Abstract:Araneoid spiders use specialized abdominal glands to produce up to seven different protein-based silks/glues that have various mechanical properties. To date, the fibroin sequences encoding egg case fibers have not been fully determined. To gain further understanding of a recently reported spider silk protein gene family, several novel strategies were utilized in this study to isolate two full-length cDNAs of egg case silk proteins, cylindrical silk protein 1 (CySp1, 9.1 kb) and cylindrical silk protein 2 (CySp2, 9.8 kb), from the wasp spider, Argiope bruennichi. Northern blotting analysis demonstrated that CySp1 and CySp2 are selectively expressed in the cylindrical glands. The amino acid composition of raw egg case silk was closely consistent with the deduced amino acid composition based on the sequences of CySp1 and CySp2, which supports the assertion that CySp1 and CySp2 represent two major components of egg case silk. CySp1 and CySp2 are primarily composed of remarkable homogeneous assemble repeats that are 180 residues in length and consist of several complex subrepeats, and they contain highly homologous C-termini and markedly different N-termini. Our results suggest a possible link between CySp1 and CySp2. In addition, comparisons of stress/strain curves for dragline and egg case silk from Argiope bruennichi showed obvious differences in ultimate strength and extensibility, and similarities in toughness.

Abstract:Silkworms have been developed over thousands years to optimize folding and crystallization of fibroin under highly controlled conditions which have resulted in their efficient fiber formation. In this paper, we reconstructed the three-dimensional architecture of the spinneret of a wild silkworm Samia cynthia ricini from approximately 1000 optical micrographs of the semi-thin cross sections. The chitin plates and muscles were observed in the silk press part together with large change in the diameter of the spinneret lumen at the press part by large shear stress. This is similar to the case of the spinneret of Bombyx mori silkworm, indicating that the structural change in the silk fibroin of S.c. ricini silkworm occurs exclusively at the silk press part due to large shear stresses. Molecular dynamics (MD) calculations were then performed to study the structural change that occurs in the crystalline region of S.c. ricini silk fibroin under shear stress. Namely, using the peptide AGGAGG(A)12GGAGAG as a model of the crystalline part of the silk fibroin under different shear stresses in the presence of water molecules and followed by molecular mechanics (MM) calculation after removal of water molecules. The simulation indicates that the Ala residues in the model peptides adopt a predominantly β-sheet structure under shear stresses of above 1.0GPa.

Abstract:Silk has a long history of use in medicine as sutures. To address the requirements of a mechanically robust and biocompatible material, basic research to clarify the role of repeated sequences in silk fibroin in its structures and properties seems important as well as the development of a processing technique suitable for the preparation of fibers with excellent mechanical properties. In this study, three silk-like protein analogs were constructed from two regions selected from among the crystalline region of Bombyx mori silk fibroin, (GAGSGA)(2), the crystalline region of Samia cynthia ricini silk fibroin, (Ala)(12,) the crystalline region of spider dragline silk fibroin, (Ala)(6), and the Gly-rich region of spider silk fibroin, (GGA)(4). The silk-like protein analog constructed from the crystalline regions of the spider dragline silk and B. mori silk fibroins, (A(6)SCS)(8), that constructed from the crystalline regions of the S. c.ricini and B. mori silk fibroins, (A(12)SGS)(4), that constructed from and the crystalline region of S. c.ricini silk fibroin and the glycine-rich region of spider dragline silk fibroin, (A(12)SGS)(4),were expressed their molecular weights being about 36.0 kDa, 17.0 kDa and 17.5 kDa, respectively in E. coli by means of genetic engineering technologies. (A(12)-SCS)(4) and (A(12)SGS)(4) undergo a structural transition from alpha-helix to beta-sheet on a change in the solvent treatment from trifluoroacetic acid (TFA) to formic acid (FA). However, (A(6)SCS)(4) takes on the P-sheet structure predominantly on TFA treatment and FA treatment. Structural analysis was performed on model peptides selected from spider dragline and S. c.ricini silks by means of C-13 CP/MAS NAIR.
From Structure To Mechanical Properties Of Spider Silk: Surviving From Severe Collision

