Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.

Abstract:How to construct three-dimensional oriented chitosan scaffolds with improved mechanical property is essential to further design bone-regenerative scaffolds. Materials with multi-layer structures involve excellent mechanical properties. In this research, a facile approach was adopted to construct three-dimensional oriented chitosan scaffolds. The chitosan gel with multi-layer structure was first prepared with in-situ precipitation, and then lyophilization was applied to obtain porous scaffolds. SEM images indicated that the porous scaffolds had spoke-like framework in cross-section and multi-layer structure in vertical-section. Compared with the disordered scaffolds prepared with lyophilization method, the scaffolds prepared with in-situ precipitation method showed a significantly improved compressive strength. With chitosan concentration of 4–5% and drying time of 60min, the scaffold showed the best comprehensive property with a suitable porosity and a high compressive strength. This novel porous scaffold with three-dimensional oriented structure might have potential application in bone tissue engineering.

Abstract:Bone regeneration scaffolds loaded with osteoblast-related cells or cytokines exhibit outstanding therapeutic potential during large-scale bone defect repair. However, limited sources of cells, opportune choosing of growth factors and their concentration, as well as immunological rejection, seriously hinder its clinical application. Developing a scaffold that can effectively recruit MSCs in situ and achieve endogenous bone regeneration is a viable strategy. Herein, we report a chitosan-calcium carbonate scaffold with high mineral content and centripetal pore arrangement using a simple in situ mineralization method. In vivo results first time demonstrate that the scaffold with high calcium carbonate content can effectively recruit MSCs near the defect area, induce their osteogenic differentiation, and ultimately accelerate the process of bone regeneration. Considering the accessible preparation and excellent osteogenicity, the chitosan-calcium carbonate scaffold possesses high potential for the therapeutics of massive bone defects.
Biodegradable and Biocompatible High Elastic Chitosan Scaffold is Cell-Friendly Both in Vitro and in Vivo.

Abstract:Biodegradable and biocompatible macromolecule chitosan has been favored for a variety of clinical applications. We reported herein the fabrication of a novel chitosan scaffold with high elasticity. This scaffold can be easily compressed and thus enable the insertion of such scaffold into surgical lesions during minimal invasive surgeries. In addition, this novel scaffold can restore its shape when released. We evidenced that this high elastic scaffold has better biocompatibility than traditional chitosan scaffold. Therefore, this new chitosan material might lead to the manufacture of a variety of novel biodegradable biomedical materials and devices.
Preparation and Characterization of Bionic Bone Structure Chitosan/hydroxyapatite Scaffold for Bone Tissue Engineering.

Abstract:Three-dimensional oriented chitosan (CS)/hydroxyapatite (HA) scaffolds were prepared via in situ precipitation method in this research. Scanning electron microscopy (SEM) images indicated that the scaffolds with acicular nano-HA had the spoke-like, multilayer and porous structure. The SEM of osteoblasts which were polygonal or spindle-shaped on the composite scaffolds after seven-day cell culture showed that the cells grew, adhered, and spread well. The results of X-ray powder diffractometer and Fourier transform infrared spectrometer showed that the mineral particles deposited in the scaffold had phase structure similar to natural bone and confirmed that particles were exactly HA. In vitro biocompatibility evaluation indicated the composite scaffolds showed a higher degree of proliferation of MC3T3-E1 cell compared with the pure CS scaffolds and the CS/HA10 scaffold was the highest one. The CS/HA scaffold also had a higher ratio of adhesion and alkaline phosphate activity value of osteoblasts compared with the pure CS scaffold, and the ratio increased with the increase of HA content. The ALP activity value of composite scaffolds was at least six times of the pure CS scaffolds. The results suggested that the composite scaffolds possessed good biocompatibility. The compressive strength of CS/HA15 increased by 33.07% compared with the pure CS scaffold. This novel porous scaffold with three-dimensional oriented structure might have a potential application in bone tissue engineering.

