The surface of NPs is covered by proteins upon contacting with biological systems (e.g. serum), resulting in the formation of a protein “corona” that is strongly associated with the NPs' surface and defines how cells “see” the NPs in a biological medium[2],[3]. Although many studies have demonstrated that surface modification of AuNPs with biomolecules endows them with various biological functionalities, the impact on living organisms of the chirality of the surface molecules at nanoscale has not been disclosed. Herein the effects of AuNPs capped with different chiral forms of poly(acryloyl-L(D)-valine) (L(D)-PAV) on uptake by A549 cells are examined. Experiment Section: The 15nm AuNPs were incubated with 1 mg/mL L(D)-PAV at room temperature overnight. The obtained solution was centrifuged and followed by dialysis in water for 4 d. Then the protein adsorption, cellular uptake amount and intracellular distribution of PAV-AuNPs were studied by µBCA, ICP-MS and laser scanning confocal microscopy, respectively. Results and Discussion: The diameter of the PAV-AuNPs was about 16 nm. The only difference between L-PAV-AuNPs and D-PAV-AuNPs was their chirality. The PAV-AuNPs were mainly distributed in cytoplasm, regardless of chirality. With the concentration of FBS increasing, the adsorbed amount of proteins on the PAV-AuNPs increased, and the internalized amount of PAV-AuNPs by A549 cells was decreased. In DMEM and 10% FBS/DMEM, A549 cells took up higher amount of D-PAV-AuNPs. However, in 50% FBS/DMEM and 20 mg/mL BSA/DMEM, the internalized amount of L-PAV-AuNPs and D-PAV-AuNPs was chirality-independent. This finding discloses the importance of molecular chirality on the protein adsorption and thereby the cellular uptake of nanomaterials. Conclusion: The molecular chirality on AuNPs acts as a direct regulator of cellular uptake, but the effect of chirality on cellular uptake disappears when the NPs are covered by proteins, especially BSA. Identification of this chirality-dependent cellular uptake of NPs provides a new idea that chiral effect can act as a novel strategy for designing bio-interface materials and may open a new avenue for further development of gold nanoparticles for biomedical applications. Influence of Albumin Configuration by the Chiral Polymer-Grafted Gold Nanoparticles

Abstract:The interaction between nanoparticles (NPs) and proteins is a topic of high relevance for the medical application of NPs. This study reveals the molecular chirality on NP surfaces as an indirect regulator of the interaction between proteins and NPs. Poly(N-acryloyl-valine) (PAV) polymers with d- and l-configurations were conjugated onto gold NPs with a size of 5 nm to obtain the l-PAV-AuNPs and d-PAV-AuNPs, respectively. They had same chemical composition and surface grafting density but different surface chirality. The isothermal titration calorimetry results showed that adsorption of bovine serum albumin onto the l-PAV-AuNPs and d-PAV-AuNPs was primarily driven by electrostatic interaction. Dynamic light scattering, circular dichroism spectroscopy, fluorescence quenching, and isothermal titration calorimetry characterizations revealed that bovine serum albumin molecules adopted both side-on and end-on configurations on the d-PAV-AuNPs, whereas only end-on configuration on the l-PAV-AuNPs.

Abstract:The chain length of ligands plays an important role in many biomedical applications. Yet the interaction of nanoparticles with surface-capped ligands of different length (especial the hydrophobic chains) and cells is neglected, and thereby the underlying mechanism influencing cellular uptake is unclear. Herein the role of alkyl length on AuNPs in mediating serum protein adsorption and the subsequent cellular uptake by A549 cells was studied. The AuNPs were modified with carboxylic alkyl thiols of different chain length (3MA, 11MA and 16MA), resulting in MA-AuNPs with a size of 5 nm in dry state and several tens of nm in media. The MA-AuNPs possessed a negatively charged surface, whose zeta potential was around -20 mV in serum-containing medium regardless of the length of ligands. Whereas the total amount of adsorbed serum proteins had no significant difference, the relative amounts of adhesion mediators especially vitronectin (Vn) depended on the alkyl length significantly. The 16MA-AuNPs adsorbed the largest amount of Vn, and were internalized by A549 cells with the largest quantity and good distribution in cytoplasm. Taking account all the results, the protein adsorption and its role in linking alkyl length on AuNPs to cellular uptake are figured out.
Surface-anchored Poly(acryloyl-L(d)-valine) with Enhanced Chirality-Selective Effect on Cellular Uptake of Gold Nanoparticles.

