Abstract:The present study was conducted by using laying ducks of 6 lines from 2 native breeds in Zhejiang and 50 eggs of each line.Each egg was measured for egg shape index,shell thickness,shell strength,egg specific gravity,albumen height,Hough unit,yolk weight and yolk percentage.Moreover,multiple comparisons for egg quality traits were conducted in different breeds and lines and their heretability were estimated.The results showed that eggshell quality in Shaoxing duck was significantly higher than that of Jinyun duck,while,albumen and yolk quality of Jinyun duck were superior to Shaoxing duck;the comparison among different lines showed that eggshell quality were the highest in Shaobai line and blue shell line of Shaoxing duck,quality of albumen and yolk among 3 strains of Jinyun duck existed only little difference;the results of estimation of heretability for egg quality traits revealed that egg specific gravity and shell thickness had the highest heritability values(0.72 and 0.47,respectively),and the heritability of Hough unit was the lowest(0.06).Our finding suggested that egg specific gravity and shell thickness could be improved by genetic breeding,while albumen height,Hough unit and yolk percentage might be promoted through environmental control.

Abstract:Khaki-Campbell duck and 4 strains of Shaoxing duck (Shaobai of high production strain, Shaobai of large egg strain, Shaohong of high production strain, Shaohong of large egg strain) were studied for the analysis of variance and estimation of least squares means based on the data of following GPT and GOT. The results indicated that the influence of the effect of location on the GOT, the effect of breed (strain) on the GPT were all highly significant (P0.01). The effect of interaction between breed (strain) and sex on GOT were significant. The significant effects were also estimated for the breed(strain), age, sex and body weight on GOT as well as the age on GPT. The least squares means (LSM) were calculated for GOT (99.97±(51.58)) Karmen and GPT (72.63±44.84) Karmen.
Path Coefficient Analysis on the Effect Factor of Meat Production Performance of Shaoxing Duck

Abstract:Ninety Shaoxing mature duck were used to analysis correlation coefficient and path coefficient between 8 quantitative traits and 3 traits of meat production from original farm of Shaoxing duck.The result indicated that more than 32 percent of dressing weight was decided by pelvis width,chest width and shank girth;pelvis width,chest width and semi-diving length together directly affected 32 percent of eviscerated weight.Semi-diring length,chest width and shank girth were key factors influencing meat weight.Therefore,these phenotypes can be used to predict meat production of mature Shaoxing duck and applied for indirect breeding of new duck line from the population of worse reproductionperformance duck.Also it was useful to understand the relationship between morphometric traits and meat produc-tion traits clearly.
Effect of Ω-3Pufa at Different Levels in Dietary to Egg Performance and Egg Quality

Abstract:Feeding experiment were conducted on 480 commercial Shaoxing Ducks at 300-day-old with similar body size and weight,they were randomly divided into four groups feeding with four different diets.Compared with the control,the group I,II and III were fed with basal diets adding 1%,2% and 3% concentrations of fish oil respectively.The results showed that the ducks in the group with the addition of 2% fishoil in diet got the highest laying rate,and its average daily laying rate reached to 83.75%.Group III(3%) took the lowest ratio of feed to egg production,which was 8.97% lower than the control.There were no significant difference among groups with respect to their daily feed intakes,broken egg rates,death-out rates etc.The colors of the yolks deepened with the increase of the addition of fish oil.Compared with the control,the groups added with fish oil had significantly lower cholesterol content(P0.05) and lipid peroxidation content(P0.05),except the group I(P0.05).The input-output analysis showed that the group with the addition of 2% fish oil in diet got the highest economic efficiency.From the above,it came to the conclusion that the optimal addition of fish oil to the diets for the ω-3PUFA-rich egg production would be 2%.
Analysis on meat production and quality of main mallard breeds in Zhejiang province

