P1‐326: Abnormal Organization of White Matter Networks in Subjective Cognitive Decline

Abstract:Subjective cognitive decline (SCD) exhibits an increased risk of progression to mild cognitive impairment (MCI) and Alzheimer’s disease (AD). More and more evidence indicates AD is a disconnection syndrome and the network architecture were disrupted in mild cognitive and dementia stage of AD . Whereas it remains unknown how it changed in SCD patients. Here, thirty-six SCD and 51 NC underwent diffusion tensor imaging and fibers were reconstructed by deterministic fiber tracking. Then networks were constructed based on brain region and fiber number. Relevant network parameters including network strength, clustering coefficient, characteristic path length, local efficiency, global efficiency and nodal efficiency were compared. Characteristic of connectivity were accessed by network based analysis (NBS). The accuracy of diagnosing were performed by receiver operating characteristic curve. And finally correlation between network properties and behavior were accessed. In SCD patients, as compared to NC, significant differences were found in strength, characteristic path length, local efficiency and global efficiency. Our result showed decreased nodal efficiency predominantly located in prefrontal cortex. Widespread changes were also found in connectivity and it presented high sensitivity and specificity in distinguishing SCD and NC. Furthermore, by partial correlation, we found the rich-club connectivity and local connectivity were positively correlated with delay memory. We demonstrated disrupted topographic organization of structural connectivity in SCD. And it helps early diagnosis and explain clinical conditions. Our results could provide new sights in the pathological mechanism of AD and indicated network a potential biomarker.
