Abstract:Optical trapping using plasmonic nanoapertures has proven to be an effective means for the contactless manipulation of nanometer-sized particles under low optical intensities. These particles have included polystyrene and silica nanospheres, proteins, coated quantum dots and magnetic nanoparticles. Here we employ fluorescence microscopy to directly observe the optical trapping process, tracking the position of a polystyrene nanosphere (20 nm diameter) trapped in water by a double nanohole (DNH) aperture in a gold film. We show that position distribution in the plane of the film has an elliptical shape. Comprehensive simulations are performed to gain insight into the trapping process, including of the distributions of the electric field, temperature, fluid velocity, optical force, and potential energy. These simulations are combined with stochastic Brownian diffusion to directly model the dynamics of the trapping process, that is, particle trajectories. We anticipate that the combination of direct particle tracking experiments with Brownian motion simulations will be valuable tool for the better understanding of fundamental mechanisms underlying nanostructure-based trapping. It could thus be helpful in the development of the future novel optical trapping devices.

Abstract:We demonstrate the trapping of single 20 and 40 nm polystyrene spheres at the cleaved end of a fiber optic with a double nanohole aperture in gold and without any microscope optics. An optical transmission increase of 15% indicates a trapping event for the 40 nm particle, and the jump is 2% for the 20 nm particle. This modular technique can be used to replace those used with current optical trapping setups that require complicated free space optics and frequent calibration, with one that is modular and requires no free space optics. This simple arrangement with the potential for fiber translation is of interest for future biosensor and optical nano-pipette devices.

Abstract:Optical trapping is a promising technique which involves holding and manipulating small particles in a non-destructive way. Conventional trapping methods are able to trap dielectric particles with size greater than 100 nm. Using a double-nanohole in a metal film (with sharp tips where the holes meet) has enabled us to trap dielectric particles such as quantum dots and single proteins. This has been achieved even while using low laser power. Since the refractive index of the particle is larger than the surrounding environment, the aperture appears larger when the particle enters the aperture. This allows for more light transmitted through the aperture. The change in transmission changes the light momentum, and by Newton's third law, there will be a force which will push back the particle to the equilibrium position. The change in light transmission also allows for facile detection of the trapping event. In this work, we use the double-nanohole to trap encapsulated quantum dots. Quantum dots are practically useful for several purposes including computing, biology and electronic devices. The ability to manipulate these particles with precision is critical to development of quantum dots usage in these fields. The CdS quantum dots, which are used in this work, are coated with a polymer shell, with a total size between 20 nm to 22 nm. The trapping and manipulation of quantum dots is promising for nanofabrication technologies that seek to place a quantum dot at a specific location in a plasmonic or nanophotonic structure. The next step in this research will be imaging of quantum dots using their fluorescence while trapping is occurring, so that a clear indication of trapping event will be available.

Abstract:Optical tweezers have found many applications in biology, but for reasonable intensities, conventional traps are limited to particles >100 nm in size. We use a double-nanohole in a gold film to experimentally trap individual nanospheres, including 20 nm polystyrene spheres and 12 nm silica spheres, at a well-defined trapping point. We present statistical studies on the trapping time, showing an exponential dependence on the optical power. Trapping experiments are repeated for different particles and several nanoholes with different gap dimensions. Unusually, smaller particles can be more easily trapped than larger ones with the double-nanohole. The 12 nm silica sphere has a size and a refractive index comparable to the smallest virus particles and has a spherical shape which is the worst case scenario for trapping.

Abstract:Optical trapping techniques have been of great interest and have advantages that enable the direct handling of nanoparticles. However, stable trapping of low-refractive-index nanoparticle remains challenging because the conventional two-dimensional hollow beams are only capable of trapping nanoparticle in the transverse plane. In this work, we propose a novel strategy to optically trap low-refractive-index nanoparticle in three-dimensional space with a hollow dark spherical focal spot, which is generated by 4Pi focusing of radially polarized first-order Laguerre-Gaussian beam. With the assumption that the laser power is 100 mW, the nanoparticles can be stably trapped with the maximal optical force of 0.3 pN, potential depth of 10 KBT and stiffness of 80 pN/mu m. Moreover, both the number and the position of the focal spot can be controlled by modulating the focusing condition and the gradient phase of the illumination respectively, enabling the simultaneously trapping of multiple nanoparticles with complex motion trajectory. The technique demonstrated in this work may open up new avenues for optical manipulation and their applications in various scientific fields.