Abstract:Four duck breeds such as fellows spot-mouth mallard(Group Ⅰ),green-head mallard crossing Shaoxin duck(Group Ⅱ),Xihu mallard(Group Ⅲ),and green-head mallard crossing Xihu mallard(Group Ⅳ)in Zhejiang province are slaughtered at the age of 70 days old to determinate their meat production and quality.Results indicate that breed type is a key factor affecting meat production. The meat quality of carcass is mainly determined by physical properties of chest muscle(CM)and thigh muscle(TM).By comparison,Group Ⅱ is the worst in eviscerating weight,but the best in ratio of CM/TM,meat tenderness,CM weight,and meat color.As far as meat production and quality are concerned,Group Ⅱ ranked first,following by GroupⅠ,Group Ⅲ,and Group Ⅳ.
Genome-Wide Association Studies and Haplotype-Sharing Analysis Targeting the Egg Production Traits in Shaoxing Duck

Abstract:Age at first egg (AFE) and egg number (EN) are economically important traits related to egg production, as they directly influence the benefits of the poultry industry, but the molecular genetic research that affects those traits in laying ducks is still sparse. Our objective was to identify the genomic regions and candidate genes associated with AFE, egg production at 43 weeks (EP43w), and egg production at 66 weeks (EP66w) in a Shaoxing duck population using genome-wide association studies (GWASs) and haplotype-sharing analysis. Single-nucleotide polymorphism (SNP)-based genetic parameter estimates showed that the heritability was 0.15, 0.20, and 0.22 for AFE, EP43w, and EP66w, respectively. Subsequently, three univariate GWASs for AFE, EP43w, and EP66w were carried out independently. Twenty-four SNPs located on chromosome 25 within a 0.01-Mb region that spans from 4.511 to 4.521 Mb were associated with AFE. There are two CIs that affect EP43w, i.e., twenty-five SNPs were in strong linkage disequilibrium region spanning from 3.186 to 3.247 Mb on chromosome 25, a region spanning from 4.442 to 4.446 Mb on chromosome 25, and two interesting genes, ACAD8 and THYN1, that may affect EP43w in laying ducks. There are also two CIs that affect EP66w, i.e., a 2.412-Mb region that spans from 127.497 to 129.910 Mb on chromosome 2 and a 0.355-Mb region that spans from 4.481 to 4.837 Mb on chromosome 29, and CA2 and GAMT may be the putative candidate genes. Our study also found some haplotypes significantly associated with these three traits based on haplotype-sharing analysis. Overall, this study was the first publication of GWAS on egg production in laying ducks, and our findings will be helpful to provide some candidate genes and haplotypes to improve egg production performance based on breeding in laying duck. Additionally, we learned from a method called bootstrap test to verify the reliability of a GWAS with small experimental samples that users can access at https://github.com/xuwenwu24/Bootstrap-test .

Abstract:Several studies have documented the process of early embryonic development in poultry; however, the molecular mechanisms underlying its developmental regulation are poorly understood, particularly in ducks. In this study, we analyzed differential gene expression of embryos 6 and 25 h following oviposition to determine which genes regulate the early developmental stage in ducks. Among 216 randomly selected clones, 39 protein-encoding cDNAs that function in metabolism, transcription, transportation, proliferation/apoptosis, cell cycle, cell adhesion, and methylation were identified. Additionally, the full-length cDNA of the Nanog gene, encoding a 302-amino acid protein, was obtained. Quantitative real-time polymerase chain reaction analyses were performed to detect expression levels of the selected genes during early and late embryonic stages, which revealed that these genes are expressed in a particular spatial and temporal pattern. These results indicate that these genes may play pivotal roles in the process of area pellucida formation through a complex and precise regulatory network during development in duck embryos.
Immunohistochemical Study of the Development of P Cells in the Proventriculus of Shao Ducks

Abstract:Immunohistochemical method (ABC method) was employed todemonstrate the postnatal development of bombesin cells (P cells) in proventriculus of Shao duck by observing its character and counting its number in every stage. The results presented that the P cells were mainly oval and polygonal, and some with short cytoplasmic processes. Most of P cells disperse in the central area of gastric glands and only a few located in the epithelial surface of lumen. The P cells' growth peak of gastric gland appeared during 0-6 weeks, and the epithelial did during 0-10 weeks. Following the growth peak, both gastric P cells and epithelial decreased quickly. The results also showed that the gastric P cells were remarkably more than the epithelial. This paper presented the postnatal development of the gastric P cells in Shao ducks. It also provided basic data for studying the dynamic change of the gastroenteropancreatic endocrine system.
Comparative Analysis of the Duodenum Microbial Communities of Zhedong White Goose (anser Cygnoides) at Different Reproductive Stages by High-Throughput Sequencing

