Abstract:Determination of squamous cell carcinoma antigen (SCCA) in human serum plays an important role in diagnosis of cervical squamous cell carcinoma. In this work, Au/Ag/Au core/double shell nanoparticles (Au/Ag/Au NPs) were prepared by a simple approach and were used as novel enzyme-mimetic labels for development of a sandwich-type electrochemical immunosensor for SCCA. The nanostructure of Au/Ag/Au NPs could be well confirmed by transmission electron microscope (TEM) and UV-vis spectra. Au NPs decorated mercapto-functionalized graphene sheets (Au@SH-GS) were used as platform for immobilization of primary antibody (Ab(1)), while Au/Ag/Au NPs were employed as labels of secondary antibody (Ab(2)). Due to the excellent electrocatalytic activity of Au/Ag/Au NPs towards the reduction of hydrogen peroxide (H2O2), electrochemical amperometric responses to SCCA were achieved after the immuno-reaction. Under optimum conditions, the electrochemical immunosensor exhibited a wide linear range from 0.5 pg/mL to 40 ng/mL with a low detection limit of 0.18 pg/mL for SCCA. Abstract:Squamous cell carcinoma antigen (SCCA) is a good specific antigen for cancer diagnosis specifically for squamous cell carcinomas. In this study, 3-amine-N-[3-(N-pyrrole) propyl] imidazole bromide (APPIBr) ionic liquid was successfully synthesized and characterized by 1H NMR, HPLC-MS and FTIR. APPIBr ionic liquid is a unique functional material with a pyrrole moiety which can be polymerized by using electrochemical technique and an amine group for immobilizing biomolecules; thus, it is ideal for the fabrication of biosensors. Using chloroauric acid as precursor and N-dodecyl imidazole as functional monomer, gold nanoroots (AuNRs) were fabricated and characterized with TEM, SEM and XRD. An immunosensor was built on a glassy carbon electrode (GCE), through the steps of forming the poly(APPIBr)/AuNRs/GCE interface by electrodeposition of APPIBr, anti-SCCA immobilization, and several optimization steps to achieve a sensitive, accurate, precise, and selective anti-SCCA/poly(APPIBr)/AuNRs/GCE for the electrochemical immunosensing SCCA. It was found that poly(APPIBr)/AuNRs nanointerface can improve the sensing performance of the immunosensor. Under the optimized experimental conditions, there existed two linear regimes relating the peak current variation to the concentration of squamous cell carcinoma antigen in the range of 0.001-0.1 ng mL(-1) and 0.1-5.0 ng mL-1. The detection limit was calculated to be 0.3 pg mL(-1). The developed sensor was demonstrated its capability in quantitative analysis of squamous cell carcinoma antigen in human serum with recoveries of 97.3%, 102.4% and 107.4%.

Abstract:Recently early diagnosis of squamous cell carcinoma antigen (SCCA) as a tumor maker of various cancers has increasingly attracted a lot of attention with heightening of incidence rate of cancer. The SCCA with low concentration in human serum should be diluted before detecting. Thus, an immunoassay with high sensitivity is significant for early detecting SCCA. Therefore, a nonenzymatic sandwich-type electrochemical immunosensor herein was conducted to quantitative detection of squamous cell carcinoma antigen (SCCA). The amino functionalized cobaltosic oxide @ ceric dioxide nanocubes with core-shell morphology were prepared to combine sea-urchin like gold @ platinum nanoparticles (Co3O4@CeO2-Au@Pt), and used as labels to conjugate with secondary antibodies for signal amplification. Due to the synergetic effect, excellent electrochemical property and superior auxiliary catalytic activity of Co3O4@CeO2-Au@Pt, high electrocatalytic current responses toward the reduction of hydrogen peroxide (H2O2) were achieved. Besides, the electrodeposited gold nanoparticles (D-Au NPs) which were modified on glassy carbon electrodes (GCE) were used as antibodies carriers and sensing platforms. With the well cooperation of Co3O4@CeO2-Au@Pt and D-Au NPs, a broad linear range from 100fg/mL to 80ng/mL with a low detection limit of 33 fg/mL for detecting SCCA was achieved. In addition, the immunosensor displayed with good reproducibility, high selectivity and stability. The results are satisfactory when the proposed method has been applied to analyze human serum samples, indicating that the potential application is promising in clinical monitoring of tumor markers.
Sensitive Photoelectrochemical Immunosensor for Squamous Cell Carcinoma Antigen Based on MoSe2 Nanosheets and Hollow Gold Nanospheres

