Abstract:In this paper, a new method of calculating the diffraction efficiency for diffraction / refraction infrared hybrid system is given. By using this method, an automatic measurement system is designed. This system covers the wavelength range of 1 similar to 3 mu m, 3 similar to 5 mu m and 8 similar to 12 mu m. We use a blackbody as the energy source, put the lens which is to be measured above a computer controlled turntable in order to gain different results in different filed of view. We also have several relay lenses to cope with long or negative focal length. We use two types of detectors to deal with wavelength range of 1 similar to 3 mu m and 3 similar to 12 mu m. Both the detector and preamplifier are assembled together so we can easily change them for different wavelength ranges. To improve the accuracy of the measurements, a new theory of measuring the main energy is developed to minimize the negative impacts of other diffraction orders and the environment. We use an adjustable slit cooperating with a two-axis stage to scan the focal point and by adjusting the width of the slit we can reduce the energy of other diffraction orders and the environment that may come into our detector. During the experiment process we can manually control the slit and two-axis stage to improve the efficiency and use our computer program to improve the accuracy. The energy modulator and lock-in amplifier also help to improve the accuracy of our system. We designed two standard lenses for the 3 similar to 5 mu m wavelength range. Both of them are made of the same material, have the same focal length of 200mm and their shape is almost the same. The only difference between them is that one of them is pure refraction piece which acts as a reference and the other one has a diffraction surface to achromatic aberration. The result fits our theory well and the possible causes of the differences are discussed. This system has a practical meaning in the quality evaluating of infrared hybrid system.
A Method for the Quantitative Analysis of a Key Component in Complex Mixtures Using Raman Spectroscopy Based on Peak Decomposition

Abstract:Raman spectroscopy has been increasingly used in industrial production processes for analytical purposes. However, the quantitative analysis of complex mixtures based on their Raman spectra remains a problem, especially when not all of the spectra of pure components in the mixture are available. In this work, a method for the quantitative analysis of a key component in complex mixtures based on peak decomposition is introduced. The proposed method only requires the spectrum of the main component in the mixture before analysis. The spectra of the mixture are decomposed to the contributions of the main component and a group of parametric Lorentzian functions. Then, the representative peaks of the key component are extracted. Finally, a calibration model is established. This method is applied to a complex mixture containing m -xylene, o -xylene, p -xylene, ethylbenzene, methylbenzene, three trimethylbenzenes, non-aromatic hydrocarbons, etc. to determine the concentration of o -xylene in the mixture. Experiments demonstrate that the proposed method performs better than the commonly-used partial least squares (PLS), especially when the number of training samples is very small.
A Study on the Use of Near-Infrared Spectroscopy for the Rapid Quantification of Major Compounds in Tanreqing Injection

Abstract:In this paper we describe the strategy used in the development and validation of a near infrared spectroscopy method for the rapid determination of baicalin, chlorogenic acid, ursodeoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), and the total solid contents (TSCs) in the Tanreqing injection. To increase the representativeness of calibration sample set, a concentrating-diluting method was adopted to artificially prepare samples. Partial least square regression (PLSR) was used to establish calibration models, with which the five quality indicators can be determined with satisfied accuracy and repeatability. In addition, the slope/bias (S/B) method was used for the models transfer between two different types of NIR instruments from the same manufacturer, which is contributing to enlarge the application range of the established models. With the presented method, a great deal of time, effort and money can be saved when large amounts of Tanreqing injection samples need to be analyzed in a relatively short period of time, which is of great significance to the traditional Chinese medicine (TCM) industries.
Rapid Quantitative Determination of Fritillaria Thunbergii Miq Mixed into Fritillaria Cirrhosa D. Don by Near- Infrared Diffuse Reflectance Spectroscopy