Abstract:The primary purpose of this study is to characterize the intestinal microbial communities in Zhedong White geese (a local breed in China) at different reproductive stages and to investigate the relationships between intestinal microflora and geese reproductive states. We performed 16S rRNA sequencing of geese duodenum content samples at three different stages (laying, broody, and recovery stage). Taxonomic analysis revealed that Firmicutes, Actinobacteria, and Proteobacteria were the three most abundant phyla in the duodenum of the laying geese. Cyanobacteria predominated in broody geese, followed by Firmicutes, Proteobacteria, and Spirochaetes. The microbial communities of the recovery geese were dominated by Spirochaetes, Firmicutes, Cyanobacteria, and Proteobacteria. Additionally, the microbial diversity and richness were significantly higher in the laying group than the broody and recovery groups. These results provide a basic profile of Zhedong white goose microbial community for the further study on goose broodiness and microbiology.
Effects of Rearing Systems on Laying Performance, Egg Quality, and Serum Biochemistry of Xianju Chickens in Summer

Abstract:The objective of this study was to compare the laying performance, egg quality, and serum biochemistry of hens maintained in conventional cage rearing system (CRS), flat net-rearing system (NRS), and free range system (FRS) under summer conditions. Indigenous Xianju chickens (n = 540) were randomly allocated into cages or pens of rearing system groups, within each system there were 5 replicates with 36 hens in each replicate. The experiment lasted between 21 and 29 wk of age. Hen-day egg production (P = 0.00) and egg mass (P = 0.00) were higher in the CRS but were similar in the NRS and FRS. Lowest egg weight (P = 0.02), yolk weight (P = 0.00) and yolk ratio (P = 0.01), and feed intake (P = 0.01) were observed from the FRS, whereas lowest feed conversion ratio (FCR) was recorded from the CRS (P = 0.01). Rearing systems had negligible effect on egg quality. Serum Ca (P = 0.04) and total protein (P = 0.03) levels were found to be higher in the CRS but were lower in the FRS. Serum levels of glucose (P = 0.01), cholesterol (P = 0.00), and triglyceride (P = 0.00) in the CRS increased compared with the NRS and FRS groups, whereas serum levels of high density lipoprotein cholesterol (HDL-C; P = 0.01) in the CRS decreased. It can be concluded that under summer conditions, Xianju chickens from CRS had an advantage in terms of productivity parameters, but exhibited higher levels of serum lipids and glucose.

Abstract:The Zhongshan duck (ZSP) is a duck breed indigenous to China and is known for its moderate body size, strong disease resistance, tender meat, and little subcutaneous fat. However, the genomic basis of such excellent breeding characteristics remains poorly understood. Therefore, we generated whole-genomes of 58 ZSPs and 180 other indigenous Chinese ducks (60 Jinding ducks, 60 Shan Partridge ducks, and 60 Liancheng white ducks) and identified 10 560 032 single nucleotide polymorphisms and 1 334 893 structural variants. Based on genetic diversity and population structure indices, our results confirm that the ZSP is a unique germplasm resource. In addition, three reproduction-related genes (i.e., OAZ, AMH, and RLF) were located in highly differentiated regions between the ZSP and the other three duck breeds (Jinding duck; Liancheng White duck; Shan Partridge duck), suggesting that these genes may have a strong influence on egg production. Among these genes, AMH may have introgressed from an unknown species of the Anatidae family. We also identified other significant genes in the significantly differentiated window (i.e., 1% cut-off), some of which are responsible for growth and development (SEMA5B and MIB1), metabolism (EDEM3 and Xylb), skeletal system morphogenesis (bglap and MGP), and egg shape (ITPR2). These findings highlight the genetic characteristics of the ZSP that shape an array of its morphological traits. Overall, this study should facilitate a more fine-scale approach towards improving the ZSP and other indigenous ducks in China and even all over the world.
Comparative Study of Reproductive Organs and Egg Quality Characteristics in Hyline and Domestic Layer Breeds