Abstract:A photoelectrochemical immunosensor was fabricated for squamous cell carcinoma antigen (SCCA) assay with high performances by using MoSe2 nanosheets and hollow gold nanospheres as photosensitive materials. MoSe2 nanosheets were simply synthesized with a hydrothermal method using 1,3-di-(3-thioacetylpropyl)-imidazolium bromide (DITAPIBr) ionic liquid as a morphological controlling reagent, selenium powder, and sodium molybdate as raw materials. Hollow gold nanospheres (HGNs) were prepared by using 1,3-di-(3-bromopropyl)-imidazolium bromide ionic liquid (DIBPIBr) as functional monomer, and NaBH4 as a reductant to interact with the HAuC1(4 )solution. Subsequently, HGNs were integrated with MoSe 2 nanosheets to provide a HGNs-MoSe(2 )nanocomposite. The photocurrent intensity of MoSe2 nanosheets was significantly enhanced by the hybridization of HGNs. Furthermore, HGNs-MoSe2 nanocomposites were coated onto a glassy carbon electrode surface to construct a platform for the immobilization of SCCA antibody (anti-SCCA), resulted in a photoelectrochemical immunosensing system for determining SCCA. Under optimized conditions, the fabricated immunosensor shows high selectivity and sensitivity towards SCCA, and exhibits a detection limit of 0.21 pg mL(-1) and a linear range from 1.0 pg mL(-1) to 50 ng mL(-1). Meanwhile, the practicability of the immunosensor was demonstrated by determining SCCA in clinical serum samples with high accuracy.

Abstract:Squamous cell carcinoma antigen (SCCA) is one of the common squamous cell carcinomas (SCC) in women, which usually works as a tumor biomarker for cervical cancer in diagnostic applications. Herein, bimetallic PtCo highly branched nanocrystals (PtCo BNCs) acted as electrode substrates to construct sandwich-typed electrochemical immunosensor for ultrasensitive detection of SCCA, by using dendritic mesoporous SiO 2 @AuPt nanoparticles (DM-SiO 2 @AuPt NPs) to adsorb electroactive thionine (Thi) as a signal label. The PtCo BNCs enlarged the loading of the primary antibody (Ab 1 ), showing effective improvement in conductivity and sensitivity. The DM-SiO 2 had abundant pores to incorporate more Thi, on which the decorated AuPt NPs created a great number of active sites to immobilize the secondary antibodies (Ab 2 ), thereby obviously amplifying the detection signals. The prepared sensor exhibited a broader linear range (0.001–120 ng mL –1 ) and a lower detection limit (0.33 pg mL –1 , S / N = 3), combined with high reproducibility, a low relative standard deviation (below 2.5%) and acceptable recovery (from 98.5 to 110.0%) even in diluted human serum samples. This research provides a substantial platform for clinical diagnosis of SCCA in practice.

Abstract:Designing and developing the electrochemical analytical equipment based on biomimetic materials and supramolecular chemistry has gained wide attention in recent years because the incorporation of molecular recognition and nanomaterials can effectively enhance the analytical performance and robustness of electrochemical/bio sensors. Herein, an ultrasensitive and robust sandwich-type electrochemical immunosensor has been fabricated for quantitative determination of squamous cell carcinoma antigen (SCCA), a well-defined biomarker of squamous cell carcinoma in human serum. The water-soluble pillar [S]arene functionalized Pd/MoS2 nanosheets are prepared to immobilize the electroactive thionine molecules through host-guest interaction, and employed as labels to capture detection antibodies for signal amplification. Due to the outstanding electrochemical properties, large specific surface area, and excellent catalytic activity of Thi@WP5-Pd/MoS2 nanocomposites, the robust current response towards the oxidation of thionine are achieved. Besides, the Au nanoparticles decorated TiO2 nanosheets are applied to primary antibodies binding via Au-N bond. Under the optimized conditions, the prepared immunosensor offers a wide linear range from 0.001 to 10 ng mL(-1) with a low detection limt of 0.14 pg mL(-1). Abstract:A simple nonenzymatic immunosensor was developed for sensitive and selective determination of squamous cell carcinoma antigen (SCC-Ag) based on bimetallic gold–silver nanoclusters (AuAg NCs) and reduced graphene oxide-tetraethylene pentamine (rGO-TEPA). rGO-TEPA was used as substrate material to immobilize antibodies since rGO-TEPA has large surface area and excellent conductivity to enhance the electrochemical response signal. AuAg NCs synthesized through simple mortar grinding route were used as catalytic labels of secondary antibodies; meanwhile, AuAg NCs could efficiently enhance the electron transfer. Under the optimal conditions, the electrochemical immunoassay exhibited a wide linear range of 0.005–20ng/mL with a low detection limit of 1.3pg/mL for SCC-Ag. The proposed sensing strategy enriches the electrochemical immunoassays and has a promising application in bioassay analysis.

