Abstract:A reliable method to separate and identify glucosinolates in rapeseed has been reported. Roughed product would be got after extraction and ionic exchange, and then the product was purified with liquid chromatography. One of the best separated component would be confirmed as 4-pentenyl glucosinolate by means of mass spectrometry, nuclear magnetic resonance spectrometry, ultraviolet spectroscopy and infrared spectrometry. This research will be used as an effective method for getting special glucosinolate compounds.

Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.

Abstract:The quantitative relationship between the structure of solutes and their chromatographic retention behavior has been studied for many years, This paper discussed mainly the retention behavior of hydroxy compound glucuronides, The samples were incubated in vitro. In conclusion, the retention behavior of glucuronides can be predicted by two ways: (1) prediction of capacity factor (K') for hydroxy compounds and their glucuronides is based on chemi-physical constants; (2) prediction of glucuronides capacity factor is based on the capacity factor of parent compound, The paper solved a knotty problem, the lack of glucuronide reference by using biological synthesis method, It is very useful for the study on drug phase II metabolism, Predicting retention behavior of metabolites based on the parent compounds is also a new idea for studying QSRR for drug metabolites.

Abstract:Cruciferous crops are rich in glucosinolate, a diverse class of sulphur- and nitrogen-containing secondary metabolites related to the flavour and quality of these crops. To date, glucosinolate biosynthetic pathway has been elucidated in model plant Arabidopsis, and regulations of glucosinolate composition and content in cruciferous crops have been extensively investigated with an aim to enhance the content of health-promoting glucosinolate profiles, reduce undesirable ones, and finally meet the requirement of human nutrition and health. This chapter presents an overview of the biological functions of glucosinolates and their degradation products, biosynthesis and metabolic engineering of glucosinolates, as well as the regulation of glucosinolate accumulation along the whole food chain in cruciferous crops.

Abstract:Glucosinolates, a crucial group of secondary metabolites in Brassica vegetables, present significant chromatographic separation challenges due to their anionic form, structure diversities, and co‐existence of other phenolic compounds. This study comparatively investigated the retention and separation of seven glucosinolates using a mixed‐mode reversed‐phase/weak anion‐exchange column and a conventional reversed‐phase C18 column. Separation factors for each glucosinolate with its adjacent peaks were over 1.0 on the mixed‐mode column, while co‐eluting was observed on the C18 column. The effects of mobile phase additives and pH on the separation and retention of glucosinolates were also investigated. Results showed that glucosinolate retention was inversely related to both buffer concentration and pH. The optimized method for the mix‐mode column was applied to the complex Brassica vegetable samples. In addition to the 17 well‐resolved glucosinolate peaks, 34 peaks for phenolic compounds were identified in broccoli microgreen, suggesting the successful application scenarios for qualitative analysis in comparison with the single mode reverse phase C18 column. This study demonstrates that the mixed‐mode reversed‐phase/weak anion‐exchange column can be used as a promising separation tool for organic anions in a complex sample matrix.

Abstract:OBJECTIVE To investigate the QSMR of glucuronidition of flavonoids catalyzed by human UGT1A3.METHODS The relationship between flavonoid structures and Km for UDP-glucuronosyltransferases1A3(UGT1A3) was studied for 11 flavonoids compounds by MOPAC-AM1 method.RESULTS Combining their calculated results with experimental data,a QSAR(pKm=-2.207 07+0.002 56HOF+3.212 90 QC5) was obtained with stepwise regression.CONCLUSION The heat of formation(HOF) and QC5 have conspicuous contributions to the values of Km for UGT1A3.

Abstract:Glucosinolates are a well-studied and highly diverse class of natural plant compounds. They play important roles in plant resistance, rapeseed oil quality, food flavoring, and human health. The biological activity of glucosinolates is released upon tissue damage, when they are mixed with the enzyme myrosinase. This results in the formation of pungent and toxic breakdown products, such as isothiocyanates and nitriles. Currently, more than 130 structurally different glucosinolates have been identified. The chemical structure of the glucosinolate is an important determinant of the product that is formed, which in turn determines its biological activity. The latter may range from detrimental (e.g., progoitrin) to beneficial (e.g., glucoraphanin). Each glucosinolate-containing plant species has its own specific glucosinolate profile. For this reason, it is important to correctly identify and reliably quantify the different glucosinolates present in brassicaceous leaf, seed, and root crops or, for ecological studies, in their wild relatives. Here, we present a well-validated, targeted, and robust method to analyze glucosinolate profiles in a wide range of plant species and plant organs. Intact glucosinolates are extracted from ground plant materials with a methanol-water mixture at high temperatures to disable myrosinase activity. Thereafter, the resulting extract is brought onto an ion-exchange column for purification. After sulfatase treatment, the desulfoglucosinolates are eluted with water and the eluate is freeze-dried. The residue is taken up in an exact volume of water, which is analyzed by high-pressure liquid chromatography (HPLC) with a photodiode array (PDA) or ultraviolet (UV) detector. Detection and quantification are achieved by conducting comparisons of the retention times and UV spectra of commercial reference standards. The concentrations are calculated based on a sinigrin reference curve and well-established response factors. The advantages and disadvantages of this straightforward method, when compared to faster and more technologically advanced methods, are discussed here.