Abstract:Spiders and silkworms, the two major sources of silk in nature, have developed the capacity by evolution to spin silk independently, however their fibers have distinct mechanical properties. Spider dragline silk, the main structural web silk, is exceptionally strong and extensible. Its toughness is far superior to that of commercial polyaramid filaments which are used in materials for bulletproof vests and reinforced composites for aircraft panels. As a remarkable biomaterial that has been optimized by millions of years of evolution, spider dragline silk is not only extremely light and strong, but also displays the function of adsorbing the energy of a severe collision. The property of stretch-hardening of spider silk plays an important role in capturing flying insects. It is observed that the spider web can snag a bee flying at 30 km/hour without breaking. However, such a severe collision to the web could knock down the spider from the web if the silk fibers are rigid. On the other hand, the property of stretch-hardening makes spider silk can dissipate the shocking energy quickly, and effectively prevents a drastic impact to the web. Our results show that compared to silkworm silk, spider dragline can acquire extra toughness as a stretch-hardening material. In contract to spider dragline silk, the silkworm silk from a cocoon shell layer is meant to protect the moth pupa against any possible external attack. Therefore, the property of stretch-hardening is not required for the silkworm silk fiber. In this talk, I will discuss the structures leading to the different properties of spider silk and silkworm silk.

Abstract:Atomic force microscopy was used to study the three-dimensional nanometer-scale structure of the dragline silk of Nephila clavipes from microtomed sections of the silk. Contrary to a previously proposed model of randomly distributed protein crystallites interspersed in amorphous regions, a highly organized skin-core structure of the fiber was observed. The skin appeared to be thin with no discernible distinct features. The core consists of pleated fibril-like structures, which are arranged in two concentric cylinders. Upon stretching, the pleats were smoothed out substantially. The mechanical properties of spider silk can quite straightforwardly be related to the newly observed structures.

Abstract:Spider dragline silk is of practical interest because of its excellent mechanical properties. However, the structure of this material is still largely unknown. In this article, we report what we believe is a new model of the hierarchical structure of silk based on scanning electron microscope and atomic force microscope images. This hierarchical structure includes β-sheet, polypeptide chain network, and silk fibril. It turns out that an exceptionally high strength of the spider dragline silk can be obtained by decreasing the size of the crystalline nodes in the polypeptide chain network while increasing the degree of orientation of the crystalline nodes. Based on this understanding, how the reeling speed affects mechanical properties of spider dragline silk can be understood properly. Hopefully, the understanding obtained in this study will shed light on the formation of spider silk, and consequently, on the principles for the design of ultrastrong silk.

Abstract:Spider dragline silk is one of the toughest materials known and understanding the hierarchical structure is a critical component in the efforts to connect structure to function. In this paper, we take the first step in elucidating the hierarchical fractal structure of $\beta$-sheet nano-crystallites, which form a robust self-similar network exhibiting an non-linear mechanical property. A combined small angle X-ray scattering (SAXS) and wide-angle X-ray scattering (WAXS) study of the nano-crystalline component in dragline silk fibers from several species of spiders including, Latrodectus hesperus, Nephila clavipes, Argiope aurantia and Araneus gemmoides is presented. SAXS structure factors exhibit a `lamellar peak' in the q-range from 0.60 to 0.82 nm$^{-1}$ for various spider dragline silk fibers, indicating the presence of strong nano-crystal ordering on the $>$10 nm length-scale. The stochastically reconstructed electron density maps indicate that the $\beta$-sheet crystals are hierarchically structured as mass fractals and that nano-crystals tend to form 10 to 50 nm sized clusters with long-range crystalline ordering. This nano-crystal ordering along the fiber axis also helps to explain the difference between axial and radial sound velocities recently measured by Brillouin spectroscopy.

