Abstract:Angiotensin II (AngII) has been shown to play a critical role in diabetic nephropathy and vasculopathy. Although it is well recognized that an angiotensin-converting enzyme (ACE)-dependent AngII-generating system is a major source of intrarenal AngII production, it is here reported that the chymase-dependent AngII-generating system is upregulated in the human diabetic kidney. This becomes particularly strong in those with hypertension. In the normal kidney, while ACE was constitutively expressed by most kidney cells, chymase was weakly expressed by mesangial cells (MC) and vascular smooth muscle cells (VSMC) only. In the diabetic kidney, while ACE expression was significantly upregulated (1 to 3-fold) by tubular epithelial cells (TEC) and infiltrating mononuclear cells, there was also markedly increased chymase expression (10 to 15-fold) by both MC and VSMC, with strong deposition in the collagen-rich extracellular matrix including both diffuse and nodular glomerulosclerosis, tubulointerstitial fibrosis, and vascular sclerosis. Interestingly, while ACE expression showed no difference in patients with or without hypertension, upregulation of chymase in hypertensive patients was much stronger than that seen in those without hypertension (4 to 7-fold, P < 0.001). Correlation analysis showed that, in contrast to the ACE expression, upregulation of chymase correlated significantly with the increase in BP and the severity of collagen matrix deposition within the glomerulus, tubulointerstitium, and arterial walls (all with P < 0.001). In conclusion, the present study demonstrates that chymase, as an alternative AngII-generating enzyme, is markedly upregulated in the diabetic kidney and may be associated with the development of diabetic/hypertensive nephropathy. In addition, differential expression of ACE and chymase in the diabetic kidney indicates that both ACE and chymase may be of equal importance for AngII-mediated diabetic nephropathy and vascular disease. Results from this study suggest that blockade of both AngII-generating pathways may provide additional beneficial effect on diabetic nephropathy.

Abstract:Angiotensin-converting enzyme (ACE) is best known for the catalytic conversion of angiotensin I to angiotensin II. However, the use of gene-targeting techniques has led to mouse models highlighting many other biochemical properties and actions of this enzyme. This review discusses recent studies examining the functional significance of ACE tissue-specific expression and the presence in ACE of two independent catalytic sites with distinct substrates and biological effects. It is these features which explain why ACE makes important contributions to many different physiological processes including renal development, blood pressure control, inflammation, and immunity.
[Effects of Angiotensin Converting Enzyme Inhibitor on the Expression of Angiotensin Converting Enzyme 2 in Atrium of Patients with Atrial Fibrillation].

Abstract:OBJECTIVE:To investigate the expression of angiotensin converting enzyme 2 (ACE2) and the changes treated with angiotensin converting enzyme inhibitor (ACEI), and its signal transduction pathway.METHODS:Atrial tissues were obtained from 47 patients with RHD undergoing cardiac surgery. The mRNA of ACE2 and ACE were semi-qualified by RT-PCR and normalized to the gene beta-actin. Western blot analysis was employed to examine the expressions of ACE2, ACE, ERK1/2 and phosphorylated ERK (pERK1/2). The atrial tissue angiotensin II (Ang II) content was determined by radioimmunoassay detection.RESULTS:The expression of ACE2 was significantly decreased (P < 0.05), the expression of ACE and pERK1/2 were significantly increased (P < 0.05), and the level of atrial tissue Ang II was significantly increased in patients with chronic atrial fibrillation group (CAF) compared with sinus rhythm group (SR) (P < 0.05). Compared with CAF patients treated without ACEI, the expression of ACE2 significantly increased (P < 0.01), and the relative activity of ERK1/2 significantly decreased (P < 0.05), whereas the expression of ACE and the level of atrial tissue Ang II remained unchanged in CAF patients treated with ACEI.CONCLUSIONS:The study suggested that the dysequilibrium of ACE/ACE2 might play an important role in the process of atrial fibrillation, which may be related to the activation of ERK1/2 pathway. The clinical effect of long-term treatment of ACEI maybe associated with elevated ACE2 expression but not ACE expression.

