Abstract:A facile and sensitive detection of L-cysteine is essential for biosensors, environmental monitoring and medical diagnosis. In this study, a simple approach has been demonstrated to prepare Prussian blue (PB)/polypyrrole ( PPy) hybrid nanofibers as efficient peroxidase-like mimics for the determination of L-cysteine. alpha-Fe2O3 nanofibers act as both templates and a source of Fe3+ to polymerize pyrrole in an acidic environment, resulting in the formation of PB/PPy hybrid nanofibers. The as-prepared PB/PPy hybrid nanofibers show a superior catalytic activity as peroxidase mimics over alpha-Fe2O3 nanofibers. Due to the high catalytic efficiency of the resultant PB/PPy hybrid nanofibers, we have developed a simple colorimetric approach for the rapid and sensitive determination of L-cysteine with a detection limit of about 16 nM over a wide linear range from 1 to 20 mu M. In addition, the detection system displays a high selectivity toward L-cysteine over other amino acids, glucose, and common ions. This work provides a new strategy for the preparation of peroxidase-like nanomaterials with improved catalytic activity, which shows great promise for application in highly sensitive detection.

Abstract:Nanozymes have attracted much attention as a new type of artificial enzyme with great development potential in recent years. Here, cubic Co and N co-doped porous carbon material (Co-HPNC@NC) was synthesized through the calcination of PDA-modified bimetallic Co-Zn ZIFs. Co-HPNC@NC exhibits exceptionally high oxidase-mimicking activity and effectively oxidize the colorless 3,3',5,5'-tetramethylbenzidine (TMB) to blue-colored oxidized TMB (oxTMB) without H2O2. However, the oxTMB was inhibited by the addition of L-cysteine (L-Cys). Therefore, the blue color faded to colorless, causing a notable decrease in absorbance. Furthermore, Co-HPNC@NC showed good selectivity and anti-interference ability toward the detection of L-Cys. As a result, a sensitive and efficient colorimetric method has been developed to determine L-Cys with a low detection limit of 0.17 μM (3σ). It is believed that this colorimetric method for the detection of L-Cys based on Co-HPNC@NC will show potential applications in biosensors and medical diagnosis.

Fabrication of ternary MoS2-polypyrrole-Pd nanotubes as peroxidase mimics with a synergistic effect and their sensitive colorimetric detection of l-cysteine

Abstract:Over the past few years, nanomaterials-based enzymatic mimics have attracted tremendous attention due to their excellent catalytic activity and environmental stability. In this work, ternary MoS2-polypyrrole (PPy)-Pd nanotubes have been prepared through a hydrothermal reaction and in situ redox polymerization process between pyrrole monomer and Na2PdCl4. The prepared MoS2-PPy-Pd nanotubes exhibited a higher peroxidase-like catalytic activity than individual MoS2, MoS2-PPy, PPy-Pd and MoS2-Pd nanocomposites due to the synergistic catalytic effect between the three components. The catalytic kinetic of MoS2-PPy-Pd nanotubes follows Michaelis-Menten behaviors, exhibiting a good affinity to both 3,3',5,5'-tetramethylbenzidine (TMB) and H2O2 substrates. On the basis of high peroxidase-like catalytic efficiency of the MoS2-PPy-Pd nanotubes, a simple and convenient colorimetric strategy for the rapid and sensitive detection of l-cysteine with a detection limit of 0.08 μM has been developed. In addition, a high selectivity towards the detection of l-cysteine is achieved. This work present an opportunity of the prepared ternary MoS2-PPy-Pd nanotubes for promising potential applications in biosensing, environmental monitoring, and medical diagnostics.
Ultrasmall Pt Nanozymes Immobilized on Spherical Polyelectrolyte Brushes with Robust Peroxidase-like Activity for Highly Sensitive Detection of Cysteine

