Abstract:<p>The present work aimed to assemble a simple, portable and economical L-junction microfluidic device to realize the adjustment and tunability of homogeneous round-shaped particles synthesis. In this study, we synthesize two kind of microparticles, including magnetic alginate microparticles (MAM) and chitosan-coated magnetic alginate (CMAM) used for controlling the drug release under a mild condition. Comparing to the traditional method, the MAM synthesized via this microfluidic approach has uniform size distribution, adjustable diameter as well as tunable magnetism. By exploring the amoxicillin as model drug, the MAM displays excellent pH-sensitive release, the effect of particle size on the drug release rate was investigated as well. The results show the smaller particles (220 μm) show a faster release rate than the bigger materials (1000 μm) due to their larger specific area, providing more frequency to interact with the reaction solution. The positive polyelectrolyte, chitosan, coated on the magnetic alginate surface endows CMAM time extension in drug release by two times, successfully achieving drug controlled and sustained release via the kinetics analysis. In summary, this microfluidic approach provides a convenient and efficient fluidic design for the well-controlled synthesis of micro-and nanoscale particles, which is a potential choice used for controlled and sustained drug release.</p>

Abstract:Facile synthesis and smart designs of multifuctional nanocarriers would make them more potential in the practical application of drug delivery for cancer treatment. In this study, we synthesized hollowmesoporous cobalt ferrite (H-mCo(x)Fe(3-x)O(4)) nanoparticles by a cetyltrimethylammonium bromide (CTAB)-assisted solvothermal method. The resulting monodisperse CoxFe3-xO4 microspheres are 280 nm in diameter with fine uniformity and hollow-mesoporous structure, and they possess a fairly high surface area of 58.03 m(2)/g, a pore volume of 0.13 cm(3)/g, an average pore size of 7.21 nm, and are suitable for drug loading. This material has a high magnetization saturation value (42.2 emu/g) for drug targeting and its sodium chloride suspension could raise to 50 degrees C from room temperature in 180 s. The drug release process shows that the release rate under the microwave irradiation was faster than that under stirring condition, and about 64% VP16 was released after six on/off microwave treatment cycles. So these multifunctional CoxFe3-xO4 nanoparticles with single material are expected to provide the research foundation for using as drug carrier themselves for drug loading, magnetic targeting, and microwave triggered controllable release. Design, Fabrication and Characterization of Magnetic Porous PDMS As an On-Demand Drug Delivery Device

Abstract:We introduce a novel, on-demand drug delivery device based on a biocompatible magnetic sponge. The sponge is made of a porous polydimethylsiloxane (PDMS) mixed with carbonyl iron (CI) particles. The sponge is deformed under a magnetic field and consequently leads to releasing its contents. As a proof of concept study, three different CI/PDMS wt% ratios of 50%, 100%, and 150% were selected where, the 100% showed the most deformation under various magnetic fields. Although this sponge can solely be used as a potential drug delivery agent, a separate reservoir has been fabricated to protect the sponge and control the release rate. The final device has a diameter of 6 mm with a thickness of 2 mm. Controlled release of methylene blue (MB) and docetaxel (DTX) have been investigated to demonstrate the consistency and flexibility in adjusting the release rate from the device to suit different treatment requirements. Ex vivo tissue implantation has also been accomplished. This device is able to be implanted and deliver therapeutic agents at prescribed dosages.

Abstract:Fabrication of magnetic particles (MPs) with high magnetization and large surface area simultaneously is critical for the application of MPs in bioseparation and drug delivery but remains a challenge. In this article, we describe an unprecedented approach to synthesize mesoporous magnetic colloidal nanocrystal clusters (MCNCs) stabilized by poly(γ-glutamic acid) (PGA) with high magnetization, large surface area (136 m(2)/g) and pore volume (0.57 cm(3)/g), excellent colloidal stability, prominent biocompatibility, and acid degradability. This result provides the important step toward the construction of a new family of MCNCs and demonstrates its capacity in a "magnetic motor" drug delivery system. Here, as an example, we explore the applicability of as-prepared mesoporous MCNCs as hydrophobic drug delivery vehicles (paclitaxel as model drug), and the resultant loading capacity is as high as 35.0 wt %. The antitumor efficacy measured by MTT assay is significantly enhanced, compared with free drugs. Thus, combined with their inherent high magnetization, the mesoporous MCNCs pave the way for applying magnetic targeting drug carriers in antitumor therapeutics.
Controlled Rupture of Magnetic Polyelectrolyte Microcapsules for Drug Delivery.