Abstract:Two datasets(azole antifungal agents and organic phosphonate esters),that contain 15 compounds respectively,and separated on the cellulose tris(3,5dimethylphenyl carbamate) chiral stationary phase by HPLC, were subjected to a quantitative structure-activity relationship study by hologram QSAR.HQSAR is a new technology for characterizing the structure of molecules,and offers the ability to rapidly generate QSAR models and easily be repeated as compared with conventional QSAR methods.The structures based on the low energy conformation to generate HQSAR models were built and good results were obtained.The cross-validation value q~2 of azole antifungal agents HQSAR model were 0.797(log k_1)、0.541(log k_2),respectively,and the q~2 of phosphonate esters were(0.797)(log k_1)、0.541(log k_2),respectively.These models expounded the effect factors in the separation of azole antifungal agents and phosphonate esters on the CDMPC chiral stationary phase,and afforded as theoretic support for this kind of separating experiments.

Abstract:Solute-related parameters of the sulfur alcohol compounds were calculated by molecule mechanics and quantum chemistry method.The quantitative structure retention relationships(QSRR) of sulfur alcohol on different polar stationary phases were set by multiple linear regression.The results show that there is excellent linear relationship between solute-related parameters and topological indexes.The QSRR models have good stability and accuracy for prediction of retention indexes of sulfur alcohol compounds on the different polar stationary phases.

Abstract:High resolution mass spectrometry (HRMS)-based analytical technique promotes the discovery and development of new bioactive molecules from natural sources. However, challenges for MS analysis of natural products include their structural diversity, numerous trace components, as well as the interference from complex matrices that limits the rapid detection and identification of all target analytes in the extracts. Herein, we presented an integrated analytical approach to obtain profiles of glucosinolates (GLSs) in Eutrema yunnanense, a perennial herb, which is used as a condiment (Wasabi), by ultra-high-pressure liquid chromatography coupled with Orbitrap high resolution mass spectrometry (UHPLC-Orbitrap/HRMS). The intelligent AcquireX deep scan greatly improved the detection efficiency and coverage of data-dependent acquisition (DDA) mode, and enhanced structurally significant product ion interrogation by generating exhaustive MS/MS spectra with more informative fragmentation. Massive HRMS data mining for searching GLSs was then achieved by a modified Kendrick mass defect filter (MKMDF), which enabled the visualization of their homologous characteristics and reduced the complicacy of data post-processing. Ultimately, a total of 175 GLSs were tentatively identified or characterized based on the MS fragmentation patterns, including 52 potentially new compounds among which 37 malonylated GLSs were first discovered. These compounds were then applied to analyse the chemical differentiation between the rhizome and leaf of E. yunnanense. This study provides a feasible approach for screening and confident structure characterization of GLSs and has practical implications for profiling other natural bioactive homologous compounds.

Abstract:INTRODUCTIONGlucosinolates (1-5) are important secondary metabolites found in Isatis indigotica roots. Due to their high hydrophilic and ionic nature, purified glucosinolates often contain salt impurities and moisture. Accurate assessment of their purities is important for glucosinolates being utilised as chemical markers.OBJECTIVETo develop and validate quantitative proton (1 H) nuclear magnetic resonance (qHNMR) methods for purity assessments of aliphatic and indole glucosinolates (1-5).METHODSeveral NMR parameters such as pulse program, relaxation time, and delay time were optimised. Three qHNMR methods were developed using gluconapin (3), neoglucobrassicin (4), and sinigrin (5) for method validation and with maleic acid as internal standard.RESULTSThe quantification was based on the integrated area ratios of an olefinic proton (H-4 for 1-3; H-6 for 4; and H-3 for 5) of the side chain from glucosinolates relative to the olefinic proton from the internal standard using deuterated water (D2 O) as the solvent. The qHNMR methods were successfully applied for purity assessments of four aliphatic glucosinolates (1-3 and 5: progoitrin, epiprogoitrin, gluconapin, and sinigrin), and an indole glucosinolate (4: neoglucobrassicin).CONCLUSIONThe purity of glucosinolates isolated from I. indigotica and commercial sinigrin was accurately assessed using the developed qHNMR method. The qHNMR provides a reliable and superior means to determine the purity of glucosinolates.
Dynamic profiling of intact glucosinolates in radish by combining UHPLC-HRMS/MS and UHPLC-QqQ-MS/MS