Abstract:Distinct platinum (Pt) nanozymes as peroxidase mimics have received extensive interest owing to their outstanding catalytic activity, high environmental tolerance, lower consumption, and great potential in replacing natural enzymes. However, easy agglomeration of Pt nanoparticles (Pt NPs) resulting from the high surface free energy significantly decrease their peroxidase-like activity. Herein, spherical polyelectrolyte brush (SPB)-stabilized ultrasmall Pt NPs (SPB@Pt NPs) were prepared by a novel synthetic strategy where the SPB not only performed as a nanoreactor for the synthesis of ultrasmall Pt NPs but also greatly stabilized Pt NPs against aggregation. The well-defined SPB@Pt NP nanozymes exhibited outstanding peroxidase-like activity for the catalytic oxidation of colorless 3,3 ',5,5 '-tetramethylbenzidine (TMB) to blue oxidized TMB and were then used to establish a colorimetric sensor for rapid detection of cysteine, giving a limit of detection of 0.11 mu M. Moreover, the colorimetric detection system was demonstrated with outstanding performance in sensitive and selective detection of cysteine in the presence of several interference molecules. From these results, SPB@Pt NPs have been regarded as promising peroxidase mimics for a large number of applications such as in biosensing, biomedicine, the food industry, and environmental chemistry.

Abstract:Although nanozyme-based colorimetric assays have been broadly used for biosensing, some limitations such as low catalytic activity of nanozyme, poor sensitivity to analytes and lack of understanding the structure-activity relationship remain unsolved. In this work, we developed an ultrasensitive colorimetric method for biothiols detection based on density functional theory-assisted design of janus Pd-Fe3O4 nanozyme. The Pd-Fe3O4 dumbbell-like nanoparticles (DBNPs) prepared by seed-mediated approach shows a uniform heterodimeric nanostructure. Ultrasensitive biothiols detection is achieved from two aspects. On one hand, due to the synergistic effect between Pd and Fe3O4 in the dumbbell structure, Pd-Fe3O4 DBNPs show enhanced peroxidase-mimic activity compared to the individual components. On the other hand, when the target biothiols molecule is present, its inhibition effect on the janus Pd-Fe3O4 nanozyme is also significantly enhanced. The above results are confirmed both in experiment and theoretical calculation. Based on the rational design, a simple, highly selective and urtrasensitive colorimetric and quantitative assay for biothiols is developed. The limit of detection (LOD) can reach as low as 3.1 nM in aqueous solution. This assay is also successfully applied to the detection of biothiols in real urine samples. Moreover, the Pd-Fe3O4 nanozyme is used to discriminate biothiols levels in normal and cancer cells with high sensitivity at the cell density of 15,000/mL, which demonstrates its great potential in biological and clinical analysis. This work not only shows the great promise of janus bimetallic nanozymes' excellent functionalities but also provides rational guidelines to design high-performance nanozymes for biosensing and biomedical applications.

Abstract:We present a simple, sensitive, and specific colorimetric using the peroxidase properties method based on Pt doped WO3 nanosheets to detect the cysteine. Pt@WO(3)NSs were synthesized by hydrothermal method and characterized by Fourier transform infrared (FTIR), Transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDX), and X-ray diffraction patterns (XRD) methods. The response surface methodology (RSM) method based on the central composite design (CCD) was used to optimize test parameters such as pH, nanosheet concentration, and temperature. When cysteine is present in the environment due to its competition with 3,3', 5,5'-Tetramethylbenzidine (TMB) in the use of hydrogen peroxide, the blue discoloration is reduced compared to the absence of cysteine and leads to its detection. We have favorably created a peculiar approach for sensing cysteine based on the colorimetric method in solution and paper with linear range 0.01-15 mu M, 0.005-14 mu M and R-2 = 0.9887 and R-2 = 0.9871 respectively. The detection limit for solution-based is 1.2 nM and for paper-based is 1 nM.
Tailoring cysteine detection in colorimetric techniques using Co/Fe-functionalized mesoporous silica nanoparticles

Abstract:Over the past decade, there has been a dramatic increase in the number of studies focused on sensors for cysteine (Cys) as a crucial factor in physiological function and disease diagnosis. Among those sensors, nanomaterial-based peroxidase mimetics have received particular attention from researchers. This study introduces a new series of mesoporous silica nanoparticles (MSNs) incorporated with iron and cobalt (Co/Fe-MSN) with a molar ratio of Si/Fe = 10 and cobalt species at 1, 3, and 5 wt% that have great potential in the sensing application. These nanomaterial characterization was investigated by FTIR spectroscopy, SEM, TEM, XRD, and nitrogen adsorption-desorption. The peroxidase activity of these nanomaterials was studied through kinetic analysis. The findings revealed that Co/Fe-MSN (1%) showed higher peroxidatic activity than the others towards the common chromogenic substrate 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) diammonium salt. Based on the enzymatic activity of Co/Fe-MSN (1%), a colorimetric sensing platform was designed to detect H2O2 and Cys. The limit of detection (LOD) for H2O2 and Cys was determined to be 1.1 μM and 0.89 nM, respectively. The results indicated that the proposed enzyme mimic exhibited excellent potential as a sensor in medical diagnostics and biological systems.
Colorimetric Sensing of Bithiols Using Photocatalytic UiO-66(NH2) As H2O2-free Peroxidase Mimics