Abstract:Squamous cell carcinoma antigen (SCCA) is an important biomarker in the diagnose of cancer, so developing effective methods for its detection is of great significance. In the present work, a novel sandwich-like electrochemical immunosensing assay (STEM) of SCCA was constructed by preparing gold nanoparticle/reduced graphene-oxide (Au NPs/rGO) to immobilize primary antibody (PAb) of SCCA and UIO-66-NH2 MOFs structure to immobilize second antibody (SAb) and electroactive toluidine blue (Tb) probe. In this designed STEM assay, the prepared Au NPs/rGO nanohybrid offers the advantages in large surface area and conductivity as carrier, and UIO-66-NH2 provided an ideal platform to accommodate SAb and a large number of Tb molecules as signal amplifier. In the existence of SCCA, the current peaks of Tb from the formed STEM structure increase with the increase of SCCA level. After optimizing the related control factors, a wide linear range (0.01 pg mL-1 and 15.0 ng mL-1) and very low detection limit (0.003 pg mL-1) of SCCA was achieved, it's thus expected the designed STEM strategy has important applications for the detection of SCCA.

Determination of the Cervical Cancer Biomarker SCCA Using CMK-3@UiO-66@AuNPs Composite-Modified Glassy Carbon Electrodes

Abstract:In order to detect the cervical cancer biomarker squamous cell carcinoma antigen (SCCA) in human serum, an electrochemical immunosensor was constructed. Zirconium 1,4-dicarboxybenzene MOF (UiO-66), with a high specific surface area, was coupled with gold nanoparticles (AuNPs), creating the UiO-66@AuNPs nanomaterial. This coupling increased the sensitivity of the electrochemical immunosensor by providing many biological binding sites for antibodies to bind to Au -S bonds. Then, mesoporous carbon (CMK-3) was added to the UiO66@AuNPs nanomaterial to obtain a CMK-3@UiO-66@AuNPs nanocomposite. This further increased the electron transfer proficiency of the material and enhanced the electrochemical signal. Finally, the sensitive detection of squamous cell carcinoma antigen (SCCA) was made possible by unique immunological responses between antigens and antibodies. Transmission electron microscopy (TEM) analysis of CMK-3@UiO-66@AuNPs revealed embedded surfaces and voids; cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) confirmed the successful assembly of the electrochemical immunosensor. The developed electrochemical immunosensor demonstrated good linearity (R12 = 0.999) from 0.01 pg/mL to 10,000 pg/mL, which is the concentration range of SCCA in human serum. The limit of detection was 0.0034 pg/mL (at an S/N ratio of 3). The immunosensor exhibited excellent precision, stability and selectivity, with a high rate of recovery (98.26-101.8 %). The experimental validation studies demonstrated that the electrochemical immunosensor was effective for the detection of SCCA in human serum, offering a novel method for the early diagnosis and dynamic surveillance of cervical cancer.

Abstract:Ultrathin-layered carbon intercalated MoS2 hollow nanospheres (C/MoS2) were prepared with a hydrothermal method combined with high-temperature annealing using cetyltrimethylammonium bromide (CTAB) as the morphological control agent and ammonium molybdate as the molybdenum source. The ultrathin-layered C/MoS2, possessing a direct band-gap of 1.8 eV, can promote the visible light absorption and the charge-hole separation ideal for photoelectrochemical (PEC) sensing. Then, the gold nanoparticles (AuNPs) were integrated onto the C/MoS2 surface by in-situ reduction of chloroauric acid without adding any reductant to produce AuNPs/C/MoS2 nanocomposites. Subsequently, AuNPs/C/MoS2 nanocomposites and SCCA antibody (antiSCCA) were sequentially modified on a glassy carbon electrode (GCE) surface to prepare a PEC immunosensing platform which was denoted as anti-SCCA(BSA)/AuNPs/C/MoS2/GCE. Under optimized conditions, the PEC immunosensor shows a broad linear range from 0.005 ng mL(-1) to 8 ng mL(-1) and a low detection limit of 1.8 pg mL(-1) for determining SCCA. For the assay of SCCA in clinical serum samples, the results obtained from the PEC immunosensor are comparable to the generally accepted ELISA method from a commercial immunoassay kit.

