412 Searching for the Homologue of the Human Testis-Specific Gene TDRG1 (testis Developmental Related Gene 1) in Rhesus Macaque

Abstract:Testis developmental related gene 1 (TDRG1) is a human testis-specific gene that may play an important role in spermatogenesis. But its exact roles remain unclear so far. Exploration and research of TDRG1’s homologous gene in animal model will provide help to elucidate and deduce possible function of TDRG1. The homologous sequences of TDRG1 were searched in the genomes of many kinds of animals such as mice, rats, non-human primates, etc, by BLASTN (NCBI). The identified homologous gene sequence was amplified by RT-PCR. Its mRNA was sequenced and then compared with the sequence of human TDRG1 to learn their identity with BLASTN (NCBI). RT-PCR and immunohistochemistry were used to clarify the gene and protein expression pattern of TDRG1’s homologue in multiple tissues and organs. No homologous sequences of TDRG1 were found in the genomes of mice and rats as searched by BLASTN (NCBI). However, they were identified in the genomes of rhesus macaques and chimpanzees. It was verified that the homologue of TDRG1 in rhesus macaque was 666 bp, which shared the identity of 94% with TDRG1. Its transcript has an open reading frame that encodes a protein consisting of 42 amino acids with the theoretical molecular weight of 4.83 KD. It was demonstrated by RT-PCR and immunohistochemistry that TDRG1 and its homologue were only expressed in testes of humans and rhesus macaques, and their coding proteins were both expressed in cytoplasm of spermatogenic cells.