Abstract:Significant difference in microscopic structure of cocoon filaments from three silkworm varieties which shows extremely fine,coarse or normal size of silk were obseved in the present paper by scanning electron microscope.Results attained in this study showed that the hybrid Xinmiao×Mingri was not only much thicker in filament size but also better in tensile strength,which were confirmed by the physical properties of raw silk and fabric test.

Abstract:This study investigates the structural and optical properties in the cribellate silk of the sheet web spider Stegodyphus sarasinorum Karsch (Eresidae) and the combined dragline, viscid silk of the orb-web spiders Argiope pulchella Thorell (Araneidae) and Nephila pilipes Fabricius (Nephilidae). X-ray diffraction (XRD), Fourier transform infra-red (FTIR), Ultraviolet-visible (UV-Vis) and fluorescence spectroscopic techniques were used to study these three spider silk species. X-ray diffraction data are consistent with the amorphous polymer network which is arising from the interaction of larger side chain amino acid contributions due to the poly-glycine rich sequences known to be present in the proteins of cribellate silk. The same amorphous polymer networks have been determined from the combined dragline and viscid silk of orb-web spiders. From FTIR spectra the results demonstrate that, cribellate silk of Stegodyphus sarasinorum, combined dragline viscid silk of Argiope pulchella and Nephila pilipes spider silks are showing protein peaks in the amide I, II and III regions. Further they proved that the functional groups present in the protein moieties are attributed to α-helical and side chain amino acid contributions. The optical properties of the obtained spider silks such as extinction coefficients, refractive index, real and imaginary dielectric constants and optical conductance were studied extensively from UV-Vis analysis. The important fluorescent amino acid tyrosine is present in the protein folding was investigated by using fluorescence spectroscopy. This research would explore the protein moieties present in the spider silks which were found to be associated with α-helix and side chain amino acid contributions than with β-sheet secondary structure and also the optical relationship between the three different spider silks are investigated. Successful spectroscopic knowledge of the internal protein structure and optical properties of the spider silks could permit industrial production of silk-based fibres with unique properties under benign conditions.

Abstract:This manuscript reviews work from our laboratory that addresses the orientation, secondary structure, wet-spinning, and molecular basis for supercontraction of spider silk. It identifies the poly(alanine) runs as the crystalline regions, establishes the degree of orientation of these regions, and identifies the secondary structural elements of the conserved L-G-X-Q (X = G, S, or N) regions. It also describes methods for spinning very small amounts of protein polymers and it sets forth several molecular-level hypotheses concerning supercontraction.

Abstract:Antheraea pernyi (A. pernyi) silk is produced and used by "wild" silkworms to construct a cocoon, but the primary structure of its protein is rather similar to that of spider major ampullate silk used to build web and dragline. Studies on this specific silk may provide valuable knowledge about the structure-property relationship for the whole animal silk family. In this work, A. pernyi silk fibers with few macroscale defects are obtained by forcibly reeling, and are investigated in detail. It is found that such silk fibers display breaking stress and toughness of the same magnitude as spider major ampullate silks and forcibly reeled mulberry silk. The other mechanical properties, such as elasticity, supercontraction, and the effect of water on modulus are between those of spider major ampullate silks and mulberry silk. Therefore, an interpretation of the connection between the primary structures of silk proteins and the mechanical properties of silks is proposed here based on the ordered fraction, which in turn is determined by both the protein sequence and spinning process of the silk.

Abstract:Spider dragline silk, which is produced in spider major ampullate gland, is a composite protemacious fiber with highly repetitive Ala-Gly-rich domain. The unique combination of both high tensile strength and high elasticity makes spider dragline silk superior to almost any other natural or synthetic fibers. Cloning of the genes reveals that the silk is composed of at least two major proteins. Each protein component contains multiple repeats of modular structures that alternate between Ala-rich domains and Gly-rich domains. Molecular engineering not only opens a door to the production of spidroins but also provides a valuable experimental system to test and further establish the relationship between modular structures. and mechanical properties. Here, based on our own studies, we review the latest progress of the modular structure and genetic engineering and outline the future prospects.