Abstract:A novel sensitive sandwich-type nonenzymatical electrochemical immunosensor was fabricated for quantitative determination of the carcino-embryonic antigen (CEA). β-cyclodextrin/multiwalled carbon nanotubes (β-CD/MWCNTs) that were used as electrochemical sensing platform improved the surface of electrode to immobilize first antibodies (Ab1) and enhanced the electrical conductivity. The synthetic silver nanoparticles-carbon nanotube/manganese dioxide (Ag NPs-MWCNTs/MnO2) was used as labels of secondary antibodies (Ab2) can improve the surface of electrode and enhance the electrochemical signal. MnO2 and Ag NPs achieve dual signal amplification to improve the sensitivity effectively. Under the optimal conditions, the electrochemical immunosensor exhibited a wide linear range of 0.0001–0.5ng·mL−1 and 0.5–10ng·mL−1 respectively with a low detection limit of 0.03pg·mL−1 for CEA. The immunosensor was also successfully applied to detect the actual sample. This novel method may find huge potential application for the detection of CEA in the clinical diagnosis.
Ionic Liquid and Spatially Confined Gold Nanoparticles Enhanced Photoelectrochemical Response of Zinc-Metal Organic Frameworks and Immunosensing Squamous Cell Carcinoma Antigen

Abstract:Metal-organic framework nanocrystal (Zn-MOF) was synthesized by using 3,3'-{(propane-1,3-diyl)bis[1-(4-carboxybenzyl)-1H-imidazol-3-ium]} hexafluorophosphate ionic liquid as the functional monomer and Zn( )(2+)as the central metal ion under hydrothermal conditions. Spatially confined gold nanoparticles (Au-NP) were prepared by in-situ reduction of chloroauric acid in the nanopores of Zn-MOF using acetic acid as the reducing agent to fabricate Au-NP@Zn-MOF nanocomposites. Au-NPs@Zn-MOF was further functionalized with 1H-imidazolium-1,3-bis(2-aminoethyl)bromide ionic liquid (IBABr) to prepare IBABr-Au@Zn-MOF nanocomposites. All abovementioned nanomaterials were thoroughly characterized by TEM, SEM, XPS, FTIR, and nitrogen-adsorption surface area analysis. IBABr-Au@Zn-MOFnanocomposites were then deposited onto a glassy carbon electrode and used as the photoactive element to fabricate a label-free photoelectrochemical (PEC) immunosensor by immobilizing anti-squamous cell carcinoma antigen (anti-SCCA). The PEC sensing principle is based on the photocurrent decline due to the blocking effect of SCCA on the electron and mass transfer after binding SCCA to anti-SCCA. The photocurrent variation related to the specific recognition of SCCA shows a linear relationship to the logarithm of SCCA concentration in the range of 5.0 pg mL(-1) to 15.0 ng mL(-1). The detection limit is as low as 2.34 pg mL(-1). Such a signal-off PEC immunosensor is highly selective, sensitive, stable, and reproducible towards SCCA detection. Its performance is comparable to enzyme-linked immunosorbent assay from the studies on clinical samples. This immunosensor is promising for the label-free determining SCCA in clinical human serum samples.

Abstract:A new magneto-controlled microfluidic device for direct electrochemical determination of squamous cell carcinoma antigen (SCC-Ag) in serum was designed by using anti-SCC antibody (SCC-Ab)-functionalized magnetic mesoporous nanogold/thionine/NiCo2O4 hybrid nanostructures as immunosensing probes (P1-Ab) and horseradish peroxidase-SCC-Ab conjugates-labeled nanogold/graphene nanosheets as signal tags (P2-Ab). In the presence of the analyte SCC-Ag, the sandwich immunocomplex was formed between the immunosensing probes and the signal tags. With the aid of an external magnet, the formed immunocomplex was attached to the microfluidic device. The assay was implemented in newborn calf serum (NBCS) containing 2.5mM H2O2 based on the labeled peroxidase on the P2-Ab toward the catalytic reduction of H2O2. Under optimal conditions, the increase in the current was proportional to the concentration of SCC-Ag from 2.5pg/mL to 15ng/mL. The detection limit (LOD) was 1.0pg/mL SCC-Ag at 3sB. The electrochemical immunoassay displayed an acceptable precision, selectivity and stability. Clinical serum specimens were assayed with the method, and the results were in acceptable agreement with those obtained from the referenced electrochemiluminescent method. Importantly, the method can be suitable for on-line use in the mass production of miniaturized lab-on-a-chip devices and open a new opportunity for protein diagnostics and biosecurity.

