Abstract:A series of amphiphilic pH-sensitive hybrid polypeptide copolymers, poly(ethylene glycol)-b-poly-(L-lysine)-b-poly(L-phenylalanine) (PEG-PLL-PLP) were synthesized. The copolymers could self-assemble into micelles with PLP as the hydrophobic core and PEG-PLL as the hydrophilic shell, as evidenced by (HNMR)-H-1 and TEM. These micelles exhibited obvious pH response in hydrodynamic diameter and pH-dependent drug release behavior, attributed to the protonation/deprotonation of amino groups in PLL segments. The copolymers could further condense plasmid DNA efficiently. Importantly, the polymer/DNA complexes showed high transfection efficiency in 293T cells under optimized conditions. This study suggested the copolymers may have great potential in both drug and gene delivery.

Abstract:Self-assembled peptide nanofibers have been widely studied in cancer nanotherapeutics with their excellent biocompatibility and low toxicity of degradation products, showing the significant potential in inhibiting tumor progression. However, poor solubility prevents direct intravenous administration of nanofibers. Although water-soluble peptide precursors have been formed via the method of phosphorylation for intravenous administration, their opportunities for broad in vivo application are limited by the weak capacity of encapsulating drugs. Herein, we designed a novel restructured reduced glutathione (GSH)-responsive drug delivery system encapsulating doxorubicin for systemic administration, which achieved the intracellular restructuration from three-dimensional micelles into one-dimensional nanofibers. After a long blood circulation, micelles endocytosed by tumor cells could degrade in response to high GSH levels, achieving more release and accumulation of doxorubicin at desired sites. Further, the synergistic chemotherapy effects of self-assembled nanofibers were confirmed in both in vitro and in vivo experiments.

Abstract:A series of poly(l-lactide)-grafted gelatin (Gel-g-PLLA) copolymers are synthesized by coupling the amino groups of gelatin with different molar masses and the carboxyl endgroup of poly(l-lactide) in the presence of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride and N-hydroxysuccinimide. Fourier transform infrared and nuclear magnetic resonance (H-1 NMR) data confirm the successful bonding between gelatin and PLLA. Self-assembled micelles of copolymers are prepared by using the direct dissolution method. The size and micellar morphology are determined from dynamic light scattering as well as transmission electron microscopy measurements. Meanwhile, in vitro drug release reveals that Gel-g-PLLA micelles show excellent sustained-release properties when used as the carrier of the anticancer drug paclitaxel. A burst release of about 70% is observed in the first 24 h. All these features, together with the outstanding biocompatibility, make Gel-g-PLLA micelles a promising drug carrier for cancer therapy.
Supramolecular polymeric prodrug micelles for efficient anticancer drug delivery

Abstract:Polymeric prodrugs have attracted great interest in the field of anticancer drug delivery owing to their integrated advantages of prodrugs and nanoparticles. However, the ambiguous chemical composition of polymeric prodrugs is still a major obstacle to their clinical applications. In this study, an amphiphilic poly(ethylene glycol) derivative with multiple thiols was first synthesized on a large scale through the melt co-polycondensation of oligo(ethylene glycol), 1,4-butanediol and mercaptosuccinic acid. Then an adamantane modified doxorubicin (DOX) with a pH-sensitive hydrazone bond and a beta-cyclodextrin derivative with multiple acryloyls were precisely synthesized. Finally, novel supramolecular polymeric prodrug (SPP) micelles with a precise drug molecular structure and drug loading content were facilely prepared using the above three components via simultaneous self-assembly, a thiol-ene "click" reaction and a host-guest supramolecular interaction in water. The resultant pH-sensitive SPP micelles with a stable core-crosslinked structure and strong drug "bonding" show long blood circulation time, effective cellular uptake and fast DOX release in tumor cells, which significantly improved the antitumor efficacy and security compared with free DOX. This work established a promising drug delivery platform for cancer chemotherapy.

