Abstract:Highly emissive inorganic-organic nanoparticles with core-shell structures are fabricated by a one-pot, surfactant-free hybridization process. The surfactant-free sol-gel reactions of tetraphenylethene- (TPE) and silole-functionalized siloxanes followed by reactions with tetraethoxysilane afford fluorescent silica nanoparticles FSNP-1 and FSNP-2, respectively. The FSNPs are uniformly sized, surface-charged and colloidally stable. The diameters of the FSNPs are tunable in the range of 45-295 nm by changing the reaction conditions. Whereas their TPE and silole precursors are non-emissive, the FSNPs strongly emit in the visible vision, as a result of the novel aggregation-induced emission (AIE) characteristics of the TPE and silole aggregates in the hybrid nanoparticles. The FSNPs pose no toxicity to living cells and can be utilized to selectively image cytoplasm of HeLa cells.

Abstract:Aggregation generally quenches the light emissions of chromophoric molecules. In this review, we demonstrate that 1,1-disubstituted 2,3,4,5-tetraphenyl siloles and 2,5-difunctionalized siloles as well as their polymers exhibit the opposite behaviors. Instead of quenching, aggregation has greatly boosted their photoluminescence quantum yields by up to two orders of magnitude, turning them from faint fluorophores into strong emitters. Such “abnormal” phenomenon of “aggregation-induced emission (AIE)” is attributed to restricted intramolecular rotations of the peripheral phenyl rings against the central silole core, which block the nonradiative channel via the rotational energy relaxation processes and effectively populate the radiative decay of the excitons. Utilizing such a novel effect, siloles and their polymers find an array of applications as: sensors for chemicals, explosives, pH, and biomacromolecules (proteins, DNAs and RNAs), indicators for determining CMC and monitoring layer-by-layer self-assembling, biocompatible fluorogens for cell imaging, visualizing agent for DNA gel electrophoresis, biolabels for immunoassay, stimuli-responsive organic nanomaterials, magnetic fluorescent nanoparticles for potential bio-imaging and -separation, and outstanding materials for efficient OLEDs and PV cells.
Preparation and optical property of fluorescent silica nanoparticles entrapping pyridinium dyes

Abstract:Fluorescent silica nanoparticles entrapped in two different pyridinium dyes were prepared by reverse microemulsion method. The nanoparticles are mono-dispersed, spherical and the particle average size is 30 nm. Their ultraviolet and fluorescent properties in chloroform, ethanol, N, N-Dimethylformamide (DMF), dimethyl sulfoxide (DMSO) and water were investigated. It is found that the fluorescence of the nanoparticles is not influenced by solvent polarity and a concentration quenching effect is inhibited; the anti-photobleaching capability is also enhanced. After 25 h irradiation of 254 nm excited light, the nanoparticle dispersion's absorbency remains at 68% in the wavelength of 460 nm; however, for the pyridinium dye solution it is only 11% under the same conditions. It implies that these silica nanoparticles can be used as a new kind of nano-probe in highly-sensitive biological detection and biological imaging.

Abstract:A general and facile approach was developed for the synthesis of almost monodisperse fluorescent silica nanoparticles (NPs) doped with inert dyes, which are organic fluorophores that are strongly fluorescent but are hydrophobic or lack a covalent binding group. The prepared NPs were mesoporous and the dye molecules were encapsulated in the pores via hydrophobic interaction with the CTAB template. The NPs were stable and highly fluorescent in aqueous solution, and have potential applications in bioanalysis and fluorescence encoding.

Abstract:Quantum dot (QD) fluorescent spheres have captivated many scientists because of their many potential applications in biomedical research. In this work, QD nanospheres were prepared using a novel method: incorporating QDs into nano-liposomes and then synthesising a silica shell using a lipid membrane as the template. The results showed that the nanocomposites obtained were spherical in shape, and each nanosphere contained a silica shell and the cores consisted of a large amount of QDs. Ultrathin sections of the spheres showed that the thickness of the silica shell was about 50–60 nm. Because the QD cores were coated with liposome and thick silica shell, the bright field of the silica sphere suspension was close to milk white in colour, which was different from that of the red-coloured QD solution. Although the quantum yield of the silica spheres (2.27%) was lower than that of the QDs (23.52%), these nanospheres still emitted a bright fluorescence, and there was no obvious difference between the fluorescent colour of the nanosphere suspension and the QD solution.

