Effects of Selenium on the Morphology and NSE IGFR Expression in Rats' Cerebral Cortex Neurons in Vitro

Abstract:AIM: To study and observe the effects of se le nium (Se) on the growth, development, and differentiation in cultured neurons in vitro. METHODS: Primary neuron culture neonatal rats' cortex with Se in vitro was used to measure the diameter of the c ells and the length of the neurites and the NSE and IGFR expressions in cultured cells were detected by immunohistochemical method. RESULTS: Newborn rats' cerebral neuron could grow and develop in vitro. Low and midd le concentrations of Se (0 0625~ 0 50 mg·L -1 )could increase the di a meter of the cell and the length of the neurite of the cultured neurons in vitro (from 3 to 7 days, the diameter of cells of control group were 19,19 and 21 μ m,the average diameter of experimental groups were 26,25 and 26 μm,the lengt h of the neuritis of control groups were 49,64 and 64 μm,the minimum length o f experimental groups were 59,78 and 76 μm, P0 05), and could promote NSE expressions in comparison with that of control group(the average of control group was 14 1, the minimum of experimental group was 21 4 , P0 05), and could promote IGFR expressions in compa rison with that of control group(the average of control group was 16 6 , the m inimum of experimental groups was 21 9, P0 05). High concentrations of Se (0 75~1 0 mg·L -1 )were toxic to cell culture. CONCLUSION: The results indicate that selenium has an important biological part in the neuron growth, development and differentiation in early stage in vitro.