Abstract:In this study, a novel and ultrasensitive sandwich-type electrochemical immunosensor was designed for the quantitative detection of squamous cell carcinoma antigen (SCCA) based on the β-cyclodextrin functionalized graphene nanosheet (CD-GN) and the ternary hollow Pt/PdCu nanocube anchored on three-dimensional graphene framework (Pt/PdCu-3DGF). CD-GN exhibited high specific surface area and good dispersibility and stability in water, which were beneficial to fix captured antibodies (Ab1) through the supramolecular host-guest interaction between CD and Ab1. The abundant oxygen-containing functional groups on 3DGF provided binding sites for anchoring noble metal nanoparticles. Pt/PdCu-3DGF could capture detected antibodies via the interaction of Pd-NH2 and Pt-NH2. Furthermore, the ternary metal nanoparticles exhibited high electrocatalytic activity toward the reduction of hydrogen peroxide. Under optimal conditions, the fabricated immunosensor showed a sensitive response to SCCA with two linear ranges. The linear ranges are 0.0001–1ng/mL and 1–30ng/mL with a detection limit of 25fg/mL. Additionally, the proposed immunosensor showed good reproducibility and stability.

Abstract:A signal-on sandwich-like electrochemical immunosensor was built for determination of cytokeratin 19 fragments 21-1 (CYFRA 21-1) in non-small cell lung cancer (NSCLC) by confining electroactive dye (e.g., methylene blue, MB) as a probe for amplifying signals. Specifically, core-shell gold@rhodium dendritic nanocrystals (Au@Rh DNCs) behaved as a substrate for primary antibody and accelerate interfacial electron transfer. Besides, hollow carbon spheres (HCSs) were subsequently modified with polydopamine (PDA) and PtPd nanoparticles for sequential integration of the secondary antibody and confinement of MB as a label, termed as MB/PtPd/PDA/HCSs for clarity. The built sensors showed a broad linear range (100 fg mL-1 ~ 100 ng mL-1) for detection of CYFRA 21-1 with an ultra-low detection limit (31.72 fg mL-1, S/N = 3), coupled with satisfactory performance in human serum samples. This work can be explored for assays of other proteins and provides some constructive insights for early and accurate diagnosis of NSCLC.
SERS spectroscopy using Au-Ag nanoshuttles and hydrophobic paper-based Au nanoflower substrate for simultaneous detection of dual cervical cancer-associated serum biomarkers

Abstract:Ultrasensitive detection of specific biomarkers in clinical serum is helpful for early diagnosis of cervical cancer. In this paper, a surface-enhanced Raman scattering (SERS)-based immunoassay was developed for the simultaneous determination of squamous cell carcinoma antigen (SCCA) and osteopontin (OPN) in cervical cancer serum. Au-Ag nanoshuttles (Au-AgNSs) as SERS tags and hydrophobic filter paper-based Au nanoflowers (AuNFs) as capture substrate were constructed into a sandwich structure which served as an ultrasensitive SERS-based immunoassay platform. Finite difference time domain simulation confirmed that the electromagnetic field coupled between the AuNFs had a prominent SERS signal enhancement effect, which improved the detection sensitivity. SERS mapping showed that hexadecenyl succinic anhydride hydrophobic treatment could prevent the analyte from being quickly absorbed by the filter paper and increase the retention time to be more evenly distributed on the filter paper substrate. The immunoassay platform was verified to have good selectivity and reproducibility. With this method, the detection limits of SCCA and OPN in human serum were as low as 8.628 pg/mL and 4.388 pg/mL, respectively. Finally, in order to verify the feasibility of its clinical application, the serum samples of healthy subjects; cervical intraepithelial neoplasia I (CINI), CINII, and CINIII; and cervical cancer patients were analyzed, and the reliability of the results was confirmed by enzyme-linked immunosorbent assay experiments. The constructed SERS-based immunoassay platform could be used as a clinical tool for early screening of cancers in the future.

