Abstract:[Objective] In order to providea reference for effective prevention and control of bacterial disease of Procypris merus,the study was conducted to isolate and identify the pathogen of bacterial disease in Procypris merus in the Feijiping fingerling farm in Quanzhou Guangxi,the pathogenicity,antibiotic resistance and six kinds of virulence genes of the pathogen were tested.[Method] The pathogen was isolated from parts such as the liver,heart and spleen tissues in the diseased Procypris merus in this study.An artificial infection was measured on the pathogenicity of the isolated strains.Bacterial identification was finished by API biochemical analysis combined with 16S rRNA gene sequencing.The K-B paper diffusion method was used to test antibiotic resistance of the pathogen,presence of virulence genes was determined by using the PCR method.[Result] Two virulent pathogenic strains TH1 and TH3 were isolated from diseased Procypris merus.The two strains both contributed a fatality rate of 100.00 ％ to healthy Procypris merus carps.Moreover,the strains were both identified as Aeromonas hydrophila,and had the closest genetic relationship relatives with the standard strain ATCC 7966 (NR 118944) of A.hydrophila,sharing homology of 99.80 ％.The two strains were highly sensitive to five kinds of antibiotics including neomycin sulphate,however,they were insensitive to four kinds of antibiotics including penicillin G.Strain TH1 carried six kinds of virulence genes including hly,Aer,Alt,Act,ahal and ahp.Whereas strain TH3 carried five virulence genes including hly,Aer,Alt,Act,ahal.[Conclusion] The pathogens causing fulminant death of Procypris merus were A.hydrophila in the Feijiping fingerling farm in Quanzhou,Guangxi.The genotypes of the pathogenic strain TH1 and TH3 were hly + Aer + Alt + Act + ahal + ahp + andh ly + Aer +Alt + Act + ahal + ahp-,respectively,and the pathogens contributed a fatality rate of 100.00 ％ to healthy Procypris merus.The pathogens were highly sensitive to five kinds of antibiotics including neomycin sulphate,however,they were insensitive to four kinds of antibiotics including penicillin G.