Abstract:A facile colorimetric sensing method for biothiols was developed, based on photocatalytic property of metal-organic frameworks (MOFs), UiO-66(NH2) nanoparticles (NPs), as peroxidase mimics under light irradiation. By the irradiation of a light emitting diode (LED) source, the colorless chromogenic substrate, 3,3',5,5'-tetramethylbenzydine (TMB), was oxidized into blue oxTMB with the aid of the catalytic UiO-66(NH2) NPs. With the existence of biothiols, the oxidization was prohibited, with the blue color paled and absorbance intensity decreased with the concentration of biothiols in a linear manner. Real samples of cysteine, glutathione, and homocysteine were analyzed under the optimized conditions, with high sensitivity (the limit of detection was calculated as 306nM, 310nM, and 330nm respectively) and selectivity. The recovery ranged from 93% to 107% with good precisions (RSD%≤5%). This photocatalytic property of UiO-66(NH2) as peroxidase mimics was studied based on steady-state kinetics, and the mechanism of oxidization process was also briefly discussed. This developed MOFs-based colorimetric sensing method demonstrated advantages over others for biothiols sensing, including high photo-catalytic activity compared to other nanomaterials, oxidation without H2O2, ease of regulation with the LED source, and low cost without expensive instrument and technically demanding.

Abstract:In the present work, a simple and sensitive colorimetric sensing strategy based on Cu-MOF nanosheet for the detection of cysteine (Cys) is developed. Cu-MOF can catalyze the color change of the H2O2-TMB detection system from colorless to blue, and produce an obvious UV absorption peak at 652 nm. In the presence of Cys, it can effectively inhibit the color development of H2O2-TMB. Under optimal conditions, the absorbance value at 652 nm and the Cys concentration range from 0.2 mM to 2.8 mM show a linear relationship. The detection limit of the sensor is 0.03 mM, and it can effectively distinguish Cys from 9 other amino acids and its metabolites. In addition, we also assessed the Cys content in milk, which has practical significance for detection.

Abstract:In order to establish a rapid, accurate, and simple method for monitoring and analyzing the biological physiological state, we constructed a colorimetric sensor array based on the peroxidase-like activity of nanozymes (CoMoO4 and Por-CoMoO4) for the determination of five kinds of biothiols. The peroxidase-like activity of CoMoO4 has been greatly enhanced by the introduction of porphyrin (Por), due to the synergistic interaction between porphyrin and CoMoO4. Experimental data of the radical scavenger and fluorescence probe verify the generation of a superoxide radical (•O2 –) during the catalytic reaction. Because different biothiols have different inhibiting effects on the catalytic activity of two nanozymes, each biothiol has a unique fingerprint with the aid of a chromogenic substrate (TMB). Through linear discriminant analysis, captopril (CAP), cysteine (Cys), homocysteine (Hcy), dithiothreitol (DTT), and glutathione (GSH) can be well identified in a certain range of concentrations. Different concentrations of biothiols and different proportions of mixed biothiols can be identified. It is expected that this study will provide a simple and rapid method for the identification of structural analogues, contributing to the monitoring of human life and health.

Colorimetric Determination of Cysteine Based on Inhibition of GSH-Au/Pt NCs As Peroxidase Mimic