Abstract:Objective: To set up a new method for rapid quantitative determination of Fritillaria thunbergii Miq (FTM) mixed into Fritillaria cirrhosa D. Don (FCD) by combining near-infrared diffuse reflectance spectroscopy (NIRS) with Partial Least Square (PLS). Methods: Collect 41 powder samples with different rate mixed with FTM and FCD, and scan their NIR spectra. Then use Partial Least Square (PLS) to correlate these data and get the bestfitted model. Final use this model to analysis prediction samples. Results: The correlation coefficient (r) for the prediction samples is 0.9997,the recovery is 97.96%-100.9% ,the RSD is 0.81%. Conclusions: The new method proposed could be used directly to determine the mixture of Chinese traditional medicine materials which are similar in appearance and chemical components. It is fast, convenient and accurate.

Abstract:A Raman quantitative analysis algorithm based on genetic algorithm ( GA) and linear additive model was developed. When the Raman spectrum of one main substance in a batch of mixed samples was known, the characteristic peak position of each component could be obtained by correlation analysis. Then genetic algorithm ( GA) was used to fit and optimize the corresponding spectral matrix of the pure substance. Finally, the obtained spectral matrix was used to establish the Raman quantitative analysis model of various substances in the mixture. This method only required a large amount of calculation when optimizing model parameters in the model training process, and only needed to perform simple matrix operations in actual prediction, showing a small real-time calculation amount. The developed method was suitable for continuous online applications, and could be applied to the quantitative analysis of liquid and gas mixture. By taking natural gas as an example, when the Raman spectrum of the main component in methane was known, based on a set of natural gas training samples whose composition and Raman spectrum were known, a quantitative analysis model based on Raman spectrum for main components in natural gas ( CH4, C2H6, C3H8, CO2, N-2, H-2, CO) was developed. For the test sample set, the corresponding root mean square error ( RMSE) were 0. 488%, 0. 103%, 0. 358%, 0. 079%, 0. 087%, 0. 126% and 0. 022%, and the multiple correlation coefficient ( R-2) were 0. 995, 0. 991, 0. 990, 0. 991, 0. 998, 0. 999 and 0. 999, respectively. Although the result was just slightly inferior to the result when the spectra of all pure substances were known, the algorithm presented here provided a solution for the case when some pure substance spectra were unknown.

Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.

Abstract:A non-invasive analysis method for bottled Tanreqing injection was developed based on near infrared (NIR) spectroscopy. The NIR trans-mission spectra were collected using a home-made accessory. The quality indicators of Tanreqing injection, which include baicalin, ursodesoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), chlorogenic acid (CA), forsythin, total amino acids (TAA) and soluble solid content (SSC), were determined with standard methods. Several outlier samples were eliminated using the Dixon test, leverage and studentised residual test and analogous samples were amalgamated. Partial least squares regression (PLSR) was used for establishing the calibration models with the pre-treated spectra and the reference values. The root mean square errors of prediction ( RMSEP) values of external validation samples were 0.098 g L−1, 0.059 g L−1, 0.140 g L−1, 0.038g L−1, 0.007 g L−1, 0.053 g L−1 and 0.538 gL−1 for baicalin, CA, UDCA, CDCA, forsythin, TAA and SSC, respectively, which indicated that the established models possessed satisfactory predictive abilities. The presented method can reduce the workload of product inspection to a large extent. This will allow the sampling proportion to be increased and enable improved quality control during Tanreqing injection production.

Abstract:Objective:To establish a new method of assaying rapidly of active components in steviol glycoside solution. Method: The near infrared transmittance spectroscopy was used to quantify the different batches of steviol glycoside. According to selected different preprocessions and wave bands for information extraction and analysis from spectrums, the best factors and wave bands for quantitative analysis were determined. The calibration models were established using the PLS (partial least squares). Result: The correlation coefficients(R2) for steviolbioside(SX), dulcoside A(DA), stevioside (STV), rebaudioside C(RC), Rebaudioside A(A3) and steviosides(STS) were 0.9271, 0.9355, 0.9759, 0.9706, 0.9500 and 0.9870, and the RMSEP were 0.0004, 0.0015, 0.0150, 0.0043, 0.0170 and 0.0199, respectively. Conclusion: The proposed method was simple and fast, and it provides a novel efficient and environmental friendly approach for the fast determination of the active components in steviol glycoside.
A Novel Calibration Technique for Quantitative Analysis of FTIR Spectra