Abstract:Chirality is one of the ubiquitous phenomena in biological systems. The left handed (L-) amino acids and right handed (D-) sugars are normally found in proteins, and in RNAs and DNAs, respectively. The effect of chiral surfaces at the nanoscale on cellular uptake has, however, not been explored. This study reveals for the first time the molecular chirality on gold nanoparticles (AuNPs) functions as a direct regulator for cellular uptake. Monolayers of 2-mercaptoacetyl-L(D)-valine (L(D)-MAV) and poly(acryloyl-L(D)-valine (L(D)-PAV) chiral molecules were formed on AuNPs surface, respectively. The internalized amount of PAV-AuNPs was several times larger than that of MAV-AuNPs by A549 and HepG2 cells, regardless of the chirality difference. However, the D-PAV-AuNPs were internalized with significantly larger amount than the L-PAV-AuNPs. This chirality-dependent uptake effect is likely attributed to the preferable interaction between the L-phospholipid-based cell membrane and the D-enantiomers.

Abstract:Despite intense research on biological and biomedical applications of nanoparticles, our understanding of their basic interactions with the biological environment is still incomplete. Systematic variation of the physicochemical properties of the nanoparticles is widely seen as a promising strategy to obtain further insights. In view of the key role of the protein adsorption layer forming on nanoparticles in contact with biofluids, we systematically varied the surface charge of proteins adsorbing onto nanoparticles by chemical modification so as to examine the effect of Coulomb forces in modulating nano-bio interactions. We chose human serum albumin (HSA) as a model protein and ultra-small, negatively charged fluorescent gold nanoclusters (AuNCs) as model nanoparticles. By using fluorescence and CD spectroscopies, we measured binding affinities and structural changes upon binding of the HSA variants. The strengths of the protein-nanoparticle interactions were found to change substantially upon modifying the surface charge of HSA. Furthermore, by using inductively coupled plasma optical emission spectroscopy, confocal fluorescence microscopy, scanning transmission electron microscopy and cell viability assays, we observed that cellular interactions of the AuNCs, including their adherence to cell membranes, uptake efficiency and cytotoxicity, depended markedly on the different surface charges of the HSA variants adsorbed onto the nanoparticles. These results illustrate vividly that the cellular responses to nanoparticle exposure depend on the specific properties of the proteins that adsorb onto nanoparticles from biofluids.
Preferential adsorption of fetal bovine serum on bare and aromatic thiol-functionalized gold surfaces in cell culture media

Abstract:Intracellular uptake of serum-coated gold nanoparticles (AuNPs) in a single mammalian cell was examined in order to investigate the interactions of cell culture media and aromatic thiol-functionalized gold surfaces using micro-spectroscopic tools. The AuNPs modified by the aromatic thiols of para-aminobenzenethiol (ABT), para-hydroxy benzenethiol (HBT), and para-carboxylic benzenethiol (CBT, para-mercaptobenzoic acid) bearing NH(2), OH, and COOH surface functional groups are presumed to adsorb the serum proteins as indicated from the compiled quartz crystal microbalance (QCM) data. The QCM results indicate that among the constituents, fetal bovine serum (FBS) should be the major adsorbate species on AuNPs incubated in Roswell Park Memorial Institute (RPMI) medium. The functionalized AuNPs were found to be internalized as an aggregation state in mammalian cells as evidenced by transmission electron microscopy (TEM) images. We monitored such cellular uptake behaviors of aromatic thiol-modified AuNPs using dark-field microscopy (DFM)-guided confocal surface-enhanced Raman scattering techniques in order to identify the three-dimensional localization inside the single cell. We found that the uptake amounts of ABT, HBT, and CBT were similar by counting up to 70 particles inside the cells incubated in the solution mixture of the aromatic thiol and 1,4-phenylenediisocyanide (PDIC) as a reference. This result indicates for the short aromatic thiol compounds, the AuNPs should enter the cell after the serum-coating regardless of the surface functional groups. Considering that the aromatic thiols have little effect on the serum coating, the DFM/SERS method is an effective tool for monitoring the localization of AuNPs inside a single cell.
Cytotoxicity of gold nanoparticles with different structures and surface-anchored chiral polymers

