-cell image analysis which reveals cellular heterogeneity, for example, in the cell-death processes demonstrated in this work-the information generally masked in non-imaging flow cytometry. Therefore, this platform empowers not only efficient large-scale single-cell measurements, but also detailed mechanistic analysis of complex cellular processes.

Abstract:Abstract By virtue of the combined merits of flow cytometry and fluorescence microscopy, imaging flow cytometry (IFC) has become an established tool for cell analysis in diverse biomedical fields such as cancer biology, microbiology, immunology, hematology, and stem cell biology. However, the performance and utility of IFC are severely limited by the fundamental trade-off between throughput, sensitivity, and spatial resolution. Here we present an optomechanical imaging method that overcomes the trade-off by virtually freezing the motion of flowing cells on the image sensor to effectively achieve 1000 times longer exposure time for microscopy-grade fluorescence image acquisition. Consequently, it enables high-throughput IFC of single cells at >10,000 cells s −1 without sacrificing sensitivity and spatial resolution. The availability of numerous information-rich fluorescence cell images allows high-dimensional statistical analysis and accurate classification with deep learning, as evidenced by our demonstration of unique applications in hematology and microbiology.

Abstract:Flow cytometry is a vital tool in biomedical research and laboratory medicine. However, its accuracy is often compromised by undesired fluctuations in fluorescence intensity. While fluorescence lifetime imaging microscopy (FLIM) bypasses this challenge as fluorescence lifetime remains unaffected by such fluctuations, the full integration of FLIM into flow cytometry has yet to be demonstrated due to speed limitations. Here we overcome the speed limitations in FLIM, thereby enabling high-throughput FLIM flow cytometry at a high rate of over 10,000 cells per second. This is made possible by using dual intensity-modulated continuous-wave beam arrays with complementary modulation frequency pairs for fluorophore excitation and acquiring fluorescence lifetime images of rapidly flowing cells. Moreover, our FLIM system distinguishes subpopulations in male rat glioma and captures dynamic changes in the cell nucleus induced by an anti-cancer drug. FLIM flow cytometry significantly enhances cellular analysis capabilities, providing detailed insights into cellular functions, interactions, and environments.

Imaging cytometry, capable of visualizing the morphology and dynamics of the circulating cells at high spatiotemporal resolution, is highly desired. Current wide-field based image cytometers are limited in the imaging depth and provide only two-dimensional resolution. For deep tissue imaging, laser scanning two-photon fluorescence microscopy (TPM) is widely adopted. However, for applications in flow cytometry, the axial scanning speed of current TPMs is inadequate to provide high-speed cross-sectional imaging of vasculature. We have integrated an optical phase-locked ultrasound lens into a standard TPM and achieved microsecond-scale axial scanning. With a galvo scanner for transverse scanning, we achieved kHz cross-sectional frame rate. Here we report its applications for in vivo deformability cytometry and in vivo imaging flow cytometry, and demonstrate the capability of imaging dynamical morphologies of flowing cells, distinguishing cells and cellular clusters, and simultaneously quantifying different cell populations based on their fluorescent labels.