Abstract:For comparison of the reproductive organs and the egg quality in hens, a case study was undertaken on traditional Chinese Zhenning and commercial Hyline hen. A total of 15 hens from each breed were selected to study lipogenesis, serum lipids, and organ body weight ratios of liver, ovary and oviduct to compare their lipid synthesis and transportation mechanism. Moreover we also examined the histomorphological comparison of the egg forming organs and egg quality parameters between two breeds. Results showed significantly higher values of peroxisome proliferator-activated receptors gamma (PPAR gamma), estrogen receptor alpha (ER alpha) and estrogen receptor beta (ER beta) mRNA expressions in the liver of Zhenning than Hyline hen. Same difference was also observed in the vitellogenin (Vtg) protein levels. Triglyceride levels were higher in the Hyline hens. Besides this, liver, ovary and oviduct body weight ratio were significantly higher in Hyline hen than Zhenning hen. Moreover, Hyline hen had significantly higher oviduct weights. Ovary weight and hierarchy was also bigger in the Hyline hen. Finally, egg quality parameter comparison showed significantly lower values in the Zhenning eggs than the White Hyline eggs. These findings suggest that Hyline and Zhenning hen differ significantly due to selection in terms of lipogenesis, increase in body weight ratio of liver, ovary and oviduct morphology, while Hyline hens have also superior egg quality than Zhenning hen eggs. Identifying Genetic Differences Between Dongxiang Blue-Shelled and White Leghorn Chickens Using Sequencing Data

Abstract:The Dongxiang Blue-shelled chicken is one of the most valuable Chinese indigenous poultry breeds. However, compared to the Italian native White Leghorn, although this Chinese breed possesses numerous favorable characteristics, it also exhibits lower growth performance and fertility. Here, we utilized genotyping sequencing data obtained via genome reduction on a sequencing platform to detect 100,114 single nucleotide polymorphisms and perform further biological analysis and functional annotation. We employed cross-population extended haplotype homozygosity, eigenvector decomposition combined with genome-wide association studies (EigenGWAS), and efficient mixed-model association expedited methods to detect areas of the genome that are potential selected regions (PSR) in both chicken breeds, and performed gene ontology (GO) enrichment and quantitative trait loci (QTL) analyses annotating using the Kyoto Encyclopedia of Genes and Genomes. The results of this study revealed a total of 2424 outlier loci (p-value <0.01), of which 2144 occur in the White Leghorn breed and 280 occur in the Dongxiang Blue-shelled chicken. These correspond to 327 and 94 PSRs containing 297 and 54 genes, respectively. The most significantly selected genes in Blue-shelled chicken are TMEM141 and CLIC3, while the SLCO1B3 gene, related to eggshell color, was identified via EigenGWAS. We show that the White Leghorn genes JARID2, RBMS3, GPC3, TRIB2, ROBO1, SAMSN1, OSBP2, and IGFALS are involved in immunity, reproduction, and growth, and thus might represent footprints of the selection process. In contrast, we identified six significantly enriched pathways in the Dongxiang Blue-shelled chicken that are related to amino acid and lipid metabolism as well as signal transduction. Our results also reveal the presence of a GO term associated with cell metabolism that occurs mainly in the White Leghorn breed, while the most significant QTL regions mapped to the Chicken QTL Database (GG_4.0) for the Dongxiang Blue-shelled breed are predominantly related to lesions, bone mineral content, and other related traits compared to tibia length and body weight (i.e., at 14, 28, 42, and 70 d) in the White Leghorn. The results of this study highlight differences in growth, immunity, and egg quality traits between the two breeds, and provide a foundation for the exploration of their genetic mechanisms.
Meat quality, fatty acid composition and sensory evaluation of Cherry Valley, Spent Layer and Crossbred ducks

