Abstract:The micellar electrokinetic capillary chromatography(MECC) combined with gas chromatography(GC)were used to determine the reactants,intermediates and end-products of liquid phase oxidation of p - xylene to terephthalic acid at high temperature. p -Xylene(PX)?4-methylbenzyl alcohol (TALC)? p - tolualdehyde (TALD)?4-carboxybenzaldehyde (4-CBA)? p -toluic acid(PT) and terephthalic acid(TA) were analyzed quantitatively within 8 minutes.The method was rapid,convenient and accurate.

Abstract:Summary A method for dead time determination in reversed-phase capillary electrochromatography (RP-CEC) with methacrylate-based monolithic stationary phases has been presented based on homologue series method. The dead times measured with homologue series method and thiourea, a commonly used electroosmotic flow (EOF) marker are compared under various conditions, such as the type and the concentration of organic modifier, ionic strength, and applied voltage. Results show that the former one is larger than the latter one. Under high ionic strength and low applied voltage, EOF velocity measured by thiourea is close to that determined by homologue series method.
Determination of residual in Erdosteine by headspace gas chromatography

Abstract:OBJECTIVE To develop a headspace gas chromatography method for determination residual organic solvents-Tetrahydrofuran and Isopropylether in erdosteine.METHODS Headspace gas chromatography was used with Angilent INNOWAX capillary column(30m×0.32mm,0.25μm) and temperature programming.The initial column temperature was 70℃ maintained for 3min and then raised(40℃·min-1) to 160℃ for 1min.with nitrogen gas as carrier and FID as detector,The flow of carrier is 1.0mL·min-1,The injection port and detector temperature were 200℃ and 250℃ respectively.RESULTS With external standard method,the solvent-Tetrahydrofuran and Isopropylether residuals in samples were less than the limits of ChP.CONCLUSION The method could be used for the determination of residual solvents in Erdosteine.

Abstract:The dead time was obtained from thiourea and calculated from retention time of benzene homologues,and they were compared.Solute retention equation for high performance liquid chromatography was applied to capillary electrochromatography based on this.Relation curve of retention value with content of organic modifier in binary mobile phase system was obtained.The experimental results showed some polar compounds that are not charged could migrate under electric field due to their polarity and their retention value could be changed.The change was influenced by content of organic modifier in mobile phase.

Abstract:Objective:To establish a method of headspace capillary gas chromatography for the detection of residual solvents,including chloroform,ethyl acetate,hexane,ethanol,benzene,methyl benzene,dimethyl benzene,ben- zene vinyl and divinyl - benzene in extracts of Herba Epimedii.Methods:The nine residual solvents were separated by Agilent INNOWAX capillary column (30 m×0.320 mm,0.25μm) with an FID detector.The carried gas was nitrogen.The column temperature was programmed from 60℃(5 min isothermal) to 120℃ at a rate of 5 ℃· min~(-1),holding 2 min,then raised to 180℃ with a rate of 15℃·min~(-1), this finished temperature was main- tained for 3 min.Results:All the nine residual solvents were well separated in 26 minutes.The recoveries of residu- al solvents were between 82.26%-117.00%,with RSD less than 10.32% (n=3).The night solvents' limits of detection were among 0.00001706%-0.004890%.Only ethyl acetate and ethanol were detected in the extracts of Herba Epimedii,and the contents are all lower than 0.15%.Conclusion:The established method can be used for the detection of residual solvents in Extracts of Herba Epimedii,and the contents of these residual solvents are all meet the standards of ChP.

Abstract:A theoretical study on the velocity of electroosmotic flow (EOF) and the retention times of neutral solutes under multiple-step gradient of capillary electrochromatography (CEC) was carried out, focusing on that with three kinds of mobile phases. Through the model computations, the detaining time of the second kind of mobile phase in the column was proved to play an important role in affecting EOF. The variation speed of EOF was shown to be determined by the differences among dead times in different steps. In addition, the prediction of the retention times of 13 aromatic compounds under gradient mode was performed with the deduced equations. A relative error below 3.3% between the calculated and experimental values was obtained, which demonstrated the rationality of the theoretical deduction. Our study could not only improve the comprehension of stepwise gradient elution, but also be of significance for the further optimization of separation conditions in the analysis of complex samples.

