A Bifunctional Fe (Iii)-Coordinated Nanoprobe for Electrochemical Detection of Sphingosine Kinase 1 Activity

Abstract:Kinase analysis is of fundamental significance in biochemical research. Here, we report on an electrochemical method to improve the simplicity of kinase analysis taking advantage of a novel bifunctional Fe (III)-coordinated nanoprobe. The Fe (III)-immobilized nanoprobe is designed to be able not only to recognize phosphorylated sites with high specificity, but also to generate electrochemical signal through NTA-Fe (III) catalyzed H2O2 redox. In this way, the phosphorylation recognition and signal output can be realized by a one-off solution, which significantly improves the simplicity of the kinase assay. Moreover, owing to the enriched NTA-Fe (III) on each nanoprobe and the continuous catalytic ability of each NTA-Fe (III), this method ensures very high sensitivity by dual signal amplification. Taking sphingosine kinase 1 (SphK1) as a model kinase, a limit of detection as low as 7.404pmolmin−1mg−1 is achieved while keeping the kinase in its lipid-simulating catalytic environment. As a result, this assay method can act as a sample method for lipid kinase detection, which may also hold great potential in clinical analysis of other vital kinases.