Abstract:The ability to optically trap nanoscale particles in a reliable and noninvasive manner is emerging as an important capability for nanoscience. Different techniques have been introduced, including plasmonic nanostructures. Nano-optical tweezers based on plasmonics face the problem of Joule heating, however, due to high losses in metals. Here we experimentally demonstrate the optical trapping and transport of nanoparticles using a nonplasmonic approach, namely, a silicon nanoantenna. We trap polystyrene nano particles with diameters of 20 and 100 nm and use fluorescence microscopy to track their positions as a function of time. We show that multiple nanoparticles can be trapped simultaneously with a single nanoantenna. We show that the infrared trapping laser beam also produces fluorescent emission from trapped nanoparticles via two-photon excitation. We present simulations of the nanoantenna that predict enhanced optical forces with insignificant heat generation. Our work demonstrates that silicon nanoantennas enable nanoparticles to be optically trapped without deleterious thermal heating effects.

Abstract:We demonstrate an optical trapping technique that integrates the light guiding of an optical fiber with the field localization of a nanoaperture in a gold film. A key innovation of our technique is to use template-stripping for easy planar fabrication without the need for nanofabrication on the tip itself. As a proof of principle, we demonstrate the trapping of 20 nm and 30 nm polystyrene nanoparticles in solution, as observed by a jump in the transmitted laser intensity through the aperture. We use the finite difference time domain technique to simulate this intensity jump with the addition of a nanoparticle in the aperture, showing reasonable agreement with the experimental data. This simple nano-aperture optical fiber tip eliminates the need for a microscope setup while allowing for trapping nanoparticles, so it is anticipated to have applications in biology (e.g. viruses), biophysics (e.g. protein interactions), physics (e.g. quantum emitters), and chemistry (e.g. colloidal particles).

Abstract:Double-nanoholes fabricated by colloidal lithography were used for trapping single colloidal particles and single proteins. A gap separation of 60 nm between the cusps of the double-nanohole was achieved in a gold film of 70 nm thickness sputter coated onglass. The cusp separation was reduced steadily down to 10 nm by plasma etching the colloidal particles prior to sputter coating. Scanning electron microscopy was used to locate a particular double-nanohole and it was registered for later microscopy experiments. 30 nm polystyrene particles, the rubisco protein and bovine serum albumin were trapped using a laser focused through the aperture. Compared to other methods that require top-down nanofabrication, this approach is inexpensive and produces high-quality samples.

Abstract:We have used a gold nanohole array to trap single polystyrene nanoparticles, with a mean diameter of 30 nm, into separated hot spots located at connecting nanoslot regions. A high trap stiffness of approximately 0.85 fN/(nmmW) at a low incident laser intensity of ~0.51 mW/um2 at 980 nm was obtained. The experimental results were compared to the simulated trapping force and a reasonable match was achieved. This plasmonic array is useful for lab-on-a-chip applications and has particular appeal for trapping multiple nanoparticles with predefined separations or arranged in patterns in order to study interactions between them.

Abstract:We present optical trapping with a 10 nm gap resonant coaxial nanoaperture in a gold film. Large arrays of 600 resonant plasmonic coaxial nanoaperture traps are produced on a single chip via atomic layer lithography with each aperture tuned to match a 785 nm laser source. We show that these single coaxial apertures can act as efficient nanotweezers with a sharp potential well, capable of trapping 30 nm polystyrene nanoparticles and streptavidin molecules with a laser power as low as 4.7 mW. Furthermore, the resonant coaxial nanoaperture enables real-time label-free detection of the trapping events via simple transmission measurements. Our fabrication technique is scalable and reproducible, since the critical nanogap dimension is defined by atomic layer deposition. Thus our platform shows significant potential to push the limit of optical trapping technologies.