Abstract:We have developed a tumor-specific phosphatase-triggered co-assembly strategy of indocyanine green (ICG)-doped nanofibers for cancer theranostics (1). This strategy relies on targeted peptide self-assemblies to improve the critical tumor uptake of ICG and its photoacoustic and photothermal properties. We demonstrated complete tumor elimination by photo-thermal therapy with high therapeutic accuracy within 24-48 hours post-injection. We have conducted extensive AFM investigations in vitro to delineate the fundamental understanding of the phosphatase-instructed formation of nanofibers. This process is crucial to its theranostic capability, and can be generalized to the self-assembly pathways of other triggered peptide hydrogelators. Our AFM results reveal a coexistence of multiple conformational structures such as the nanosheets and nanofibers with distinct nanomechnical properties. Additionally, AFM results also reveal that water can induce conformational change from nanosheets to nanofibers, which shows the dominant hydrophobic interactions during the self-assembly process. Peptide hydrogels are widely used in various biological applications, such as drug delivery and tissue engineering owing to their fibrous structure that provides the advantages of high water content and biocompatibility. This work illustrates that AFM is a powerful biophysical tool in nanomedicine for probing the structure and mechanical properties of complex biopolymer systems. Drug‐Bearing Supramolecular MMP Inhibitor Nanofibers for Inhibition of Metastasis and Growth of Liver Cancer

Abstract:Treatment of hepatocellular carcinoma (HCC) requires sustained suppression of tumor cell growth and metastasis for long-term efficacy. However, traditional intratumoral drug delivery system always exhibits burst release with less therapeutic outcomes. Here, a new self-assembling amphiphilic peptide drug conjugate (SAAPDC) is fabricated as a "two-in-one" nanofiber system comprising a hexapeptide as a matrix metalloproteinases (MMP) inhibitor and doxorubicin (DOX) for the treatment of HCC. The results indicate that doxorubicin-conjugated peptide (DOX-KGFRWR) self-assembles to form long nanofibers showing sustained release property for inhibiting the enzymatic activities of MMP-2 and MMP-9. This nanofiber not only inhibits tumor growth in situ but also effectively prevents pulmonary metastasis in an SMMC7721 cell line-based mouse model. In summary, this hexapeptide-based supermolecule system represents a promising nanoscale platform to sustain drug release with high loading capacity for intratumoral administration. Moreover, the delivery of chemotherapeutic drugs via drug-bearing supramolecular MMP inhibitor nanofibers simultaneously inhibits metastasis and tumor growth to achieve synergistic effects for metastatic HCC therapy.
A Self-Assembling Amphiphilic Peptide Dendrimer-Based Drug Delivery System for Cancer Therapy

Abstract:Despite being a mainstay of clinical cancer treatment, chemotherapy is limited by its severe side effects and inherent or acquired drug resistance. Nanotechnology-based drug-delivery systems are widely expected to bring new hope for cancer therapy. These systems exploit the ability of nanomaterials to accumulate and deliver anticancer drugs at the tumor site via the enhanced permeability and retention effect. Here, we established a novel drug-delivery nanosystem based on amphiphilic peptide dendrimers (AmPDs) composed of a hydrophobic alkyl chain and a hydrophilic polylysine dendron with different generations (AmPD KK2 and AmPD KK2K4). These AmPDs assembled into nanoassemblies for efficient encapsulation of the anti-cancer drug doxorubicin (DOX). The AmPDs/DOX nanoformulations improved the intracellular uptake and accumulation of DOX in drug-resistant breast cancer cells and increased permeation in 3D multicellular tumor spheroids in comparison with free DOX. Thus, they exerted effective anticancer activity while circumventing drug resistance in 2D and 3D breast cancer models. Interestingly, AmPD KK2 bearing a smaller peptide dendron encapsulated DOX to form more stable nanoparticles than AmPD KK2K4 bearing a larger peptide dendron, resulting in better cellular uptake, penetration, and anti-proliferative activity. This may be because AmPD KK2 maintains a better balance between hydrophobicity and hydrophilicity to achieve optimal self-assembly, thereby facilitating more stable drug encapsulation and efficient drug release. Together, our study provides a promising perspective on the design of the safe and efficient cancer drug-delivery nanosystems based on the self-assembling amphiphilic peptide dendrimer.