Abstract:Nanoparticles (NPs) can have profound effects on cell biology. However, the potential adverse effects of gold nanoparticles (AuNPs) with different surface chirality and structures have not been elucidated. In this study, monolayers of poly(acryloyl-l(d)-valine (l(d)-PAV) chiral molecules were anchored on the surfaces of gold nanocubes (AuNCs) and nanooctahedras (AuNOs), respectively. The l-PAV-AuNCs and d-PAV-AuNCs, or the l-PAV-AuNOs and d-PAV-AuNOs, had identical physicochemical properties in terms of size, morphology and ligand density except of the reverse molecular chirality on the particle surfaces, respectively. The l-PAV capped AuNCs and AuNOs exhibited larger cytotoxicity to A549 cells than the D-PAV coated ones, and the PAV-AuNOs had larger cytotoxicity than PAV-AuNCs when being capped with the same type of enantiomers, respectively. The cytotoxicity was positively correlated with the cellular uptake amount, and thereby the production of intracellular reactive oxygen species (ROS).
Protein adsorption onto Fe3O4 nanoparticles with opposite surface charge and its impact on cell uptake

Abstract:Nanoparticles (NPs) engineered for biomedical applications are meant to be in contact with protein-rich physiological fluids. These proteins are usually adsorbed onto the NP surface, forming a swaddling layer called protein corona that influences cell internalization. We present a study on protein adsorption onto different magnetic NPs (MNPs) when immersed in cell culture medium, and how these changes affect the cellular uptake. Two colloids with magnetite cores of 25 nm, same hydrodynamic size and opposite surface charge were in situ coated with (a) positive polyethyleneimine (PEI-MNPs) and (b) negative poly(acrylic acid) (PAA-MNPs). After few minutes of incubation in cell culture medium the wrapping of the MNPs by protein adsorption resulted in a 5-fold size increase. After 24 h of incubation large MNP-protein aggregates with hydrodynamic sizes 1500 to 3000 nm (PAA-MNPs and PEI-MNPs respectively) were observed. Each cluster contained an estimated number of magnetic cores between 450 and 1000, indicating the formation of large aggregates with a "plum pudding" structure of MNPs embedded into a protein network of negative surface charge irrespective of the MNP_core charge. We demonstrated that PEI-MNPs are incorporated in much larger amounts than the PAA-MNPs units. Quantitative analysis showed that SH-SY5Y cells can incorporate 100 per cent of the added PEI-MNPs up to about 100 pg per cell, whereas for PAA-MNPs the uptake was less than 50 percent. The final cellular distribution showed also notable differences regarding partial attachment to the cell membrane. These results highlight the need to characterize the final properties of MNPs after protein adsorption in biological media, and demonstrate the impact of these properties on the internalization mechanisms in neural cells.