Abstract:Angiotensin converting enzyme (ACE) in the endoplasmic reticulum has been revealed as the major carboxypeptidase involved in MHC class I antigen processing. ACE is a type I membrane protein and are also present on the surface of cytoplasmic membrane. This study is to understand whether ACE is involved in MHC class II antigen presentation. We find that the macrophages expressing null, normal and excessive ACE have different abilities in presenting model antigens ovalbumin (OVA) and hen egg lysozyme (HEL) in vitro. The differences were observed in the presentation of either the whole antigen proteins or their MHC class II epitopes. The mice with ACE over‐expressed in macrophages are superior over wild‐type (WT) mice in producing CD4+ T cell responses and antibody responses when immunized with OVA in vivo. In addition, ACE can be localized in the endosomic pathway where MHC class II antigen processing and loading take place. The efficiency of macrophages to present MHC class II antigens can be altered by transfecting ACE‐expressing vector or using ACE inhibitors. These data strongly suggest that ACE affects the presentation of MHC class II antigen.

Abstract:Purpose To investigate the influence on the cell proliferation and AngⅡ generation by bloc- king chymase path. Methods The human umbilicus artery smooth muscle cells were cultured and divided into five groups:① DMEM group(blank control group); ② AngⅠgroup;③ AngⅠ plus captopril group; ④ AngⅠ plus chymostatin group; ⑤ AngⅠ plus valsatan group.They are designed to block angiotensin-converting enzyme,chymase and AngⅠrespectively.Each group was undergone cell counting, cell proliferation index and AngⅡ concentration in the culture medium tests. Results The cells'numbers were decreased in the drug interfered groups compared with the Ang Ⅰ group (P0.01); the cell proliferation of chymostatin and valsatan group were obviously inhibited(P0.01);the concentration of AngⅡ in chymostatin group was reduced significantly(P0.01), while it was raised in valsatan group(P0.01). Conclusions Chymase plays an important role on generation of AngⅡ and proliferation of cultured human vascular smooth muscle cells.

Abstract:Angiotensin-converting enzyme (ACE) plays an important role in blood pressure control. ACE also has effects on renal function, reproduction, hematopoiesis, and several aspects of the immune response. ACE 10/10 mice overexpress ACE in monocytic cells; macrophages from ACE 10/10 mice demonstrate increased polarization toward a proinflammatory phenotype. As a result, ACE 10/10 mice have a highly effective immune response following challenge with melanoma, bacterial infection, or Alzheimer disease. As shown in ACE 10/10 mice, enhanced monocytic function greatly contributes to the ability of the immune response to defend against a wide variety of antigenic and non-antigenic challenges.

Abstract:Background Angiotensin-(1–12) [Ang-(1–12)] is a chymase-dependent source for angiotensin II (Ang II) cardiac activity. The direct contractile effects of Ang-(1–12) in normal and heart failure (HF) remain to be demonstrated. We assessed the hypothesis that Ang-(1–12) may modulate [Ca2+]i regulation and alter cardiomyocyte contractility in normal and HF rats. Methods and results We compared left ventricle (LV) myocyte contractile and calcium transient ([Ca2+]iT) responses to angiotensin peptides in 16 SD rats with isoproterenol-induced HF and 16 age-matched controls. In normal myocytes, versus baseline, Ang II (10−6 M) superfusion significantly increased myocyte contractility (dL/dtmax: 40%) and [Ca2+]iT (29%). Ang-(1–12) (4 × 10−6 M) caused similar increases in dL/dtmax (34%) and [Ca2+]iT (25%). Compared with normal myocytes, superfusion of Ang II and Ang-(1–12) in myocytes obtained from rats with isoproterenol-induced HF caused similar but significantly attenuated positive inotropic actions with about 42% to 50% less increases in dL/dtmax and [Ca2+]iT. Chymostatin abolished Ang-(1–12)-mediated effects in normal and HF myocytes. The presence of an inhibitory cAMP analog, Rp-cAMPS prevented Ang-(1–12)-induced inotropic effects in both normal and HF myocytes. Incubation of HF myocytes with pertussis toxin (PTX) further augmented Ang II-mediated contractility. Conclusions Ang-(1–12) stimulates cardiomyocyte contractile function and [Ca2+]iT in both normal and HF rats through a chymase mediated action. Altered inotropic responses to Ang-(1–12) and Ang II in HF myocytes are mediated through a cAMP-dependent mechanism that is coupled to both stimulatory G and inhibitory PTX-sensitive G proteins.

