Effect of hypertonic saline resuscitation on intestinal mucosal barrier function in rats with traumatichemorrhagic shock

Abstract:Objective To investigate the effect and underlying mechanism of lactated Ringerr′s solution and hypertonic saline resuscitation on the function of intestinal mucosal barrier in rats with traumatic-hemorrhagic shock.Methods A total of 80SD rats was randomly divided into four groups of A(sham operated,8rats),B(traumatic-hemorrhagic shock model,8rats),C(resuscitated with lactated Ringerr′s solution,32rats)and D(resuscitated with 7.5% hypertonic saline followed by addition of lactated Ringerr′s solution,32rats).Plasma levels of diamine oxidase(DAO)and D-lactate were determined by spectrophotometry,and the morphologic changes of small intestine tissues were examined under light microscope.Results Compared with group A,DAO activity,D-lactate concentration and intestinal mucosal damage index were all significantly increased at different times in groups of C and D,especially in group C(P0.05).Compared with group A,small intestinal villous height and intestinal mucosal thickness were obviously decreased at 3and 6hafter resuscitation in groups of C and D,especially in group C(P0.05).Conclusion Application of 7.5% hypertonic saline during early resuscitation of traumatic hemorrhagic shock shows fewer dosage,faster infusion speed and better blood pressure maintenance than traditional lactated Ringerr′s injection,and is able to diminish the damage of intestinal mucosal barrier and maintain the morphology and structure of intestinal mucosa in rats.
