Abstract:Indoleamine 2, 3-dioxygenase 1 (IDO1) is a rate-limiting enzyme that metabolizes an essential amino acid tryptophan (Trp) into kynurenine (Kyn), and it promotes the occurrence of immunosuppressive effects by regulating the consumption of Trp and the accumulation of Kyn in the tumor microenvironment (TME). Recent studies have shown that the main cellular components of TME interact with each other through this pathway to promote the formation of tumor immunosuppressive microenvironment. Here, we review the role of the immunosuppression mechanisms mediated by the IDO1 pathway in tumor growth. We discuss obstacles encountered in using IDO1 as a new tumor immunotherapy target, as well as the current clinical research progress.

Abstract:Abstract Background: Indoleamine 2,3-dioxygenase 1 (IDO1) degrades tryptophan (TRP) into kynurenine (KYN), driving the development, stabilization and activation of regulatory T cells, and suppressing anti-tumor T-cell responses. Patients and methods: We investigated IDO1 expression and function in 22 children with acute myeloid leukemia (AML; median age at diagnosis: 8 years, range 1-16). IDO expression was assessed with Western blotting; TRP and KYN levels were measured with RP-HPLC. Results: Leukemia blasts from 10 out of 21 evaluable AML cases up-regulated IDO1 expression and function after in vitro challenge with IFN-γ, as indicated by higher KYN levels in supernatants of stimulated (24.3 μM/L, range 10.8-37.7) compared with unstimulated AML cells (1.3 μM/L, 0.9-2.7; p=0.0022). IDO induction was abrogated by STAT3 inhibitors; furthermore, co-immuno-precipitation assays suggested the occurrence of physical interactions between IDO and STAT3 in leukemia cells. IDO-expressing AML blasts restrained Th1 cytokine production by allogeneic CD8+ T cells, an effect that was partially abrogated by D,L-1-methyl-tryptophan, an IDO inhibitor. A Kaplan-Meier estimate of the 3-year overall survival indicated a clear benefit for IDO- AML cases compared with IDO+ ones (68% versus 33%). Conclusions: Blast cells from childhood AML express functional IDO1. From a therapeutic standpoint, interference with IDO1 activity may restore anti-leukemia Th1 immune responses and improve disease outcome.

Abstract:Over expression of indoleamine 2,3-dioxygenase(IDO)is an important mechanism that is responsible for immune tolerance of tumor.The expression of IDO is known to be induced by interferon-mediated effects of the signal transducer and activator of transcription 1α(STAT1α)and interferon regulatory factor(IRF)-1.Moreover,IDO expression can also be induced by LPS in human PBMC and THP-1 cells through an interferon independent mechanism,which may be related to the p38 MAPK and NF-κB pathway.Clarifying these signaling pathways and interactions of relevant cytokines might help to improve tumor therapy.

Abstract:Indoleamine 2,3-dioxygenase (IDO) has been well defined as one of the important immunosuppressive properties for T(H)1 cell-mediated immune responses, but its function in T(H)2 dominant system is poorly understood. Recently, an appreciable number of publications suggest that the role of IDO in T(H)2 cell regulation may be different from that of T(H)1 immune responses. Here we review the evidence on the regulatory function of IDO and tryptophan metabolites in T(H)1/T(H)2 differentiation. We propose that IDO-kynurenine pathway can serve as a negative feedback loop for T(H)1 cells but it may play a distinct role in up-regulating T(H)2 dominant immune responses.