Abstract:Glucosinolates (GSLs) and their degradation products in radish confer plant defense, promote human health, and generate pungent flavor. However, the intact GSLs in radish have not been investigated comprehensively yet. Here, an accurate qualitative and quantitative analyses of 15 intact GSLs from radish, including four major GSLs of glucoraphasatin (GRH), glucoerucin (GER), glucoraphenin (GRE), and 4-methoxyglucobrassicin (4MGBS), were conducted using UHPLC-HRMS/MS in combination with UHPLC-QqQ-MS/MS. Simultaneously, three isomers of hexyl GSL, 3-methylpentyl GSL, and 4-methylpentyl GSL were identified in radish. The highest content of GSLs was up to 232.46 μmol/g DW at the 42 DAG stage in the 'SQY' taproot, with an approximately 184.49-fold increase compared to the lowest content in another sample. That the GSLs content in the taproots of two radishes fluctuated in a similar pattern throughout the five vegetative growth stages according to the metabolic profiling, whereas the GSLs content in the '55' leaf steadily decreased over the same period. Additionally, the proposed biosynthetic pathways of radish-specific GSLs were elucidated in this study. Our findings will provide an abundance of qualitative and quantitative data on intact GSLs, as well as a method for detecting GSLs, thus providing direction for the scientific progress and practical utilization of GSLs in radish.

Abstract:As chemical drugs, separation and quantification of the specific enantiomer from the chiral compounds in herbal medicines are becoming more important. To clarify the chemical characterization of chiral glucosinolates—the antiviral active ingredients of Radix Isatidis, an optimized efficient method of HPLC-UV-CD was developed to simultaneously separate and quantify the four main chiral glucosinolates: progoitrin, epiprogoitrin, and R,S-goitrin. The first step was to determine progoitrin, epiprogoitrin, and R,S-goitrin using HPLC-UV, and then determine the R-goitrin and S-goitrin by coupling with CD detection. Subsequently, through the linear relations between anisotropy factor (g factor) and the percent optical purity of R-goitrin, the contents of R-goitrin and S-goitrin from the R,S-goitrin mixture were calculated separately. Furthermore, the chemical composition features of the four chiral glucosinolates in 37 samples from crude drugs, decoction pieces, and granules of R. Isatidis were conducted. The total content of the four glucosinolates was obviously higher in crude drugs, and the variance character of each glucosinolate contents was different. In summary, the accurate measurement method reported here allows for better control of the internal quality of R. Isatidis and its granules and provides a powerful approach for the analysis of other chiral components in traditional Chinese medicines.
Quantitative Analysis of Glucosinolate of Chinese Kale by HPLC-external Standard Method

Abstract:The feasibility of glucosinolate quantitative analysis by Sinigrin as external standard and the glucosinolates in bolting stem of Chinese kale were studied. It was found that there was a positive correlation between the concentration of Sinigrin and peak area under the condition of 14.2 U sulfatase and HPLC measurement. The regression equation was y =3E -07x +0.001 and coefficient of regression is 0.9994. The standard recovery and comparison experiments indicated that Sinigrin external standard method is precise and can be used instead of the expensive internal standard Glucotropaeolin. 11 GS compounds in Chinese kale bolting stems were separated by HPLC and confirmed by electrospray mass spectrometry, including 7 kinds of aliphatic GSs and 4 kinds of indolyl GSs which accounted for 63.41% and 36.59% of total GSs content, respectively. Among them, Glucoerucin was detected in Chinese kale for the first time. The highest content GS in bolting stem of Chinese kale was Gluconapin, while the lowestcontent of GS was Glucoerucin.
QSPR Analysis of Amino Acid Conjugates of Jasmonic Acid