Abstract:Nanofibrils play a pivotal role in spider silk and are responsible for many of the impressive properties of this unique natural material. However, little is known about the internal structure of these protein fibrils. We carry out polarized Raman and polarized Fourier-transform infrared spectroscopies on native spider silk nanofibrils and determine the concentrations of six distinct protein secondary structures, including β-sheets, and two types of helical structures, for which we also determine orientation distributions. Our advancements in peak assignments are in full agreement with the published silk vibrational spectroscopy literature. We further corroborate our findings with X-ray diffraction and magic-angle spinning nuclear magnetic resonance experiments. Based on the latter and on polypeptide Raman spectra, we assess the role of key amino acids in different secondary structures. For the recluse spider we develop a highly detailed structural model, featuring seven levels of structural hierarchy. The approaches we develop are directly applicable to other proteinaceous materials.

Abstract:A unique cuboid spider silk from the outer egg sac of Nephila pilipes, with an unusual square cross-section, is disclosed. The structure-function relationships within this silk are first studied through structural characterization, mechanical measurement, protein conformation, and polypeptide signature of silk proteins. This silk maintains the higher stiffness property of egg sac silks, and also shows a species difference. Environmental response of the mechanical properties within this silk are observed. Synchrotron FTIR microspectroscopy is used to monitor the silk protein conformation in a single natural silk. The beta-sheet structure aligns parallel to the fiber axis with a content of 22% +/- 2.6%. The de novo resulting polypeptide from the solid silk fibers are novel, and an abundant polar amino acid insertion is observed. Short polyalanine (A(n), n <= 3), alternating serine and alanine (S/A)X, and alternating glycine and alanine (G/A)X, GGX, and SSX dominates in the resulting de novo polypeptide. This accords with the composition pattern of other egg sac silk proteins, besides the rarely observed GGX. This study broadens the library of egg sac spider silks and provides a new perspective to uncover structure-function relationships in spider silk.

Abstract:The hierarchical structure of spider dragline silk is composed of two major constituents, the amorphous phase and crystalline units, and its mechanical response has been attributed to these prime constituents. Silk mechanics, however, might also be influenced by the resistance against sliding of these two phases relative to each other under load. We here used atomistic molecular dynamics (MD) simulations to obtain friction forces for the relative sliding of the amorphous phase and crystalline units of Araneus diadematus spider silk. We computed the coefficient of viscosity of this interface to be in the order of 10(2) Ns/m(2) by extrapolating our simulation data to the viscous limit. Interestingly, this value is two orders of magnitude smaller than the coefficient of viscosity within the amorphous phase. This suggests that sliding along a planar and homogeneous surface of straight polyalanine chains is much less hindered than within entangled disordered chains. Finally, in a simple finite element model, which is based on parameters determined from MD simulations including the newly deduced coefficient of viscosity, we assessed the frictional behavior between these two components for the experimental range of relative pulling velocities. We found that a perfectly relative horizontal motion has no significant resistance against sliding, however, slightly inclined loading causes measurable resistance. Our analysis paves the way towards a finite element model of silk fibers in which crystalline units can slide, move and rearrange themselves in the fiber during loading.

Abstract:Raman spectroscopy is used to elucidate the effect of spinning conditions upon the structure and mechanical properties of silk spun by Nephila spiders from the major ampullate gland. Silk fibers produced under natural spinning conditions with spinning rates between 2 and 20 mm s −1 differed in microstructure and mechanical properties from fibers produced either more slowly or more rapidly. The data support the “uniform strain” hypothesis that the reinforcing units in spider silk fibers are subjected to the same strain as the fiber, to optimize the toughness. In contrast, in the case of synthetic high-performance polymer fibers, the both units and the fiber experience uniform stress, which maximizes stiffness. The comparison of Nephila major and minor ampullate silks opens an intriguing window into dragline silk evolution and the first evidence of significant differences between the two silks providing possibilities for further testing of hypotheses concerning the uniform strain versus uniform stress models. Impact statement It is well established that the microstructure and mechanical properties of engineering materials are controlled by the conditions employed to both synthesize and process them. Herein, we demonstrate that the situation is similar for a natural material, namely spider silk. We show that for a spider that normally produces silk at a reeling speed of between 2 and 20 mm s −1 , silk produced at speeds outside this natural processing window has a different microstructure that leads to inferior tensile properties. Moreover, we also show that the silk has a generic microstructure that is optimized to respond mechanically to deformation such that the crystals in the fibers are deformed under conditions of uniform strain. This is different from high-performance synthetic polymer fibers where the microstructure is optimized such that crystals within the fibers are subjected to uniform stress. Graphic abstract

