Abstract:Spiroplasma eriocheiris, a novel pathogen of Chinese mitten crab is a wall-less and helical bacterium which can swim up to 5 μm/s without flagella. Its distinct morphology and unique swimming manner are caused by its internal structure. In this study, we identified a unique and well-defined internal structure by Triton X-100 treatment. The internal structure was composed of a rod-like tip connected by a flat ribbon which traces the shortest line in the helical cell shape from a cell pole to the other. The structural unit of the ribbon was ring-like tetramers with about 10 nm wide and 8.2 nm long. We identified 12 proteins as the components of the internal structures by MALDI-TOF including four types of MreBs, i.e. bacterial actin and fibril, a Spiroplasma -specific protein [ 1 ]. Furthermore, pili-like structures about 6 nm long were observed on the surface of the cell pole. The combination of the tip and the ribbon may act as the internal motor for the Spiroplasma swimming.