Abstract:Background Little is known about the role of dual angiotensin Ⅱ forming pathways during heart failure. In the present study, the changes of chymase and angiotensin converting enzyme (ACE) expressions in the failing hearts of hamsters were analysed.Methods Heart failure was induced by ligation of left anterior descending branch of the coronary artery. Chymase, ACE and angiotensin Ⅱ type 1 receptor (AT1R) mRNA levels were analysed by reverse transcription polymerase chain reaction (RT-PCR). The activities of chymase and ACE were determined by radioimmunoassay (RIA). Myocardial collagen fibre analysis was performed under optical microscope.Results Left ventricular systolic pressure (LVSP) and maximum left ventricular developed pressure increase rate (dp/dtmax , mmHg/s) gradually moved lower at 2, 3, 4 and 8 weeks after operation. On the other hand, left ventricular end-diastolic pressure (LVEDP) increased gradually after operation. Compared with the control group (3.55±0.06, 4.79±0.70), the heart weight/body weight ratio in operation group had increased significantly at 4 weeks and 8 weeks (4.28±0.43, 6.17±0.73) (P<0.01). Collagen staining showed that the quantity of myocardial collagen fibre increased significantly in the operation group. RT-PCR showed that the chymase mRNA level in the operation group was consistently greater than that in the control group. AT1R mRNA level was also increased significantly at 3 weeks and 4 weeks, both being 1.3 times that of the control group (P<0.01), whereas ACE mRNA level was not changed. Higher activity of chymase was detected in operation group, being 4, 8, 13 and 19 times that of the control group at 2, 3, 4 and 8 weeks (P<0.01), respectively. ACE activity was also significantly higher at the same time, being 7, 10, 10 and 3.5 times that of the control (P<0.01). Angiotensin Ⅱ (Ang Ⅱ) level in operation group increased significantly, being 2.5, 2.7, 3.5 and 2 times that of the control group at 2, 3, 4 and 8 weeks, respectively (P<0.01).Conclusions A dual Ang Ⅱ forming pathway from both ACE and chymase in the hamster hearts plays an important role during the development of heart failure. At the decompensatory stage, the reduction of AngII level may be associated with the decrease of ACE activity.