Abstract:In this study, a magnetic-sensitive microcapsule was prepared using Fe 3O 4/poly(allylamine) (Fe 3O 4/PAH) polyelectrolyte to construct the shell. Structural integrity, microstructural evolution, and corresponding release behaviors of fluorescence dyes and doxorubicin were systematically investigated. Experimental observations showed that the presence of the magnetic nanoparticles in the shell structure allowed the shell structure to evolve from nanocavity development to final rupture of the shell under a given magnetic stimulus of different time durations. Such a microstructural evolution of the magnetic sensitive shell structure explained a corresponding variation of the drug release profile, from relatively slow release to burst-like behavior at different stages of stimulus. It has proposed that the presence of magnetic nanoparticles produced heat, due to magnetic energy dissipation (as Brown and Neel relaxations), and mechanical vibration and motion that induced stress development in the thin shell. Both mechanisms significantly accelerated the relaxation of the shell structure, causing such a microstructural evolution. With such a controllable microstructural evolution of the magnetic-sensitive shell structure, active substances can be well-regulated in a manageable manner with a designable profile according to the time duration under magnetic field. A cell culture study also indicated that the magnetic-sensitive microcapsules allowed a rapid uptake by the A549 cell line, a cancerous cell line, suggesting that the magnetic-sensitive microcapsule with controllable rupturing behavior of the shell offers a potential and effective drug carrier for anticancer applications.
Encapsulation of drug microparticles with self-assembled Fe 3O 4/alginate hybrid multilayers for targeted controlled release

Abstract:The magnetic-targeted drug-delivery system was developed with the Fe3O4 nanoparticles as the self-assembling material to form the hybrid multilayer shells. Layer-by-Layer stepwise self-assembly of the natural polyelectrolytes sodium alginate and the Fe3O4 nanoparticles was used to create the magnetic hybrid multilayers around drug microparticles (similar to 400 nm in diameter) for the purpose of targeted controlled release. The obtained drug microparticles encapsulated with hybrid multilayers were characterized with dynamic light scattering, transmission electron microscope, and vibrating sample magnetometer. UVvis spectroscopy was employed to monitor the drug releasing behaviors in pH 1.8 and 7.4 buffer solutions. It was found that the release of drug molecules from the pH-sensitive magnetic-targeted drug-delivery system was mainly dependent on the following factors: the permeability of the hybrid multilayer shells and the solubility of the drug molecules in the bulk solutions. Abstract:Remarkable evolution of drug-loaded microrobots has aroused growing interest in targeted therapy. However, existing microrobots encounter a number of challenges including maneuverability, biodegradability, and drug sustained release capabilities in medical applications. Designed double-layer drug-loaded microrobots (TDMs) were made of outer layer calcium alginate hydrogel and inner layer magnetic chitosan microspheres (mCSs). TDMs precisely deliver drug-loaded mCSs following the expected route towards the predefined destinations under a vision-guided magnetic driving system. In acidic conditions the outer layer of TDMs can protect mCSs, while in alkaline conditions calcium alginate hydrogel dissolves at an appropriate rate to render mCSs be continuously released. The mCSs fulfill drug sustained release tasks in vitro. The new designed low-cost microrobots will be competent for intestinal cancer targeted therapy after oral administration and bridge the gap between scientific research and market demand.

Abstract:Drug delivery systems that can control drug release profile to ensure a high therapeutic efficacy and reduced side effects are highly desired in pharmaceutical and biomedical fields. Microparticles are the most commonly used drug delivery systems, because they can be easily administrated to patients, and be engineered with different structures and functions for keeping drug stability, delivering drugs to a desired location, and releasing drugs with a predetermined rate in a well‐controlled manner. Microfluidic techniques show great power for controllable generation of highly monodisperse multiple emulsion droplets with unprecedented structural diversity. Microfluidics‐templated emulsions allow elaborately design and controllable generation of highly uniform microparticles with well‐controlled sizes, shapes, compositions, and structures, and integrated functions for controlled drug release. This review highlights recent progress on controllable microfluidic fabrication of monodisperse emulsion templates and the resultant polymeric microparticles with well‐tailored structures and functions for flexible encapsulation and controlled release of drugs. Especially, a comprehensive overview of the recent biomedical applications of these microparticles with diverse release mechanisms is provided. Finally, perspectives on further advancing the microfluidic techniques for fabricating functional microparticles from lab scale to industrial scale are discussed.
Magnetic Mesoporous Silica Microspheres with Thermo-Sensitive Polymer Shell for Controlled Drug Release