Abstract:The aim of this research was to compare meat quality characteristics of Cherry Valley (CV), Spent Layer (SL) and Crossbred (CB) ducks. Meat quality, proximate and fatty acid composition were measured in breast and thigh muscles from CV, SL and CB, as well as sensory evaluation of duck soup made from three breeds. The results showed SL contained a highest percentage of protein but lowest moisture than those of CV and intermediate CB (P < 0.05). The L* and b* value of SL ducks were lowest among three breeds (P < 0.05). The breast of SL had higher water-holding capacity compared to CV (P < 0.05), although CV was the most tender among the three breeds (P < 0.05). SL had lower contents of saturated fatty acid and higher contents of unsaturated fatty acids with a more acceptable P/S ratio of 0.57 and n-6/n-3 ratio of 1.52 (P < 0.05). Intramuscular fat and polyunsaturated fatty acids were involved in producing intense aroma and flavor of duck meats. Differences of color and tenderness in meats were attributed to values of L* and cooking loss of muscles. In conclusion, it would be suggested that SL was more suitable for producing duck soup among the three breeds.

For the reason of out - of - orderly traditional mode of production and mates , the community gene of white chest duck tends to be identical. So we can' t breed a good breed with advantage of crossbreed . The author of this article analyses the value of breed selection for this species resources of ducks from the angle of breed selection in systematism, and points out it is of much significance to develop species resources of ducks in the area east of Liaoning for enriching the gene storage of duck breed in the world, enhancing the production performance , and boosting the characteristic of local education.
Effects of Probiotic (bacillus Subtilis) on Laying Performance, Blood Biochemical Properties and Intestinal Microflora of Shaoxing Duck

Abstract:The study was conducted to evaluate the effects of probiotic supplementation (Bacillus subtilis) in diet on performance, blood properties and microflora of Shaoxing duck. A population of 200 laying ducks (160 days old) was divided into two groups each having five replications. The control (T0) were fed on basal diet while T1 with Bacillus subtilis 1 x 10 CFU/kg in addition to basal diet for thirty five days. The results 8 showed that for T1 group, the egg laying rate increased (3.79%) significantly, while a decrease of about 12.60%, 23.52% and 40.14% was observed in egg triglyceride, total cholesterol and malondialdehyde, respectively. However, the blood glucose content and alkaline phosphatase activity increased by 15.13% and 53.41%, significantly. Moreover, there was a decrease of blood cholesterol, uric acid and ammonia in T1 by 28.86%, 22.39% and 62.5%, respectively. As compared to T0, T1 showed a significant increased in the amount of caecal microflora, i.e. general aerobic bacteria (18.68%) and Lactobacillus (9.92%). In conclusion, supplementation of Bacillus subtilis in diets had significant effects on some aspects of egg composition, biochemical properties of blood and few microflora of Shaoxing ducks.
Exploring differentially expressed key genes related to development of follicle by RNA-seq in Peking ducks (Anas Platyrhynchos)

Abstract:Duck follicles enter different reproductive phases throughout life, and follicle gene expression patterns differ according to these phases. In particular, differentially expressed genes and related to development of follicle (mRNAs) play an important role to explore the key genes in this process; however, the expression profiles of these genes remain unclear. In this study, transcriptome sequencing was used to investigate the expression levels of duck ovarian genes, and comparative transcriptional analysis was carried out to identify differential genes, and cluster them into groups and function identification. The results showed differential expression of 593 coding genes between young and laying ducks, and of 518 coding genes between laying and old ducks. In further GO analysis, 35 genes from the comparison between old ducks and laying ducks have significant been changed involved in hormones related to follicle development. They include up-regulated genes StAR, CYP17, EPOX, 3β-HSD, CYP1B1 CYP19A1 and down-regulated genes SR-B1 in laying ducks hormone synthesis than old ducks. Among which EPOX is a key gene for time special highly expression during egg laying stage, and other key regulatory genes' highly expression showed in young and laying stage, and lower expression showing with follicular development stopping. Therefore, EPOX is a key regulator for duck follicle development in laying period, its expression level increase 100 times higher than in youth and decrease 98% than stop laying period in duck life cycle.