Abstract:Nanoparticles (NPs) are widely applied as targeted delivery vehicles for drugs. During transport in the body, proteins in serum are rapidly adsorbed onto the NP surface, creating a protein corona which impacts on the fate of NPs extensively. The objective of this study is to find how the charged groups on the NP surfaces affect the interactions among NPs, protein corona and cells, determining the cellular uptake of NPs consequently. We synthesized two types of polystyrene NPs, carrying carboxyl and amine groups on the surface, respectively, with similar size by emulsion polymerization. We quantified the protein (BSA) adsorption on the two types of NPs, characterized the configuration of the adsorbed proteins, and identified the change in physiochemical properties of NPs coated by protein corona. Despite the different charged groups of the NPs, we detected similar protein adsorption amounts and similar secondary structures of BSA adsorbed on the NPs, and we also found the surface properties (zeta potential and hydrophilicity) and particle size of the NPs became undifferentiated after adsorbing proteins. But we uncovered a significant difference in protein adsorption thermodynamics between the two types of NPs. HepG2 cell uptake studies were conducted. We are the first to make clear that the protein corona and the nature of NP charged surface groups both affected the uptake of NPs, but in different ways. Protein corona transformed the dynamic profile of uptake, turned the endocytosis pathway and may further impact on intracellular transport of NPs; while the charged groups of NPs significantly altered the uptake percentage via the possible mechanism that charged groups influenced configuration of the protein corona.
Gold Nanoparticles with Different Amino Acid Surfaces: Serum Albumin Adsorption, Intracellular Uptake and Cytotoxicity

Abstract:Gold nanoparticles with aspartate, glycine, leucine, lysine, and serine surfaces were produced from the mixed solutions of HAuCl4 and respective amino acids via UV irradiation. The amino acids bind to the nanoparticle surfaces via amine groups and their carboxylic groups extend out to stabilize the nanoparticles. The nanoparticles have diameters of 15-47 nm in pH 7.4 aqueous solution and have diameters of 62-73 nm after 48 h incubation in cell culture containing serum. The nanoparticles adsorb human and bovine serum albumins on their surfaces by specific interactions, characterized by the intrinsic fluorescence quenching of the albumins. The albumin adsorption effectively decreases the aggregation of the nanoparticles in cell culture and also decreases the intracellular uptake of the nanoparticles. The gold nanoparticles produced from leucine and lysine, which have amphiphilic groups on their surfaces, present better biocompatibility than the other gold nanoparticles.
The Effects of Serum Albumin Pre-Adsorption of Nanoparticles on Protein Corona and Membrane Interaction: A Molecular Simulation Study

Abstract:As a platform to deliver imaging and therapeutic agents to targeted sites in vivo , nanoparticles (NPs) have widespread applications in diagnosis and treatment of cancer. However, the poor in vivo delivery efficiency of nanoparticles limits its potential for further application. Once enter the physiological environment, nanoparticles immediately interact with proteins and form protein corona, which changes the physicochemical properties of nanoparticle surface and further affects their transport. In this study, we performed molecular dynamics simulations to study the adsorption mechanism of nanoparticles with various surface modifications and different proteins ( e.g. , human serum albumin, complement protein C3b), and their interactions with cell membrane. The results show that protein human serum albumin prefers to interact with hydrophobic and positively charged nanoparticles, while the protein C3b prefers the hydrophobic and charged nanoparticles. The pre-adsorption of human serum albumin on the nanoparticle surface obviously decreases the interaction of nanoparticle with C3b. Furthermore, the high amount of protein pre-adsorption could decrease the probability of nanoparticle-membrane interaction. These results indicate that appropriate modification of nanoparticles with protein provides nanoparticles with better capability of targeting, which could be used to guide nanoparticle design and improve transport efficiency.

Abstract:Nanoparticles (NP) in biological fluids almost invariably become coated with proteins to form protein coronas. It is the NP–protein corona rather than the bare nanoparticle that determines the nanoparticle's bio-behavior. Here, ultrasmall gold nanoparticles (AuNPs) coated by a human serum albumin (HSA) corona were studied by Fourier transform infrared spectroscopy, denature experiments, fluorescence quenching. Moreover, the intracellular fate of AuNPs and the AuNP–HSA corona has also been investigated. The results show that HSA corona undergo a conformational transition (partial β-sheet changed to α-helicity) when they adsorb on AuNPs, which lead to an enhanced thermal stability. Importantly, we observed that the conformation-transited protein corona–AuNP complex could induce cell apoptosis. Meanwhile, for the first time, the conformation-transited HSA on the AuNPs surface are shown to disrupt living cell membranes. The results obtained here not only provide the detailed conformational behavior of HSA molecules on nanoparticles, but also reveal the structure–function relationship of protein corona, which is of utmost importance in the safe application of nanoscale objects in living organisms.
Influence of chirality on protein corona formation of low-fouling chiral poly(2-oxazoline) coated nanoparticles