Abstract:Currently, combination therapy has become a popular research topic in cancer therapy. However, the outcome of the current combination therapy strategy does not satisfy the demand for clinical applications because of factors such as their overlapping toxicities, poor drug payload, and inevitable therapy resistance. Supramolecular self-assembly of peptides holds great potential for solving relevant problems and achieving superior therapeutic effects. In this study, we aimed to propose supramolecular self-assemblies based on a combination therapy strategy, consisting of a heptapeptide A7R for the inhibition of angiogenesis, the chemotherapeutic drug HCPT for treating numerous tumors with high efficacy, and an efficient self-assembling molecular FFY to improve drug loading and cell permeability. We regulated the secondary structure of nanomaterials by optimizing the pathways of self-assembly and significantly improved the affinity of A7R peptide to NRP-1. Endothelial tube formation in Matrigel and a tumor mouse model (in vivo) were then performed to demonstrate the superior antitumor effects of our supramolecular self-assemblies both in vitro and in vivo. In summary, this work not only provides a promising platform for the development of effective combination therapy, but also offers a useful strategy to prepare self-assembled nanomaterials with optimized performance.

Abstract:Doxorubicin (Dox) and Taxol can be covalently bonded to the same peptide segment via proper structural modification. Doxorubicin-maleimide derivative links to peptide via Michael addition reaction and Taxol-Succi-NHS active ester connects to the same peptide backbone through ester-amide exchange reaction. Enzymatic transformation, as an inherent biological process, is applied here to trigger the formation of nanofiber networks from the as prepared hydrogelator precursor. The precursor which loads equal molar ratio of Dox and Taxol can self-assemble to form a red stable hydrogel after dephosphorylation reaction catalyzed by alkaline phosphatase (ALP). This hydrogel could maintain sustained release of drugs and show strong anticancer effect. This work, as a new strategy to build a codelivery system of covalently linked Dox and Taxol, owns the potential to serve as an injectable hydrogel for therapeutic applications.

Abstract:Poor nuclear delivery and accumulation are the main reasons for the reduced drug efficacy of many anticancer drugs that target DNA or enzymes in the nucleus, and it is a major obstacle to successful cancer therapy. To address this problem, developing practical drug delivery systems for nuclear delivery is urgently needed. Here we develop a supramolecular hydrogel by conjugating the anticancer agent 10-hydroxycamptothecine (HCPT) and macrocyclic polyamine cyclen to a self-assembling peptide. The cyclen fragment possesses nuclear localization and ATP hydrolysis properties, which can provide a synergistic therapeutic effect for cancer treatment. The HCPT-FFFK-cyclen nanofibers showed improved nuclear accumulation and inhibition capacity in cancer cells including drug-resistant cancer cells in vitro. The nanofibers also exhibited favorable ATP consuming ability in vitro. Moreover, the obtained nanomedicine showed enhanced anticancer efficiency and favorable biocompatibility in vivo when administered to mice via tail vein injection. This constructed self-delivery drug system significantly improved the delivery efficiency of the small molecule agents into the nucleus and showed favorable ATP consuming ability, offering new strategies for developing nanomedicines for cancer combination therapy.