Abstract:In this work, a microfluidic chip using Au@SiO2 array-based highly active SERS substrates was developed for quantitative detection of squamous cell carcinoma antigen (SCCA) and carcinoembryonic antigen (CEA) associated with cervical cancer. The chip consisted of six functional units with pump-free design, enabling parallel detection of multiple samples in an automatic manner without external pumps and improving the portability. Ag nanocubes (AgNCs) were labeled with Raman reporters and coupled with antibodies (labeling) to prepare SERS tags, while the Au nanoparticle-modified SiO2 microsphere (Au@SiO2) array was conjugated with antibodies (coating) to generate the highly SERS-active capturing substrate. In the presence of target biomarkers, they were captured by SERS tags and capturing substrate, resulting in the formation of "sandwich" structures which were trapped in the detection chamber. As the immune reaction proceeded, a large number of "hot spots" were generated by the proximity of the Au@SiO2 array substrate and AgNCs, greatly amplifying SERS signals. With this chip, the limits of detection of the SCCA and CEA levels in human serum were estimated to be as low as 0.45 pg mL-1 and 0.36 pg mL-1, respectively. Furthermore, the good selectivity and reproducibility of this chip were confirmed. Finally, clinical serum samples were analyzed by this chip, and the outcomes were consistent with those of enzyme-linked immunosorbent assay (ELISA). Thus, the proposed microfluidic chip can be potentially applied for the clinical diagnosis of cervical cancer.

Abstract:Many studies confirm that the aberrant expression of Cytokeratin 19 fragment 21-1 (CYFRA21-1) is highly correlated with non-small cell lung cancer (NSCLC), especially for squamous cell carcinoma. Herein, we report a sandwich-type electrochemical immunosensor based on signal amplification strategy of multiple nanocomposites to test CYFRA21-1 selectively and sensitively. The proposed immunosensor fabricated by three-dimensional graphene (3D–G), chitosan (CS) and glutaraldehyde (GA) composite on the glass carbon electrode (GCE) with a large surface area is prepared to immobilize primary antibodies (Ab1) and provide excellent conductivity. To further amplify the electrochemical signal, the trace tag on the foundation of gold nanoparticles (AuNPs) is coated with amino-functionalized carbon nanotube (MWCNT-NH2) nanocomposite through thionine linking, which provides more amino groups to capture more horseradish peroxidase-labeled antibodies (HPR-Ab2) and enhances the conductivity. Under optimal conditions, the developed immunosensor exhibits excellent analytical performance for the determination of CYFRA21-1 with a wide linear range from 0.1 to 150ng·mL−1 and a low detection limit (LOD) of 43pg·mL−1. Furthermore, satisfactory results are obtained for the determination of CYFRA21-1 in real clinical serum samples, indicating the potential of the immunoassay to be applied in clinical analysis.

Abstract:Herein, a novel and sensitive sandwich-type electrochemical immunosensor was fabricated for quantitative detection of carcinoembryonic antigen (CEA). In order to construct the base of the immunosensor, the gold nanoparticles (Au NPs) were immobilized on the surface of bare glassy carbon electrode (GCE) through electrochemical reduction of HAuCl4 solution. The electrodeposited gold nanoparticles (D-Au NPs) not only effective improve immobilization of primary anti-CEA antibody (Ab(1)) but also accelerate the electron transfer on the electrode interface due to the high specific surface area, good biocompatibility and superior electrical conductivity. Moreover, the amino functionalized magnetic graphene loaded gold and silver core-shell nanoparticles to adsorb nickel ion (GS-Fe3O4/Au@Ag/Ni2+), which were used as a novel label to load the secondary anti-CEA antibody (Ab(2)). The resultant nanocomposites possess high specific surface area, excellent electrochemical property, good biocompatibility and superior auxiliary catalytic activity due to the synergetic effect. The signal amplification strategy, using the synergetic effect present in Au@Ag/Fe3O4-GS/Ni2+ to improve immobilization of Ab(2) and increase the reduction ability of the nanocomposites towards H2O2, improved the sensitivity of the immunosensor. Under the optimal conditions, a linear relationship between current signals and the concentrations of CEA was obtained in the range from 0.1 pg/mL to 100 ng/mL and the detection limit of CEA was 0.0697 pg/mL (signal-to-noise ratio of 3). Furthermore, the as-proposed immunosensor showed excellent performance in detection of human serum samples. The results suggest that the proposed immunosensor will be promising in the diagnostics application for accurately quantitative detection of CEA.
