Polycomb group protein Yin Yang 1 controls quiescence and self-renewal potential in hematopoietic stem cells

Abstract:Yin Yang 1 (YY1) is a ubiquitous, multifunctional transcription factor that plays important functions in early embryonic development, lineage differentiation, cell growth control and oncogenesis. YY1 is also a crucial mammalian Polycomb Group Protein (PcG). It can orchestrate the PcG network by recruiting other PcG members to sequence-specific DNA binding sites and results in consequent histone modifications. Our prior data show that the elevated YY1 expression results in a significant long-term hematopoietic stem cell (LT-HSC) enrichment in bone marrow (BM) transplanted mice. However, the precise regulatory role of YY1 in hematopoietic stem cell (HSC) is still largely unknown. Here we generated Yy1f/f Mx1-Cre mice and assessed the impact of YY1-null phenotype on HSCs by conditionally knocking out YY1 in hematopoietic system. Compared with Mx1-Cre control mice, Yy1f/f Mx1-Cre mice were pancytopenic, and had decreased total cell counts of both bone marrow (BM) and spleen. Yy1f/f Mx1-Cre mice had significantly decreased number and frequency of LSK, LT-HSC, short-term (ST) HSC, myeloid progenitor (MP), and multipotent progenitor (MPP) cells. To assess YY1 impact on HSC long-term self-renewal and differentiation, serial competitive BM transplantation and colony formation assay were performed. Our data show there was a significant reduction of donor derived cells (CD45.2+) in peripheral blood (PB), BM and spleen of Yy1f/f Mx1-Cre transplanted mice. LT-HSCs (Lin-Scal+C-kit+Cd48-CD150+) sorted from Yy1f/f Mx1-Cre mice had a decreased capacity of forming colonies compared with Mx1-Cre control. Yy1-deficient adult LT-HSCs had markedly decreased quiescence and reduced Kit expression. Chromatin immunoprecipitation (ChIP) assay revealed that YY1 bound at intron 7 of Kit. We conclude that PcG protein YY1 is essential for LT-HSCs self-renewal and lineage differentiation. YY1 maintains LT-HSCs in a quiescent state potentially by direct regulation of kit.