Abstract:Low-fouling polymers play a crucial role in the development of therapeutic nanoparticles (NPs), which are commonly described as nanomedicines. These hydrophilic polymers are typically designed to favourably alter the protein corona that forms around a NP by reducing the binding of opsonins, resulting in reduced immune recognition and clearance. The polymer specific proteome of the corona is mainly dictated by the physicochemical properties of the polymer and several parameters such as hydrophilicity and charge have been explored to regulate its composition. To date, there has been little attention given to the influence of the chirality of the hydrophilic polymer on the composition of the corona. Water-soluble poly(2,4-dialkyl-2-oxazoline)s (PdOx) are a unique synthetic polymer class to explore the effect of chirality as they can be rendered chiral if enantiopure monomers are used for their synthesis. Herein we prepared two water-soluble poly(2,4-dimethyl-2-oxazoline)s (PdMeOx) of different handedness, alongside racemic PdMeOx and the well-established low-fouling structural isomer poly(2-ethyl-2-oxazoline) (PEtOx). All polymers were equipped with sulphur-containing xanthate moieties with high end-group fidelity, which enabled anchoring to gold nanoparticles (AuNPs) as a useful handle for evaluation of the polymer-specific fouling. It was revealed that a hard corona formed after exposing the coated AuNP to human plasma proteins as investigated by dynamic light scattering, SDS-PAGE and Pierce TM bicinchoninic acid protein assay (BCA) revealing a similar protein adsorption between PEtOx and all three PdMeOx. Moreover, a detailed proteomics study using liquid chromatography coupled to mass spectrometry (LC–MS/MS) was performed and analysed by label-free quantitation (LFQ). Overall, the PdMeOx systems showed similar human plasma protein corona profiles compared to the PEtOx control. However, between the R- and S-PdMeOx enantiomers differences in protein population abundances were observed, suggesting the propensity for specific interaction of chiral PdOx with different elements of the proteome. Overall, the present study introduces PdMeOx as a chiral, low fouling polymer system, which can reduce protein fouling in a similar magnitude to the PEtOx control. Importantly, we provide a first insight into how changes in the chirality of PdMeOx can influence the components of the protein corona, which will inform the future design of PdOx nanotherapeutics.

Green Synthesis of Biocompatible Chiral Gold Nanoparticles

Abstract:Chiral gold nanoparticles (Au NPs) have been investigated widely and have shown great potential in biomedical applications, such as biosensing, combating bacterial infections and tissue regeneration. However, some stabilizers and reducing agents for the synthesis of chiral Au NPs can produce toxicity in living organisms. Therefore, it is interesting to design green methods to prepare chiral gold nanoparticles that are nontoxic, environment-friendly, and low-cost. Herein, novel biocompatible chiral Au NPs with a diameter of 54.4 ± 14.9 nm were prepared by the in situ reduction of HAuCl4 with alginates as the green reducing agent and chiral-inducing and stabilizing agent. XPS, TGA, UV-Vis and CD analyses demonstrated that alginate-stabilized chiral Au NPs (ALG-Au NPs) were successfully prepared, while biocompatibility assessment showed that cell viability was 116.0% when the concentration of ALG-Au NPs arrived at 300 μg/mL, which indicated that ALG-AuNPs showed excellent biocompatibility. Furthermore, the ALG-Au NPs can respond to metal ions, such as Ca2+, Cu2+, Mn2+ and so on, implying potential application for biosensing.