Abstract:[Aims] QSPR analysis of amino acid conjugates of jasmonic acid was investigated with quantum-chemical descriptors for octyl alcohol/water partition coefficient(logP). [Methods] Genetic function approximation(GFA), multiple linear regression(MLR) and partial least square(PLS) based on rational classification of the compounds into training and test set using principal component analysis coupled with cluster analysis were employed to establish the QSPR models.[Results] Optimal model generated by GFA were cross-validated with r2 of 0.846, r2cv of 0.792. [Conclusions] The results can provide a preliminary valuable guidance for continuing search for signal transduction mechanism as well as design and synthesis of amino acid conjugates of jasmonic acid as novel environmental-friendly pesticide.
New RP-HPLC Method for Precise Determination of Chemical Stability of Rhg-Csf Products

Abstract:To set up an efficient analytic method for the determination of chemical stability of rhG-CSF,especially for the precise detection of rhG-CSF oxidation products,two different types of RP-HPLC columns(C4 and C18 column) were used.The methods used in Chinese Pharmacopeia(CP 2010 edition),European Pharmacopoeia(EP6.3 edition) and European Pharmacopoeia(EP6.8 edition) were evaluated by the new RP-HPLC method.The chromatographic variables such as column temperature,load volume,column length on the resolution were also investigated.Different time-course samples of rhG-CSF were applied to validate the difference of purity at different column temperatures.The optimized operation conditions for the new RP-HPLC method were column temperature 60℃,load volume 20~50μL,column length 15cm etc.To check the validity of the new method,the limit of detection,specialization and robustness were measured.In contrast to C18 column,C4 column exhibited better sensitivity and resolution.As for the determination of rhG-CSF chemical stability,the new RP-HPLC method exhibited better resolution and accuracy than those in Chinese Pharmacopeia(CP 2010 edition),European Pharmacopoeia(EP6.3 edition) and European Pharmacopoeia(EP6.8 edition).
Quantitative Structure–chromatographic Retention Relationship for Polycyclic Aromatic Sulfur Heterocycles

Abstract:Polycyclic aromatic sulfur heterocycles (PASHs) are of concern in petroleum geochemistry and environmental chemistry. In the present study, geometrical optimization and electrostatic potential calculations have been performed for 114 PASHs reported previously at the HF/6-31G* level of theory. A group of 25 statistically based parameters have been extracted. Linear relationships between gas-chromatographic retention index (RI) and the structural descriptors have been established by stepwise linear regression analysis. The result shows that two quantities derived from positive electrostatic potential on molecular surface, Vs+¯ (the average value of the positive electrostatic potentials on molecular surface) and σ+2 (a measure of dispersion tendency of positive electrostatic potential), together with Vmc (the molecular volume) and EHOMO (the energy of the highest occupied molecular orbital) can be well used to express the quantitative structure–retention relationship (QSRR) of PASHs. Predictive capability of the model has been demonstrated by leave-one-out cross-validation with the cross-validated correlation coefficient (RCV) of 0.992. Furthermore, when splitting the 114 PASH samples into calibration and test sets in the ratio of 2:1, a similar treatment yields an equation of almost equal statistical quality and very similar regression coefficients, validating the robustness of our model. Predictions for six PASHs from other source have also been made. The QSRR model established may provide a new powerful method for predicting chromatographic properties of aromatic organosulfur compounds.

Abstract:In this work a multi-objective quantitative structure-property relationship (QSPR) analysis approach was reported based on the study on three partition properties of 50 aromatic sulfur-containing carboxylates. Here multi-objectives (properties) were taken as a vector for QSPR modeling. The quantitative correlations for partition properties were developed using a genetic algorithm-based variable-selection approach with quantum chemical descriptors derived from AM1-based calculations. With the QSPR models, the aqueous solubility, octanol/water partition coefficients and reversed-phase HPLC capacity factors of sulfur-containing compounds were estimated and predicted. Using GA-based multivariate linear regression with cross-validation procedure, a set of the most promising descriptors was selected from a, pool of 28 quantum chemical semi-empirical descriptors, including steric and electronic types, to integrally build QSPR models. The selected molecular descriptors included the net charges on carboxyl group (Q(OC)), the 2nd power of net charges on nitrogen atoms (Q(N)(2)), the net atomic charge on the sulfur atoms (Q(S)), the van der Waals volume of molecule (V), the most positive net atomic charge on hydrogen atoms (Q(H)) and the measure of polarity and polarizability (pi), which were main factors affecting the distribution processes of the compounds under study. The statistically best QSPR models of six descriptors were simultaneously obtained by GA-based linear regression analysis. With the selected descriptors and the QSPR equations, mechanisms of partition action of the Sulfur-containing carboxylates were able to be investigated and interpreted.