Abstract:A controlled drug release system was designed based on the combination of three advantages into one entity, which was composed of Fe3O4 magnetic nanoparticle as the core, mesoporous silica as the sandwiched layer, and thermo-sensitive P(NIPAM-co-NHMA) copolymer as the outer shell. The hydrophilic comonomer content affected the volume phase-transition temperature (VPTT) of this composite microsphere and the behavior of the temperature-triggered drug release. Zn(II) phthalocyanine tetrasulfonic acid (ZnPcS4), a well-known photodynamic therapy (PDT) drug, was used as a model drug to assess the release system. The results demonstrated that the drug release behavior was dependent on the temperature and had a close correlation with the VPTT. Above the VPTT, the drug release rate was much faster than that below the VPTT, which showed a great potential application in tumor therapy.

Abstract:In this work, we present a novel class of uniform eccentric magnetic microcapsules based on polydimethylsiloxane (PDMS) for magnetic resonance imaging (MRI)-guided local injection and pH-regulated drug release. The microcapsules contained magnetic nanoparticles in their PDMS shells, which allowed them to be easily tracked by MRI during administration. Besides, they showed pH-dependent drug release profiles due to dissolution of the embedded magnetic nanoparticles in acidic solutions. Moreover, by tuning the mass fraction of the magnetic nanoparticles, we could further regulate the release rate of drug molecules from them. As a demonstration, we investigated the delivery of cis-platinum using the microcapsules through an in vitro cell test, which confirmed the pH-controlled release of the drug in phantom tissues. Our study suggests that this type of eccentric magnetic microcapsules could be simultaneously employed as a potential imaging contrast and a smart drug delivery system, holding great potential for guided local therapy.

Abstract:Magnetic nanoparticles (MNPs) with excellent monodispersity, colloidal stability and biocompatibility are important for the fabrication of magnetic targeted drug delivery system. In this study, magnetic lipid microcapsules (MLMs) containing lipid-coated MNPs were fabricated. 5-(6)-carboxyfluorescein (CF) release from MLMs triggered through thermal activation of MNPs was investigated in detail upon application of an alternating magnetic field (AMF). The experimental results revealed that coating lipid on MNPs effectively reduced the MNPs aggregation, enhanced the dispersion, stability and biocompatibility but did not affect their heat transfer. Abstract:<span>To overcome the shortcoming of conventional transarterial chemoembolization (cTACE) like high systemic release, a novel droplet-based flow-focusing microfluidic device was fabricated and the biocompatible poly(lactic-co-glycolic acid) (PLGA) magnetic drug-eluting beads transarterial chemoembolization (TACE) microspheres with tunable size and shell thickness were prepared via this device. Paclitaxel, as a model active, was loaded through O/O/W emulsion method with high efficiency. The size and the shell thickness vary when adjusting the flow velocity and/or solution concentration, which caters for different clinical requirements to have different drug loading and release behavior. Under the designed experimental conditions, the average diameter of the microspheres is 60 ± 2 μm and the drug loading efficiency has reached 6%. The drug release behavior of the microspheres shows the combination of delayed release and smoothly sustained release profiles and the release kinetics differ within different shell thickness. The microspheres also own the potential of magnetic resonance imaging (MRI) visuality because of the loaded magnetic nanoparticles. The microsphere preparation method and device we proposed are simple, feasible, and effective, which have a good application prospect.</span>

Abstract:The synthesis of magnetite nanoparticles coated with pH-sensitive poly((2-dimethylamino) ethyl methacrylate) (PDMAEMA) via atom transfer radical polymerization (ATRP) for use as novel potential carriers for targeted drug delivery and controllable release is reported. The organic/inorganic hybrid nanoparticles were obtained with a narrow molecular weight distribution. The pH-sensitivity of the nanoparticles was investigated by the measurement of the pH dependence of hydrodynamic radius and the superparamagnetism was illustrated by vibrating sample magnetometer (VSM). The behavior of model drug phenolphthalein released from the nanoparticles indicated that the rate of drug release could be effectively controlled by altering the pH values of the environment.