Abstract:A number of dye-doped silica nanoparticles (DDSNs) have been developed by encapsulating various fluorophores into silica nanomatrixes. These DDSNs have shown great potential as highly sensitive fluorescent labeling agents for biosensing and bioimaging. The silica nanomatrix affects the radiative properties and reactivity of doped fluorophores and endows the DDSN surface properties of silica. Compared with molecular fluorophores, the DDSNs exhibit advantageous properties such as high fluorescence intensity, good photostability, and biological compatibility. In this chapter, methods for silica nanoparticle synthesis and approaches for doping dye molecules within the silica nanomatrixes are over-viewed. Various advantageous properties of DDSNs are discussed including reaction kinetics, solubility, photostability, quantum yield, lifetime, and toxicity.

Abstract:A method for the fabrication of an improved class of controllable fluorescence intensity, highly sensitive and photostable porphyrin doped silica nanoparticles (PDSNPs) was demonstrated. The fluorescence intensity of PDSNPs could be controlled by finely tuning self-dissociated time of the precursor of porphyrin-bridged silsesquioxane. Porphyrin was well dispersed into the silica matrix due to the covalent attachment in the system, thus entirely avoiding the porphyrin leakage and fluorescence quenching effects. The resultant PDSNPs with a narrow size-distributed region and a regular spherical structure can be attained.

Abstract:Effects of surfactant/water volume ratios and dye amounts on the properties of micelles and fluorescence silica nanoparticles were studied in microemulsions containing nonionic surfactant Triton X-100, hexanol as co-surfactant, cyclohexane as organic solvent, and metal organic dye (tris(2,2′-bipyridyl)dichlororuthenium) via fluorescence probe technique, TEM, and XPS. Fluorescence probe measurements show that the micelle microenvironment becomes stable at the surfactant/water volume ratio higher than 3.5 and the incubation time longer than 10 h. The data suggest that the silica shell, which is formed at the surfactant/water ratio of 3.5, yields an efficient protection of dye molecules against the e-beam irradiation and result in high photostability of fluorescent silica. We pioneered the localization of dye molecules on the surface of dye-doped silica and found that an increase of dye amounts, beyond a threshold, in the microemulsion cannot enhance the fluorescence intensity of dye-doped nanoparticles. These results are of significant importance for optimizing the synthesis of fluorescent nanoparticles with high photostability and low cost.

Abstract:Fluorescent silica nanoparticles (FSNPs) have been extensively investigated for various biomedical applications in recently years. However, the aggregation of organic dyes in silica nanoparticles also leads the significant fluorescence quenching owing to the aggregation caused quenching effects of organic dyes. Herein, we developed a rather facile strategy to fabricate FSNPs with desirable fluorescent properties through non-covalent incorporation of fluorophores with aggregation-induced emission (ALE) feature into silica nanoparticles, which were subsequently modified with functional polymers. The resultant FSNPs polymer nanocomposites (named as FSNPs-poly(IA-co-PEGMA)) exhibited uniform spherical morphology, high water dispersiity, and bright red fluorescence. Cytotoxicity results indicate that FSNPs-poly(IA-co-PEGMA) possess excellent biocompatibility. Cell uptake behavior suggests FSNPs-poly(IA-co-PEGMA) are of great potential for biological imaging applications. Taken together, we have reported a facile method for the fabrication of FSNPs through non-covalent encapsulation using an AIE-active dye. These FSNPs can be further functionalized with functional polymers through ring-opening reaction and the resultant FSNPs-poly(IA-co-PEGMA) showed great potential for biological imaging. More importantly, we believe that many other functional components could also be integrated into these FSNPs through the facile ring-opening reaction. Abstract:Modified fluorescent nanoparticles continue to emerge as promising candidates for drug delivery, bioimaging, and labeling tools for various biomedical applications. The ability of nanomaterials to fluorescently label cells allow for the enhanced detection and understanding of diseases. Silica nanoparticles have a variety of unique properties that can be harnessed for many different applications, causing their increased popularity. In combination with an organic dye, fluorescent nanoparticles demonstrate a vast range of advantageous properties including long photostability, surface modification, and signal amplification, thus allowing ease of manipulation to best suit bioimaging purposes. In this study, the Stöber method with tetraethyl orthosilicate (TEOS) and a fluorescent dye sulfo-Cy5-amine was used to synthesize fluorescent silica nanoparticles. The fluorescence spectra, zeta potential, quantum yield, cytotoxicity, and photostability were evaluated. The increased intracellular uptake and photostability of the dye-silica nanoparticles show their potential for bioimaging.