Abstract:The influence factors of electroosmotic flow (EOF) in reversed-phase capillary electrochromatography were studied on a 75 μm I. D. × 20 cm capillary column packed with 3 μm ODS-silica. The factors of separation on EOF were systematically examined, including the composition of the mobile phase, the kind and percentage of organic modifiers, the kind, concentration and pH value of electrolyte buffers. The effects of the applied voltage and column temperature on the electroosmotic flow were also investigated. A rapid separation electrochromatogram of six substituted benzenes under the optimum condition was obtained.

Abstract:Summary Polymethacrylate-based monolithic columns were prepared for capillary electrochromatography (CEC) by in situ copolymerization of butyl methacrylate (BMA), 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS), and ethylene dimethacrylate (EDMA) in the presence of a porogen in fused-silica capillaries of 100 μm I.D. The abnormal phenomenon that retention factors for neutral species decreases with applied voltage in CEC was observed. Capillary electrophoresis (CE) instruments usually require a period of time to increase voltage from 0 kV to desired value, which is called as ramp time. Such ramp time and any error in the determination of dead time should be taken into account during the accurate calculation of retention factors. After the correction of the retention factors, the plots of the corrected factors for alkylbenzene versus applied voltage were made, the absolute value of the plot slopes are less than 1.8×10 −4 , which indicates that the corrected retention times for neutral species do not show any dependence on applied voltage. Further, the plats of the corrected retention times for acidic and basic compounds versus the reciprocal of applied voltage were drawn, where the target compounds were eluted in neutral form. The very nice linearity of the plots was obtained. The linear correlation coefficients are over 0.999. Here, the slopes of the plots represent L_eff· L_tot·( l + k)/μ eo , which indicates that retention factors for the test compounds are constants under a given separation condition.

Abstract:In capillary electrochromatography (CEC), the larger the internal diameter of a capillary column is, the higher the Joule heat is, the easier-the bubbles are gotten. In this paper, a new method was given to solve this problem. We got higher column efficiency in separating benzyl substitution on 320 mum CEC column by using lower concentration of organic salt as a buffer than that from pressurized CEC. General principle was given that's how to control the components of the mobile phase by choosing a suitable buffer.

Abstract:A phenomenological approach was presented to describe the retention behaviors of solutes in capillary electrochromatography (CEC). Equations for calculation of the retention time and actual chromatographic retention factor for ionic solutes, weak monoprotic acid and weak monoprotic base were derived, which can be described by two general expressions regardless the charge status of the solute. The general expressions enable calculation of the retention time and retention factor in CEC from chromatographic and electrophoretic data, which were experimentally verified with a variety of compounds and a variety of CEC modes. Based on this approach, the chromatographic retention and the electrophoretic migration in the CEC systems studied were found to be two independent processes. The validity of this approach was also discussed.
Separation and Determination of Nycodenz and Its Impurities by Capillary Electrophoresis

Abstract:Separation and determination of nycodenz (a new radiography reagent) and its impurities by capillary electrophoresis was investigated. Since the analytes were polyols, Na2B4O7-H3BO3 was chosen as the buffer system. The analytes were negatively charged by complexing with borate. Then cetyltrimethylammonium bromide was used to reverse electroosmotic mobility in order to shorten the analysis time. 2-Propanol was added to optimize the separation further. Acceptable results of linearity, precision of corrected peak area and migration time were obtained. The RSD(n = 6) of corrected peak area and migration time were 0.73% and 1.4% respectively. The results obtained by CE were directly compared to those obtained by high performance liquid chromatography.

