Functional Analysis of Arabidopsis Exocyst Complex Subunit SEC8 Gene in Programmed Cell Death in Primary Root

Abstract:[Objectives] Exocyst is a vesicle tethering complex that exits in most eukaryotic organisms.In Arabidopsis thaliana,exocyst complex participates in many vital processes such as auxin polar transport,cell polar growth,cytokinesis,autophagy,etc.To understand the molecular roles of exocyst complex in detail,further exploration of its physiological functions is necessary.[Methods] In this study,an exocyst subunit SEC8 gene sporophytic mutant PRsec8ml was generated by pollen rescued of sec8m1.Trypan blue staining,DHCF-DC staining,DAPI staining,TUNEL assay and genetic strategy were used to analyze the primary root growth defects in this mutant.[Results] We found that PRsec8m1 mutant primary roots underwent programmed cell death (PCD).Blocking salicylic acid (SA)signal pathway could not rescue this phenotype.[Conclusions] Exocyst complex subunit SEC8 was essential for primary root development.Mutation in SEC8 gene led to PCD in primary root,which might be independent on SA signal pathway.