The differentiation of preadipocytes and gene expression related to adipogenesis in ducks (Anas platyrhynchos)

Abstract:Meat quality is closely related to adipose tissues in ducks, and adipogenesis is controlled by a complex network of transcription factors tightly acting at different stages of differentiation especially in ducks. The aim of this study was to establish the preadipocyte in vitro culture system and understand the biological characteristics of expansion of duck adipocyte tissue at the cellular and molecular level. We isolated pre-adipocytes from the subcutaneous fat of three breeds of duck and differentiated them into mature adipocytes using a mixture of insulin, rosiglitazone, dexamethasone, 3-isobutyl-1-methylxanthine, and oleic acid over 0,2, 4, 6, and 8 days. Successful differentiation was confirmed from the development of lipid droplets and their response to Oil Red O, and increasing numbers of lipid droplets were stained red over time. The expression of key marker genes, including peroxisome proliferator activated receptor γ (PPARγ), CCAAT/enhancer binding protein-α (C/EBPα), adipocyte fatty acid binding protein 4 (FABP4), and fatty acid synthetase (FAS), gradually increased during pre-adipocyte differentiation. Furthermore, it was verified by interference experiments that the knockdown of PPARγ directly reduced lipid production. Meanwhile we analyzed the role of unsaturated fatty acids in the production of poultry fat using different concentrations of oleic acid and found that lipid droplet deposition was highest when the concentration of oleic acid was 300 μM. We also compared the level of differentiated pre-adipocytes that were isolated from Jianchang ducks (fatty-meat duck), Cherry Valley ducks (lean-meat duck) and White-crested ducks (egg-producing duck). The proliferation and differentiation rate of pre-adipocytes derived from Jianchang ducks was higher than that of White-crested ducks. These results provide the foundation for further research into waterfowl adipogenesis.

Abstract:The study was designed to evaluate the effects of probiotics (Bacillus subtilis) to enhance immunity, antioxidation, digestive enzymes activity and hematological profile of Shaoxing duck. A population of 200 laying ducks (160 days old) was divided into two groups each further divided in five replications. The control (G(1)) were fed on basal diet and (G(2)) with B. subtilis 1 x 10(8) cfu/ kg in addition of basal diet for thirty five days. The results showed that, ducks were treated with probiotics (B. subtilis), their serum IL-2 increased and IL-10 decreased (P<0.05). The concentrations of IgG, IgA and sIgA were observed significantly higher in (G(2)) as compared to (G(1)). Treatment group (G(2)), showed significantly improvement in (SOD), T-AOC and ASAFR activity in serum and liver. However, digestive enzymes amylase and trypsin activity also improved (P<0.05) in (G(2)). The blood chemistry analysis showed significant decrease in FT3 and no other significant change observed in hematological profile as compared to (G(1)). In conclusion, application of B. subtilis (1 x 10(8) cfu/kg) may be beneficial to improve antioxidation response, supportive in innate immunity and digestibility of fowls (Shaoxing duck). Whole-genome resequencing provides insights into the population structure and domestication signatures of ducks in eastern China

Abstract:Abstract Background Duck is an ancient domesticated animal with high economic value, used for its meat, eggs, and feathers. However, the origin of indigenous Chinese ducks remains elusive. To address this question, we performed whole-genome resequencing to first explore the genetic relationship among variants of these domestic ducks with their potential wild ancestors in eastern China, as well as understand how the their genomes were shaped by different natural and artificial selective pressures. Results Here, we report the resequencing of 60 ducks from Chinese spot-billed ducks ( Anas zonorhyncha) , mallards (Anas platyrhnchos ), Fenghua ducks, Shaoxing ducks, Shanma ducks and Cherry Valley Pekin ducks of eastern China (ten from each population) at an average effective sequencing depth of ~ 6× per individual. The results of population and demographic analysis revealed a deep phylogenetic split between wild (Chinese spot-billed ducks and mallards) and domestic ducks. By applying selective sweep analysis, we identified that several candidate genes, important pathways and GO categories associated with artificial selection were functionally related to cellular adhesion, type 2 diabetes, lipid metabolism, the cell cycle, liver cell proliferation, and muscle functioning in domestic ducks. Conclusion Genetic structure analysis showed a close genetic relationship of Chinese spot-billed ducks and mallards, which supported that Chinese spot-billed ducks contributed to the breeding of domestic ducks. During the long history of artificial selection, domestic ducks have developed a complex biological adaptation to captivity.