Abstract:The role of the tryptophan-catabolizing enzyme, indoleamine 2,3-dioxygenase (IDO1), in tumor escape and metastasis formation was analyzed using two pairs of Ido1+ and Ido1− murine breast cancer cell lines. Ido1 expression in 4T1 cells was knocked down by shRNA, and Ido1 expression in NT-5 cells was upregulated by stable transfection. Growth of Ido1− tumors and spontaneous metastasis formation were inhibited in immunocompetent mice. A higher level of cytotoxic T lymphocytes was generated by spleen cells from mice bearing Ido1− tumors than Ido1+ tumors. Tumor and metastatic growth was enhanced in immunodeficient mice, confirming an intensified immune response in the absence of Ido1 expression. However, Ido1+ tumors grow faster than Ido1− tumors in immunodeficient SCID/beige mice (lacking T, B, and NK cells) suggesting that some Ido1 -controlled nonimmunological mechanisms may be involved in tumor cell growth regulation. In vitro experiments demonstrated that downregulation of Ido1 in tumor cells was associated with decreased cell proliferation, increased apoptosis, and changed expression of cell cycle regulatory genes, whereas upregulation of Ido1 in the cells had the opposite effects. Taken together, our findings indicate that Ido1 expression could exert immunological and nonimmunological effects in murine breast tumor cells.

Of all originally identified articles, 50 specially addressed the stated purpose.Results Recent work has revealed IDO at high levels in tumors and in tumor-draining lymph nodes and a close relationship between IDO activity and the regulatory T cells.Conclusion Up-regulation of IDO is proven to be a mechanism of acquired tolerance in tumors, in which the closely coupled positive feedback system between IDO and regulatory T cells may be considered to play an important role. Abstract:Tumorigenesis is a complex multifactorial and multistep process in which tumors can utilize a diverse repertoire of immunosuppressive mechanisms to evade host immune attacks. The degradation of tryptophan into immunosuppressive kynurenine is considered an important immunosuppressive mechanism in the tumor microenvironment. There are three enzymes, namely, tryptophan 2,3-dioxygenase (TDO), indoleamine 2,3-dioxygenase 1 (IDO1), and indoleamine 2,3-dioxygenase 2 (IDO2), involved in the metabolism of tryptophan. IDO1 has a wider distribution and higher activity in catalyzing tryptophan than the other two; therefore, it has been studied most extensively. IDO1 is a cytosolic monomeric, heme-containing enzyme, which is now considered an authentic immune regulator and represents one of the promising drug targets for tumor immunotherapy. Collectively, this review highlights the regulation of IDO1 gene expression and the ambivalent mechanisms of IDO1 on the antitumoral immune response. Further, new therapeutic targets via the regulation of IDO1 are discussed. A comprehensive analysis of the expression and biological function of IDO1 can help us to understand the therapeutic strategies of the inhibitors targeting IDO1 in malignant tumors.

Abstract:Indoleamine 2, 3-Dioxygenase 1 (IDO1) in cancer cells plays a critical role in tumor immunosuppression. However, the precise mechanisms regulating tumoral IDO1 expression in tumor milieus remain unclear. Here, we reported that IDO1 expression in tumor cells of hepatocelluar carcinomas (HCC), displayed a discrete rather than uniform pattern. In vitro culture, human hepatoma cell lines did not constitutively express IDO1. Interestingly, co-culture with peripheral blood mononuclear cells (PBMC) significantly induced and maintained IDO1 expression in these tumor cells, predominantly through IFN-gamma. Mechanistically, we showed that IDO1 expression in tumor cells was only induced when co-cultured with both T lymphocytes and monocytes. Moreover, the cooperation between T lymphocytes and monocytes played an indispensable role on the tumoral IDO1 expression in immunocompromised mice. Taken together, our data supported the notion that IDO1 expression in tumor cells might serve as a counter-regulatory mechanism regulated by immune system, and provided new insights into the collaborative action of different inflammatory cells in tumor immunosuppression.
High indoleamine 2,3-dioxygenase transcript levels predict better outcome after front-line cancer immunotherapy

