Stage-specific Expression of Jmjd2 Histone Demethylases in Early Mouse Embryos.

Abstract:Dynamic chromatin modifications occur more frequently and have been proposed important for early embryonic development. The methylation modification on lysine residues in core histones has been considered as an irreversible process. Until recently, Jmjc domain containing proteins have been discovered as histone demethylases. Jmjd2 family is believed to specifically demethylate H3K9 and H3K36 methylation, whether this family participating in regulating early embryo development remains unknown. In the present study, we first analyzed the expression pattern of jmjd2 family in early mouse embryos by RT-PCR and quantitive real-time PCR. Oocytes and early embryos were collected from the B6D2F1 female mice. The results indicated that jmjd2a and jmjd2b expressed from MII to 2-cell stage, and then the mRNA transcripts decreased dramatically. Interestingly, jmjd2c expressed from 2-cell stage and reached the peak level at 4-cell stage, which expressed right after diminishing of jmjd2a and jmdj2b. From these results, we found that, jmjd2 family exhibited a stage-specific expression profile in mouse oocyte and early embryos. We further detected the expression profile of jmjd2 family in somatic cell nuclear transfer (SCNT)-embryos. Similar results were observed in SCNT embryos compared to normal embryos. Based on the stage-specific expression of jmjd2 demethylases in early embryos and other published reports, we suspected that oct4, nanog and other pluipotency maintaining genes might be regulated by jmjd2 family. We designed double-stand RNA(dsRNA) and shRNA specifically against jmjd2a to perform knockdown assay in occyte, early embryos and cell line respectively. We injected dsRNA into MII oocyte then parthenogenetic activation was performed, the embryos were collected and RNA extracted to perform real-time PCR 24h after dsRNA injection. We found that jmjd2a expression decreased 80% after injection and interestingly, Oct4, which is an important pluripotency marker gene, also decreased about 50%. This result was confirmed in p19 cell line by the knockdown assay through shRNA transfection. Besides Oct4 down-regulation, we detected the altered expression of some other important genes in mouse development, including nanog, sox2, klf4, and c-myc. We found that all these genes were up-regulated compared to down regulation of Oct4 in jmjd2a knockdown P19 cells. Particularly, klf4 increased about 7 folds. Klf4 is characterized as an oncogene and also a tumor repression gene. It can regulate cell cycle by activating p21 and inhibiting p53. So, we further detected the p53 expression in knockdown cell line. We found that expression of p53 was decreased about 80% after jmjd2a knockdown. Our study indicated that jmjd2 family, the newly discovered histone demthylases, was expressed in early embryos with stage-specific and they might play important roles in early embryo development by regulating some pluripotency genes. Conditional knockout mice have been generated and the function of jmjd2a in embryo development will be elucidated in future studies. [Supported by National High Technology Project 863 (2005AA210930)]