Abstract:Fourier transform infrared spectroscopy (FTIR) can be used for the continuously on-line quantitative determination of the concentrations of several gas components in smoke gas. The process of acquiring accurate calibration of spectra is one of the most important steps in the quantitative analysis. In the present paper, a novel calibration technique was proposed and evaluated by the results of CO and HCl. Data obtained by this new technique were compared to those of conventional concentration-based calibration. The results showed that this calibration technique is feasible, and its reliability and validity were also confirmed. Further more, it has several other advantages, which will also be discussed here. Finally, the calibration problem that the substances are liquid at ambient temperature was solved by this technique.
A Modified Standard Addition Method and Its Application in ICP Emission Spectrometry

Abstract:When a small volume (<50 μL)of liquid is sampling,it is easy to produce large sampling error.In order to reduce the uncertainty of analytical results,the quantification based on the sample volume is usually replaced by the quantification based on the sample mass.The conventional standard addition method (SAM),in which the quantification is based on the sample vol-ume,cannot be applied when the quantification is based on the sample mass.Therefore,a modified SAM with mass-based quan-tification is proposed.As a demonstration,the modified SAM has been applied to determine low or trace elements Hg,Mo and Rh in complex solution with ICP-OES.The aim of the SAM is to correct the matrix effect,while the matrix effect includes two kinds of interferences:constant interference and proportional interference.Only the proportional interference can be corrected with the modified SAM,which can characterize the extent of the proportional interference with a quantitative index k :k = 1 means there is no proportional interference;while the larger difference between k and 1 means greater proportional interference existing in the sample.As for constant interference,it can be reduced or eliminated by applying the background correction func-tion of the ICP-OES instrument.The uncertainty of analytical results mainly comes from background correction,and depends on the signal-to-background ratio of the analytical line selected.The lower the signal-to-background ratio is,the greater the uncer-tainty of background correction is.Therefore,in the actual analysis,we should try to choose the analytical line which has as high signal-to-background ratio as possible.Otherwise,the final analytical result maybe contains very large uncertainty,even though the background correction has been made in advance.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Abstract:Due to the advantages of fast and noninvasive, requiring almost no sample preparation, supplying chemical and physical information, near infrared (NIR) spectroscopy is widely used in the pharmaceutical industry. In the life cycle of an established analytical method, rigorous validation should be performed before routine use. However,there is no detailed experimental standards or decision rules for the NIR methods validation currently, and various approaches were applied to validate the established NIR quantification models in literatures.In this paper, progress on the validation of NIR methods developed for the pharmaceutical application was reviewed, concentrating on the standard documents and main ideas of validation.It will provide references for the standardized application of NIR spectroscopy in the pharmaceutical industries.
Study on the Quantitative Analysis of Multi-Component Toxic Gases Based on FTIR