Abstract:Indoleamine 2,3-dioxygenase 1 (IDO1), which catabolizes tryptophan, is a potential target to unlock the immunosuppressive tumor microenvironment. Correlations between IDO1 and immune checkpoint inhibitor (ICI) efficacy remain unclear. Herein, we investigated IDO1 transcript expression across cancers and clinical outcome correlations. High IDO1 transcripts were more frequent in uterine (54.2%) and ovarian cancer (37.2%) but varied between and within malignancies. High IDO1 RNA expression was associated with high expression of PD-L1 (immune checkpoint ligand), CXCL10 (an effector T cell recruitment chemokine), and STAT1 (a component of the JAK-STAT pathway) (all multivariable p < 0.05). PIK3CA and CTCF alterations were more frequent in the high IDO1 group. High IDO1 expression was an independent predictor of progression-free survival (adjusted HR = 0.44, 95% CI 0.20-0.99, p = 0.049) and overall survival (adjusted HR = 0.31, 95% CI 0.11-0.87, p = 0.026) after front-line ICIs. IDO1 expression warrants further exploration as a predictive biomarker for immunotherapy. Moreover, co-expressed immunoregulatory molecules merit exploration for co-targeting.

Abstract:Indoleamine 2,3-dioxygenase 1 (IDO1) is a heme enzyme involved in catalyzing the conversion of tryptophan (Trp) into kynurenine (Kyn) at the first rate-limiting step in the kynurenine pathway of L-tryptophan metabolism. It has been found to be involved in several biological functions such as aging, immune microorganism, neurodegenerative and infectious diseases, and cancer. IDO1 plays an important role in immune tolerance by depleting tryptophan in the tumor microenvironment and inhibiting the proliferation of effector T cells, which makes it an important emerging biomarker for cancer immunotherapy. Therefore, the research and development of IDO1 inhibitors are of great importance for tumor therapy. Of interest, IDO activity assays are of great value in the screening and evaluation of inhibitors. Herein, we mainly review the biological functions of IDO1, immune regulation, key signaling molecules in the response pathway, and the development of IDO1 inhibitors in clinical trials. Furthermore, this review provides a comprehensive overview and, in particular, a discussion of currently available IDO activity assays for use in the evaluation of IDO inhibitors in human blood. We believe that the IDO activity is a promising biomarker for the immune escape and laboratory evaluation of tumor immunotherapy.

Abstract:Indoleamine 2, 3-dioxygenase (IDO) is an intracellular hemeprotein enzyme which catalyses the essential amino acid tryptophan. Accumulating evidence has demonstrated that tryptophan depletion and its toxic metabolites expression in tissue microenvironment can suppress local allogeneic T cell proliferation and activation. Ever since the discovery that IDO was involved in the maintenance of fetal–maternal tolerance, numerous studies have confirmed that IDO is a potent regulator of immune cell function. Importantly, IDO+dendritic cells (DCs) might interact with regulatory T cells (Tregs) to form an immunomodulatory network to promote immune tolerance induction. Moreover, it has been reported that overexpression of IDO in transplanted organs can prolong allograft survival, suggesting a possible peripheral tolerogenic pathway with important implications in transplantation. However, the underlying mechanism for the beneficial effects of IDO in transplantation remains unclear. In this review, we attempt to summarize our current understandings about IDO as a mediator of immunity in transplantation and provide an overview of IDO as a new paradigm in transplantation.

Abstract:Indoleamine 2,3-dioxigenase 1 (IDO1) acts in pathogenic inflammatory processes and engender immune tolerance to tumor antigens. IDO1 can decrease the tryptophan and produce a series of toxic kynurenine metabolites to promote the immune toleration via GCN2 pathway, mTOR pathway, toxic effect of kynurenine and favoring differentiation of Tregs. IDO1 can be induced in most human cells, especially APCs and cancer cells through canonical and non-canonical NF-κB and Jak/STAT pathways, as well as PKC and TGF-β signaling pathways. A series of human cancers over-express IDO1 in a constitutive way. Thus, IDO1 is likely to be an attractive target for developing inhibitors of tumor treatments. Many preclinical and clinical trials have been underway and suggest that IDO1 inhibitor maybe an effective tool against a wide range of cancers. However, the IDO1 inhibitor alone had been verified that to be disappointment in achieving effective antitumor efficacy. Concentrating on its molecular mechanism in immune toleration and complex environments of cancer, IDO1 inhibitor could cooperate with chemotherapies and other immune target inhibitors to lessen the tumor.