Abstract:In this work, we reported a facile and highly sensitive strategy for colorimetric detection of cysteine (Cys) based on the inhibition of catalytic activity of bimetallic nanoclusters induced by Cys. Glutathione-modified gold-platinum nanoclusters (GSH-Au/Pt NCs) with different Au/Pt molar ratios were prepared via one-pot approach and utilized as peroxidase mimics to catalyze the oxidation of 3,3',5,5'-tetramethyl benzidine (TMB) by H2O2. It has been found that Cys could inhibit the peroxidase-like activity of GSH-Au/Pt NCs efficiently, which leads to a decrease of the absorption intensity of the system at 652 nm with a fading of the blue color. These findings provide a worthy method for visualization and quantitative detection of Cys with different concentrations in the range from 0.5 to 30 mu M, and the detection limit is 0.154 mu M. Abstract:Biological thiols play a critical role in biological processes and are involved in a variety of diseases. The discrimination detection of biological thiols is of increasing importance in clinical diagnosis. In this paper, a novel nanosensor was developed to discriminate cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH) with multiple signals: colorimetric, photoluminescence (PL), and up-conversional photoluminescence (UCP). The nanosensor (NC-dots/AuNPs) was constructed by nitrogen-doped carbon dots (NC-dots) and gold nanoparticles (AuNPs) through assembling NC-dots "shell" on AuNPs and showed the obvious different response to Cys, Hcy, and GSH with colorimetric, PL, and UCP signals. The discrimination effect for Cys is originated from conformations and interaction difference of the thiols groups in Cys and Hcy and/or GSH with AuNPs. Among them, only Cys can quickly penetrate into the NC-dots "shell" of the composite and induce the dispersing of the aggregated NC-dots/AuNPs, which lead to the color change from purple to red and the recovery of PL and UCP of NC-dots. This assay was successfully applied for the detection of Cys in human serum with the detection limit of 4 nM.

Abstract:Many diseases in the human body are related to the level of L-cysteine. Therefore, it is crucial to establish an efficient, simple and sensitive platform for L-cysteine detection. In this work, we synthesized platinum palladium bimetallic nanoparticles (Van-Ptm/Pdn NPs) using vancomycin hydrochloride (Van) as a stabilizer, which exhibited high oxidase-like catalytic activity. In addition, the catalytic kinetics of the Van-Pt1/Pd1 NPs followed the typical Michaelis–Menten equation, exhibiting a strong affinity for 3,3′,5,5′-tetramethylbenzidine substrates. More importantly, we developed a simple and effective strategy for the sensitive colorimetric detection of L-cysteine using biocompatible Van-Pt1/Pd1 NPs. The detection limit was low, at 0.07 μM, which was lower than the values for many previously reported enzyme-like detection systems. The colorimetric method of the L-cysteine assay had good selectivity. The established method for the detection of L-cysteine showed promise for biomedical analysis.

Abstract:Biothiol assays offer vital insights into health assessment and facilitate the early detection of potential health issues, thereby enabling timely and effective interventions. In this study, we developed ultrasmall CuMn-Histidine (His) nanozymes with multiple enzymatic activities. CuMn-His enhanced peroxidase (POD)-like activity at neutral pH was achieved through hydrogen bonding and electrostatic effects. In addition, CuMn-His possesses laccase (LAC)-like and superoxide dismutase (SOD)-like activities at neutral pH. Based on three different enzyme mimetic activities of CuMn-His at neutral pH, the colorimetric sensing array without changing the buffer solution was successfully constructed. The array was successfully used for the identification of three biothiols, glutathione (GSH), cysteine (Cys), and homocysteine (Hcy). Subsequently, excellent application results were shown in complex serum and cellular level analyses. This study provides an innovative strategy for the development of ultrasmall bimetallic nanozymes with multiple enzymatic activities and the construction of colorimetric sensing arrays.

Abstract:Covalent organic frameworks (COFs), as a novel porous organic polymer with periodic and highly ordered structure, are ideal carrier matrix for metal nanoparticles due to high specific surface area, good stability, controllable pore size, and structural tunability. In this work, COFs are used as a carrier to in-situ grow ultrasmall rhodium nanoparticles (Rh NPs, ∼2.4 nm), which are uniformly distributed in the pores and on the surfaces of the COFs. The formed composite (COF-Rh) shows excellent peroxidase-mimetic activity benefiting from the good catalytic activity of ultrafine and highly dispersed Rh NPs as well as the high affinity of COFs to organic molecules (i.e., catalytic substrates). Cysteine (Cys) can inhibit the peroxidase-like activity of COF-Rh due to the interaction of -SH in Cys with Rh and the reduction of oxidized peroxidase substrate by Cys. By regulating the peroxidase-like activity of the system, a colorimetric method is successfully developed for Cys detection. Using smartphone as a readout, a portable strategy is further proposed for rapid and visual sensing of Cys.
Nanozyme based on CoFe2O4 modified with MoS2 for colorimetric determination of cysteine and glutathione