Abstract:A novel asymmetric membrane based on small intestinal submucosa (SIS) was fabricated. The microstructure, physicochemical properties, cytological characterization and effects on bone defect repair in vivo were investigated. Compared with a traditional SIS-based dense membrane, the asymmetric membrane had a bilayer structure with dense and loose layers, and better mechanical properties and wettability. The loose layer was favorable for the three-dimensional proliferation of human bone mesenchymal stem cells, and thus led to better osteogenic effects in vivo. Given its extensive material sources and simple preparation process, the asymmetric SIS membrane is expected to be a promising candidate for guided bone regeneration.

Abstract:Natural bone exhibits a complex anisotropic and micro-nano hierarchical structure, more importantly, bone extracellular matrix (ECM) presents liquid crystal (LC) phase and viscoelastic characteristics, providing a unique microenvironment for guiding cell behavior and regulating osteogenesis. However, in bone tissue engineering scaffolds, the construction of bone-like ECM microenvironment with exquisite microstructure is still a great challenge. Here, we developed a novel polysaccharide LC hydrogel supported 3D printed poly(l-lactide) (PLLA) scaffold with bone-like ECM microenvironment and micro-nano aligned structure. First, we prepared a chitin whisker/chitosan polysaccharide LC precursor, and then infuse it into the pores of 3D printed PLLA scaffold, which was previously surface modified with a polydopamine layer. Next, the LC precursor was chemical cross-linked by genipin to form a hydrogel network with bone-like ECM viscoelasticity and LC phase in the scaffold. Subsequently, we performed directional freeze-casting on the composite scaffold to create oriented channels in the LC hydrogel. Finally, we soaked the composite scaffold in phytic acid to further physical cross-link the LC hydrogel through electrostatic interactions and impart antibacterial effects to the scaffold. The resultant biomimetic scaffold displays osteogenic activity, vascularization ability and antibacterial effect, and is expected to be a promising candidate for bone repair.

Abstract:A novel long-bone-like Structural scaffold was fabricated by freeze-drying and alkalisation process. The structure of the hydroxyapatite/chitosan scaffolds was composed of outer dense shell and inner porous sponge with aperture of 300 similar to 500 mu m. An advantage of the scaffolds is that the dense shell could Support the main loading while it was pressed, and the inner pores and wherein cells and tissues Could be protected. The compressive strength and compressive Modulus were 6.85 MPa and 65.09 MPa, respectively. Hence, the scaffolds were suitable for bone replacement or bone tissue engineering.

Abstract:Degenerated cartilage tissues remain a burgeoning issue to be tackled, while bioactive engineering products available for optimal cartilage regeneration are scarce. In the present study, two-dimensional (2DS) poly(L-lactide-co-epsilon-caprolactone)/silk fibroin (PLCL/SF)-based scaffolds were fabricated by conjugate electrospinning method, which were then cross-linked with chondroitin sulfate (CS) to further enhance their mechanical and biological performance. Afterwards, three-dimensional (3D) PI.CL/SF scaffolds (3DS) and CS-crosslinked 3D scaffolds (3DCSS) with tailored size were successfully fabricated by an in-sini gas foaming in a confined mold followed by freeze-dried. Gas-foamed scaffolds displayed high porosity, rapid water uptake, and stable mechanical properties. While all of the scaffolds exhibited good cytocompatibility in vitro; 3DCSS showed better cell seeding efficiency and chondro-protective effect compared to other scaffolds. Besides, 3DCSS scaffolds supported the formation of more mature cartilage-like tissues along with the best repair outcome in a rabbit articular cartilage defect model in vivo, as well as less expression level of pro-inflammatory cytokines, including interleukin (IL)-1 beta and tumor necrosis factor (TNF)-alpha than that of the other groups. Taken together, 3DCSS may provide an alternative therapeutic option for cartilage tissue repair.
Fiber-Based Chitosan Tubular Scaffolds for Soft Tissue Engineering: Fabrication and in Vitro Evaluation