Abstract:The biomedical applications of nanoparticles are still impeded by the non-specific adsorption of proteins, cells, or others biological species in vivo/in vitro. In this work, poly(N-hydroxyethyl acrylamide) was hired to modify a solid polymer core, polystyrene (PS) nanoparticles, via surface-initiated photo-emulsion polymerization to form nano-sized spherical poly(N-hydroxyethyl acrylamide) brush (PS@PHEAA). Its antifouling ability and stability were investigated by dynamic light scattering (DLS), turbidimetric titration, and isothermal titration calorimetry (ITC). The size of PS@PHEAA was constant as a function of pH, while slightly changed with ionic strength in single protein solution. ITC data confirmed that protein was slightly adsorbed on PS@PHEAA and the ionic strength influenced the adsorption. All characterizations demonstrated that PHEAA layer reduced the interaction between nanoparticles and proteins. Thus, these nanoparticles ideal candidates for future applications in the biomedical field.
Understanding the Cellular Uptake of Ph-Responsive Zwitterionic Gold Nanoparticles: A Computer Simulation Study

Abstract:Surface functionalization of nanoparticles (NPs) with stealth polymers (e.g., hydrophilic and zwitterionic polymers) has become a common strategy to resist nonspecific protein adsorption recently. Understanding the role of surface decoration on NP biomembrane interactions is of great significance to promote the application of NPs in biomedical fields. Herein, using coarse-grained molecular dynamics (CGMD) simulations, we investigate the interactions between stealth polymer-coated gold nanoparticles (AuNPs) and lipid membranes. The results show that AuNPs grafted with zwitterionic polymers can more easily approach the membrane surface than those coated with hydrophilic poly(ethylene glycol) (PEG), which can be explained by the weak dipole dipole interaction between them. For zwitterionic AuNPs which can undergo pH-dependent charge conversion, different interaction modes which depend on the polymer protonation degree are found. When the protonation degree is low, the particles just adsorb on the membrane surface; at moderate protonation degrees, the particles can directly translocate across the lipid membrane through a transient hydrophilic pore formed on the membrane surface; the particles are fully wrapped by the curved lipid membrane at high protonation degrees, which may lead to endocytosis. Finally, the effect of polymer chain length on the cellular uptake of zwitterionic polymer-coated AuNPs is considered. The results demonstrate that longer polymer chain length will block the translocation of AuNPs across the lipid membrane when the protonation degree is not high; however, it can improve the transmembrane efficiency of AuNPs at high protonation degrees. We expect that these findings are of immediate interest to the design and synthesis of pH-responsive nanomaterials based on zwitterionic polymers and can prompt their further applications in the field of biomedicine.

Abstract:Chirality is one of the basic, unique, and most appealing features of biological molecules; however, many intriguing chiral phenomena in biological world remains insufficiently revealed yet. In this research, we fabricated chiral surfaces by assembling natural chiral amino acids-cysteine of opposite configurations (D- and L-) onto gold surfaces, respectively, and investigated the adhesion of the L929 fibroblast on them. No significant differences were observed in the density of adherent cells under serum-free culture condition; while in serum-containing condition, significantly more cells adhered on the L-Cys assembled surfaces. This phenomenon suggested that serum protein might play an important role in mediating the selective adhesion of cells on chiral surfaces. Hence, we adopted both radiolabeling and surface plasmon resonance (SPR) techniques to monitor protein adsorption onto the above surfaces. The results evidently showed more proteins adsorbed onto surfaces assembled with L-Cys. We propose that the difference in protein adsorption on chiral surfaces as demonstrated in this paper might not only shed light on the ensuing investigation of bio-related chirality phenomena, but also provide a novel strategy for the rational design and fabrication of novel biomaterials and bio-related devices based on chiral effects.
Nanoparticle Size and Surface Chemistry Determine Serum Protein Adsorption and Macrophage Uptake