Abstract:A nanozyme based on CoFe2O4 modified with MoS2 was constructed for colorimetric determination of cysteine (Cys) and glutathione (GSH). Firstly, ferrite CoFe2O4 is synthesized, and it is then modified by MoS2 to form a flower-like polymer (MoS2@CoFe2O4). In the presence of H2O2, a redox interaction takes place, and the resulting hydroxyl promoted a colorimetric conversion from colorless to blue in the presence of 3,3′,5,5′-tetramethylbenzidine (TMB). However, once Cys or GSH is added, they are capable to compete with the interaction of the hydroxyl with TMB, resulting in an inhibition of the colorimetric conversion. The colorimetric distinction is sensitive to the amount of target. The results obtained proved that the catalytic efficiency of MoS2@CoFe2O4 is 4.4-fold and 1.8-fold to that of MoS2 and CoFe2O4. Meanwhile, the Km values to TMB and H2O2 are 0.067 and 0.048 mM, respectively, which are 6.5-fold and 77-fold, respectively smaller than those of natural peroxidase such as HPR. This indicates that the MoS2@CoFe2O4 possesses a favorable interaction affinity. Additionally, the colorimetric distinction caused by the competition between TMB and cysteine or glutathione is obvious. The signal responses to cysteine and glutathione are linear in the range 0.5~15 μM and 0.5~35 μM, and the LODs are 0.10 and 0.21 μM, respectively. In practical assay of Cys in serum, the RSD of the sample tests is 4.6%, and the recoveries for the spiked assays are 95.3% and 96.0% with the RSD of 2.1% and 4.2%, respectively.

Abstract:A H2O2-free colorimetric protocol based on urchin-like Au@Pt nanoparticles (Au@Pt NPs) has been developed for the sensitive and selective determination of cysteine (Cys). We verified the intrinsic oxidase-like activity of the Au@Pt NPs. They can act as artificial mimic oxidases to catalyse the oxidization of 3,3′,5,5′-tetramethylbenzidine (TMB) with the assistance of dissolved oxygen, avoiding the use of H2O2 in the colorimetric determination of Cys. In addition, the discrimination of Cys from the other two biothiol analogues, homocysteine and glutathione, can be easily realized through a simple ageing process. HNO3 is added to this colorimetric system to terminate the reaction by oxidizing ox-TMB (oxidized form of TMB) to diphenoquinone (DPQ), thus generating a characteristic absorption peak of DPQ at 450 nm. By recording the absorbance at 450 nm, interference from the aggregated Au@Pt NPs (absorption peak at 670 nm) when 650 nm (the characteristic absorption peak of ox-TMB) is used as the absorption wavelength can be eliminated. We investigated this H2O2-free colorimetric protocol and obtained high sensitivity, with a detection limit of 1.5 nM and relatively high selectivity. The analytical performance for real samples was further explored. The Au@Pt NP–based H2O2-free colorimetric protocol is of great significance for the sensitive and selective determination of Cys in practical samples in different scenarios.Graphical abstract

Abstract:In this work, PtCo bimetallic nanoparticles supported on multi-walled carbon nanotubes (PtCo@MWCNTs) nanohybrid was prepared simply and used for the first time as a novel nanozyme in the colorimetric sensing of L-cysteine (L-Cys) and Cu 2+ . Due to its strong enzyme-like catalytic activity, the prepared PtCo@MWCNTs nanohybrid can catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) to form ox-TMB without H 2 O 2 . Interestingly, the oxidase-like active of PtCo@MWCNTs was effectively suppressed by L-Cys, which could reduce ox-TMB to colorless TMB and lead to a pronounced blue fading, and the absorbance at 652 nm gradually decreased with increasing L-Cys concentration. On the other hand, the nanozyme activity of PtCo@MWCNTs can be recovered due to the complexation between L-Cys and Cu 2+ . Therefore, a colorimetric method based on PtCo@MWCNTs nanozyme was established to detect L-Cys and Cu 2+ . The results show that the assay platform has simple, rapid, sensitive performance and good selectivity. The detection limits for L-Cys and Cu 2+ are 0.041 μM and 0.056 μM, respectively, coupled with the linearities of 0.01 ~ 60.0 μM and 0.05 ~ 80.0 μM. The successful first application of PtCo bimetal-based nanozyme in colorimetric sensing herein opens a new direction for nanozyme and colorimetric analysis, showing great potential applications.