Abstract:ACE2, the first known human homologue of angiotensin-converting enzyme (ACE), was identified from 5′ sequencing of a human heart failure ventricle cDNA library. ACE2 has an apparent signal peptide, a single metalloprotease active site, and a transmembrane domain. The metalloprotease catalytic domains of ACE2 and ACE are 42% identical, and comparison of the genomic structures indicates that the two genes arose through duplication. In contrast to the more ubiquitous ACE, ACE2 transcripts are found only in heart, kidney, and testis of 23 human tissues examined. Immunohistochemistry shows ACE2 protein predominantly in the endothelium of coronary and intrarenal vessels and in renal tubular epithelium. Active ACE2 enzyme is secreted from transfected cells by cleavage of the N-terminal to the transmembrane domain. Recombinant ACE2 hydrolyzes the carboxy terminal leucine from angiotensin I to generate angiotensin 1-9, which is converted to smaller angiotensin peptides by ACE in vitro and by cardiomyocytes in culture. ACE2 can also cleave des-Arg bradykinin and neurotensin but not bradykinin or 15 other vasoactive and hormonal peptides tested. ACE2 is not inhibited by lisinopril or captopril. The organ- and cell-specific expression of ACE2 and its unique cleavage of key vasoactive peptides suggest an essential role for ACE2 in the local renin-angiotensin system of the heart and kidney. Abstract:Objective To investigate the changes in the content of angiotensin Ⅱ(ATⅡ) and the gene expressions of chymase and angiotensin-converting enzyme (ACE) in diabetic hamsters' heart, so as to understand the role of chymase and ACE in the development of diabetic cardiomyopathy. Methods ATⅡ levels in plasma and heart were determined in normal control group (n=10) and diabetic group (n=10). Gene expressions of chymase and ACE were detected by using fluorescent quantitative polymerize-chain-reaction (FQ-PCR).Levels of type Ⅰand Ⅲ collagens in heart were measured by immunohistochemical method.Levels of blood glucose, glycated serum protein, insulin and lipid profile were determined. Results Compared with normal control group, levels of collagen Ⅰand Ⅲ in diabetic group were 2.71 and 1.68 folds higher separately, and the ratio of collagen Ⅰ/collagen Ⅲ was also significantly higher.Level of ATⅡ in diabetic hamsters' heart was higher than that in normal control.Gene expressions of chymase but not ACE in diabetic group were higher than those in control group. Conclusions Diabetic cardiomyopathy is manifested by elevated level of ATⅡ in hearts, which might be produced by route of chymase conversion.And the inhibiting of chymase may have some therapeutic values.

Abstract:Background: Angiotensin-(1-12) [Ang-(1-12)], a new member of the renin-angiotensin system (RAS) has been identified as a chymase-dependent source for Angiotensin II (Ang II) inotropic activity. Recent observations suggest that activation of this non-canonical chymase/Ang-(1-12) pathway may modulate cardiac function bypassing the inhibitory effects of RAS blockade with ACEI and ARBs in heart failure (HF). However, the direct cardiac effects of Ang-(1-12) and Ang II are unclear. Moreover, whether and how HF alters Ang-(1-12) and Ang II cardiac responses are undefined. We assessed the hypothesis that HF is associated with a reduced action of Ang-(1-12) on myocyte contractility and [Ca 2+ ] i regulation. Methods: We compared LV myocyte contractile and calcium transient ([Ca 2+ ] iT ) responses to angiotensin peptides in 12 SD rats with isoproterenol induced HF (2 months after isoproterenol 170 mg/kg sq for 2 days) and 16 age-matched controls. Results: In normal myocytes, versus baseline, Ang II (10 -6 M) superfusion significantly increased myocyte contraction (dL/dt max ) (40%, 184.6 vs 132.1 μm/s), relaxation (dR/dt max ) (34%, 141.3 vs 107.0 μm/s) and [Ca 2+ ] iT (29%, 0.31 vs 0.24). Superfusion of Ang-(1-12) (4x10 -6 M) caused similar increases in dL/dt max (34%) dR/dt max (25%) and [Ca 2+ ] iT (25%). Compared with the changes in normal myocytes, in HF myocytes, Ang II and Ang-(1-12) caused similar but significantly attenuated positive inotropic actions with about 42% to 50% less increases in dL/dt max , dR/dt max and [Ca 2+ ] iT . The Ang-(1-12)-mediated effects were abolished by prior exposure of myocytes to chymostatin in both normal and HF. The Ang II-induced inotropic effects were completely prevented in the presence of an inhibitory cAMP analog, Rp-cAMPS (10 –4 M 2 h) in both normal and HF myocytes, but were further augmented only in HF after the incubation of myocytes with the G i inhibitor, pertussis toxin (PTX, 2 μg/ml, 36°C, 5h). Conclusions: Ang-(1-12) stimulates LV myocyte contractile function and [Ca 2+ ] iT in both normal and HF rats through a chymase mediated action. Altered inotropic responses to Ang-(1-12) and Ang II in HF myocytes is mediated through a cAMP-dependent mechanism that is coupled to both stimulatory G and inhibitory PTX-sensitive G proteins.