Abstract:Porous, two-ply tubular chitosan conduits for guided tissue regeneration were fabricated by combining the textile technique (inner layer) with the thermally induced phase separation process (outer layer). A hollow chitosan tube was prepared using an industrial warp knitting process with chitosan yarns. Then, an appropriate diameter mandrel was inserted into the pre-fabricated tube. The tube and the mandrel were dipped into the chitosan solution together, taken out, and freeze-dried. After being neutralized in alkaline solution and dried at room temperature, the mandrel was removed to create the chitosan tubular scaffold. Scanning electron micrographs show that the resulting tubes have a biphasic wall structure, with a fibrous inner layer and a semipermeable outer layer. The swelling properties and the mechanical strength before and after in vitro degradation were investigated. The biocompatibility of the scaffolds was also investigated by co-culturing neuroblastoma cells (N2A, mouse) with the scaffolds. The results suggest that these chitosan tubular scaffolds are useful for the regeneration of tissues requiring a tubular scaffold.

Abstract:The fabrication process for a novel three-dimensional (3D) chitosan/gelatin scaffold with predefined multilevel internal architectures and highly porous structures is presented combining solid freeform fabrication (SFF), microreplication and lyophilization techniques. The computer model of the scaffold is designed with biological data such as branching angle in liver vascular cast incorporated. Stereolithography (SL), known as a SFF technique, is utilized to build the resin mould, based on which poly-dimethylsilicone (PDMS) mould is produced by microreplication. The chitosan/gelatin hybrid solution is then cast onto the PDMS mould for pre-freeze and the monolayer porous structures with organized internal morphology are produced upon lyophilization. The 3D scaffold can be constructed via stacking these monolayer structures. The properties of porous structure, such as porosity, pore size and micromorphology as well as wall thickness, were investigated. Scanning electron microscopy (SEM) demonstrated that the scaffold possesses multilevel organized internal morphologies including vascular systems (portal vein, artery and hepatic vein) and parenchymal component (hepatocyte chamber). These organized structures enable orderly arrangement of hepatocyte and hepatic nonparenchymal cells and co-culture in the same 3D scaffold to guide liver regeneration in a controlled manner. Cell culture experiment in vitro showed that hepatocytes perform better in the well-defined chitosan/gelatin scaffold than in porous scaffold. This approach makes it flexible to investigate the relationship between internal scaffold microstructure and hepatocyte behavior in vitro. It also provides a new way to fabricate complex 3D scaffold using various natural biomaterials for vital organ engineering.
Patient-specific Scaffolds with a Biomimetic Gradient Environment for Articular Cartilage-Subchondral Bone Regeneration.

Abstract:Functional articular repair is known to be hampered by tissue degradation, which occurs in the defective local inflammatory environment that is also characterized by disrupted angiogenesis. The advanced fabrication of scaffolds with designed chemical and physical cues, which provide a biomimetic environment for tissue regeneration, holds considerable promise to circumvent the problem and thus allows functional articular repair. Herein, we developed scaffolds with controllable shapes with hydroxybutyl chitosan (HBC) and oxidized chondroitin sulfate (OCS) hydrogels, whose chemical composition was similar to that of the cartilage extracellular matrix (ECM). By optimizing the concentration of OCS, the functional cross-linker, we achieved a hydrogel promoting proliferation, adhesion, and ECM formation of chondrocytes and inhibiting tube formation of endothelial cells. Using a hydration procedure and bioactivation of mesenchymal stem cells (MSCs), we obtained mesoporous silicate-doped calcium phosphate cement (MS/CPC) scaffolds with a bioactive surface similar to that of bones, with improved osteogenesis and vascularization properties. Personalized cartilage-subchondral repair scaffolds with stable combination were successfully fabricated based on the self-cross-linking properties of the Schiff-based HBC/OCS hydrogel and the macroporous structure of MS/CPC scaffolds with the aid of a 3D printing technique. This study proposes a strategy to design individualized tissue repair biomimetic gradient scaffolds. Further assessments of their osteochondral defect repair properties in vivo should be performed.

