Analysis on Hyphal Form of Candida Albicans by Randomly Amplified Polymorphic DNA

Abstract:Objective To compare the randomly amplified polymorphic DNA (RAPD) map difference among susceptible,susceptible dose-dependent and resistant hyphal form of Candida albicans strains with fluconazole.Methods DNAs extracted from hyphal form of C. albicans were amplified by PCR with the same arbitrary sequence decamer primer 5′-TCACCACGGT-3′,and the PCR products were separated by denaturing polyacrylamine gel electrophoresis (PAGE) and photoed.Results The electrophoresis bands of CA-1-CA-13 were identical,they all had 6 bands;The electrophoresis bands of CA-14-CA-17 were identical,they all had 7 bands;a band of about 450bp was specific to substains CA-14-CA-17.Conclusion There was difference on RAPD map between resistant and susceptible substrains hyphal form of C.albican.