Abstract:Angiotensin-converting enzyme 2 (ACE2) is a monocarboxypeptidase of the renin-angiotensin-system (RAS) which is known to cleave several substrates among vasoactive peptides. Its preferred substrate is Angiotensin II, which is tightly involved in the regulation of important physiological functions including fluid homeostasis and blood pressure. Ang 1–7, the main enzymatic product of ACE2, became increasingly important in the literature in recent years, as it was reported to counteract hypertensive and fibrotic actions of Angiotensin II via the MAS receptor. The functional connection of ACE2, Ang 1–7, and the MAS receptor is also referred to as the alternative axis of the RAS. In the present paper, we describe the recombinant expression and purification of human and murine ACE2 (rhACE2 and rmACE2). Furthermore, we determined the conversion rates of rhACE2 and rmACE2 for different natural peptide substrates in plasma samples and discovered species-specific differences in substrate specificities, probably leading to functional differences in the alternative axis of the RAS. In particular, conversion rates of Ang 1–10 to Ang 1–9 were found to be substantially different when applying rhACE2 or rmACE2 in vitro . In contrast to rhACE2, rm ACE2 is substantially less potent in transformation of Ang 1–10 to Ang 1–9.

Abstract:The renin-angiotensin system (RAS) is a key player in the control of the cardiovascular system and hydroelectrolyte balance, with an influence on organs and functions throughout the body. The classical view of this system saw it as a sequence of many enzymatic steps that culminate in the production of a single biologically active metabolite, the octapeptide angiotensin (ANG) II, by the angiotensin converting enzyme (ACE). The past two decades have revealed new functions for some of the intermediate products, beyond their roles as substrates along the classical route. They may be processed in alternative ways by enzymes such as the ACE homolog ACE2. One effect is to establish a second axis through ACE2/ANG-(1–7)/MAS, whose end point is the metabolite ANG-(1–7). ACE2 and other enzymes can form ANG-(1–7) directly or indirectly from either the decapeptide ANG I or from ANG II. In many cases, this second axis appears to counteract or modulate the effects of the classical axis. ANG-(1–7) itself acts on the receptor MAS to influence a range of mechanisms in the heart, kidney, brain, and other tissues. This review highlights the current knowledge about the roles of ANG-(1–7) in physiology and disease, with particular emphasis on the brain.