Strong Fluorescence Enhancement with Silica-Coated Au Nanoshell Dimers

Abstract:Fluorescence intensity is vital for fluorescence sensing and imaging because it determines the sensing sensitivity and imaging brightness. This study reports plasmon-enhanced fluorescence by engineering plasmonic nanostructures, that are SiO 2 -coated Au nanoshell dimers with a high yield exceeding 60 %. With this elaborately designed nanostructure, we show that the thin SiO 2 shell can conveniently distance the fluorophore from the underneath metal, thereby effectively avoiding fluorescence quenching. Meanwhile, the inner Au nanoshell dimers create abundant hot spots at particle-particle junctions and enable near-infrared fluorescence enhancement. The largest fluorescence enhancement achieved is 69 times for the design with a 9 nm external SiO 2 shell, as is also confirmed by three-dimensional finite-difference time-domain simulations. This dramatically increased fluorescence has great significance in fluorescence-based sensing and imaging.

Abstract:The fluorescence-doped silica nanoparticles (FDS-NP) were synthesized in a microemulsion system of polyoxyethylene nonylphenyl ether (OP-10)/ cyclohexane/ammonium hydroxide. To elucidate the character of FDS-NP, techniques of electron microscopy, analysis of spectrum and collisional quenching of fluorescence were used. In order to study the fluorescent signal and intensity of FDS-NP in the cultured cells, laser-scanning confocal microscope and the analysis of flow cytometry were utilized. The results showed that the size of FDS-NP was very small (about 20 nm), the distribution was narrow, the shape was circular and lubricity, and the fluorescence was photostable. Cultured cells could efficiently uptake FDS-NP, and yield high fluorescent signal and intensity. All these indicated that FDS-NP is an important nanoparticulate biomarker, and is applicable to many areas of modern biochemical analysis, such as ultrasensitive detection, immunoassys, as well as cellular biology.

Abstract:Aggregation-induced emission (AIE) dyes have recently attracted much attention for biomedical applications for their remarkable AIE properties. However, the hydrophobic nature of AIE dyes made them difficult to be dispersed in physiological solution and problematic for biomedical application directly. Great efforts have been made to overcome this problem, and different strategies for preparation of water dispersible AIE based nanoprobes had been explored previously. However, a facile and effective strategy is still highly desirable and of great importance for the biomedical applications of AIE dye based on nanoprobes. In this work, the fabrication of amphiphilic hyperbranched fluorescent organic nanoparticles with a core-shell structure based on an AIE dye [tetraphenylethene acrylate (TPE-O-E)] and a hyperbranched polyamino compound [polyethylene imine (PEI)] through Michael addition reaction is described for the first time. The AIE dye as well as the final product PEI-TPE-O-E was characterized in detail by a number of techniques. To test their biomedical application potential, the cell viability as well as cell imaging properties of the PEI-TPE-O-E was also examined. The results showed that the PEI-TPE-O-E organic nanoparticles presented high water dispersiblity, ultrabright fluroerescence, low cytotoxicity and excellent biocompatibility, making them promising for biological imaging and gene delivery applications.
A Facile Surface Modification Strategy for Fabrication of Fluorescent Silica Nanoparticles with the Aggregation-Induced Emission Dye Through Surface-Initiated Cationic Ring Opening Polymerization.

Abstract:Fluorescent silica nanoparticles (FSNPs) have attracted great interest for potential applications in biological and biomedical fields because they possess higher fluorescence quantum yield and better fluorescence stability as comparison with small organic fluorescent molecules. The encapsulation of covalent linkage with fluorescent organic dyes or fluorescent metal complexes has demonstrated to be the commonly adopted strategies for fabrication of FSNPs previously. However, it is still challengeable to obtain FSNPs based polymer composites with intensive fluorescence and good water dispersibility through a one-pot surface modification strategy. In this paper, we developed a facile method to fabricate novel FSNPs based polymer composites (PhE@MSNs-PEtOx) through introducing the aggregation-induced emission (AIE) dye (PhE-OH) and poly(2-ethyl-2-oxazoline) (PEtOx) onto mesoporous silica nanoparticles (MSNs) based on cationic ring opening polymerization (CROP). The resulting PhE@MSNs-PEtOx composites possess strong fluorescence emission, excellent hydrophilicity and biocompatibility. These features make the final FSNPs based polymer composites great potential for biomedical applications. Taken together, we have developed for the first time that FSNPs based polymer composites can be facilely prepared through the one-pot introduction of AIE dyes and hydrophilic PEtOx on MSNs. Moreover, the novel FSNPs based composites could also be utilized for other biomedical applications considered their properties.

