Abstract:An efficient method to construct congested 4-phenyl-4-hydroxyquinazolin-2-thiones 3 via solvent-free and catalyst-free addition of aryl isothiocyanates 2 and with o-aminobenzophenone 1 under thermal conditions to provide new congested 4-phenyl-4-hydroxyquinazolin-2-thiones 3 in practically quantitative yields via simple purification by filtration/recrystallization. Extension of these conditions to o-aminoacetophenone 4 in place of o-aminobenzophenone 1 led to 4-methylenequinazoline-thione 5 as a result of a subsequent dehydration. Quinazoline derivatives represent a pharmaceutically important class of heterocycles in a variety of drug molecules1 and occur frequently in natural and synthetic pharmaceutical products.2 Among this family, quinazolin-2-thione analogs received particular attention not only because of their potential pharmacological activity (Scheme 1)3 but also because of their versatile transformations to other quinazoline derivatives thanks to rich chemistry of the thione group.4 The reported synthesis of such compounds remains challenging and involves multi-step reactions, high-temperature, prolonged heating, strong base/acid promoters/catalysts,5 transition metals6 with frequently unsatisfactory overall efficiency and yields.7 During the course of our development of a more environmentally friendly sulfur chemistry,8 including isothiocyanate one,9 we have been focusing our effort on boosting the reactivity of a reaction in simple and costless manners. One of the most frequently used techniques, which are also in line with the objective of reducing waste generated during the transformations, is to perform the reaction under solvent-free conditions. We have exemplified in many cases that such approach was particularly efficient in accelerating the reaction rate while limiting the amounts of waste unnecessarily generated during the reaction. Thanks to unique nature of these conditions in which molecules could closely interact under high concentration, reaction time could be shortened with lower heating temperatures. In many circumstances, the expected products could be readily crystallized out from the reaction mixtures, thus facilitating the purification by simple trituration, filtration and drying sequence.