Abstract:The paper is concerned about a measuring method developed by the present author based on FTIR for measuring the concentrations of components in the toxic gases harmful both to human health and the environment quality.The said experimental system was established based on the use of Fourier transform infrared spectrometer, gas compounding equipment,vacuum pump as well as other devices. To make the quantitative analysis successful,we have chosen eight kinds of classic toxic gases with low concentration,including CO, CO_2,NO,NO_2,SO_2,HCl,HBr and HCN.The best calculated spectrum regions of the 8 components were analyzed in hoping to avoid the interference of vapor and carbon dioxide from the outside environment as the result of a series of trials and appropriate choice. The sample of calibration set has been tested and verified by the method of Mahalanobis distance (M) for the model to be soundly established. For example,we have deleted two samples due to their being less purposeful for our research goals.Then,two data matrixes were adopted to establish our model,one of which is infrared spectrum data matrix and the other of which is the gas concentration data matrix.And,finally,the partial least square (PLS) regression model was established after choice of an appropriate spectrum region,data pre-treating,standards selection and parameters setting.With the result of each component verified by the P1.S model and regulated by the multinomial,the fitting regression coefficient between the actual concentrations and predicted concentrations of each component proved to be higher than 0.99,and the Root-mean Standard F.rror of Calibration (S_(RMSEC)) proves lower than 15 × 10~(-6).With the expected performance of the model well verified by the standards in validation set,the error of the concentration prediction proves lower than ±2% FS and the Root-mean Standard Error of Prediction (S_(RMSEP)) is lower than 20 × 10~(-6).Thus,it can be seen that the proposed qualitative analysis of such toxic gases based on FTIR is a new valuable method for gas detection,which can be applied for environmental monitoring, chemical plants' quality-inspection,fire-hazards detection,anti-terrorist activities and other areas concerned.

Abstract:Near infrared spectroscopy combined with chemometrics was investigated for the fast determination of total organic carbon (TOC) and soluble solids contents (SSC) of Tanreqing injection intermediates. The NIR spectra were collected in transflective mode, and the TOC and SSC reference values were determined with Multi N/C UV HS analyzer and loss on drying method. The samples were divided into calibration sets and validation sets using the Kennard–Stone (KS) algorithm. The Dixon test, leverage and studentized residual test were studied for the sample outlier analysis. The selection of wavebands, spectra pretreated method and the number of latent variables were optimized to obtain better results. The quantitative calibration models were established with 3 different PLS regression algorithms, named linear PLS, non-linear PLS and concentration weighted PLS, and the net result was defined as the average of the predicted values of the different calibration models. The overall results indicated that the presented method is more powerful than single multivariable regression method, characterized by higher mean recovery rate (MRR) of the validation set, and can be used for the rapid determination of TOC and SSC values of Tanreqing injection intermediates, which are two important quality indicators for the process monitoring.

Abstract:The feasibility of the method to determine the content of water in polymerization liquid by NIR spectroscopy was explored.The standard data for mathematical model establishment came from water content by Microcoulomb equipment.By first derivative,the sensitivity and accuracy of the method were improved.The test results by NIR and standard method had little difference.NIR′s relative deviation was less than 5%.The results of the paired t test also showed that there was no obvious difference between NIR method and standard method.It was showed that the presented method had a good repeatability and stability and could be applied to online detection in industrial process.

Abstract:Quantitative analysis of trace gases is an important research field in analytical chemistry. The terahertz electronic spectrometer is one of the most powerful tools for detecting trace gas. Here, a terahertz spectrometer based on frequency multiplier chain and heterodyne detection was presented. The rotational spectra of acetonitrile (CH3CN) gas were measured in the 290-370 GHz frequency band with 100 kHz spectral resolution. The spectrometer demonstrated excellent spectral specificity and the extrapolated limit of detection for CH3CN gas of 1.4 ppm. Furthermore, a novel quantification method of trace gas was proposed based on broadening mechanisms. The CH3CN self-and nitrogen (N-2)-collisional broadening coefficients were obtained experimentally for verifying the method. The CH3CN concentration of the validation group was calculated, and the relative error was 0.1%. The error analysis of the different number of measurements of the method was carried out. Abstract:In FTIR film thickness measurement system, traditional process for determining film thickness is optical interference method or absorption method. Tn practice, both interference effect and absorption effect make a partial contribution to spectrum respectively. When both of the two effects have influence on measurement, neither interference method nor absorption method can be individually used to determine the film thickness accurately (e.g. film thickness is about 5 mu m similar to 50 mu m). A new mathematical correction method is described for FTIR film thickness measurement in this case. Employing this new mathematical correction method, the effects of interference and absorption can be apart from each other effectively. The film thickness measurement accuracy of lambda/100 has been achieved.