Abstract:D-Peptides, as the enantiomers of the naturally occurring L-peptides, usually resist endogenous proteases and are presumably insensitive to most enzymes. But, it is unclear whether or how a phosphatase catalyzes the dephosphorylation from D-peptides. In this work, we examine the formation of the nanofibers of D-peptides via enzymatic dephosphorylation. By comparing the enzymatic hydrogelation of L-peptide and D-peptide based hydrogelators, we find that the chirality of the precursors of the hydrogelators affects little on the enzymatic hydrogelation resulted from the removal of the phosphate group from a tyrosine phosphate residue. The attachment of a therapeutic agent (e.g., taxol) or a fluorophore (e.g., 4-nitro-2,1,3-benzoxadiazole) to the D-peptide based hydrogelators affords a new type of biostable or biocompatible hydrogelators, which may find applications in intratumoral chemotherapy or intracellular imaging, respectively. This work, as the first comprehensive and systematic study of the unexpected enzymatic dephosphorylation of D-peptides, illustrates a useful approach to generate supramolecular hydrogels that have both biostability and other desired functions.
Supramolecular Nanodrugs Based on Covalent Assembly of Therapeutic Peptides toward In Vitro Synergistic Anticancer Therapy

Abstract:Therapeutic peptides have attracted numerous attentions in clinical applications due to their advantages of biological origination and good biocompatibility. However, the therapeutic performances of peptides are usually hindered by their short half‐lives in blood and inferior activities. Herein, supramolecular nanodrugs of therapeutic peptides are constructed by covalent assembly of chemotherapeutic peptides through genipin cross‐linking. The resulting nanodrugs have intense absorbance in the near‐infrared region and high photothermal conversion efficiencies, leading to the possibility of photothermal therapy. The combination of photothermal therapy and chemotherapy using the nanodrugs shows synergistic therapeutic effects on cancer cells. Hence, covalent assembly not only maintains the chemotherapeutic activities of the peptides but also triggers supramolecular photothermal effects, demonstrating that the covalent assembly of therapeutic peptides through genipin cross‐linking is an efficient approach for constructing supramolecular nanodrugs toward synergistic anticancer therapy.

Abstract:Supramolecular hydrogels of self-assembling peptide-drug conjugates have been considered as effective self-delivery drug systems for cancer therapy in recent years. Here, a novel self-assembling peptide-based supramolecular hydrogel was developed by simultaneously conjugating small-molecule drug chlorambucil (CRB) and peptide drug tyroservatide (YSV) to the self-assembling peptide. The resulting hydrogel with a nanofiber structure showed enhanced stability against proteinase K degradation and an improved cellular uptake performance in comparison with the free molecules. As a consequence, it exhibited enhanced antitumor efficiency both in vitro and in vivo with favorable biocompatibility. This biocompatible self-delivery drug system could not only significantly improve the delivery efficiency of the small-molecule drugs but also adequately synergize the antitumor effect of CRB and YSV, inspiring the design of new strategies of cancer combination therapy.
Hydrogen-bonded cytosine-endowed supramolecular polymeric nanogels: Highly efficient cancer cell targeting and enhanced therapeutic efficacy

Abstract:We demonstrate that cytosine moieties within physically cross-linked supramolecular polymers not only manipulate drug delivery and release, but also confer specific targeting of cancer cells to effectively enhance the safety and efficacy of chemotherapy—and thus hold significant potential as a new perspective for development of drug delivery systems. Herein, we successfully developed physically cross-linked supramolecular polymers (PECH-PEG-Cy) comprised of hydrogen-bonding cytosine pendant groups, hydrophilic poly(ethylene glycol) side chains, and a hydrophobic poly(epichlorohydrin) main chain. The polymers spontaneously self-assemble into a reversibly hydrogen-bonded network structure induced by cytosine and directly form spherical nanogels in aqueous solution. Nanogels with a high hydrogen-bond network density (i.e., a higher content of cytosine moieties) exhibit outstanding long-term structural stability in cell culture substrates containing serum, whereas nanogels with a relatively low hydrogen-bond network density cannot preserve their structural integrity. The nanogels also exhibit numerous unique physicochemical characteristics in aqueous solution, such as a desirable spherical size, high biocompatibility with normal and cancer cells, excellent drug encapsulation capacity, and controlled pH-responsive drug release properties. More importantly, in vitro experiments conclusively indicate the drug-loaded PECH-PEG-Cy nanogels can selectively induce cancer cell-specific apoptosis and cell death via cytosine receptor-mediated endocytosis, without significantly harming normal cells. In contrast, control drug-loaded PECH-PEG nanogels, which lack cytosine moieties in their structure, can only induce cell death in cancer cells through non-specific pathways, which significantly inhibits the induction of apoptosis. This work clearly demonstrates that the cytosine moieties in PECH-PEG-Cy nanogels confer selective affinity for the surface of cancer cells, which enhances their targeted cellular uptake, cytotoxicity, and subsequent induction of programmed cell death in cancer cells.

