Abstract:A metal-free process for the C-3 alkylation of imidazopyridines have been developed. Various alkylation products including alkyl ester-, cyano-, ketone- and amide-substituted imidazopyridines were prepared in good yields.

Abstract:A general and straightforward method for the regioselective construction of C-3 heteroaryl-containing imidazo[1,2-a]pyridines via cross-dehydrogenative coupling under transition-metal-free conditions has been reported, utilizing N,N-dimethylaniline as the methylenation source and furnishing the C(sp(2))-C(sp(3)) functionalized products in good to excellent yields. Mechanism studies indicate that a radical pathway is responsible for this transformation.

Abstract:Molecular oxygen‐mediated C‐H bond alkylation of cyclic aldimines has been reported, which avoids the use of transition metals and additional oxidants. This method allows straightforward access to a series of cyclic aldimine derivatives through a free radical course. Furthermore, this transformation features mild conditions, excellent yields and good functional group tolerance.

Abstract:A metal-free sequential dual oxidative amination of C(sp(3))-H bonds under ambient conditions was the first developed, affording imidazo[1,5-a]pyridines in good to excellent yields. The reaction was involved in two oxidative C-N couplings and one oxidative dehydrogenation process with six hydrogen atoms removed.

Abstract:Electrochemical oxidative cross-coupling represents one of the most atom-economical and green approaches to synthesize various useful compounds. In this work, an electrochemical C3-sulfonylation of imidazo[1,2-a]pyridines with sodium benzenesulfinates as sulfonylation reagents has been developed. This transformation provides an effective and straightforward metal-free protocol towards the synthesis of biologically and synthetically useful 3-(arylsulfonyl)imidazo[1,2-a]pyridine. Up to 94% yields were obtained. The scale-up experiment gave 1.56 g of the desired product in 90% yield.

Abstract:The direct oxidative N-acylation reaction of primary amides with aryl/alpha,beta-unsaturated aldehydes was achieved in the presence of azolium salt C3 and an inorganic base using 3,3',5,5'-tetra-tert-butyldipherioquinone as the oxidant, thus providing an efficient approach for the synthesis of three types of imide compounds including N-sulfonylcarboxamides, N-sulfinylcarboxamides, and dicarboxyimides in good yield.

Abstract:In the presence of KI and (diacetoxyiodo)benzene (PIDA), primary amines bearing an alpha-hydrogen can be efficiently converted to fluoroalkylated imidazoles by reacting with fluoroalkylated alkynes and sodium azide via a cascade Michael addition/azidation/cycloamination process. KI plays a crucial role in this reaction by generating iodine(I) species to promote the cyclization. Substrate scope was explored, and the application of this protocol was demonstrated. Additionally, a plausible reaction mechanism was proposed.

Abstract:Alkynyl amides play crucial roles in organic synthesis in the production of bioactive compounds and valuable heterocycles. Despite numerous studies on their synthesis, challenges persist due to the necessity of harsh or hazardous conditions and the use of costly or unstable reagents. Herein, we present a one-pot method for the synthesis of all three bonds of the alkyne under transition-metal free conditions. An important feature of this chemistry is the use of readily available feedstock chemicals, such as methyl esters and acetamides. This approach offers efficient access to a wide range of aryl and alkyl alkynyl amides and demonstrates excellent tolerance towards various functional groups in a sustainable and cost-effective manner.

Abstract:The one-pot sequential reaction of various amines, carbodiimides, and acyl dichlorides has been achieved for the first time under metal-free conditions to provide symmetric cyclic di-oxoguanidines via an unexpected 2,2-dichloro-imidazolidindione intermediate. Acyl dichlorides have a dual function: to serve as the third component and to activate carbodiimides. In sharp contrast, the AlMe(3)-catalyzed sequential reaction from the same substrates gives the isomer.

Abstract:A novel and convenient transformation for the regiospecific synthesis of functionalized imidazo[1,2-a]pyridine aldehydes/ketones and 3-vinyl imidazo[1,2-a]pyridines has been developed via copper(I)- and palladium(II)-catalyzed cyclization. The one-pot reaction proceeds smoothly with commercially available catalysts and affords the products in moderate to good yields. It represents an efficient approach for the formation of C-N, C═O, and C═C bonds under mild conditions.

Abstract:A novel, efficient, and practical method for the synthesis of imidazopyridine derivatives has been developed through the copper-catalyzed aerobic oxidative CH functionalization of substituted pyridines with N-(alkylidene)-4H-1,2,4-triazol-4-amines. The procedure occurs by cleavage of the NN bond in the N-(alkylidene)-4H-1,2,4-triazol-4-amines and activation of an aryl CH bond in the substituted pyridines. This is the first example of the preparation of imidazopyridine derivatives by using pyridines as the substrates by transition-metal-catalyzed CH functionalization. This method should provide a novel and efficient strategy for the synthesis of other nitrogen heterocycles.

Abstract:A metal-free and oxidant-free electrochemically enabled strategy for C-3 formylation of imidazopyridines using trimethylamine as a one-carbon source has been established. This conversion has high functional group compatibility under mild conditions and ensures late-stage functionalization of pharmaceutical molecules. Furthermore, unexpected hexafluoroisopropoxylation products have been observed in some cases. Mechanistic studies using cyclic voltammetry and control experiments reveal the key intermediate of the formylation process.

Abstract:Herein, a multi‐component tandem arylation reaction was established under transition‐metal‐free conditions to synthesize structurally diverse imidazo[1,2‐a]pyridines. As N,N‐dimethylaniline is not only a reactant but also a methylation reagent, its use affords an alternative route for methylation. This approach features mild reaction conditions and tolerant functional group compatibility, providing 29 compounds in acceptable yields.

Abstract:A simple, efficient, and environmentally benign method for the direct C-3 alkoxycarbonylation of 2H-indazoles using alkylcarbazates has been developed under metal-free conditions at room temperature. This current protocol represents a facile access to C-3-carboxylic ester-derived 2H-indazoles with wide functional group tolerance in good to excellent yields. The mechanistic studies suggest that the reaction proceeds through a radical pathway.

Abstract:The addition reaction of alkynes to N-heterocycles by simply heating in DMSO with potassium phosphate is reported. Good yields with high stereoselectivity could be achieved for a range of substrates. The scope is quite general for both amines and phenylacetylenes. In addition, internal alkynes and α-bromostyrene were also examined in this reaction. This process is efficient and useful for the synthesis of (Z)-N-(1-alkenyl)imidazoles and related Z products. Thus, the reaction is useful because of the importance of the imidazole scaffold.
