Human Papillomavirus (HPV) E7 Oncogene Mediated Squamous Cell Malignancy of the Oropharynx and Cervix in K14E7 Fancd2-/- Mice and Also Causes Hematopoietic Cell Radiation Sensitivity and Malignant B Cell Transformation

Abstract:HPV positive squamous cell tumors are often radiocontrollable at lower doses. The mechanism is unknown. K14E7 Fancd2 -/- transgenic mice (FVB/N X Sv/129 background) express the E7 oncogene controlled by K14 promoter in oral cavity/oropharynx, esophagus, and cervix. The oral carcinogen 4-nitroquinoloine N-oxide induces oral and esophageal cancer (Park, et al. Cancer Research, 70(23): 9959, 2010). We tested the radiosensitivity of hematopoietic cells and whether the oxidative stress of long term bone marrow cultures (LTBMCs) induced E7 expression and malignant transformation, comparing Fancd2+/+ (SV129), Fancd2-/- (SV129), K14E7 transgenic (FVB/n)Fancd2+/+(Sv/129) with K14E7 Fancd2-/- mice. LTBMCs were established by flushing bone marrow into T-25 flasks from which bone marrow stromal cell lines and IL3-dependent hematopoietic cell lines were derived from nonadherent cell harvests. Clonal cell lines and whole bone marrow were analyzed by irradiation survival curves, Western analysis for E7 protein, and binding of induced E7 to nuclear p53. K14E7 Fancd2-/- fresh bone marrow and expanded Interleukin-(IL)3 dependent clonal cell lines were tested for radiosensitivity in clonogenic survival curve assay and for tumorigenicity by injecting 1 X 106into the flanks of K14E7 Fancd2+/+ mice. Tumors were examined by histopathology. K14E7 Fancd2-/- LTBMCs, like those from Sv/129 Fancd2-/- showed reduced duration of hematopoiesis compared to those from K14E7 Fancd2+/+ or Fancd2+/+ mice. Marrow stromal cell lines from SV129 Fancd2+/+, and K14E7 Fancd2+/+ were radioresistant compared to SV129 Fancd2-/- or K14E7 Fancd2-/- cell lines. Hematopoietic IL-3 dependent cell lines from K14E7 Fancd2+/+ and Sv/129 Fancd2+/+ cell lines were radiosensitive compared to those from Sv/129 Fancd2-/- mice (Do = 2.15 ± 0.4 and 1.92 ± 0.06, P = 0.6469, respectively, confirming Berhane et. al, Rad Res 182: 35, 2014. The Sv/129 Fancd2-/- IL-3 dependent cell lines were radioresistant compared (Do = 2.12 ± 0.12 and 1.92 ± 0.06, respectively, P = 0.0236). In striking contrast, K14E7 Fancd2-/- IL-3 dependent cell lines lost the radioresistance (Do=1.44 ± 0.13) compared to K14E7 Fancd2-/- (Do= 2.15 ± 0.28, respectively, P = 0.0498). Fresh whole bone marrow from K14E7 Fancd2-/- was also radiosensitive (Do 1.46+/-0.08 compared to Sv/129 Fancd2-/- or K14Fancd2+/+ bone marrow. Sv/129 Fancd2+/+ bone marrow (Do = 1.85 ± 0.06) respectively, P = 0.0223). K14E7 Fancd2-/- whole bone marrow was radiosensitive compared to K14E7 Fancd2+/+ bone marrow (Do = 1.47 ± 0.04 compared to 1.84 ± 0.11, respectively, P = 0.0386). Cells grown in IL-3 from K14E7 Fancd2-/-. Expression of the E7 oncogene in K14E7 Fancd2-/- fresh marrow and culture derived hematopoietic progenitor cell lines induces radiosensitivity, and malignant transformation. The DNA stress response defect in Fancd2-/- marrow extends the spectrum of K14E7 oncogene transformation capacity to the plasma cell lineage.