Abstract:Chitosan-based porous scaffolds are of great interest in biomedical applications because of their biodegradability and biocompatibility. However, the poor mechanical properties of these scaffolds hinder their broad utility. In the present study, a novel compression method was developed to fabricate chitosan scaffolds with high mechanical strength and tuneable topography, based on the ionic strength and pH-dependent solubility of chitosan. When the compressive ratio increases from 1 to 8, the compressive elastic modulus of the scaffold increases from 5.2kPa to 520kPa and the porosity decreases from 94.1% to 82.5%. Furthermore, the number of human adipose-derived stem cells adhering to the scaffolds increases as the compressive ratio increases, owing to the high density of the chitosan fibres. This method does not require external cross-linker agent, sophisticated instrumentation and/or technical proficiency and could be extended to other polysaccharides.

Abstract:The development of guided bone regeneration (GBR) technique brings a promising alternative for bone defects and fracture healing. In this study, an asymmetric nano-hydroxyapatite/chitosan (n-HA/CS) composite GBR membrane was fabricated by means of solution-blending and solvent-evaporating in vacuum. The membranes were characterized using SEM, XPS and contact angle. It was found that the composite membrane displayed an asymmetric structure, in which the upper surface was CS and the under surface was a complex of n-HA and CS, and some interactions between n-HA and CS were also confirmed to exist. The contact angle testing showed that the under surface was more hydrophilic than the upper surface. The in vivo experiments demonstrated that the asymmetric composite membrane had the ability to make osteoblasts mineralize and promote loose bone calcified, and then accelerate the bone regeneration. Compared with CS membrane, the asymmetric composite membrane displays a better bone regeneration ability and is suitable for GBR membrane.

Abstract:Repair of bone defects with irregular shapes or at soft tissue insertion sites faces a huge challenge. Scaffolds capable of adapting to bone cavities, generating stiffness gradients, and inducing osteogenesis are necessary. Herein, a superelastic 3D ceramic fibrous scaffold is developed by assembly of intrinsically rigid, structurally flexible electrospun SiO2 nanofibers with chitosan as bonding sites (SiO2 NF-CS) via a lyophilization technique. SiO2 NF-CS scaffolds exhibit excellent elasticity (full recovery from 80% compression), fast recovery rate (>500 mm min(-1)), and good fatigue resistance (>10 000 cycles of compression) in an aqueous medium. SiO2 NF-CS scaffolds induce human mesenchymal stem cell (hMSC) elongation and differentiation into osteoblasts. In vivo self-fitting capability is demonstrated by implanting compressed SiO2 NF-CS scaffolds into different shaped mandibular defects in rabbits, with a spontaneous recovery and full filling of defects. Rat calvarial defect repair validates enhanced bone formation and vascularization by cell (hMSC) histomorphology analysis. Further, subchondral bone scaffolds with gradations in SiO2 nanofibers are developed, leading to a stiffness gradient and spatially chondrogenic and osteogenic differentiation of hMSCs. This work presents a type of 3D ceramic fibrous scaffold, which can closely match bone defects with irregular shapes or at different implant sites, and is promising for clinical translation.