Abstract:When gold nanoparticles meet strongly fluorescent dye molecules, the fluorescence of the molecules is quenched. On page 581, organo-soluble thiol monolayer-protected gold nanoparticles of different shapes and sizes are homogeneously mixed with perylene dye molecules by Q. Li and co-workers, in both solution and solid states. The nanoparticles modify the fluorescence properties of perylene depending on the particle size and shape. This demonstrates the potential to introduce gold nanoparticles into practical applications such as antifraud technologies.
One-pot Ultrafast Preparation of Silica Quantum Dots and Their Utilization for Fabrication of Luminescent Mesoporous Silica Nanoparticles.

Abstract:Silica quantum dots (SiQDs) and their luminescent composites have displayed great potential for biomedical applications owing to their chemical inert and low cost. In this work, we report a facile, cost-effective and ultrafast strategy to prepare a stable luminescent SiQDs using N-[3-(trimethoxysilyl)propyl]ethylenediamine (EDAS) and salicylaldehyde as precursors for the first time. These luminescent SiQDs were further utilized for fabrication of luminescent mesoporous silica nanoparticles (MSNs) through direct encapsulation of SiQDs by MSNs. The novel synthetic and modified SiQDs uses commercial raw materials and the entire reaction can be completed within 30 s. The successful preparation of SiQDs and SiQDs@MSNs were characterized by various characterization equipments. The cell viability as well as cell uptake behavior of SiQDs@MSNs were also examined to evaluate their potential for biomedical applications. We demonstrated that these SiQDs@MSNs are low toxicity and of great potential for biological imaging. Based on the above results, we believe that these SiQDs@MSNs should be novel and promising candidates for biomedical applications owing to their intense fluorescence, biocompatibility and high specific surface areas.

Abstract:Highly fluorescent CdSe/CdS/ZnS core/shell/shell QD@SiO2 NPs have been synthesized via a reverse microemulsion approach with a novel post-treatment process by combining annealing with photoactivation. The QD@SiO2 NPs have a high photoluminescent quantum yield of up to 56% and were successfully applied to cellular uptake by HeLa cells.

Abstract:To develop a class of robust and colloidally stable silica nanocapsules for bioimaging, we have investigated the interfacial hydrolysis and condensation of silicon alkoxides confined between the PPO core and PEO corona of self-assembled Pluronic (R) polymeric micelles. The interfacial hydrolysis and condensation, which are completed under the benign conditions of near-neutral pH and at room temperature, give rise to the decoration of silica nanocapsules with a layer of PEO chains dangling on their surface. This feature enables them to be intrinsically stable in aqueous and physiological environments. To demonstrate the silica nanocapsules as effective nanocarriers in bioimaging as well as their biocompatibility, fluorescent conjugated polymer MEH-PPV is encapsulated into the silica nanocapsules and used for in vivo studies with zebrafish as the vertebrate model. When microinjected into the transgenic zebrafish line TG(fli1:EGFP), the cellular internalization of these fluorescent silica nanocapsules does not appear to interfere with larval development nor affect vessel growth. Their biostability is verified by the continued presence of fluorescent-labeled cells in the zebrafish larvae over a long period after the microinjection of fluorescent silica nanocapsules.

Abstract:After microwave-induced hydrosilylation reaction with 10-undecenoic acid, luminescent porous silicon nanoparticles (LPSiNPs) showed excellent fluorescence stability under physiological conditions. Fluorescence quenching in as-prepared LPSiNPs was highly sensitive with the concentration of Cu2+ at mmol L-1 level, which could be further applied to the detection of intracellular Cu2+.

In this work, AIE luminogens based on salicylaldehyde hydrazones were embedded in silica nanoparticles through a facile noncovalent approach, which afforded AIE-FSNPs emitting much brighter fluorescence than that of some commercial fluorescein-doped silica and polystyrene nanoparticles. These AIE-FSNPs displaying multiple fluorescence colors were fabricated by a general method, and they underwent much less fluorescence variation due to environmental pH changes compared with fluorescein-hybridized FSNPs. In addition, a DNA aptamer specific to nucleolin was functionalized on the surface of AIE-FSNPs for targeted cell imaging. Fluorescent microscopy and flow cytometry studies both revealed highly selective fluorescence staining of MCF-7 (a cancer cell line with nucleolin overexpression) over MCF-10A (normal) cells by the aptamer-functionalized AIE-FSNPs. The fluorescence imaging in different color channels was achieved using AIE-FSNPs containing each of the AIE luminogens, as well as photoactivatable fluorescent imaging of target cells by the caged AIE fluorophore.