Abstract:Magnetic microcapsules with inner asymmetric structure are prepared by miniemulsion polymerization of styrene in presence of oleic acid-modified Fe3O4 as magnetic nanoparticles and n-heptane (HP) as core. The resultant microcapsules with novel structure exhibit magnetic anisotropy and cavity anisotropy. The formation mechanism of the microcapsule is illustrated in detail. The obtained results showed that the polymerization process is associated with phase-separation between polystyrene, oleic acid (OA) and HP. Based on this formation mechanism, a kind of highly asymmetric magnetic microcapsules are prepared, which is further verified the presented phase-separation mechanism. Meanwhile, four kinds of magnetic microcapsules with inner asymmetric structure, which show different shell thickness and cavity volume, are prepared for application to carrying and releasing drugs. The results reveal that the microcapsules exhibit excellent controlled drug release property and the release speed can be controlled by adjusting the thickness of the thinner-side capsule-wall. Drug loading and drug release of inner asymmetric magnetic microcapsules were from the thinner side of capsule-wall. Abstract:Bioinspired by the morphology of dandelion pollen grains, we successfully prepared a template-free solution-based method for the large-scale preparation of three-dimensional (3D) hierarchical CoFe2O4 porous microspheres. Besides, on the basis of the effect of the reaction time on the morphology evolution of the precursor, we proposed an in situ dissolution-recrystallization growth mechanism with morphology and phase change to understand the formation of dandelion pollenlike microspheres. Doxorubicin hydrochloride, an anticancer drug, is efficiently loaded into the CoFe2O4 microspheres. The magnetic nanoparticles as field-controlled drug carriers offer a unique power of magnetic guidance and field-triggered drug-release behavior. Therefore, 3D hierarchical CoFe2O4 porous microspheres demonstrate the great potential for drug encapsulation and controlled drug-release applications.

Abstract:An innovative drug delivery system based on magnetic and fluorescent multifunctional chitosan nanoparticles was developed, which combined magnetic targeting, fluorescent imaging and stimulus-responsive drug release properties into one drug delivery system. Water-soluble superparamagnetic Fe3O4 nanoparticles, CdTe quantum dots (QDs) and pharmaceutical drugs were simultaneously incorporated into chitosan nanoparticles; cross-linking the composite particles with glutaraldehyde tailored their size, morphology, surface properties and drug release behaviors. The system showed superparamagnetic and strong fluorescent properties, and was used as a controlled drug release vehicle, which showed pH-sensitive drug release over a long time. The composite magnetic and fluorescent chitosan nanoparticles are potential candidates as a smart drug delivery system.

Abstract:In this paper, we fabricated uniform polydimethylsiloxane (PDMS) magnetic microcapsules with eccentric internal structures and employed them as a novel delivery system for orientation-specific and dual stimuli-responsive controlled drug release. These eccentric microcapsules contained Fe3O4 nanoparticles in their inner cores. Because of the paramagnetic Fe3O4 nanoparticles, the eccentric microcapsules could be accurately moved by a magnetic field, leading to the precise control of the microcapsule locations. Also, due to the eccentric structures of the magnetic microcapsules, the capsules exhibited a non-uniform magnetic property; the capsules could be aligned by magnetic fields with their thin walls facing the magnet, resulting in a precise orientation-specific control of the microcapsules. More interestingly, the eccentric magnetic microcapsules demonstrated a dual stimuli-responsive controlled release of inclusions involving a sustained release under ultrasound and an intense release under laser stimulation. Furthermore, we studied the efficacy of doxorubicin (DOX) release from the microcapsules regulated by laser stimulation by performing in vitro cell tests with and without an applied magnetic field. The cell tests showed that the orientation-specific control of the microcapsules under a magnetic field (when the thin walls of the eccentric microcapsules were oriented towards the cell) improved the efficacy of the drug released from the microcapsules. The results suggested that our eccentric magnetic microcapsules hold all the properties needed for a site-specific, orientation-specific and dual stimuli-responsive delivery system, demonstrating a great potential application for multifunctional controlled drug release.