Abstract:An ideal bone repair scaffold is expected to possess superior architectural characteristics to facilitate the adhesion, proliferation, and migration of bone-repair-related cells, while excluding nonosteogenic cells and fibrous tissues from interfering with normal bone regeneration. Unfortunately, such scaffold material has rarely been reported. Herein, nanocomposite scaffolds with a radially ordered porous structure are presented, manufactured using a modified directional freeze-casting method, and are promising bone defect repair materials to satisfy this requirement. The prepared nanocomposite scaffolds consist of a natural bio-macromolecule, chitosan, and bioactive hydroxyapatite nanoparticles derived from porcine cortical bone, demonstrating favorable biocompatibility and biological functions. Both in vitro cell studies and in vivo animal studies reveal the great superiority of the radially oriented porous structure of the scaffolds in guiding bone regeneration, while simultaneously preventing the invasion of surrounding nonosteogenic cells and fibrous tissue, compared to the axially oriented porous structure. This work indicates the distinctive potential of radially oriented porous scaffolds for repairing tabular and lacunar bone defects.

Abstract:Cartilage is severely limited in self-repair after damage, and tissue engineering scaffold transplantation is considered the most promising strategy for cartilage regeneration. However, scaffolds without cells and growth factors, which can effectively avoid long cell culture times, high risk of infection, and susceptibility to contamination, remain scarce. Hence, we developed a cell- and growth factor-dual free hierarchically structured nanofibrous sponge to mimic the extracellular matrix, in which the encapsulated core-shell nanofibers served both as mechanical supports and as long-lasting carriers for bioactive biomass molecules (glucosamine sulfate). Under the protection of the nanofibers in this designed sponge, glucosamine sulfate could be released continuously for at least 30 days, which significantly accelerated the repair of cartilage tissue in a rat cartilage defect model. Moreover, the nanofibrous sponge based on carboxymethyl chitosan as the framework could effectively fill irregular cartilage defects, adapt to the dynamic changes during cartilage movement, and maintain almost 100 % elasticity even after multiple compression cycles. This strategy, which combines fiber freeze-shaping technology with a controlled-release method for encapsulating bioactivity, allows for the assembly of porous bionic scaffolds with hierarchical nanofiber structure, providing a novel and safe approach to tissue repair.
Construction of Chitosan Scaffolds with Controllable Microchannel for Tissue Engineering and Regenerative Medicine.

Abstract:Microchannels are effective means of enabling the functional performance of tissue engineering scaffolds. Chitosan, a partial deacetylation derivative of chitin, exhibiting excellent biocompatibility, has been widely used in clinical practice. However, development of chitosan scaffolds with controllable microchannels architecture remains an engineering challenge. Here, we generated chitosan scaffolds with adjustable microchannel by combining a 3D printing microfiber templates-leaching method and a freeze-drying method. We can precisely control the arrangement, diameter and density of microchannel within chitosan scaffolds. Moreover, the integrated bilayer scaffolds with the desired structural parameters in each layer were fabricated and exhibited no delamination. The flow rate and volume of the simulated fluid can be modulated by diverse channels architecture. Additionally, the microchannel structure promoted cell survival, proliferation and distribution in vitro, and improved cell and tissue ingrowth and vascular formation in vivo. This study opens a new road for constructing chitosan scaffolds, and can further extend their application scope across tissue engineering and regenerative medicine.

Abstract:In this study, a novel method was developed to create porous tubular scaffolds with desirable mechanical properties and controllable inner structure from chitosan, for nerve tissue engineering. Chitosan fiber-based yarns were first used to create porous hollow tubes, which served as the outer wall of the scaffolds, through an industrial knitting process. Then, an innovative molding technique was developed and used to produce inner matrices with multiple axially oriented macrochannels and radially interconnected micropores. Acupuncture needles were used as mandrels during molding to improve the safety and controllability of the process. In vitro characterization demonstrated that the scaffolds possessed suitable mechanical strength, porosity, swelling, and biodegradability for applications in nerve tissue engineering. In vitro cell culture experiments showed that differentiated Neuro-2a cells grew along the oriented macrochannels and the interconnected micropores were beneficial for nutrient diffusion and cell ingrowth to the scaffold's interior. Collectively, the well-defined architectural features in addition to the desirable mechanical and biological properties of the scaffolds make them promising for nerve tissue engineering.