Abstract:INTRODUCTION: Angiotensin (Ang) II is known to activate matrix metalloproteinases (MMPs), leading to degradation of extracellular matrix (ECM) proteins and myocardial remodelling. Angiotensin-converting enzyme-2 (ACE2) is a carboxypeptidase that metabolises Ang II to yield Ang-(1–7), essentially negatively regulating the renin-angiotensin system. We hypothesised that loss of ACE2 exacerbates myocardial remodelling by modulating the levels of MMPs. 10-week old male wildtype (WT, Ace2+/y) and ACE2 knockout (ACE2KO, Ace2−/y) mice received with mini-osmotic pumps (model 1002; USA) with a pressor dose of Ang II (1.5 mg kg−1 d−1) or saline for 2 weeks. Pro and cleaved forms of MMP2 and MMP9 were detected by gelatine zymography, and total collagenase and gelatinase activities were measured using fluorescent-based activity assays from EnzCheck (Molecular Probes). The membrane-anchored collagenase membrane type 1 (MT1)-MMP, the fibrotic factors transforming growth factor-β1 (TGF-β1) and fibronectin levels in heart were determined by TaqMan real-time PCR and western blotting analyses, respectively. In response to chronic stimulation by Ang II, there were significant increases in myocardial expression of MMP2, MMP9 and MT1-MMP in both WT and ACE2KO mice with elevated plasma Ang II levels. Furthermore, loss of ACE2 resulted in greater increases in Ang II-mediated expression of pro MMP2 and active MMP2 and MMP9 in ACE2KO hearts associated with enhanced expression of MT1-MMP, TGF-β1 and fibronectin and elevated activities of gelatinase and collagenase. These changes were linked with a degraded and disorganised ECM in the Ang II-treated ACE2KO heart by picrosirius red staining. ACE2 deficiency exacerbates Ang II-mediated adverse myocardial remodelling by modulating the levels of MMP2, MMP9 and MT1-MMP and enhancing the expression of TGF-β1 and fibronectin, implying a critical role of ACE2 in regulating the balance between generation and degradation of myocardial extracellular matrix and potential therapeutic approaches by enhancing ACE2 action for patients with heart diseases. Abstract:Cardiovascular diseases are predicted to be the most common cause of death worldwide by 2020. Here we show that angiotensin-converting enzyme 2 (ace2) maps to a defined quantitative trait locus (QTL) on the X chromosome in three different rat models of hypertension. In all hypertensive rat strains, ACE2 messenger RNA and protein expression were markedly reduced, suggesting that ace2 is a candidate gene for this QTL. Targeted disruption of ACE2 in mice results in a severe cardiac contractility defect, increased angiotensin II levels, and upregulation of hypoxia-induced genes in the heart. Genetic ablation of ACE on an ACE2 mutant background completely rescues the cardiac phenotype. But disruption of ACER, a Drosophila ACE2 homologue, results in a severe defect of heart morphogenesis. These genetic data for ACE2 show that it is an essential regulator of heart function in vivo.

Abstract:The renin-angiotensin system (RAS) plays a central role in the maintainance of homeostasis of blood pressure and electrolyte balance. Recently,the RAS has been implicated in the pathogenesis on the pathogenesis of respiratory disease,especially in pulmonary fibrosis,pulmonary artery hypertension.Angiotensin-converting enzyme 2 was cloned as a first homolog of human ACE and functioned as an negative regulator of RAS.Recent studies has confirmed the protective effect of ACE2 against acute lung injury.In this review we shall discuss the roles for ACE2 in disease pathogenesis and the possibility of ACE2 as a novel approach for the treatment of lung diseases.

Abstract:Abstract Background Chymase is the major angiotensin II (Ang II)-forming enzyme in cardiovascular tissue, with an important role in atrial remodeling. This study aimed to examine the association between chymase 1 gene (CMA1) polymorphisms and atrial fibrillation (AF) in a Chinese Han population. Methods This case-control study enrolled 126 patients with lone AF and 120 age- and sex-matched healthy controls, all from a Chinese Han population. Five CMA1 polymorphisms were genotyped. Results The CMA1 polymorphism rs1800875 (G-1903A) was associated with AF. The frequency of the GG genotype was significantly higher in AF patients compared with controls (p = 0.009). Haplotype analysis further demonstrated an increased risk of AF associated with the rs1800875-G haplotype (Hap8 TGTTG, odds ratio (OR) = 1.668, 95% CI 1.132–2.458, p = 0.009), and a decreased risk for the rs1800875-A haplotype (Hap5 TATTG, OR = 0.178, 95% CI 0.042–0.749, p = 0.008). Conclusions CMA1 polymorphisms may be associated with AF, and the rs1800875 GG genotype might be a susceptibility factor for AF in the Chinese Han population.

