Cellular metabolic changes in oncogene-induced senescence and its value in cancer diagnosis and treatment

Abstract:Mutation and activation of oncogenes are closely related to the tumorigenesis, but the activation of oncogenes may induce many changes in normal cells, such as DNA replication stress, DNA damage, increase in expressions of tumor suppressors p53 and p16, cell cycle arrest, and increase in activity of beta-galactose which is related to senescence. This phenomenon is called oncogene-induced senescence (OIS). The cells must overcome senescence as a barrier for tumor formation. More and more evidence has demonstrated that the change in metabolism of the cells is induced by OIS. This review summarizes the OIS-induced changes in glucose, nucleotide and lipid metabolism, and the application of cell metabolism-based strategy for diagnosis and treatment of tumors, which yields a prospect for precision-medicine.