Abstract:Optical trapping of nano-objects (i.e., the nano-tweezers) has been investigated intensively. Most of those nano-tweezers, however, were focused on the trapping of a single nanoparticle, while the interactions between them were seldom considered. In this work, we propose a nano-tweezers in a slot photonic crystal cavity supporting multiple modes, where the relative positions of two trapped nanoparticles can be tuned by selective excitation of different resonant mode. Results show that both the nanoparticles are trapped at the center of the cavity when the first order mode is excited. When the incident source is tuned to the second order mode, however, these two nanoparticles push each other and are trapped stably at two separated positions. Also, the inter-distance between them can be tuned precisely by changing the relative power of the two modes. This provides a potential method to control the interactions between two nano-objects via optically tuning the separation between them, and may have applications in various related disciplinary.

Abstract:Optical trapping of gold nanoparticles is experimentally demonstrated using radially and azimuthally polarized beams. The transverse optical trapping stiffness of gold nanoparticles is measured. The radially polarized beam exhibits a higher trapping efficiency than the azimuthally polarized beam and the Gaussian beam. The transverse stiffness of particles with different diameters is measured experimentally and calculated via the discrete-dipole approximation method, and good agreement between theory and experiment is found.

Abstract:We investigate numerically the modification of the nonlinear optical properties of a nanoantenna in the trapping of nanoparticles (NPs) by using both the discrete dipole approximation method and the finite-difference time-domain technique. The nanoantenna, which is formed by two gold nanorods (GNRs) aligned end to end and separated by a small gap, can emit strong two-photon-induced luminescence (TPL) under the excitation of a femtosecond laser light which is resonant with its longitudinal surface plasmon resonance. In addition, the excited antenna can stably trap small NPs which in turn induce modifications in the emitted TPL. These two features make it a promising candidate for building highly sensitive detectors for NPs of different materials and sizes. It is demonstrated that sensors built with antennas possess higher sensitivities than those built with single GNRs and nanorod-based antennas are more sensitive than nanoprism-based antennas. In addition, it is found that the trapping probability for a second NP is significantly reduced for the antenna with a trapped NP, implying that trapping of NPs may occur sequentially. A relationship between the TPL of the system (antenna + NP) and the optical potential energy of the NP is established, enabling the extraction of the information on the optical potential energy and optical force by recording the TPL of the system. It is shown that the sequential trapping and releasing of NPs flowing in a microfluid channel can be realized by designing two different antennas arranged closely.

Abstract:Photonic crystal (PhC) cavities have been widely utilized for the optical trapping. However, it is still challenging to achieve high-efficiency optical trapping of ultrasmall nanoparticles. In this paper, we show optical trapping of a 3 nm size single nanoparticle by using an ultrahigh Q/V bow-tie-shaped PhC nanobeam cavity. For the trapping of a single polystyrene nanoparticle with the radius as small as 3 nm, a maximum trapping force of 1.2 × 105 pN/mW is theoretically obtained, which is at least one order of magnitude higher than the previous results. Furthermore, the calculated sensitivity for the cavity is around 350 nm/RIU, which provides a valid solution for monitoring the trapping process of the nanoparticles with ultrasmall size. We believe that such structure with characteristics of extreme light confinement and high trapping efficiency will be conducive to the development of multifunctional on-chip trapping devices.

Abstract:Now, Matthew Peters and colleagues at the University of Victoria, Canada, and Nottingham Trent University, United Kingdom, have demonstrated real-time image tracking (see image) of unmodified proteins down to 14 kDa ( ACS Nanosci. Au . https://doi.org/10.1021/acsnanoscienceau.3c00045 (2023)). This was achieved by interference imaging enhanced by surface plasmons, using just an optical microscope, an 850-nm laser, a gold metal film, and without the aid of fluorescence. The team also trapped diffusing proteins with a nanoaperture on the metal film and rapidly characterized the protein via power spectral density and noise amplitude analysis.

Abstract:Based on our previous investigation of optical tweezers with dark field illumination [Chin. Phys. Lett. 25(2008) 329], nanoparticles at large trap depth are better viewed in wide field and real time for a long time, but with poor forces. Here we present the mismatched tube length to compensate for spherical aberration of an oil-immersion objective in a glass-water interface in an optical tweezers system for manipulating nanoparticles. In this way, the critical power of stable trapping particles is measured at different trap depths. It is found that trap depth is enlarged for trapping nanoparticles and trapping forces are enhanced at large trap depth. According to the measurement, 70-nm particles are manipulated in three dimensions and observed clearly at large appropriate depth. This will expand applications of optical tweezers in a nanometre-scale colloidal system.