Abstract:Abstract Recently, anti-programmed death (anti-PD) therapy has been at the forefront of cancer therapy. However, the clinical efficacy of anti-PD therapy in DLBCL remains obscure. The marginal response to anti-PD therapy in DLBCL indicates that different mechanisms of immune evasion other than the PD-1/PD-L1 pathway may be present. To investigate the mechanisms of resistance to anti-PD therapy in DLBCL, we assessed gene expression in patients with DLBCL in public database. Accordingly, we found indoleamine 2,3-dioxygenase 1 (IDO1), a key enzyme responsible for tryptophan catabolism, is highly expressed in DLBCL. IDO1 expression is upregulated by IFN-gamma produced by T cells in tumor microenvironments (TMEs) of various cancer types. Upregulated IDO1 leads to the production of kynurenine, which creates an immunosuppressive TMEs. However, it remains unknown whether IDO1 induces immune evasion in DLBCL and if so, which subsets of IDO1 expressing cells are responsible for immune evasion in DLBCL. To address these questions, we first determined the intrinsic role of IDO1 in lymphoma cells. We knocked down IDO1 expression (IDO1 KD) in a human DLBCL RC-K8 cell line using short hairpin RNA. We observed decreased cell proliferation in IDO KD lymphoma cells compared with control cells. We also found increased susceptibility to apoptosis in IDO1 KD RC-K8 cells compared with IDO1 WT control, when these were co-cultured with THP-1-derived macrophages or activated Jurkat T lymphocytes. These data suggest that lymphoma IDO1 has an intrinsic role of lymphoma progression and makes lymphoma sensitive to cytotoxic immune cells. To validate the anti-tumor effect of IDO1 deficiency in immune cells, we assessed EL4 lymphoma growth and immune TME of lymphoma in syngeneic wild type (WT) and Ido1 knock-out (KO) mice. The growth of EL4 lymphoma tumor was significantly suppressed in Ido1 KO mice compared to WT mice (Ido1 KO: 621.9mm3 vs WT: 1228mm3, N=22, P<0.001). Immune TME assessment revealed a decreased frequency of regulatory T cells, yet, the increased proportions of B cells, MHC II+ antigen-presenting cells, monocytes, and NK cells in Ido1 KO tumors compared with WT control. In summary, IDO1 is highly expressed in DLBCL. IDO1 up-regulation may drive lymphoma progression both intrinsically and extrinsically (immune evasion). This data suggests that IDO1 overexpression may be one of the mechanisms of resistance to anti-PD therapy. We will extend our studies to determine a combination effect of IDO1 inhibitors with immune checkpoint inhibitors including anti-PD therapy in DLBCL. Up-regulated Expression of Indoleamine 2,3-Dioxygenase 1 in Non-Hodgkin Lymphoma Correlates with Increased Regulatory T-cell Infiltration

Abstract:Indoleamine 2,3-dioxygenase 1 (IDO1), which is a key enzyme in tryptophan metabolism expressed in some subsets of normal and neoplastic cells, participates in tumor-induced tolerance. However, the mechanisms involved are not clearly understood. A hypothesis suggests that IDO1 may be involved in proliferation and conversion of regulatory T cells (Tregs). In this study, we evaluated the levels of IDO1 and forkhead box P3 (FoxP3) in non-Hodgkin lymphoma (NHL) tissues and performed ex vivo experiments to investigate the role of IDO1 on T-cell tolerance in NHL. The results showed that expressions of IDO1 mRNA and protein were coincidentally higher in NHL tissues than in reactive hyperplasia of lymph node tissues. Up-regulation of IDO1 was correlated with later clinical phases, larger tumors and higher serum lactate dehydrogenase (LDH), and indicated a worse prognosis. FoxP3 mRNA and protein levels were markedly increased alongside elevated IDO1 levels. Co-culture of murine CD4 + CD25- T cells with A20 cells could initiate the conversion of CD4 + CD25+ T cells, which showed a suppressive function in the mixed lymphocyte reaction. Moreover, the potent inhibitor of IDO1, 1-methyl-l-tryptophan, attenuated the conversion of CD4 + CD25- T cells into CD4 + CD25+ FoxP3 + T cells. The results suggested that up-regulation of IDO1 in NHL tissues could induce local immune tolerance by favoring development and infiltration of FoxP3 + Tregs through the conversion of CD4 + CD25- T cells into CD4 + CD25+ FoxP3 + T cells in the tumor microenvironment. This could be a novel mechanism of NHL escape from immune control.