Abstract:Co-electroosmotic capillary zone electrophoresis (CZE) with direct UV detection was developed for simultaneous determination of inorganic anions, carboxylic and aromatic carboxylic acids. These solutes were separated using a 30 mM phosphate buffer containing 1.0 mM tetradecyltrimethylammonium bromide (TTAB) and 20% (v/v) acetonitrile at pH of 6.5 and directly detected by UV at 190 nm. Calibration curves were linear in the range 0.01–2.0 mM, depending of the solutes. The detection limits ranged from 1.0 to 8.0 μM and the relative standards deviations (n=5) in range from 1.9 to 3.6% for the peak area. The proposed method was used to determine inorganic anions and carboxylic and aromatic acids in soil and plant tissue extracts.

Abstract:An equation describing the capillary electrochromatographic (CEC) retention factor is derived using the fundamental chromatographic concept of retention factor. This retention equation couples both the chromatographic retention factor and the electrophoretic velocity factor, which can explain well the charged solute retention in CEC. The known CEC retention expressions are compared. A simple experimental method for obtaining the CEC retention factors for charged solutes is demonstrated.

Abstract:A method was developed using micellar electrokinetic capillary chromatography (MECC) to analyze and determine the content of the benzene homologue compounds in marine shallow sediment samples. A fused silica capillary of 57cm×50μm (50cm to detector) was used. Running buffer was 2.0 mmol/L aqueous tetraborate solution containing 50mmol/L sodium dodecyl sulfate (pH 9.16) and 20% methanol as an organic modifier. The voltage used was 25 kV. Detection wavelength was 200nm.Benzene, toluene, ethylbenzene and xylene were separated well within 15min.Peak area was measared for quantitative analysis. The calibration curves were linear over 2～100mg/L. The minimum detectable concentrations were 0.75mg/L for benzene, 0.45mg/L for toluene, 0.39mg/L for ethylbenzene, 0.1mg/L for o-xylene and 0.31mg/L for p-xylene. Benzene homologue compounds in marine shallow sediment samples, which deposit in the prospect area of oil exploration, were analyzed using proposed method. The detection ranges obtained for these compounds were (0.0x～0.x)μg/g.
High-efficiency On-Line Concentration Technique of Capillary Electrochromatography.

Abstract:With the coexistence of the mobile phase, the stationary phase, and the electric field in capillary electrochromatography, the chromatographic zone-sharpening effect and field-enhanced sample-stacking technique were utilized to improve detection sensitivity. By the former means, with less organic modifier in the sample solution compared to that in the mobile phase, the concentration factors of neutral solutes benzoin and mephenytion were 134 and 219, respectively. Through the latter one, without electrolyte in the sample solution, the detection sensitivity of propatenene with positive charge was improved by 1600 times. While with the combination of these two methods, improvement of over 17,000 times for the sensitivity of propatenene was obtained. By the combined means, the analysis of basic pharmaceutical compounds at concentrations of nanograms per milliliter by UV detection was realized. In addition, parameters that might affect the efficiency of on-line concentration were studied and equations that described the on-line concentration procedure were deduced.

Abstract:Monolithic columns of capillary electrochromatography (CEC) with weak electroosmotic flow (EOF) have been prepared by in situ polymerization of butyl methacrylate and ethylene dimethacrylate, without any charged groups in the reaction mixture. The reproducibility of such columns has been proved good no matter whether they are prepared in the same batch or in different batches. In the case of BMA-EDMA monoliths, besides the traditional ternary mixture - 1-propanol, 1,4-butanediol, and water, binary porogenic solvents with only alcohols have also been adopted. Compared with ternary porogenic solvents, the design with binary ones allows for fine control of the pore diameter and the formation of the specific surface of the monolithic polymers. The composition of porogenic reagents has also been shown to have an effect on EOF in the column systems. In addition, the Joule heat effect in such columns has been studied by varying the inner diameter of columns. Through the separation of acidic compounds, monolithic columns with low EOF have shown potential in the analysis of charged samples.