Abstract:E-cadherin/catenin complex is crucial for cancer cell migration and invasion. The histidine-alanine-valine (HAV) sequence has been shown to inhibit a variety of cadherin-based functions. In this study, by fusing HAV and the classical tumor-targeting Arg-Gly-Asp (RGD) motif and Asn-Gly-Arg (NGR) motif to the apoptosis-inducing peptide sequence-AVPIAQK, a bifunctional peptide has been constructed with enhanced tumor targeting and apoptosis effects. This peptide is further processed as a nanoscale vector to encapsulate the hydrophobic drug docetaxel (DOC). Bioimaging analysis shows that peptide nanoparticles can penetrate into xenograft tumor cells with a significantly long retention in tumors and high tumor targeting specificity. In vivo, DOC/peptide NPs are substantially more effective at inhibiting tumor growth and prolonging survival compared with DOC control. Together, the findings of this study suggest that DOC/peptide NPs may have promising applications in pulmonary carcinoma therapy.

Abstract:Self-assembly supramolecular nanoparticles (SNPs) are synthesized using polyethylenimine (PEI) crosslinked by cyclodextrins (CyDs), adamantyl doxorubicin and plasmid DNA. SNPs successfully conduct the cooperative co-delivery of chemotherapeutic anticancer drug and plasmid DNA in vitro and in vivo.
An Amphiphilic PEGylated Peptide Dendron-Gemcitabine Prodrug-Based Nanoagent for Cancer Therapy

Abstract:An amphiphilic peptide dendrimer conjugated with gemcitabine (GEM), PEGylated dendron-Gly-Phe-Leu-Gly-GEM (PEGylated dendron-GFLG-GEM), is developed as a nano-prodrug for breast cancer therapy. The self-assembled behavior is observed under a transmission electron microscopy and dynamic light scattering. The negatively charged surface and hydrodynamic size of the amphiphilic nanosized prodrug supported that the prodrug can maintain the stability of GEM during circulation and accumulate in the tumor tissue. Drug release assays are conducted to monitor the release of GEM from this nanodrug delivery system in response to the tumor microenvironment, and these assays confirm that GEM released from the nanocarrier is identical to free GEM. The GEM prodrug can prevent proliferation of tumor cells. The therapeutic effect against breast cancer is systematically investigated using an in vivo animal model. Immunohistochemical results are aligned with the significantly enhanced anticancer efficacy of GEM released from the prodrug. This self-assembled amphiphilic drug delivery nanocarrier may broaden the application for GEM and other anticancer agents for breast cancer chemotherapy.

Abstract:Pharmaceutical science based on biological nanotechnology is developing rapidly in parallel with the development of nanomaterials and nanotechnology in general. Pectin is a natural polysaccharide obtainable from a wide range of sources. Here, we show that doxorubicin (DOX)-conjugated hydrophilic pectin (PET) comprising an amphiphilic polymer loaded with hydrophobic dihydroartemisinin (DHA) self-assemble into nanoparticles. Importantly, conjugated DOX and DHA could be released quickly in a weakly acidic environment by cleavage of the acid-sensitive acyl hydrazone bond. Confocal microscopy and flow cytometry confirmed that these PET-DOX/DHA nanoparticles efficiently delivered DOX into the nuclei of MCF-7 cells. Significant tumor growth reduction was monitored in a female C57BL/6 mouse model, showing that the PET-DOX/DHA nanoparticle-mediated drug delivery system inhibited tumor growth and may improve therapy. Thus, we have demonstrated that pectin may be useful in the design of materials for biomedical applications.