Abstract:Indoleamine 2, 3-dioxygenase (IDO) is the first and rate limiting catabolic enzyme in the degradation pathway of the essential amino acid tryptophan. By cleaving the aromatic indole ring of tryptophan, IDO initiates the production of a variety of tryptophan degradation products called "kynurenines" that are known to exert important immuno-regulatory functions. Because tryptophan must be supplied in the diet, regulation of tryptophan catabolism may exert profound effects by activating or inhibiting metabolism and immune responses. Important for survival, the regulation of IDO biosynthesis and its activity in cells of the immune system can critically alter their responses to immunological insults, such as infection, autoimmunity and cancer. In this review, we assess how IDO-mediated catabolism of tryptophan can modulate the immune system to arrest inflammation, suppress immunity to cancer and inhibit allergy, autoimmunity and the rejection of transplanted tissues. Finally, we examine how vaccines may enhance immune suppression of autoimmunity through the upregulation of IDO biosynthesis in human dendritic cells.

Abstract:Indoleamine 2,3-dioxygenase (IDO) has been implicated in preventing the fetus from undergoing maternal T cell-mediated immune responses, yet the mechanism underlying these kinds of IDO-mediated immune responses has not been fully elucidated. Since the CD4 molecule plays a central role in the onset and regulation of antigen-specific immune responses, and T cell is sensitive in the absence of tryptophan, we hypothesize that IDO may reduce cell surface CD4 expression. To test this hypothesis, an adenoviral vector-based construct IDO-EGFP was generated and the effect of IDO-EGFP on CD4 expression was determined on recombinant adenoviral infected C8166 and MT-2 cells, by flow cytometry and/or Western blot analysis. The results revealed a significant downregulation of cell membrane CD4 in pAd-IDOEGFP infected cells when compared to that of mock-infected cells or infection with empty vector pAd-EGFP. Further experiments disclosed that either an addition of tryptophan or IDO inhibitor could partly restore CD4 expression in pAd-IDOEGFP infected C8166 cells. Our findings suggest that downregulation of CD4 by IDO might be one of the mechanisms through which IDO regulates T cell-mediated immune responses.

Abstract:Abstract Indoleamine 2,3-dioxygenase 1 (IDO1) is a heme enzyme that catalyzes the oxidation of L -tryptophan. Functionally, IDO1 has played a pivotal role in cancer immune escape via catalyzing the initial step of the kynurenine pathway, and overexpression of IDO1 is also associated with poor prognosis in various cancers. Currently, several small-molecule candidates and peptide vaccines are currently being assessed in clinical trials. Furthermore, the “proteolysis targeting chimera” (PROTAC) technology has also been successfully used in the development of IDO1 degraders, providing novel therapeutics for cancers. Herein, we review the biological functions of IDO1, structural biology and also extensively summarize medicinal chemistry strategies for the development of IDO1 inhibitors in clinical trials. The emerging PROTAC-based IDO1 degraders are also highlighted. This review may provide a comprehensive and updated overview on IDO1 inhibitors and their therapeutic potentials.

Abstract:Indoleamine 2,3-dioxygenase(IDO)is an intracellular hemeprotein enzyme which catalyses the essential amino acid tryptophan.Accumulating evidences have demonstrated that tryptophan depletion and its toxic metabolites can suppress local allogeneic T cell proliferation and lead to apoptosis.With the study of the mechanism of immune regulation of IDO goes further,it have emerged as a proming enzyme in organ transplantation and tumor treatment.We reviewed in the article the immune regulative mechanism of IDO and its effects in transplantation tolerance and tumor escape.