Abstract:In this letter, we adopt a new approach combining theoretical modeling with silk stretching measurements to explore the mystery of the structures between silkworm and spider silks, leading to the differences in mechanical response against stretching. Hereby the typical stress-strain profiles are reproduced by implementing the newly discovered and verified "$\beta$-sheet splitting" mechanism, which primarily varies the secondary structure of protein macromolecules; our modeling and simulation results show good accordance with the experimental measurements. Hence, it can be concluded that the post-yielding mechanical behaviors of both kinds of silks are resulted from the splitting of crystallines while the high extensibility of spider dragline is attributed to the tiny $\beta$-sheets solely existed in spider silk fibrils. This research reveals for the first time the structural factors leading to the significant difference between spider and silkworm silks in mechanical response to the stretching force. Additionally, the combination of theoretical modeling with experiments opens up a completely new approach in resolving conformation of various biomacromolecules.

Abstract:In this chapter we review the properties and characterization of spider silk, particularly dragline silk, which is attracting increasing interest due to its marvelous properties. The spider's unique protein dope and natural spinning process play important roles in providing this unrivalled material with its characteristic structure that determines its strong and tough properties. In general, dragline silk is a semicrystalline material composed of crystallites embedded in an amorphous matrix. We introduce the widely applied techniques used to probe the structural characteristics of dragline silk. The inherent excellent mechanical properties of dragline silk, as well as the influence of silking conditions its mechanical properties, are reviewed. Models relating the structure to the mechanical properties are discussed, aiming to unravel the structural base for the unique mechanical properties. Supercontraction and cyclic contraction are two other interesting inherent properties of dragline silk; the former can maintain tension in a web after deformation by natural elements and the latter has the ability to perform work by subjecting the silk fibre to periodic cycling at high and low humidity. Current advances in possible applications of dragline silk in the biomedical and clinical fields using biomaterials of different forms, based on recombinant and reconstituted silk proteins, are discussed finally.

Abstract:Spider dragline silk, as a type of high-performance natural fiber, displays a unique combination of tensile strength and extensibility that gives rise to a greater toughness than any other natural or synthetic fiber. In contrast to silkworm silk, spider dragline silk displays a remarkable strain-hardening character for which the origin remains unknown. In this paper, the performance of silkworm silk and spider dragline fibers under stretching is compared based on a combined structural and mechanical analysis. The molecular origin of the strain-hardening of spider silk filaments is addressed in comparison to rubber and Kevlar. Unlike rubber, the occurrence of strain-hardening can be attributed to the unfolding of the intramolecular beta-sheets in spider silk fibrils, which serve as "molecular spindles" to store lengthy molecular chains in space compactly. With the progressive unfolding and alignment of protein during fiber extension, protein backbones and nodes of the molecular network are stretched to support the load. Consequently the dragline filaments become gradually hardened, enabling efficient energy buffering when an abseiling spider escapes from a predator. As distinct from synthetic materials such as rubber (elastomers), this particular structural feature of spider draglines not only enables quick energy absorption, but also efficiently suppresses the drastic oscillation which occurs upon an impact. The mimicking of this strain-hardening character of spider silk will give rise to the design and fabrication of new advanced functional materials with applications in kinetic energy buffering and absorption.