Abstract:The separation of selected lignin/humic substance (HS) degradation compounds by capillary electrochromatography (CEC) with a methacrylate-based monolithic column and a conventional column packed with 5 microm octadecyl silica (ODS) particles is presented. The effects of organic modifier concentration, pH of the mobile phase, ionic strength, applied voltage, and temperature on the separation were investigated to determine the optimal separation conditions. With the increase of pH in the mobile phase, some of analytes start to ionize and both chromatographic partition and electrophoresis can play roles in separation simultaneously. Accordingly, different selectivity from high-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) could be achieved. The performances of both kinds of columns were compared. The results showed that the peaks of compounds obtained on the former column were much wider than those on the latter one, although good separation efficiency of alkylbenzenes could be readily achieved; the most probable reasons for this behavior and method to solve this problem were briefly discussed. The CEC of a soil fulvic acid with a monolithic column produced partly resolved broad bands; by means of nuclear magnetic resonance (NMR) analysis a wide range of oxygen derived aromatic substitution patterns was found with prominent contributions from phenolic and carboxylic groups.

Abstract:Characteristics of electroosmotic flow (EOF) and the migration of neutral solutes under double stepwise gradient elution in capillary electrochromatography were studied systematically. EOF velocity proved to be the function of operation time changing with the introduction of the second mobile phase. Accordingly, the retention of components also changed. The migration of neutral solutes was studied under the following three situations; A, components eluted when the column was filled only with the first kind of mobile phase; B, solutes eluted still in the first kind of mobile phase while at that time two kinds of mobile phase coexisted in the column and C, samples eluted in the second kind of mobile phase. Equations to describe the retention times of components under these three kinds of conditions were deduced and applied to predict the retention times of 12 aromatic compounds. Relative errors between experimental and calculated values were below 5.0%, which proved the reliability of the equations. In addition, parameters that might affect the retention time of solutes, such as the transferring time of mobile phase vials, the capacity factors of components and EOF velocities two steps were studied systematically.
A Study on Peak Compression Effect in Ion-Exchange Capillary Electrochromatography with Relaxation Theory

Abstract:Due to the electric characteristics of the solute transport in capillary electrochromatography CEC some special phenomena might occur. An example is the peak compression effect in ion-exchange capillary electrochromatography which cannot be explained with common chromatography theory. According to the basic model of relaxation theory it is considered that there are co- and counter-solute flows in mobile phase and in stationary phase the expressions of the first moment and the second center moment of profile and column efficiency are derived by the method of Laplace transform. The relations of electrophoresis velocity electroosmosis velocity and electric dispersion velocity are also investigated with these expressions. The results show that this new model offers a better explanation for peak compression effect on the migration behaviour of charged solute in CEC. The retention time will be shorter by peak compression effect which is similar to a solvent gradients process. The peak compression effect is not steady and doesn t appear until the conditions are fitted in some special situation.

Abstract:The separation mechanisms in capillary electromatography (CEC) of bare silica - packed column was investigated under aqueous - organic mobile phases. The separations of five typical solutes, including apolar compound (benzene), polar compounds (formamide and dimethyl formamide) and basic compounds (pyridine and quinoline), showed that reverse phase retention, normal phase retention, ion-exchange and electrophoresis mechanisms were involved. The effect of the concentration of organic modifier, the concentration of buffer and the pH value on separation were investigated. Due to the hydrophilicity of bare silica, the silica-packed column exhibits the following merit, i. e., the mobile phase composition can change in rather large range of organic modifier content, buffer concentration and pH value. This means that the bubble problem that are usually met in ODS-packed column can be minimized with the silica-packed column. Additionally, we presented a novel model of CEC, termed as dynamically modified silica column CEC. Five alkyl substitued benzenes were baseline separated with a mobile phase containing cetyltrimethylammonium bromide.