Abstract:The angiotensin-converting enzyme (ACE) was discovered in 1956 and has been actively studied to date. It has a unique structure of two homologous domains, each containing a catalytic zinc ion. Domains have different substrate specificity. In terms of function, ACE is a zinc metallopeptidase widely present on the surface of endothelial and epithelial cells. The gene encoding ACE is located on the long arm of chromosome 17 (17q23) and is 21 kb long, including 26 exons and 25 introns. The structure of ACE may be the result of an ancient gene duplication that occurred approximately 700 million years ago. The main function of ACE is the conversion of AngI to the vasoconstrictor AngII, which is the main active product. In addition, ACE metabolizes bradykinin, which is a potent vasodilator. ACE is involved in the metabolism of other angiotensins, in particular Ang(1–7), forming, together with ACE 2 and other components of the renin-angiotensin-aldosterone system (RAAS), a complex balanced system for maintaining blood pressure, water and electrolyte balance, and many other components of systemic, tissue and cellular homeostasis that have not yet been fully studied. More data are accumulating confirming the role of ACE for the renal development, early hematopoiesis, normal male fertility, erythropoiesis, myelopoiesis. ACE plays important roles in the immune response, intracellular signaling.

Abstract:Mast cell-derived chymase is implicated in myocardial fibrosis (MF), but the underlying mechanism of intracellular signaling remains unclear. Transforming growth factor-β1 (TGF-β1) is identified as the most important profibrotic cytokine, and Smad proteins are essential, but not exclusive downstream components of TGF-β1 signaling. Moreover, novel evidence indicates that there is a cross talk between Smad and mitogen-activated protein kinase (MAPK) signaling cascade. We investigated whether chymase activated TGF-β1/Smad pathway and its potential role in MF by evaluating cardiac fibroblasts (CFs) proliferation and collagen synthesis in neonatal rats. MTT assay and 3H-Proline incorporation revealed that chymase induced CFs proliferation and collagen synthesis in a dose-dependent manner. RT-PCR and Western blot assay demonstrated that chymase not only increased TGF-β1 expression but also upregulated phosphorylated-Smad2/3 protein. Furthermore, pretreatment with TGF-β1 neutralizing antibody suppressed chymase-induced cell growth, collagen production, and Smad activation. In contrast, the blockade of angiotensin II receptor had no effects on chymase-induced production of TGF-β1 and profibrotic action. Additionally, the inhibition of MAPK signaling had no effect on Smad activation elicited by chymase. These results suggest that chymase can promote CFs proliferation and collagen synthesis via TGF-β1/Smad pathway rather than angiotensin II, which is implicated in the process of MF.

Abstract:In the updated concept of renin–angiotensin system (RAS), it contains the angiotensin converting enzyme (ACE)–angiotensin (Ang) II–angtiogensin type 1 receptor (AT1) axis and the angiotensin-converting enzyme-related carboxypeptidase (ACE2)–Ang-(1–7)–Mas axis. The former axis has been well demonstrated performing the vasoconstrictive, proliferative and pro-inflammatory functions by activation of AT1 receptors, while the later new identified axis is considered counterbalancing the effects of the former. The present study is aimed at observing the interaction between Ang-(1–7) and Ang II on cultured rat renal mesangial cells (MCs). RT-PCR, Western blot and immunofluorescent staining and confocal microscopy results showed that both AT1 and Mas receptor were co-distributed in rat renal MCs. Ang-(1–7) showed similar effects on Ang II in cultured MCs that stimulated phosphorylated extracellular signal-regulated kinase (ERK)1/2 phosphorylation and transforms growth factor-β1 synthesis, and cell proliferation and extracellular matrix synthesis. Co-treatment of the cell with Ang-(1–7) and Ang II, Ang-(1–7) counteracted AngII-induced effects in a concentration dependent manner, but failed to alter the changes induced by endothelin-1. The stimulating effect of Ang II was mediated by AT1 receptor while all the effects of Ang-(1–7) were blocked by Mas receptor antagonist A-779, but not by AT1 receptor antagonist losartan or AT2 receptor antagonist PD123319. These results suggest that Ang-(1–7) and Ang II specifically interact with each other on rat renal MCs via activation of their specific receptors, Mas and AT1 receptor respectively.