Abstract:Delivery and toxicity are critical issues facing nanomedicine research. Currently, there is limited understanding and connection between the physicochemical properties of a nanomaterial and its interactions with a physiological system. As a result, it remains unclear how to optimally synthesize and chemically modify nanomaterials for in vivo applications. It has been suggested that the physicochemical properties of a nanomaterial after synthesis, known as its "synthetic identity", are not what a cell encounters in vivo. Adsorption of blood components and interactions with phagocytes can modify the size, aggregation state, and interfacial composition of a nanomaterial, giving it a distinct "biological identity". Here, we investigate the role of size and surface chemistry in mediating serum protein adsorption to gold nanoparticles and their subsequent uptake by macrophages. Using label-free liquid chromatography tandem mass spectrometry, we find that over 70 different serum proteins are heterogeneously adsorbed to the surface of gold nanoparticles. The relative density of each of these adsorbed proteins depends on nanoparticle size and poly(ethylene glycol) grafting density. Variations in serum protein adsorption correlate with differences in the mechanism and efficiency of nanoparticle uptake by a macrophage cell line. Macrophages contribute to the poor efficiency of nanomaterial delivery into diseased tissues, redistribution of nanomaterials within the body, and potential toxicity. This study establishes principles for the rational design of clinically useful nanomaterials.

Abstract:Adsorption and desorption of proteins on biomaterial surfaces play a critical role in numerous biomedical applications. Spherical diblock polymer brushes (polystyrene with photoiniferter (PSV) as the core) with different block sequence, poly(acrylic acid)-b-poly(N-(2-hydroxyethyl) acrylamide) (PSV@PAA-b-PHEAA) and poly(N-(2-hydroxyethyl) acrylamide)-b-poly(acrylic acid) (PSV@PHEAA-b-PAA) were prepared via surface-initiated photoiniferter-mediated polymerization (SI-PIMP) and confirmed by a series of characterizations including TEM, Fourier transform infrared (FTIR) and elemental analysis. Both diblock polymer brushes show typical pH-dependent properties measured by dynamic light scattering (DLS) and Zeta potential. It is interesting to find out that conformation of PSV@PAA-b-PHEAA uniquely change with pH values, which is due to cooperation of electrostatic repulsion and steric hindrance. High-resolution turbidimetric titration was applied to explore the behavior of bovine serum albumin (BSA) binding to diblock polymer brushes, and the protein adsorption could be tuned by the existence of PHEAA as well as apparent PAA density. These studies laid a theoretical foundation for design of diblock polymer brushes and a possible application in biomedical fields.
From Neutral to Zwitterionic Poly(α-Amino Acid) Nonfouling Surfaces: Effects of Helical Conformation and Anchoring Orientation.

Abstract:The development of high-performance nonfouling polymer surfaces for implantable medical devices and therapeutic nanomaterials is of great importance. Elaborating the relationship of polymer structural characteristics and the resulted surface properties can offer useful guidance toward ideal biointerfaces. In this work, we investigate the effects of the helical conformation and anchoring orientation of poly(α-amino acid)s (PαAAs) to produce advanced nonfouling surfaces. By using the neutral poly(γ-(2-(2-(2-methoxyethoxy)ethoxy)ethoxy)esteryl glutamates) (P(EG3Glu)s) as a model system, the adsorption kinetics are monitored by ex-situ variable angle spectroscopic ellipsometry and in-situ quartz crystal microbalance with dissipation. It is found that the polymers adopting a rigid rod-like α-helical conformation can self-assemble more rapidly to produce denser adlayers, and generate significantly improved nonfouling surfaces compared to those flexible polymer analogues including the widely used antifouling polymer PEG. Moreover, the surface properties can be further enhanced by using the antiparallel orientated helical P(EG3Glu)s. Most importantly, the insights gained from the P(EG3Glu) model system are successfully applied to the generation of ultra-low-fouling surfaces using zwitterionic PαAAs brushes, underscoring the generality of the approach. Particularly, the surface based on the antiparallel aligned zwitterionic helical PαAAs exhibits ∼98-99% reduction of human serum adsorption relative to the bare gold, and gives almost no adhesion of mouse platelet. Taken together, this work depicts an extremely simple yet highly effective approach to manipulate surface properties for numerous applications in biomaterial interfaces, diagnostics, and biosensors.
